1.Influencing factors for tracheotomy after cervical spinal Cord injury

[Objective] To investigate risk factors and the incidence rate of tracheotomy after cervical spinal cord injuries.[Method]A retrospective analysis was done on 1185 cases suffering from cervical spinal cord injuries statistically.The following correlation factors: ages,gender,type of job,reason of injuries,degree of spinal cord injuries,segment of spinal cord injures,nutritional status during therapy were evaluated by Binary Logistic Regression to discover tile influencing factors for tracheotomy.[Result]Totally 99 cases were undergone tracheotomy(male 87 and female 14).The rate of tracheotomy was 8.35%,Ages,degree of spinal cord injuries,segment of spinal cold injuries,reason of injuries had influence to rate of tracheotomy.The other factors were not correlation to tracheotomy.With increasing of age,the rate of tracheotomy was gradually increased.Nobody was undergone tracheotomy in patients who was younger than 20 years old,and the rate of tracheotomy was 38.7% in patients who was older than 60 years old.With aggravating and segment going up of cervical spinal cord injuries,the rate of tracheotomy was gradually increased.The rate of tracheotomy for cervical spinal cord injury at Grade D was 0.5%,While the rate of tracheotomy for cervical spinal cord complete injury was 19.1%.Tile rate of tracheotomy for cervical spinal cord complete injury was 4.3,22.3,21.6 and 32.7% from C7 to C4,while the rate wasn't high in C3 and C2,because most of these patients had died before they was undergone tracheotomy.The rate of tracheotomy was higher in patients who got injuries from fall than other.[Conclusion]Elder,cervical spinal cord severe injuries,high cervical spinal cord injuries was risk factors for tracheotomy.The patients who got cervical spinal cord complete injuries in C3 or higher than C3 need tracheotomy as early as possible.
2.Determination of Heavy Metals Content in Five Kinds of Chinese Medicinal Material by ICP-MS

Objective To establish an ICP-MS method for the determination of heavy metals,including copper(Cu),arsenium(As),cadmium(Cd) and plumbum(Pb),in five kinds of Chinese medicinal material.Methods The samples were digested by closed-vessel microwave.The four heavy metals were directly analyzed by ICP-MS,with elements 72Ge,115In,and 209Bi as the internal standards.Results For all of the analyzed heavy metals,the correlative coefficient of the calibration curves was in the range of 0.999 5~ 0.999 7.The recovery rates were in the range of 92.3 % ~ 108.0 %,and RSD were in the range of 2.4 % ~ 5.8 %.Conclusion The method is convenient,rapid,accurate and can be applied to determine four heavy metals,including Cu,As,Cd and Pb,in Chinese medicinal material.
3.Allergen Specific Serum IgE Antibody in Patients with Intestinal Polyp and Irritable Bowel Syndrome:A Clinical Analysis

Background:Eosinophilic gastroenteritis is the most common gastrointestinal allergic disease,however,allergy also accounts for some gastrointestinal functional disorders. The correlation between allergic factor and intestinal polyp is not clarified. Aims:To investigate the correlation of allergic factor with intestinal polyp and irritable bowel syndrome(IBS). Methods:Ninety-five inpatients including 60 cases of intestinal polyp and 35 cases of IBS admitted from May 2012 to April 2014 were recruited;95 healthy volunteers were served as controls. Serum samples were collected to test specific IgE antibodies derived from 20 common inhalant and alimentary allergens by EUROIMMUN blotting test. Results:The positivity rates of allergen specific serum IgE antibodies in intestinal polyp group,IBS group and both two groups were 56. 67% ,74. 29% and 63. 16% ,respectively,all were significantly higher than that in control group(6. 32% ,P all <0. 05),but no statistically significant difference was seen between intestinal polyp group and IBS group(P > 0. 05). In all the 20 allergens tested,the positivity rates of Dermatophagoides,roach,crab,sea fish,freshwater fish,shrimp,and egg albumin specific serum IgE antibodies were significantly higher in patients group than in control group( P < 0. 05), especially the crab,see fish and Dermatophagoides specific IgE antibodies. Conclusions:Allergic factor might have some association with intestinal polyp and IBS.
4.Application of Cytological Examination of Cerebrospinal Fluid in Diag nosis of Central Nervous System Leukemia

Objective To investigate the diagnosis value of the cytological examination of the cerebrospinal fluid of central nervous system leukemia.Methods Adopting cell smear centrifugal machine to collect the cerebrospinal fluid cells,the cells were stained and examinated under the microscope.Results Fifty-nine children with different type of leukemia had been examinated by 438 times by cerebrospinal fluid.The positive rates of the cases and samples were 15.3% and 8.7%,respectively.Conclusion The cytological examination of cerebrospinal fluid is especially valuable for the early diagnosis ,therapy and relapse of central nervous system leukemia of monitoring.
5.The effects of electroacupuncture on learning, memory and hippocampal synaptic plasticity in vascular dementia

