1.Effect of Mcl-1 siRNA on TRAIL-induced apoptosis in gastric cancer cells

Objective To explore the effect of Mcl-1 small interference RNA ( siRNA) on tumor necrosis factor-re-lated apoptosis-inducing ligand ( TRAIL)-induced apoptosis in gastric cancer HGC-27 cells. Methods The apop-totic rates of cells treated with TRAIL and pan-caspase inhibitor ( z-VAD-fmk) alone or combination were measured by propidium iodide (PI) method using flow cytometry. The activation of caspase-3, cleavage of PARP-1, as well as the protein level of anti-apoptotic Bcl-2 proteins Bcl-2, Bcl-XL and Mcl-1 before and after TRAIL treatment were monitored by Western blot analysis. Transfection of Mcl-1 siRNA was performed using Lipofectamine 2000 reagent. The efficiency of gene silencing was quantified by Western blot and the effect of Mcl-1 siRNA on TRAIL-induced apoptosis was measured using PI method. Results HGC-27 cells were resistant to TRAIL-induced apoptosis, and z-VAD-fmk pretreatment could block apoptosis nearly completely. Activation of caspase-3 and cleavage of PARP-1 occurred in the late stage of apoptosis. The expression levels of Bcl-2, Bcl-XL and Mcl-1 were not altered after exposure to TRAIL. Transfection with Mcl-1 siRNA could obviously downregulate the expression evel of Mcl-1 in HGC-27 cells and enhanced the sensitivity of cells to TRAIL-induced apoptosis. Conclusion Overexpression of Mcl-1 may account for the resistance of HGC-27 cells to TRAIL.ownregulationof Mcl-1 by siRNA can effectively enhance the sensitivity of HGC-27 cells to TRAIL-induced apoptosis.

5.Fucosylated Glycans Associated With Development and Metastasis of Hepatocellular Carcinoma Cells

Dynamic fucosylation of glycoprotein especially 80 ku which bound to UEA and LCA during the course of rat hepatocarcinogenesis was investigated. In patient hepatocellular carcinoma, more UEA- and LCA-bound proteins were also observed in patients with high metastatic potential than those with low metastatic potential. Fueosylated glycans constitute important adhesion molecules such as Lewis antigens. A differential expression pattern of Lewis antigens was further conf'Lrmed on various metastasis potential hepatocellular carcinoma cells (HCC). High metastatic hepatocellular carcinoma cell line (HMCC97H) expressed much more Lewis x and b than low metastasis HMCC97L cells. Moreover, surface Lewis x, or b expression level declined significantly after the cells were treated by retinoie acid. Not only in experimental metastasis foei, but in HCC as well, both α1,3/1,2 and α1,6 fucosyltransferase activities were quite high. After retinoie acid treatment, α1,3/1,2 fueosyltransferase activities were significantly inhibited, Lewis x on epidermal growth factor receptor reduced, and the EGFR was less phosphorylated. These results suggested that fucosylated glycans such as Lewis x played an important role in HCC development and metastasis.
6.Optimization of preparation for cyanidenon-?-cyclodextrin inclusion compound by Box-Behnken design and response surface method

Objective To optimize the formulation of cyanidenon-?-cyclodextrin inclusion compound.Methods Cyanidenon-?-cyclodextrin inclusion compound was prepared by saturated water solution method.The effects of influence factors,such as ingredient proportion,inclusion time,inclusion temperature on inclusion rate,obtaining rate,and overall desirability were investigated by using Box-Behnken design and response surface method.The data were imitated using multi-linear equation and second-order polynomial equation.Results The latter was prior to the former considering from multiple correlation coefficients.Under the optimal conditions,inclusion rate and yield were 75% and 66%,respectively.Conclusion The optimized preparation technique for cyanidenon-?-cyclodextrin inclusion compound is stable,feasible,and has high inclusion rate.It can be used for the production of cyanidenon-?-cyclodextrin inclusion compound.
7.Nursing of patients with rheumatic valvular heart disease with atrial fibrillation treated with CoxⅢ labyrinth and simultaneous valve replacement operation

