2.Expression of microRNA-100 and its relation with prognosis of colorectal cancer

Objective The aim of this study was to investigate the expression of microRNA?100 ( miR?100) and its relation with prognosis in colorectal cancer ( CRC) . Methods The expression of miR?100 was analyzed by quantitative real?time PCR ( qRT?PCR) in 172 CRC tissue samples. The relation of miR?100 expression patterns with clinical pathological significance in CRC was analyzed. The effects of alterations of miR?100 expression and its consequences on CRC cell proliferation, apoptosis and migration were demonstrated in cells cultured in vitro. Results The relative expression of miR?100 in CRC tissues and peritumoral tissues were -6. 185 ± 1. 921 and -3. 698 ± 1. 786, respectively, with a significant difference between the two groups( P<0.01) . There was a significant difference between the relative expression of miR?100 in CRC with lymph node metastasis (-5.706±1.809) and without lymph node metastasis (-6.775± 1.902, P<0.01). The relative expression of miR?100 in tumors of different TNM stages were -7.267±1.888 in stage Ⅰ, -6.443±1.859 in stageⅡ,-5.923±1.796 in stageⅢ, and-4.639±1.516 in stageⅣ, with a significant difference among them(P<0.01). Different differentiation grades showed different expression of miR?100, i.e. -7. 389 ± 1. 828 in well differentiated tumors, -6. 095 ± 1. 843 in moderately differentiated tumors, and -5.476±2.088 in poorly differentiated tumors (P<0.01). There was no significant correlation between miR?100 expression and overall survival rates of the CRC patients (P=0.179). Overexpression of miR?100 in the CRC cell line HCT?8 inhibited cell proliferation, but promoted cell apoptosis and migration. Conclusions The expression of miR?100 is correlated with lymph node metastasis, TNM stage and differentiation grade, and may be a potential biomarker indicating the development of CRC.

Objective: To propose reasonable suggestions to promote the standardization of clinical studies by reviewing the systematic reviews and meta-analyses of acupuncture-moxibustion treatment of essential hypertension (EH). Methods: Computer retrieval was conducted through Excerpta Medica Database (EMBASE), PubMed, China National Knowledge Infrastructure (CNKI), Chongqing VIP Database (CQVIP), China Biology Medicine Disc (CBM), and Wanfang Academic Journal Full-text Database (Wanfang) to collect systematic reviews and meta-analyses relevant to treating EH with acupuncture-moxibustion therapy. The time range was from the database's inception till July, 2020. The studies were screened based on the inclusion and exclusion criteria and then data-extracted. The study's quality and evidence ratings were performed by referring to the preferred reporting items for systematic review and meta-analysis (PRISMA), a measurement tool to assess systematic reviews 2 (AMSTAR 2), and the grading of recommendations, assessment, development, and evaluation (GRADE). Results: A total of 14 studies, 10 in Chinese and 4 in English, published between 2012 and 2019, were included, involving 70 outcome measures. The methodological quality was rated as critically low, the reporting was relatively complete or had certain flaws, and the evidence strength was rated as low or very low. Conclusion: Regarding the acupuncture-moxibustion treatment of EH, the methodological quality and outcome measure evidence of existing systematic reviews and meta-analyses are relatively low, and the reporting quality also expects further improvements.

OBJECTIVEThe aim of this study was to identify six miRNAs expressed in plasma of patients with pancreatic cancer (PCa) and analyze their value as a diagnostic index of pancreatic cancer.
METHODSPlasma total RNAs were extracted from 30 PCa patients and 26 normal controls, and the abundance of six microRNAs was measured using real-time PCR. The possibility to combine them with CA19-9 as diagnostic biomarkers was analyzed.
RESULTSThe expression level of miR-21, miR-210, miR-155, miR-20a, miR-25 and miR-196a in plasma of patients with pancreatic cancer were 1.65×10(6), 5.98×10(4), 2.83×10(3), 3.47×10(6), 2.76×10(6), and 1.03×10(3) (copies/µl), while the normal controls were 4.08×10(5), 2.54×10(4), 8.55×10(2), 1.79×10(6), 9.32×10(5), and 4.67×10(2) (copies/µl), respectively, with a significant difference between the two groups (P < 0.05). The areas under the ROC curve of miR-21, miR-210, miR-155, miR-20a, miR-25 and miR-196a were 0.893, 0.810, 0.820, 0.766, 0.816 and 0.729, respectively. MiR-21 had the highest diagnostic value when it was used as diagnostic marker alone. The combination of miR-155 and miR-25 was more effective to distinguish PCa from normal than to be used alone, and the area under the ROC curve was 0.913 (95%CI 0.838-0.988) .When CA199 associated with miR -210 and miR-25, respectively, the areas under the ROC curves were 0.96 (95%CI was 0-1.0) and 0.942 (95% CI was 0.876-1.0), which were higher than CA199 alone (0.862, 95%CI was 0.748-0.975). There was a high improvement in diagnostic sensitivity and accuracy when miR-210 and miR-25 were combined with CA19-9, respectively.
CONCLUSIONSPlasma miR-21, miR-155, miR-25, miR-210 have diagnostic value for pancreatic cancer, and deserve further study.