Objective To investigate the effects of electroacupuncture (EA) on spatial learning and memory ability, long-term potentiation (LTP) and synaptophysin (Syp) levels in the dentate gyrus of the hippocampus.Methods Forty Sprague-Dawley rats were randomly divided into a sham-operated control group, a model group, an EA group and a medication group. A rat model of vascular dementia (VD) was established by repeated ischemia and reperfusion after pretreatment with sodium nitroprusside. EA was applied to the rat homologues of the human "Baihui" (GV20) and "Dazhui" (DU14) acupoints of the rats in the EA group for 20 min, once daily for 10 d.Morris water maze tests were conducted for evaluating the rats,learning and memory ability. LTP in the hippocampal dentate gyrus was recorded as an electrophysiological index for evaluating learning and memory. Antigenic Syp produced in hippocampal tissue was examined with immunohistochemical assays. Results In comparison the with model group, escape latency (EL) shortened and target-platform crossing times increased significantly in the EA and medication groups. After high frequency stimulation the population spike (PS) latency shortened in the EA group compared with the model group, and the excitatory postsynaptic potential (EPSP) slope and PS amplitude increased.All of these differences were statistically significant. Syp positive cells also increased significantly in the EA-treated rats. Conclusions EA can increase Syp expression and improve the learning and memory ability of rats with this model of VD. EA could facilitate the induction and maintenance of LTP in the dentate gyrus of the hippocampus through influencing synaptic transmission.
6.Analysis on the HIV results of clients in general hospital

Objective To investigate the infection situation of HIV in patents in hospital ,and to provide basis for the prevention of AIDS in the hospital .Methods Three kinds of enzyme‐linked immunosorbent assay(ELISA)kits(Beijing wantai ,Beijing kewei , French BIO‐RAD)were used for screening HIV antibody and antigen in 240 781 samples from Jan .2011 to Dec .2013 .All HIV re‐peated positive screening samples were confirmed by western blotting in Chongqing shapingba Center for Disease Control and Pre‐vention .Results Among 240 781 samples from 2011 to 2013 ,593 samples(0 .246% ) were HIV positive at the first screening ,558 samples(0 .231% ) were HIV positive in at the repeated screening ,29 samples(0 .012% ) were HIV indeterminate ,6 samples(0 . 002% ) were HIV negative.Male and female ratio was 3 .39:1 .Conclusion Screening in hospital patients could be an important way to discover cases with HIV infection .It is nesscessary to strengthen the promotion and propaganda of HIV detection ,and new technology of HIV detection could be used to strengthen the inspection of HIV .Moreover ,the consciousness of self protection should be promoted in the treatment of HIV patients .
8.EdCC reduces myocardial ischemia-reperfusion injury via MEK-ERK signaling pathway

AIM:To investigate the myocardial protective effect of endometrial stem cell ( EnSC)-derived cyto-kine cocktail ( EdCC) on myocardial ischemic reperfusion injury and the MEK-ERK signaling pathway.METHODS: A mouse model of myocardial ischemic reperfusion injury was established.Infarct area, cell apoptosis, and expression of cleaved caspase-3 and phosphorylatied ERK1/2 were determined by TTC/Evans blue staining, TUNEL assay and Western blot, respectively.RESULTS:The mesenchymal characteristics were observed in the EnSCs with expressing CD90 and in absence of CD34 and CD45.EdCC contained (6 811 ±312) ng/g epidermal growth factor (EGF) protein.The phospho-rylation of ERK1/2 markedly increased after injection of EdCC, but was abolished by MEK1 inhibitor PD98059 ( 5 mg/kg) .EdCC decreased the infarct area and apoptotic cell number in the border zone and inhibited caspase-3 activation. However, the effects were abolished by MEK1 specific inhibitor PD98059.EGF did not decrease the infarct area, but the EGF receptor antagonist AG-1487 (6 mg/kg) partly abolished the myocardial protective effect of EdCC.CONCLUSION:EdCC protects the myocardium from ischemic reperfusion injury via activating MEK1-ERK signaling pathway, indicating an essential role in the transmission of stem cell therapy from the cell transplantation to cytokine based strategy.
9.Expression of interferon-stimulated gene ISG20 and investigation of its antiviral effect on HCV replicon

Objective:To study the antiviral effect of ISG20 on HCV replicon.Methods:Wild type ISG20/mutated ISG20 cDNAs were obtained by RT-PCR/two step-PCR directed mutagenesis, and wild type ISG20 and dominant negative mutated ISG20 mammal expression vectors were consuructed. The constructed pISG20wt and pISG20m expressing vectors were transfected into Huh7 cells or Huh7 cells containing HCV replicon to investigate its effects on HCV replicon replication.Results:The ISG20wt/ISG20m expression vectors were constructed and the expressions of these two vectors were confirmed at both mRNA and protein levels. The effects of ISG20wt on HCV replicon replication were evaluated by Northern blot and Western blot. The results showed that expression of ISG20wt had significant inhibitory effect on HCV RNA replication.Conclusion:ISG20 participates in the anti-HCV action of IFN-? on HCV replicon system.
10.Toxicological study on Jiutong Capsule

AIM:To estimate the toxicological characterization of Jiutong Capsule(Radix Puerariae lobatae,Fructus seu Semen Hoveniae,etc.).METHODS:The acute toxicity test,micronucleus test of bone marrow cell,sperm shape abnormality test in mice,the Ames test,and 30 days feeding test in rat were performed.RESULTS:(1) The acute toxicity test showed that LD_ 50 of Jiutong Capsule was more than 10.0 g/kg.(2) The result of Ames test,micronucleus test of bone marrow cell,and sperm shape abnormality test were negative.(3) The 30 days feeding test showed that Jiutong Capsule had no cumulate toxicity in mice.CONCLUSION:The results show that Jiutong Capsule does not have toxicity,and it can't cause mutation and heredity toxicity.
