Chinese Journal of cardiovascular Rehabilitation Medicine 2014;23(4):404-407
Objective:To explore the relationship between glycosylated hemoglobin (HbA1c)and carotid artery atheroscle-rosis in aged patients with normal glucose tolerance. Methods:A total of 100 aged patients with normal glucose tolerance were selected.Their bilateral carotid arteries were measured by color Doppler ultrasonography. They were grouped accord-ing to carotid intima-media thickness (C-IMT,normal,thicken and atherosclerosis)and plaque form (no plaque, hard plaque and soft plaque);blood pressure, blood lipids and HbA1c etc.were compared among groups. Results: (1)Along with CIMT rose, there was significant increase in HbA1c level [(4.98 ± 0.55)% vs.(5.51 ± 0.42)% vs.(5.92 ± 0.39)%],and there was significant difference between any two groups (P<0.01 all);(2)HbA1c level was (5.36±0.51)%,(5.89±0.44)% and (5.97±0.2)% in no plaque group, hard plaque group and soft plaque group respectively, and there was significant difference among hard plaque group, soft plaque group and no plaque group (P <0.01 all).Conclu-sion:Glycosylated hemoglobin may become an important index assessing carotid atherosclerosis in aged people.
6. Antiaddictive indole alkaloids in Ervatamia yunnanensis and their bioactivity
Academic Journal of Second Military Medical University 2006;27(1):92-96
Objective: To investigate the constituents of indole alkaloids in Ervatamia yunnanensis and their antiaddictive bioactivities. Methods: The preliminary alkaloids (TEYA) were extracted by 95 % EtOH from the stems of Ervatamia yunnanensis. TEYA was then purified by silica gel and Sephadex LH20 column chromatography repeatedly and its structure was identified with spectral analysis. The antiaddictive effects of these indole alkaloids were evaluated by Conditioned Place Preference (CPP) experiments. Results: Nine indole alkaloids were obtained and identified as: coronaridine (1) , voacangine (2), 3-R-ethoxycoronaridine (3), 3-S-ethoxycoronaridine (4), 19-epi-heyneanine (5), heyneanine (6), 19-epi-voacristine (7), coronaridine hydroxyindolenine (8) and 12-methoxyl-voaphylline (9). Among them voacangine (2) and 19-epi-voacristine (7) were effective in prevention and treatment of addiction induced by morphine in rats (n=8). Conclusion: Compounds 3, 4, 8, and 9 have been isolated from Ervatamia yunnanensis for the first time and coronaridine-type indole alkaloids are the main antiaddictive constituents of E. yunnanensis.

Progress in Biochemistry and Biophysics 2006;0(10):-
RNA interfering (RNAi) is a process of sequence-specific, posttranscriptional gene silencing in animals and plants. In mammalian cells, duplexes of 19~25 nts (nucleotides) RNAs efficiently inhibit gene expression. The pBS/H1SP vector expressing siRNA is used which inhibit specific HIV-1 gene expression. To assess the intracellular effect of this H1 promoter-driven siRNA, a reporter plasmid pEGFP-C1-HIV protein which expresses fusion protein of enhanced green fluorescent protein (EGFP) and HIV protein was used. The expression of the reporters can be easily visualized by fluorescence microscopy in living cells. siRNA-generating vectors targeted to several HIV-1 genomes were constructed and then co-transfected with respond reporter expression vectors into HEK293 cells. Cells transfected with pHIV-siRNA exhibited a significant inhibition of pEGFP-HIV expression compared with cells transfected with control vectors. By this way, it is successfully to select effective siRNA for silencing target HIV-1 genes. Then two or three siRNA transcripts targeted to different HIV genes were expressed by one plasmid, and a relative strong inhibition effect was observed.
