1.Mechanism Study on Electroacupuncture in Reconstructing the Neurologic Function in Mice after Spinal Cord Injury

Objective To observe the effect of electroacupuncture at thoracic Jiaji points (EX-B 2) on the Behavior Measurement Scale (BMS) score, and expressions of nestin and glial fibrillary acidic protein (GFAP) in spinal cord in mice after spinal cord injury, and to explore the action mechanism of electroacupuncture in protecting nerves and recovering motor function.Method Ninety-six female C57BL/6 mice were randomized into a sham operation group, a spinal cord injury group, an electroacupuncture group, and an acupuncture group, 24 mice in each group. Except for the sham operation group, spinal cord injury at T9 level was induced in the other three groups. The electroacupuncture group was intervened by electroacupuncture at Jiaji points (EX-B 2) of T7 and T11, and the acupuncture group received manual acupuncture at the same acupoints. The intervention was conducted 15 min each time, once a day, with 1 d interval every 5 d, for a total of 28 d. The BMS score was evaluated respectively after 3 d, 7 d, 14 d, and 28 d intervention. Enzyme-linked immunosorbent assay (ELISA) was adopted to detect the expression ofnestin in spinal cord, and immunofluorescence and Western blotting were used to detect the expression of GFAP in spinal cord.Result Respectively 14 d and 28 d after spinal cord injury, the main and auxiliary scores of BMS in the acupuncture group were significantly higher than those in the spinal cord injury group (P<0.05); 28 d after spinal cord injury, the main and auxiliary scores of BMS in the acupuncture group were significantly higher than those in the spinal cord injury group (P<0.05) and the auxiliary BMS score in the electroacupuncture group was significantly higher than that in the acupuncture group (P<0.05). Respectively 3 d, 7 d, and 14 d after spinal cord injury, the contents of nestin in the electroacupuncture group were obviously increased and significantly different from those in the spinal cord injury group (P<0.05). The nestin content of the acupuncture group was significantly higher than that of the spinal cord injury group 7 d and 14 d after spinal cord injury (P<0.05). The expression of GFAP in the electro-acupuncture group was significantly higher than that in the spinal cord injury group 3 d and 7 d after spinal cord injury (P<0.05); the inhibitions on the immunoreactivity of GFAP in the electroacupuncture group and acupuncture group were more significant than the inhibition in the spinal cord injury group 28 d after the injury (P<0.05).Conclusion Electroacupuncture at Jiaji points (EX-B 2) can promote the recovery of motor function after spinal cord injury, which is related to the enhancement of the expressions of nestin and GFAP within 7 d after the injury, and the two expressions are in a positive correlation. In the early stage of treatment, electroacupuncture can boost the activity of astrocytes to act as neural stem cells and inhibit the immunoreactivity of GFAP in the later stage to benefit the reconstruction of neurologic function.

3.Study on foveal thickness and image features of macular changes observed through OCT in patients with retinitis pigmentosa

AIM:By optical coherence tomography to observe macular thickness change characteristics and foveal images of patients with retinitis pigmentosa (RP).METHODS: From September 2014 to September 2016, 74 patients (148 eyes) with retinitis pigmentosa diagnosed in our hospital were selected, and 50 healthy subjects (100 eyes) as normal controls;two groups received optical coherence tomography and fundus camera detection.The images of the macular were obtained to observe and measure the thickness of the macular fovea and at 4mm temporal side, the center of papillo macular bundle.RESULTS:The results showed that: the thickness of the macular fovea and at 4mm temporal side of RP patients were the same as normal controls without statistically significant(P>0.05);the thickness of the center of papillo macular bundle was less than that of normal control with statistically significant (P<0.05).There were five types characteristics in OCT imges in RP patients: macular edema in 19 patients 38 eyes;retinal pigment epithelium and choroidal capillary thinning in 18 patients 36 eyes;retinal pigment epithelium atrophy in 12 patients 24 eyes;normal in 12 patients 24 eyes;different thickness of pigment epithelium layer in 13 patients 26 eyes.CONCLUSION: Optical coherence tomography (OCT) can be used to detect the macular lesions in a timely manner, to help understanding the development of the disease, and provide the clinical basis for the early diagnosis and treatment of patients.

