1.A New Spot Test Method for Nitrite in Water

Objective To introduce a rapid spot test method for nitrite in water to meet the urgent need of nitrite monitoring during accidental pollution. Methods According to the drinking water legislation in Europe (threshold: 0.10 mg/L NO2-), a rapid spot test method for nitrite in water was established by using an optimized composition of reagent mixture and a binary positive/negative response, conducted as follows: a tablet of a mixture of solid reagents containing sulfanilamide, N-(1-naphthyl) ethylene diamine dihydrochloride and sodium chloride was put on a spot test plate of Teflon mould, then, a drop of 30%(V/V) HCl and a drop of water sample were added on the reagent material. Results Under these conditions, all the test samples containing ≥0.10 mg/L NO2- can produce a positive response of the characteristic red-purple color, whereas, the samples will produce a negative response when the level of NO2- was below the limit. The results of real water samples determined with this new method were confirmed by using the photometric method based on the use of the same reagents. Conclusion The method is simple, rapid , accurate, and suitable for field monitoring of nitrite in water.
3.Checking to the Proportional Hazards Assumption of the Cox′s Proportional Hazards Model.

Objective Exploring how to check the proportional hazards assumption of the Cox model,and the solutions to non-proportional hazards between the covariates and the hazard function.Methods With a example data set of Ⅲc stage ovarian serous cystadenocarcina,illustrating how to use graphical methods to check the proportional hazards assumption of the Cox model.Results the predictor of post-surgery administering medicine times violated the proportional hazards assumption of the Cox model.Conclusion when using the Cox model to analyze the predictors of survival time,checking whether the predictors violate the proportional hazards assumption of the Cox model or not should be paid attention to.
4.Observation on the effect of endoscopic sinus low-temperature plasma-assisted sinus surgery combined with antifungal drug in treatment of fungal nasal-sinusitis

Objective To study the clinical effect of surgery combined with antifungal drug in treatment of fungal nose-sinusitis.Methods The clinical data of 108 cases with fungal nose-sinusitis were retrospectively analyzed.All patients were treated with nasal endoscopic sinus surgery.According to whether the patients received antifungal drugs after low-temperature plasma-assisted surgery,the patients were divided into the treatment group and the control group.The treatment group(56 cases) was given antifungal fluconazole after low-temperature plasma-assisted endoscopic sinus surgery.52 cases in the control group were washed by 0.9% sodium chloride solution after conventional endoscopic sinus surgery.The patients were followed up for 6 months to 2 years,the therapeutic effect and recurrence were compared between the two groups.Results 56 patients of the treatment group,49 cases were cured,7 cases improved,no recurrence.52 cases in the control group,47 cases were cured,improved in 4 cases,ineffective in 1 case,3 cases of recurrence.The recurrence rate of the treatment group was significantly lower than the control group (x2 =4.82,P < 0.05).Conclusion The low-temperature plasma-assisted surgery combined with antifungal drug.in the treatment of fungal nose-sinusitis has exact efficacy,and has significant effect on the prevention and reduce the relapse rate,which worthy of clinical promotion.
5.Effect of triptolide on the proliferation and apoptosis and on Wnt/β-catenin pathway in imatanib resistant CML cell

Objective To investigate the impact of triptolide (TP) on proliferation and apoptosis of imatanib resistant CML cell (K562/G01) and its regulating effect on Wnt/β3-catenin signal pathway.Methods A series of 10,20,40,and 80 nmol/L of triptolide were used in CML cells K562/G01 for 12,24,and 48 hours.The cell proliferation was detected with methyl thiazolyl tetrazolium (MTT) test.The apoptosis was assessed with flow cytometry (FCM).The mRNA expressions of breakpoint cluster region-c-abl (BCR-ABL),β-catenin and its down-stream targets Lef-1,and cyclinD1 were analyzed with real-time quantitative polymerase chain reaction (RT-PCR),respectively.Results Triptolide significantly inhibited K562/G01 cell growth ability and induced apoptosis in a dose-dependent manner.Mter being treated with 20,40 nmol/L TP for 24 hours,the cell growth inhibition rates were (22.62 ± 1.33) %,and (51.41 ±1.39) %,respectively.The late apuptosis rates were (6.91 ± 0.14) %,and (7.64 ± 0.47) %,respectively.Meanwhile,PCR data showed that the mRNA levels of BCR-ABL were decreased,compared to the control group,the mRNA levels of β-catenin,Lef-1,and cyclinD1 were also decreased obviously after treatment.Conclusions Our data indicated that the triptolide could inhibit the proliferation and induce the apoptosis of K562/G01,and the mechanism might be related to the blockade of Wnt/β-catenin signal pathway.
6.Hypoxia/reoxygenation induced endoplasmic reticulum stress in cultured neonatal rat cardiomyocyte

