1.Common triggers of ischemic stroke

Ischemic stroke is often triggered under the effect of the basic lesions combined with a variety of incentives in daily life. Its generative mechanism is complicated, and there are more risk factors. Only according to the long-term existence of chronic risk factors, it cannot make appropriate explanation for the acute onset of this disease. At present, there are a few studies on the predisposing factors of stroke, and its related mechanism needs to be further discussed. This article aims to introduce the relationship between the common predisposing factors and ischemic stroke, discusses its possible mechanism, and provides reference for the clinical prevention work of ischemic stroke.
2.Common triggers of ischemic stroke: a case cross-over study

Objective To investigate the correlation between 6 common triggers and ischemic stroke.Methods The demographic and clinical data of the consecutive inpatients with acute ischemic stroke were collected.A questionnaire survey of the triggers of the disease was conducted within 4 d after admission.A case cross-over study was used to compare the exposure of the 6 potential triggers (overeating,anger,negative emotion,heavy physical activity,sudden posture changes as response to a startling event,drinking coffee) at 2 h before onset (dangerous period) and at 1 d before onset during the same period (control period),and exposures to potential triggers in patients according to gender,age and etiological subtypes were further analyzed.Results A total of 369 patients were enrolled.They aged 24-93 years old (mean 61.75 ±13.57),220 patients were male (59.6%) and 149 were female (40.4%).A total of 91 patients (24.7%) exposed to at least one of the triggers at 2 h before onset (odds ratio [OR] 6.1,95% confidence interval [CI] 3.7-9.9);OR for exposure to the sudden posture change in response to a startling event was 12.0 (95% CI 2.4-59.3),for heavy physical activity 10.7 (95% CI 4.2-27.6),for anger 8.0 (95% CI 2.3-27.5),and for negative emotion 4.9 (95% CI 2.3-10.3).There was no exposure to drinking coffee.There were no significant differences in the exposure to various triggers among the different gender,age,and etiological subtypes.Conclusions Sudden posture changes as response to a startling event,heavy physical activity,anger,and negative emotion are the triggers for ischemic stroke,attention should be paid to the influence of triggers in the prevention of ischemic stroke.
3.Stable expression of targeting complement inhibitor CR2-CD59 in Chinese hamster ovary cells

Objective To obtain Chinese hamster ovary (CHO) cell lines that stably express a targeting complement inhibitor CR2-CD59.Methods The recombinant plasmid PEE14.1-CR2-CD59 was constru-cted by cloning the DNA fragment CR2-CD59 into plasmid PEE14.1,and the obtained plasmid was transfected into CHO cells by FuGENE 6.The clones with stable high expression of target fragment were selected by methionine sulfoximine (MSX),the expression of CR2-CD59 was analyzed by ELISA,SDS-PAGE and Western blotting analysis.Results Several stable expression clones were obtained,and CR2-CD59 was highly expressed in the secret form in CHO cells.SDS-PAGE analysis showed that the molecular weight of the recombined protein CR2-CD59 was consistent with the predicted one.ELISA and Western blotting results revealed that the CR2-CD59 could react with both anti-human CR2 and anti-human CD59 polyclonal antibodies.Compared with serum-containing medium,the protein was highly expressed in serum-free medium (P

After its infarction, it is easy to be misdiagnosed or missed because of its various clinical manifestations and lack of specificity. This article reviews the pathogenesis, clinical manifestations, and prognosis of medullary infarction in different sites.
5.Investigation on degeneration of outer hair cells in guinea pig.

OBJECTIVE:
To investigate the degeneration mechanics of outer hair cells in guinea pig.
METHOD:
The mechanics of outer hair cells isolated by enzyme were observed under inverted microscope for 6-8 h continuously.
RESULT:
Over half of living outer hair cells could keep good conditions in 6 hours. During the degeneration there was always a longitudinal fold line from tip to base. Presence or absence of calcium, as well as lossing of stereociliary bundle, couldn't change the conditions of out hair cells.
CONCLUSION
Neither calcium nor stereociliary bundle is the decisive cause in keeping outer hair cells alive, and its degeneration may be basically related with something surrounding the cell.

6.A simple method of separating outer hair cells in Guinea pig.

