To observe the high glucose concentration on proliferation and fibronectin (FN) synthesis of glomerular mesangial cells (GMC),a culture of human GMC was established and the cell proliferation was assessed using MTT method and cell counting, and the changes in FN was analyzed with RT PCR and ELISA. It was found that high glucose concentration inhibited GMC proliferation and increased FN synthesis in dose and time dependent manner.The results suggest that hyperglycemia may have important role in the development of diabetic nephropathy.
4.The effects of diprospan on osteoarthritis by injected into the inside of knuckle

Objective To investigate the effect of infecting betamethasone dipropionate 5mg/betamethasone disodium hydrogen phosphate 2mg in articular genu cavity on osteoarthritis.Methods 100 patients were randomly divided into two groups, the treatment group and the control group. The treatment group was treated with diprospan,injected into the inside of knuckle. The control group was treated with oral administration NSAIDs. Results In treatment group the rate of amelioration was 84% after one injection,the rate of amelioration was 90% after one course of treatment. Conclusion Injected diprospan into the inside of knuckle could relieve pain of arthritides, improve the function of arthrosis.

7.Protein expression of activated Pak 1 and its correlation with chromosome segregation in mouse oocyte meiosis

Objective To observe the protein expression , subcelluar localization of activated Pak 1 and its relation-ship with chromosome separation during meiosis in mouse oocytes .Methods Western blot was applied to analyze the expression of activated Pak1 (phosphorylated Pak1 at Ser204, pPak1Ser204 ) at different stages of meiotic progres-sion in semi-quantitative manner;immunofluorescent staining was employed to detect the sub-cellular localization of pPak1Ser204 and its spatial-temporal correlation with spindle and microtubule organizing centers (MTOC) during oo-cyte meiosis.Results pPak1Ser204 expression was found upon germinal vesicle breakdown (GVBD) in mouse oo-cytes which increased along with the meiotic progression , reaching peak level at metaphase Ⅰ( MⅠ) , which re-mained up to meiosis Ⅱ(MⅡ).From pro-metaphase Ⅰ(pro-MⅠ) to MⅠand at MⅡstage, pPak1Ser204was co-localized with MTOC key components , pericentrin and γ-tubulin on spindle poles, pPak1Ser204 was also distributed on chromosomes;During anaphase Ⅰ(AⅠ) to telophase Ⅰ(TelⅠ) progression, pPak1Ser204 was detached from chromosomes and MTOC, and mainly concentrated on the cleavage furrow.Conclusions pPak1Ser204 is expressed upon meiotic resumption in oocytes .It is a MTOC-associated protein which may regulate chromosome separation through participating the formation and maintenance of spindle apparatus .
8.Serum Levels of Some Cytokines and Their Correlation with Disease Severity in Pemphigus

Objective To study serum levels of interleukin-4,10,interferon-g amma,tumor necrosis factor alpha and soluble interleukin-2 receptor in patients with pemphigus, and their correlation with disease severity. Methods Serum leve ls of different cytokines were determined by double antibody sandwich enzyme-li nked immunosorbent assay in 31 patients with pemphigus in acute and remission st ages, and in 15 normal controls. Serum autoantibody titer was determined by indi rect immunofluorescent assay (IIF). The severity of disease was recorded by the Rule of Nine for the calculation of burn surface area. Results IL-10 level was lower in the sera of patients in their acute stage than that of controls(P0.05). Conclusion There are abnormalities of serum levels of cytokine s in patients with pemphigus. Serum levels of IL-10, sIL-2R and IFN-?are cor related with the severity of the disease.
9.Partial internal sphincterectomy for the treatment of internal anal sphincter achalasia in childhood

Objective To evaluate the long-term results of partial internal sphincterectomy for the treatment of internal anal sphincter achalasia in childhood. Methods The clinical,radiographic,manometrical and histochemical data of 6 cases were reviewed retrospectively. All patients had received partial internal sphincterectomy and were followed-up for 2 to 8 years. Results All patients presented with severe constipation with or without soiling. No stenosis zone of intestine could be noted in 3 patients by barium enema examination. The rectoanal inhibition reflex on rectal balloon inflation was absent in all patients. The normal acetylcholinesterase activity on rectal biopsies was demonstrated by histochemical staining. Ganglion cells within internal anal sphincter was noted in all cases. On follow-up,all patients regained regular bowel habits and are not on any laxatives. Conclusion The long term results of partial internal sphincterectomy for the treatment of internal anal sphincter achalasia in childhood are satisfactory.
10.Association of isokinetic strength with pain and functional status in knee osteoarthritis patients aged 80 years and over

Objective To evaluate the difference in isokinetic strength of the knee muscles between knee osteoarthritis (KOA) patients aged 80 years and over and matched healthy controls,and to establish the association of isokinetic strength with pain and functional status in patients with KOA.Methods The study enrolled 32 patients aged 80 and over diagnosed with unilateral knee OA and 10 matched controls.Pain was evaluated by the visual analogue scale (VAS),the pain intensity scale and the Lequesne index for patients with knee OA.Functional status was assessed by the Timed Up and Go Test (TUGT),the Five Times Sit to Stand Test (FTSST) and the Timed 10 Meter Walk Test (TWT).Muscle strength was measured using the isokinetic dynamometer Biodex System 4 Pro.Bilateral isokinetic (concentric) knee flexion and extension with the protocol of 60 degrees/sec,180 degrees/sec was also performed.Results The difference in two angular velocities of extensor and flexor peak torques between the knee OA group and the control group was statistically significant (t=2.747,P< 0.05).The extensor peak torque was negatively correlated with age,Liquesce index scores,FTSST,TUGT and TWT (r=-0.39~~-0.75,P<0.05),but had no correlation with the VAS score or the Lequesne index pain score (r=-0.23~ 0.31,P>0.05).The regression analysis results suggested that the extensor peak torque and the VAS score together were important predictors for the Liquesce index (adjusted R2 =0.41).Conclusions The degree of knee muscle damage is closely related to the functional performance of the muscles in knee OA patients aged 80 years or over.The strength of the quadriceps and the intensity of pain are important predictors of disability for those patients.