Objective: To investigate the effect of hypoxia/reoxygenation on endoplasmic reticulum stress in cultured neonatal rat cardiomyocytes. Methods: Neonatal rat cardiac myocytes in primary culture were exposed to hypoxia for 5.5 hours and subsequently reoxygenation for 2-24 hours. Western blot and RT- PCR were applied to monitor the expression change of GRP78(glucose regulated protein 78). 2-deoxy-D-glucose(2-DG) was the positive control of this study. Then Western blot and RT- PCR were used to examine the expression of GRP78. Results: Cell viability was decreased obviously after hypoxia/reoxygenation. Compared with untreated cells, the GRP78 content of the cells had increased significantly in the hypoxia/reoxygenation cells. The level of GRP78 protein and mRNA elevated from the points of 2 hours to 24 hours after reoxygenation, and increased most obviously at the point of 4 hours after reoxygenation.(4 hours: protein level 142% of the control, mRNA level 200%). 2-DG could induce the increasing expression of GRP78 in a concentration-dependent manner from 10-50 mmol/L. Conclusion: Hypoxia/reperfusion can induce endoplasmic reticulum stress in rat cardiomyocytes.
7.Effect of Shenkangwan on TGF-?1/Smad signaling pathway in rat diabetic nephropathy

Objective To explore the mechanism of Shenkangwan in preventing the development of diabetic nephropathy(DN)through TGF-?1/Smad signaling pathway.Methods The diabetic rat models were established by intraperitoneal injection of streptozotocin(STZ)in male Wistar rats and randomly divided into 3 groups:model control group,Capoten group and Shenkangwan group.Another 8 normal Wistar rats served as normal control.After the rats had been treated for 8 weeks,the mRNA expressions of fibronetin(FN)and TGF-?1 were detected by RT-PCR and the protein expressions of Smad 2,Smad 3 by immunohistochemistry in the renal tissues.Results During the past 8 weeks,the blood glucose of the rats in the model control group was all over 16.7 mmol/L and they exhibited the symptoms of diabetes mellitus,such as excessive thirsty,extreme hunger,frequent urination and unusual weight loss.The mRNA expressions of FN and TGF-?1 in rat renal tissues of model control group were significantly higher than those in the rats of normal control group(P

Objective:The purpose of the study was to investigate the effects of enteral nutrition on stress response and gut barrier function after surgery for esophageal cancer. Methods: Patients who underwent esophagectomy were divided into two groups. Group PN (28 patients) served as the control group. Group EN (26 patients) received enteral nutrition. Results: The serum total protein, albumin, prealbumin and transferrin decreased after operation, but there were no differences between the two groups. 9.Gene expression differences between murine intestine intraepithelial lymphocytes and splenic T lymphocytes

Objective: To compare the differences in gene expression between murine intestine intraepithelial lymphocytes (iIELs) and splenic T lymphocytes, establishing the cDNA subtractive library of iIELs, analyzing the iIELs special genes. Methods: The iIELs and splenic T lymphocytes were isolated by density gradient in Percoll and nylon column respectively. cDNA subtractive library of iIELs was established by improved subtractive hybridization. The cDNA was ligated with a different adaptor. After 2 rounds of hybridization and suppression PCR amplification, the PCR products were directly inserted into the plasmid vectors. The ESTs special for iIELs were screened by reverse Northern blot. Results: Fifty ESTs with obvious difference in the cDNA subtractive library of iIELs were screened. These ESTs were squenced and submitted to Genebank. The ESTs were analyzed through BLAST, and 14 genes special for iIELs were obtained. These genes were associated with the pattern recognition of pathogen, cytokine signal transduction and proteinase inhibitors. Conclusion: iIELs are important components of intestine natural immunity, it can recognize the conservative antigens of bacteria and can be affected by cytokines and proteinase inhibitors, leading to unique cellular behavior--more resting and less activation.
10.Research on current education situation and demand of network teaching for health service management course

Objective To explore the current education situation and demand of network teaching for health service management course so as to provide recommendations for improvement.Methods Questionnaire survey was conducted among 232 undergraduates from majors of preventive medicine,stomatology and clinical medicine.Totally 204 valid questionnaires were organized and recorded into database and SPSS 12.0 was employed to make descriptive statistical analysis of the data.Statistical results were used to show the current education situation and demand of network teaching for health service management course.Results 52.94% (108/204) of the students interviewed were dissatisfied with current teaching mode.87.25% (178/204) of the students believed the course website would be beneficial for learning.For the construction of the course website,health insurance system,health policy analysis and health organization system were considered both important and difficult.59.31% (121/204) of the students showed that the course website should be updated at least once a week.Conclusions It is necessary to explore the network teaching mode for health service management course.Teaching reform depending on different professions and curriculums should be carried out for network teaching.
