A Review Many studies indicate that apoptosis is involved in the progression of congestive heart failure. At present, mechanisms that mitochondria regulates cell apoptosis is widely accepted.Cardiac myocytes have abundant mitochondria,which plays an important role in maintenance of cell physiological function. Recent studies find that cardiac energy metabolic shifts occur as a normal response to diverse physiologic and dietary conditions and as a component of the pathophysiologic processes which accompany cardiac hypertrophy, heart failure, and myocardial ischemia.Both clinical and experimental studies show that cardiac function can be improved and apoptosis is inhibited by intervention in energetic metabolism of myocytes. 2.Effects of colon hydrotherapy on lowering body temperature,serum enzymes and inflammatory cytokines of dogs with heat stroke

Objective To investigate the effects of colon hydrotherapy on lowering body temperature and serum enzymes as well as inflammatory cytokines of dogs with heat stroke.Methods 14 dogs were randomly devided into 2 groups,7 for each,as colon hydrotherapy group and conventional therapy group.After the reproduction of heat stroke model was finished(core temperature reached 43℃,maintaing for 10min),cooling therapy was commenced on dogs in circumstance with temperature at 23℃ and 65% of humidity.Conventional therapy group was lowered temperature by ice bags placed on conducting arteries,and the colon hydrotherapy group was by colon way therapeutic machine,the temperature of therapeutic liquid was maintained between 37℃~38℃.Cooling was finished while temperature in dogs reduced to 38℃.The periods of time for lowering body temperature in both groups were compared.Alanine aminotransferase(ALT),aspartate aminotransferase(AST),lactate dehydrogenase(LDH)and creatine kinase(CK)were determined before heat stroke(0h),and 6h,24h and 48h after heat stroke.Interleukin-1?(IL-1?),Interleukin-6(IL-6)and tumor necrosis factor ?(TNF-?)were determined by radioimmunity assay.Results Body temperature lowering time in dogs with colon hydrotherapy were 92.9?9.5min,which was significantly shorter than that of conventional therapy group(123.4?11.6min,P0.05).IL-1? in colon hydrotherapy group at 6h,24h and 48h after treatment were significantly lower than those in conventional therapy group(P

Objective To investigate the changes in nitric oxide synthase (NOS) activity in cardiac fibroblasts (CFs) of neonatal rat induced by aldosterone. Method CFs were isolated by trypsin digestion method. Proliferation of CF was assessed by MTT colorimetric assay. Nitric acid reductase method and spectrophotometry were used to determine the NO contents and NOS activity, respectively, at different time after CFs being treated with aldosterone. Result Aldosterone (1?10 -11 -1?10 -7 mol/L) could increase CFs proliferation in a dose-dependent manner. 4.Diagnostic value of gram-stained sputum smear in elderly patients with ventilator-associated lower respiratory infection

Objective To explore the diagnostic value of gram-stained sputum smears in elderly patients with ventilator-associated lower respiratory infection (VALRI).Methods 368 elderly patients undergoing mechanical ventilation over 48 hours in ICU were retrospectiely analyzed from Jun 2009 to Jun 2011.42 patients fulfilled the inclusion criteria (with good-quality sputa and presence of the same microorganism in sputum and blood cultures).58 gram-stained smears and sputum cultures were obtained and analyzed.Results The diagnostic sensitivity and specificity of Gram-positive bacteria and Gram-negative bacteria by gram-stained smears were 82.35% and 78.05%,80.49% and 82.35%,respectively.The positive and negative predictive value of Gram-positive bacteria and Gramnegative bacteria were 63.64% and 91.43%,91.67% and 60.87%,respectively.The coincidence rates in diagnosing Gram-positive bacteria and Gram-negative bacteria were 81.03% and 79.31%,respectively.Conclusions Gram-stained sputum smear is reliable in diagnosing ventilator-associated lower respiratory infection in elderly patients,and has a certain clinical significance in guiding to the selection of antibiotics.
5.Isolation, culture and characterization of endothelial progenitor cells from the human peripheral blood

