AIM: To investigate the protective effects and its mechanism of Jinkui Gastric Drug(JK). METHODS: The model of gastric mucosal lesion in rats induced by hydrochloride acid-aspirin was used. The indexes of gastric mucosal lesion in JK and control groups were observed. The contents of nitric oxide(NO) and the activity of nitric oxide synthase(NOS) in blood serum and gastric tissue in rats, the levels of 6-keto-PGF_(1?),thromboxane B_2(TXB_2) in blood plasma and epidermal growth factor(EGF) in blood serum were examined. RESULTS: The index of gastric mucosal lesion after administration of JK reduced obviously. The contents of NO and the activity of NOS in serum and gastric tissue increased markedly. The levels of 6-keto-PGF_(1?),TXB_2 and EGF increased significantly. CONCLUSION: JK can protect experimental gastric mucosal lesion and its mechanism may be related to increasing the protective factors.

Objective To compare detection of STAT3 and CEA by nest RT-PCR and peritoneal lavage cytology examination(PLC) in predicting peritoneal micrometastasis in gastric carcinoma patients. Methods Peritoneal washings of 66 patients with gastric carcinoma and 12 patients with benign gastric diseases were collected to detect the mRNA level of STAT3 and CEA by nest RT-PCR.PLC were carried out by H-E staining. Results The positive rates of STAT3 mRNA and CEA mRNA in peritoneal washings were 43.9% and 48.5%,respectively.The positive rates of STAT3 mRNA and CEA mRNA were significantly higher than those of PLC(31.8%).The positive rates of STAT3 mRNA and CEA mRNA were concurrently increased with the depth of tumor invasion,TNM staging and pathogenesis. Conclusion Detecting STAT3 mRNA in peritoneal washings by nest RT-PCR can improve the sensitivity of free tumor cell detection in peritoneal washings, and may be used to predict peritoneal micrometastasis of gastric carcinoma.

Objective To establish a rabbit VX2 liver tumor model by transplanting tumor tissue mass into the rabbit's liver,to analyze and observe the growing features of the liver tumor. Methods The tumor tissue mass(about 106-108 VX2 liver tumor cells)was inoculated into the left hepatic labe in 20 rabbits to establish rabbit VX-2 hepatic carcinoma model. The observation included the following two respects.(1)The tumor's volume at 7,10,14,17 and 21 days after the procedure was measured by ultrasonography and the growth rate of tumor was calculated.(2)The morphological feature of the tumor was inspected both macroscopically and microscopically. Results The growing pattern of the tumor was compatible with the exponential curve.Seventeen days after transplantation the increase rate of the tumor volume was much higher than that of the tumor diameter. Histopathologjcally, the growing pattern of the tumor took the form of infiltrative way, with its appearance being quite similar to the VX2 squamous cell carcinoma. Conclusion Transplantation of tumor tissue mass is the technique of first choice to establish the VX2 liver carcinoma model in rabbits. This experimental model is a very ideal animal form for both clinical and fundamental studies of liver carcinoma.

Objective To investigate the detection of IMP andVIM metallo-β-lactamases (MβLs)genes in clinically iso-lated gram-negative bacteria as well as bacterial resistance toβ-lactam antimicrobial agents. Methods 113 clinically isolated bacteria were performed antimicrobial susceptibility testing by Kirby-Bauer method ,drug-resistant genes IMP and VIM were detected by polymerase chain reaction (PCR),PCR products were sequenced and aligned with BLAST software. Results VIM gene was detected in 1 Pseudomonas fluorescens strain ,IMP gene was detected in 15 strains ,they were Klebsiella pneumoniae (n=6),Acinetobacter baumannii (n=3),Escherichia coli (n=2),Ralstonia picket-tii (n=1),Pseudomonas aeruginosa (n=1 ),Citrobacter amalonaticua (n=1 ),and Enterobacter cloacae (n=1 ). BLAST results showed that VIM gene was VIM-2 subtype, similarity with gene bank was 99%;all IMP genes were IMP-1 subtype, which were highly homologous ,similarity was 98%-99%.Resistant rates of IMP positive strains to ceftriaxone, cefotaxime, cefoxitin, aztreonam and imipenem were all significantly higher than negative strains (all P <0.05).Conclusion IMP genes of different strains are highly homologous, all are IMP-1 type,indi-cating that IMP genes are highly transmissible and can spread among different species of bacteria.IMP genes are related with resistance ofβ-lactam antimicrobial agents.