9.Effects of mindfulness-based parenting course on parenting burnout and parent-child relationship of adolescents' mothers

Objective To investigate the effects of mindfulness-based parenting course on parenting burnout and parent-child relationship of adolescents' mothers.Methods A total of 203 mothers who volun-teered to participate in the mindfulness-based parenting course in a middle school in Chongqing City in May 2023 were recruited as the research subjects and divided into the intervention group (n=99) and control group (n=104).The intervention group attended the eight mindfulness parenting sessions twice a week,while the control group did not attended the course.The Parenting Burnout Assessment Scale,Parent-Child Relationship Self Evaluation Scale,Five Factors of Mindfulness scale and Satisfaction with Life Scale were used to evaluate the two groups before and after intervention.Results The scores of parent-child relationship and satisfaction with life had statistical difference between the two groups (P<0.05).Compared with before intervention,the parental burnout score of the intervention group was decreased,the scores of parent-child relationship,five fac-tors of mindfulness,scores of awareness,no judgment,no reaction dimensions and life satisfaction were in-creased,the differences were statistically significant (P<0.05).Compared with the control group,the scores of parent-child relationship,five factors of mindfulness,awareness dimension and life satisfaction after inter-vention in the intervention group were higher,while the score of conscious action dimension was lower,the differences were statistically significant (P<0.05).Conclusion The 4-week mindfulness-based parenting course could could decrease the parenting burnout,and improve the parent-child relationship and satisfaction with life.
10.Regulatory and functional mechanism of angiopoietin-like protein-4 in gastric epithelial cells with Helicobacter pylori infection

Objective To explore the regulatory and functional mechanism of angiopoietin-like 4(ANGPTL4)expression in gastric epithelial cells infected with Helicobacter pylori(H.pylori).Methods H.pylori-positive gastric specimens from Department of Gastroenterology of second Affiliated Hospital of Army Medical University in November 2021 and the gastric tissues of H.pylori-infected mice were collected,and the expression locations of ANGPTL4 in these gastric tissues were detected by immunofluorescence assay.After gastric epithelial cells and gastric organoids were infected with H.pylori,the expression of ANGPTL4 was detected by real-time PCR and Western blotting.The regulatory mechanism of H.pylori-induced ANGPTL4 expression was investigated by cellular models above.Recombinant ANGPTL4 was used to stimulate gastric epithelial cells to investigate the effect of ANGPTL4 on the claudin 1(CLDN1)expression.Results H.pylori infection significantly increased the expression of ANGPTL4 in gastric epithelial cells in a CagA-dependent,bacterial concentration-dependent and infection time-dependent manner(P<0.05).Compared to uninfected cells and CagA knockout(ΔcagA)H.pylori-induced ones,the ANGPTL4 expression was significantly increased in WT H.pylori-infected human and mouse gastric organoid(P<0.05).Blocking NF-κB signaling pathway significantly inhibited the enhanced ANGPTL4 expression in H.pylori-infected gastric epithelial cells(P<0.05).Recombinant ANGPTL4 activated ERK signaling pathway to down-regulate the CLDN1 expression in gastric epithelial cells(P<0.05).Conclusion H.pylori activates NF-κB signaling pathway to up-regulate ANGPTL4 expression in gastric epithelial cells in a CagA-dependent manner,and ANGPTL4 activates ERK signaling pathway to down-regulate the CLDN1 expression in gastric epithelial cells,which may contributes to the development of gastric diseases induced by H.pylori infection.