BACKGROUND:Endothelial progenitor cel s, known as the precursor cel s of mature endothelial cel s, have the function of neovascularization and neoendothelialization. Therefore, endothelial progenitor cel s have potential applicability in many fields. Endothelial progenitor cel s can be isolated and cultured from different resources with different methods, but the biological properties and identification of endothelial progenitor cel s stil have controversies.
OBJECTIVE:To explore the methods of isolation and culture of endothelial progenitor cel s from the human peripheral blood and to identify the biological features of endothelial progenitor cel s.
METHODS:Mononuclear cel s were isolated from the human peripheral blood using density gradient centrifugation, and the cel s were resuspended in endothelial basal medium-2 supplemented with the EGM-2-MV-SingleQuots. Then, the cel s were inoculated in human fibronectin-coated culture flasks and cultured in EBM-2MV medium. The morphology of endothelial progenitor cel s was observed. The proliferation potential and surface markers of endothelial progenitor cel s were characterized careful y. Furthermore, the functional properties such as nitric oxide release and tube formation on Matrigel were also evaluated.
RESULTS AND CONCLUSION:While adherent cel s maintained, spindle-shaped cel s formed a cel cluster after 6-7 days. Then, adherent cel s developed to endothelial progenitor cel s with a cobblestone appearance after 2-3 weeks. The endothelial progenitor cel s were confluent with an outgrowth appearance. Endothelial progenitor cel s had a higher proliferation potential compared with human aortic endothelial cel s under the same culture condition. Endothelial progenitor cel s expressed CD31, CD34, CD144 and KDR, displaying an obvious endothelial phenotype. Endothelial progenitor cel s were also found to uptake DiL-acLDL and exhibit lectin binding capability. Furthermore, endothelial progenitor cel s were able to form capil ary tubes on Matrigel and had the ability to release nitric oxide. Therefore, endothelial progenitor cel s can be obtained from the human peripheral blood by density gradient centrifugation and adherent culture. A combining method for the identification of endothelial progenitor cel s should be recommended.
6.Clinical study of 40 cases with chronic hydrocephalus after traumatic subarachnoid hemorrhage

Objective To discuss the clinical features and treatment methods of chronic hydrocephalus after traumatic subarachnoid hemorrhage.Methods A total of 40 cases of chronic hydrocephalus after traumatic subarachnoid hemorrhage were analyzed retrospectively.Results Eight cases improved after treatment of non-operation,the lateral ventricle external drainage was done in 32 cases of chronic hydrocephalus.Shunt tube obstruction occurred in 3 patients,with abdominal,subcutaneous and intracranial infection.The shunting operation device was pulled out and the 3 patients received operation for the second time.Two cases appeared intracranial hemorrhage,and the hemorrhage was absorbed after conservative treatment.Total Glasgow prognosis score was good in 32 cases,with mild disability in 5 cases,moderate disability in 2 cases and severe disability in 1 case.Conclusions Traumatic subarachnoid hemorrhage after chronic hydrocephalus should be early diagnosed and early treated,CT and MRI scan is the effective means to the diagnosis and differentiaion.The lateral ventricle external drainage is good method in treatment.
7.Combined Chemotherapy of HCPT,MTX,Leucovorin and 5-Fu for Metastatic Breast Cancer

Objective:The therapeutic efficacy and side effects of combined chemotherapy of HCPT,MTX,LV and 5-Fu for metastatic or recurrent breast cancer were evaluated in our study.Methods:A total of 43 cases of advanced metastatic or recurrent breast cancer were treated with chemotherapy regimen consisting of HCPT 10mg/m2 iv gtt for dl~5,MTX 100mg iv dl,Leucovorin 150mg/m2 iv gtt for d2~4,5-Fu 500mg/m2 iv gtt for dl~5.The cycle was repeated every 4 weeks,and 2 cycles were given as one course.Results:The overall CR+PR was 47%.One year survival rate was 54% and the median survival interval was 19 months.The main side effects were bone marrow suppression and gastrointestinal reaction.Conclusion:The combined chemotherapy regimen consisting of HCPT etc is beneficial for metastatic breast cancer.
8.COMPARING CHARACTERISTICS OF ETHANOL PRODUCTION BY RESPIRATORY DEFICIENT MUTANTS AND WILD STRAIN IN SACCHROMYCES CEREVISIAE