5.Investigation on Strengthening the Supervision of Drug Stocktaking in Our Hospital

OBJECTIVE:To strengthen the supervision of drug stocktaking,and to improve drug management. METHODS:The data of drug stocktaking were extracted from our hospital from 2011 to 2014,and was analyzed in respects of accounts coinci-dence rate,drug profit and loss account rate,the entry and exit account between departments and original record,etc. A series of supervision strategies were established,and the data of drug stocktaking was analyzed during Jan.-Jun. in 2015 statistically to evalu-ated supervision effects. RESULTS:The supervision of drug stocktaking was strengthened through limiting the right of drug profit and loss on stocktaking,strengthening original stocktaking record management,establishing Key Drug List,setting up drug stock-taking supervision institutions,etc. During Jan.-Jun. in 2015,there was obvious improvement in the drug supervision skills,in terms of account coincidence rate,profit and loss account,material safety,cash flow rate,staff responsibility and stocktaking dura-tion,etc. More specially,the accounts coincidence rate had rose from about 82.8% to more than 95%;both the key drug accounts coincidence rate and the entry and exit account between departments coincidence rate had reached to 100%;the profit and loss ac-count rate had reduced from about 7.6% to less than 0.3%;the stocktaking time had reduced from 10 h to 3 h. CONCLUSIONS:Strengthening drug stocktaking supervision can not only improve the stocktaking efficiency and quality markedly,but also contrib-utes to the development of other pharmacy work in hospital.
7.Nutrition support in patients with heart insufficiency

8.Effects of presenilin-1 V97L mutation on neprilysin expressing in SH-SYSY cell

Objective To study the mechanisms of increasing amyloid [β1-42 (Aβ42) level by presenilin-1 (PS-1) mutation of V97L.Methods Expression level of neprilysin (NEP) in nontransfected, or mock vector, wild type (wt) PS-1 and V97L-PS-1-transfected SH-SY5Y cells were assessed by reverse transcriptional-polymerase chain reaction (RT-PCR) and Western blot.The level of Aβ42 was also assessed by enzyme linked immunosorbent assay (ELISA).Results The mRNA level of NEP was decreased significantly in PS-1 V97L transfected cells (0.650 ± 0.010) compared to that in normal cells (1.090 ± 0.015), wt PS-1 gene (1.040 ± 0.021) and mock (1.080 ± 0.020) stably transfected cell lines (t = 9.236, 10.452, 5.678; all P < 0.01).Although there was a decreased tendency in the protein expression of NEP in PS-Ⅰ V97L transfected cells (1.000 ± 0.126)compared to that in normal cells (1.020 ± 0.110), wt PS-1 gene (1.040 ±0.110), mock (2.130 ±0.130) stably transfected cell lines, nosignificant differences were found.The expression of Aβ42 was increased significantly in PS-1 V97L transfected cells compared to other cell lines (t = 2.109, 3.355, 3.976; all P < 0.01).Conclusions PS-1 V97L mutation can decrease the transcription of NEP, however, in contrast, this mutation also resulted in increased Aβ42.The mechanism needs further analysis.
9.Content Determination of Taxol in Taxus Yunnanensis of Different Parts and Growth Time Limits

Objective To study the content of taxol in different parts of Taxus Yunnanensis and the effects of different growth time on taxol content. Method A HPLC method was developed for the detetmination of taxol content in bark, needle, twig, root from Taxus Yunnanensi, and the effects of different growth time. Results Taxol content in bark was the highest, that in root was the lowest and that in needle and twig were higher. Taxol content was higher with the growth time in needle and twig of Taxus Yunnanensi. Conclution It was significant to take taxol from needle and twig of Taxus Yunnanensis which is more than five years growth time.
10.Determination of Stachydrine Hydrochloride in Different Parts of Herba Leonuri by HPLC

Objective To control the quality of Herba Leonuri by studying the content of stachydrine hydrochloride in different parts of it. Method A HPLC method was developed for the content detetmination of stachydrine hydrochloride in flower, stem, leafage from Herba Leonuri. Spherisorb SCX colum was used with mobile phase of 20 mmol/L sodium dihydrogen phosphate (containing 0.04% triethylamine and 0.15% phosphoric acid). The colum temperature was 25 ℃, the detective wavelength was 192 nm. Results Stachydrine hydrochloride content was the highest in leafage and the lowest in stem. Conclusion To ensure the quality of Herba Leonuri, it is significant to choose medical material with more leafage.
