OBJECTIVEA novel bHLH-like gene, designated SmbHLH1, was isolated from Salvia miltiorrhiza, in order to identify a bHLH gene in related to danshinone biosysnthesis.
METHODSmbHLH1 was isolated by RT-PCR,and Semi-quantitative RT-PCR was used to detect the gene expression level.
RESULTThe full length of SmbHLH1 cDNA has an open reading frame of 999 bp. The deduced amino acid sequence of SmbHLH1 has 332 amino acid residues which forms a 36 kDa polypeptide with a calculated pI of 5.4. SmbHLH1 gene was expressed at high level in root, but low level in stem, leaf and flower of S. miltiorrhiza. The transcripts of SmbHLH1 was suppressed when the plants were treated with exogenous MeJA, Yeast + Ag+. The transcripts of SmbHLH1 constitutively accumulated in response to exogenous ABA and low concentration of salicylic acid.
CONCLUSIONSmbHLH is a new member of the S. miltiorrhiza bHLH family, and its possible roles in brassinosteriods signaling responses.

This study was aimed to construct a prokaryotic expressing vector of Hap gene from Nontypeable Haemophilus influenzae, and express and identify the fusion proteins of Hap-His in E. coli. The gene encoding protein Hap was amplified from Nontypeable Haemophilus influenzae ATCC49247 chromosomal DNA by PCR, then it was cloned into prokaryotic expression plasmid pET-32a (+). The recombinant plasmid pET-32a(+)-Hap was transformed into E. coli BL21 and expression was induced by Isopropy-beta-D-thiogalatoside(IPTG). The Hap-His fusion protein expressed so was analyzed by SDS-PAGE and Western-blot. The results showed that the recombinant expressive plasmid pET-32a (+)-Hap was constructed successfully, and the recombinant plasmid expressed Hap-His fusion protein with relative molecule mass 176 000 and mainly existed in inclusion body. This fusion protein could combine with anti-His monoclonal antibody specifically through Western blot analysis. Successful expression of Hap-His fusion protein in prokaryotic cell could lay a basis for further study of immunocompetence of Hap protein and development of NTHi vaccine.

China Modern Doctor 2014;(35):56-59
Objective To analyze solitary fibrous tumor (solitary fibrous tumor, SFT) ’s clinical manifestations, patholog-ical features, immunohistochemical phenotype, and follow-up, discuss the diagnosis, differential diagnosis and progno-sis. Methods Ten cases of SFT were analyzed by clinical data and follow-pathological characteristics study and im-munohistochemical staining of Vimentin, CD34, Bcl-2, CD99, SMA and β-catenin. Results There were 6 males and 4 females with ranging in age from 31 to 78 years (mean 53.2 years old). SFT in four cases originated from the visceral pleura, two cases from parietal pleura, one from the skin of the chest wall, one from the neck, one from the medi-astinum, one from subcutaneously. Microscopic examination of tumor tissue was mainly composed of spindle cells, dif-ferent levels of visible cells and cell-rich areas of sparse areas, tumor cells were short spindle or oval, mitotic due to the severity varies. During the visible rich collagen fibers, may be associated with hyaline degeneration or myxoid change. Immunohistochemistry revealed the tumor cells expessed Vimentin, CD34, Bcl-2, CD99 and β-catenin, SMA focal or weakly positive. Conclusion SFT may occur in various sites of the body. The clinicopathological features and immunohistochemical profile are helpful to identify SFT. Its biological behavior is consistent with its histological mor-phology and growth the way. If tumor could be removed completely, the patients may have a good prognosis;some have low-grade malignant or malignant behaviour, manifesting as local recurrence or metastasis. Patients need long-term follow-up.

A real time PCR assay for the detection of Vibrio parahaemolyticus in seafood samples was developed using a novel specific target and a competitive internal amplification control (IAC). The specificity of this assay was evaluated using 390 bacterial strains including V. parahaemolyticus, and other strains belonging to Vibrio and non-Vibrio species. The real time PCR assay unambiguously distinguished V. parahaemolyticus with a detection sensitivity of 4.8 fg per PCR with purified genomic DNA or 1 CFU per reaction by counting V. parahaemolyticus colonies. The assays of avoiding interference demonstrated that, even in the presence of 2.1 μg genomic DNA or 10(7) CFU background bacteria, V. parahaemolyticus could still be accurately detected. In addition, the IAC was used to indicate false-negative results, and lower than 94 copies of IAC per reaction had no influence on the detection limit. Ninety-six seafood samples were tested, of which 58 (60.4%) were positive, including 3 false negative results. Consequently, the real time PCR assay is effective for the rapid detection of V. parahaemotyticus contaminants in seafood.
