Chinese Journal of Tissue Engineering Research 2015;(47):7703-7708
BACKGROUND:Currently, in pterygium excision combined with limbal epithelial transplantation, there were stil some controversies about whether the medical fibrin glue could instead of stitch fixing autologous conjunctival graft. OBJECTIVE:To compare the therapeutic effect of medical fibrin glue and stitch in pterygium excision combined with limbal epithelial transplantation using Meta analysis. METHODS: The randomized controled trials about the therapeutic effect of medical fibrin glue versus stitch in pterygium excision combined with limbal epithelial transplantation were reviewed from PubMed database, CNKI ful-text database, Chinese Biomedical Literature Database, VIP database and Wanfang database from database creation to June 2015. The operative time, complications and recurrence rate of these two groups were analyzed using Meta. RESULTS AND CONCLUSION:A total of 14 randomized controled trials, and 879 patients were enroled. Meta-analysis results showed that the operative time, incidence of postoperative complications, the relapse rate after six months and one-year in the fibrin glue group were lower than those in the suture group [MD=-15.58, 95%CI (-18.96,-12.21),P < 0.000 01]; [OR=0.39, 95%CI(0.19, 0.68),P=0.02]; [OR=0.28, 95%CI(0.12, 0.65),P=0.003]; [OR=0.23, 95%CI(0.07, 0.76),P=0.02]. These results demonstrate that the application of fibrin glue in pterygium excision combined with limbal stem cel transplantation can promote graft attachment, shorten the operation time, reduce postoperative complications and reduce the postoperative recurrence of pterygium.

Aim To explore the effects of hypertension on the contractibility of rat basilar artery and its inter-actions with the sodium pump activity. Methods The basilar artery was respectively isolated from Wistar and SHR rats, and the isometric tension of arterial rings was measured by Multi Myograph System-610M.The cont-ractibilities of arterial rings induced by KCl or 5-HT were compared between the basilar arteries of the two groups of rats to analyze the effect of hypertension on the cerebral vascular tension and the activity of sodium pump. Results In SHR rats, the concentration-re-sponse curves of the contraction of isolated basilar ar-tery rings induced by KCl and 5-HT were significantly shifted to right, and the relaxation of vascular tone in-duced by K+which was reintroduced from the external was attenuated compared with those in the WR.These results suggested that hypertension could significantly decrease the activity of the sodium pump and the con-tractile responses of KCl and 5-HT.OUA could con-tract the basilar artery in a concentration-dependent manner, and its concentration-response curve was opti-mally fitted by a two-site binding model:Kd was 1.7 ×10 -8 and 1.6 ×10 -5 mol·L-1,respectively.The results indicated that the two different function sodium pumps existed in the rat basilar artery:one with the high OUA affinity and the other with the low OUA af-finity.If the high and low affinity sodium pumps were inhibited by 5 ×10 -7 and 10 -4 mol · L-1 OUA,re-spectively,the concentration-response curves of KCl and 5-HT would shift to left in SHR rats but not in WR rats.It suggested that OUA could enhance the contrac-tion induced by KCl and 5-HT significantly, and a concentration-dependent effect was observed in the SHR vascular contraction induced by 5-HT (r =0.9393 ,P<0.05 ).When the two concentrations of OUA were applied, there was no significant difference in the shift left of the concentration-response curves in-duced by KCl in the SHR cerebral vessels. However, the marked difference was shown in the shift left in-duced by 5-HT.The results implied that only the high affinity sodium pump was involved in the contractile re-sponse of SHR cerebral vascular to KCl,whereas, the contractile response of SHR cerebral vascular to 5-HT was induced by both high and low affinity sodium pumps. Conclusion Hypertension could lower the contractile response of the basilar artery to vasocon-strictors,and the mechanism might relate to the de-creased sensitivity of the sodium pump induced by hy-pertension or the increased sensibility of the sodium pump to OUA.

ObjectiveTo investigate the expression of caspase-3 in the brain of acutely pentylenetetrazole(PTZ)-kindled rats.MethodsThe caspase-3 positive cells were revealed using immunohistochemical SP method. CMIAS image analysis system was used to analyse the expression of caspase-3 hemi-quantitatively. ResultsFollowing PTZ induced epilepsy, the expression of caspase-3 increased both in the hippocampus and in the cortex, and that was more remarkable in the hippocampus than in the cortex.ConclusionCaspase-3 may be activated during neuronal apoptosis after epilepsy. Hippocampus is more sensitive to the neuronal damage due to epilepsy than the cortex is.

AIM: To determine the concentration of nisoldipine in human plasma by HPLC-MS method and investigate the pharmacokinetics of sustained and immediate-release preparations. METHODS: A C 18 column was used to separate nisoldipine from plasma with the mobile phase of a mixture of methanol-water-acetic acid (7525 0.1) at a flow rate of 1.0 ml?min -1. MS: atmospheric pressure electronic spray ionization (AP-ESI) and ion mass spectral (m/z) of 411 were selected to quantify nisoldipine. Internal standard (IS): atmospheric pressure electronic spray ionization and m/z of 441 for nimodipine. RESULTS: The linear range of the standard curve of nisoldipine was 0.2- 50 ?g?L -1 and the determination limit was 0.15 ?g?L -1. The recovery rate was more than 70%, and intra-day relative standard deviation (RSD) and inter-day RSD were less than 10%. After being given a single dose of 10 mg nisoldipine sustained release tablet, sustained release capsule and normal tablet, the half life(t 1/2 /h) were 6.08? 1.48, 7.06? 1.80 and 3.70? 0.25, the time to peak concentration (T peak /h) were 5.4? 0.7, 5.8? 0.4 and 2.0? 0.2, the peak concentration (C max / ?g?L -1) were 3.43? 0.55, 3.71? 0.24 and 9.18? 3.78, the area under time- concentration curve (AUC 0-t / ?g?h -1?L -1) were 31.10? 5.00, 33.63? 7.16 and 32.72? 5.09. But after being given multiple doses of nisoldipine, C max/ ?g?L -1 were 5.20? 0.27, 3.91? 0.22 and 5.30? 1.04, C min / ?g?L -1 were 0.72? 0.10, 0.77? 0.07 and 0.53? 0.07, DF were 175.00%? 16.34%, 177.10%? 18.43% and 247.92%? 57.71% respectively. The bioavailability of sustained- release tablet and capsule were 96%?12% and 102%?9% respectively. CONCLUSION: The determination of concentration of nisoldipine in human plasma by HPLC-MS method is sensitive and accurate. It can be used for the investigation of the bioavailability and pharmacokinetic of nisoldipine.