Objective To summarize the effect and experience of nursing of patients with rheumatic valvular heart disease with atrial fibrillation treated with Cox Ⅲ labyrinth and simultaneous valve replace-ment operation. Methods A retrospective analysis was carried out in the nursing of 58 patients with rheumatic valvular heart disease with atrial fibrillation treated with Cox Ⅲ labyrinth and simultaneous valve replacement operation in our hospital from April 2006 to July 2008. Results One patient died of periop-erative renal failure,one died of low cardiac output,one died of infectious endoearditis.Tbe mortality rate reached 5.2%.Other patients were discharged from hospital after completely cured.The survived patients maintained sinus rate during hospitalization days after operation and short-term follow-up after being dis-charged.Among 43 patients who completed follow-up of 6 months after operation,3 patients (7.0%) damon-strated atrial fibrillation,while others still kept sinus rate. Conclusions Attention should be paid to psy-chological nursing,respiratory tract preparation,cardiac function monitoring and nursing of anticoagulation treatment for patients treated with Cox Ⅲ labyrinth and simultaneous valve replacement operation.

9.Investigaion about the mRNA expression of ASB2 and Jak3 in patients with acute lymphoblastic leuke-mia

Objective To explore the expression of ankyrin-repeat SOCS box containing protein 2 (ASB2)and Janus kinase 3 (Jak3)mRNA in bone marrow plasma mononuclear cells of patients with acute lymphoblastic leukemia (ALL)and discuss the correlation of them.Methods Bone marrow plasma was collected from 48 patients of newly diagnosed ALL (37 cases B-ALL and 11 cases T-ALL)and 34 non leukemia patients (control group),respectively.Expression levels of ASB2 and Jak3 mRNA were detected by real-time quantitative PCR.Results The expression of ASB2 mRNA in B-ALL and T-ALL were significantly different compared to control group (t =14.2,P <0.01;t =15.2,P <0.01),which were 68.5,32.7 folded more than control group,respectively.Moreover,the expression level of Jak3 mRNA in B-ALL group and T-ALL group were 2 336.1 and 7 131.5 folded more than control group (t =37.3,P <0.01;t =37.6,P <0.01). At last,the expression of ASB2 and Jak3 gene was positive correlation (r =0.523,P <0.001).Conclusion ASB2 and Jak3 are expressed abnormally in ALL patients and the correlation of them is positive.ASB2 and Jak3 may be related to the proliferation and differentiation in leukemia cells.
10.Determination of frequencies of Th17 cells in human peripheral blood by flow cytometry

Objective To investigate the effect of different stimulating culture conditions and detecting methods on the frequency of Th17 cells and to set up and optimize the experimental method for detecting the frequencies of human Th 17 cells in peripheral blood by flow cytometry (FCM ) .Methods The peripheral blood in the healthy people was performed the stimulating culture by phorbol myristate acetate (PMA) at different final concentrations(50 ,100 ,150 ,200 ,250 ,300 ng/mL) and ionomycin 1 μg/mL .The frequencies of Th17 cells were assessed by FCM after different stimulating time points (4 ,6 ,8 ,10 ,12 h) .The mean fluorescence in-tensity(MFI) and percentage of the ratio of CD4 positive cells on the cellular surface and inside cell membrane were measured be-fore and after PMA stimulation .Results The frequency of Th17 cells was highest with the good stimulation effect under the condi-tion of PMA 150 ng/mL and ionomycin1μg/mL stimulation for 6 h (P<0 .05) .MFI of CD4 inside cellular membrane after stimula-tion was lower than that in the cells with stimulation after labeling membrane surface CD 4(P<0 .05) ,and the Th17 cell frequencies and the ratio of CD4 positive cells were higher than those in the latter ,but the differences were not statistically significant (P>0 .05) .Conclusion After stimulating whole blood with the final concentration of PMA 150ng/mL and ionomycin1μg/mL for 6 h , measuring intracytoplasm CD4 and IL-17 levels is the simple and effective method for detecting the frequency of human Th 17 cells .