In order to compare the characteristics of ethano l production, a serie s respiratory deficient mutants (rho -) of Saccharomyces cerevisiae and the ir wil d parent strain (rho +) were assayed for ethanol yield, ethanol fermentation ki netics, ethanol tolerance, and activities of ethanol-dehydrogenase. The data i nd icated that: Though the average amount of ethanol produced by mutants rho - a nd by wild rho + strains were similar, some mutants produced a little more amount of ethanol than wild type. The kinetics curve of ethanol fermentation suggested t h at rho - mutants had a little higher ethanol fermenting speeds than wild type . Th e activity of ethanol-dehydrogenase per unit weight of protein of rho - mutan ts c ells were higher than that of wild strain rho +; the activity of ethanol-dehyd rogenase per unit volume of fermentation medium of rho + were higher than that rho - mutants,mainly due to the higher biomass of rho + were higher than that of rho - mutan ts. Some rho - mutants could tolerate higher ethanol concentration than wild rh o + stra in, but some not. If higher ethanol tolerance and higher ethanol yield rho - mu tants were used in fermenting process,higher ethanol yield would be expected,but some problems such as that rho - mutants grew slowly should be considered.

Switching Mechanism At 5′ end of the RNA Transcript(SMART)is a technology used in biology researching,So far,there is no review only about SMART technology.So,the aim is to investigate the research developments of principles,methods and applications of the SMART technology.Based on some researches and combined with the review of the related literature at home and abroad,it analyzed and evaluated the latest development of the research on SMART.As the applications of the SMART technology expand in many fields day after day,it has been proved that the SMART technology is a very useful and efficient skill to construct full length cDNA library.As more and more researchers know this technology,the advantages of the SMART technology become obvious,and meanwhile the disadvantages of the SMART technology also show up.That is to say,the technology needed to be improved.
10.Curative effect of epluchage combined with vacuum sealing drainage and topical oxygen therapy on chronic bone exposed wounds

Objective To explore the therapeutic effect of epluchage combined with vacuum sealing drainage and topical oxygen therapy in the treatment of chronic bone exposed wounds.Methods Thirty-two patients with chronic bone exposed wounds were enrolled in this study from December 2014 to January 2017.All these patients were treated by epluchage combined with vacuum sealing drainage and topical oxygen therapy.Observed the growth and healing time,bacteriology changes,and related preoperative imaging studies of the wounds.Results The X-ray examination showed that there were 14 cases of decrease of the local bone density,disappearance of normal bone structure,and thickening of the outer membrane;10 patients with local defects of bone cortex and cancellous;4 cases of bone density increase;and 4 cases of periosteal reaction with different degree and unclear bone tissue damage.After epluchage combined with vacuum sealing drainage and topical oxygen therapy,granulation tissue of all the 32 cases grew rapidly,and the bone exposure areas were quickly covered.Wound secretions and exudate were significantly reduced,and the wounds were crued after several dressing change,without any postoperative infection.The healing time was 16 to 30 days,with the average time of (23.6±3.6)days.After being followed-up for 6 to 12 months,it was showed that the wound healed well without ulceration and recurrence.Secretions examination showed that there were 20 cases of secretions bacterial cultured to negative,while there were 6 cases of staphylococcus aureus and 6 cases of staphylococcus epidermidis.Conclusion On the basis of definite lesion range,remove the lesions dead bone through epluchage and cure the chronic bone exposed wounds with postoperative vacuum sealing drainage and topical oxygen therapy can significantly shorten the wound healing time and avoid recurrence.