OBJECTIVE:
In order to meet the needs of electrophysiology outer hair cells, we investigate a method to separate outer hair cells simply and effectively.
METHOD:
The basal membrane was dissected combined with spiral ligament and axis of cochlea, then incubated with enzyme, and then mechanically triturated by micropipette.
RESULT:
A large number of living outer hair cells were obtained. Of 80% cells could keep good condition in 6 hours.
CONCLUSION
If increasing the enzyme concentration slightly and the large part of cochlea without outer bone was enzymes, we can get living outer hair cells easily.

7.Ginsenoside Rg1 inhibits H2O2-induced apoptosis and autophagy in human periodontal ligament cells

BACKGROUND:Our previous studies have confirmed that H2O2 and ginsenoside Rg1 can cause changes in reactive oxygen species levels in human periodontal ligament cells,but the correlation of reactive oxygen species with apoptosis and autophagy remains unclear. OBJECTIVE:To explore the effect and mechanism of ginsenoside Rg1 on H2O2-induced apoptosis and autophagy of human periodontal ligament cells. METHODS:Human periodontal ligament cells were divided into control group,H2O2 group and H2O2+Rg1 group.Ginsenoside Rg1(50 μmol/L)was pre-incubated for 24 hours and H2O2 was treated for 2 hours(500 μmol/L).CCK-8 assay was used to detect the proliferation ability of the cells.A fluorescent probe DCFH-DA was used to detect the reactive oxygen species level of the cells.qRT-PCR and western blot assay were used to detect heme oxygenase 1,apoptosis-related factor Caspase-3,Bax,anti-apoptotic factor Bcl-2,autophagy Beclin-1,P62,LC3,pathway-related factors phosphatidylinositol-3-kinase(PI3K),protein kinase B(AKT),mammalian target of rapamycin(mTOR)mRNA and protein expression. RESULTS AND CONCLUSION:(1)Compared with the control group,the proliferation activity of human periodontal ligament cells in the H2O2 group decreased.Compared with the H2O2 group,the proliferation activity of human periodontal ligament cells increased in the H2O2+Rg1 group.(2)Compared with the control group,the expression of reactive oxygen species and heme oxygenase-1 increased in the H2O2 group.Compared with the H2O2 group,the expression of reactive oxygen species and heme oxygenase-1 decreased in the H2O2+Rg1 group.(3)Compared with the control group,the expression of Caspase-3,Bax,Beclin-1 and LC3 increased,while the expression of Bcl-2 and P62 decreased in human periodontal ligament cells of the H2O2 group.Compared with the H2O2 group,the expressions of Caspase-3,Bax,Beclin-1 and LC3 decreased,and the expressions of Bcl-2 and P62 increased in the H2O2+Rg1 group.(4)Compared with the control group,the expressions of PI3K,AKT and mTOR decreased in human periodontal ligament cells of the H2O2 group.Compared with the H2O2 group,the expressions of PI3K,AKT and mTOR increased in human periodontal ligament cells of the H2O2+Rg1 group.(5)These results suggest that ginsenoside Rg1 can inhibit H2O2-induced apoptosis and autophagy in human periodontal ligament cells by reducing the content of reactive oxygen species and down-regulating the related factor expression of the PI3K/AKT/mTOR pathway.
8.Prevention and Treatment of Type 2 Diabetes Mellitus with Traditional Chinese Medicine Based on Inflammatory Signaling Pathways: A Review

Type 2 diabetes mellitus (T2DM),as a chronic disease with an increasing prevalence,has greatly affected the quality of life of patients, and aroused widespread concern in society. Previous studies have shown that inflammation is closely related to the occurrence and development of T2DM. The role of inflammatory response and its signaling pathway in the pathogenesis of T2DM has become a research hotspot,and the intervention of inflammatory response to control the progression of T2DM has become a new approach for treatment. With the deepening in the research field of traditional Chinese medicine (TCM),the generation of inflammation is attributed to turbid phlegm and blood stasis, whose mutual transformation results in the impairment of body functions. However,the monomers and compounds of Chinese medicine in the treatment of T2DM based on inflammatory signaling pathways are advantageous in multiple targets,multiple pathways, and few side effects,which can significantly relieve the symptoms of T2DM. In recent years,there has been an increasing number of studies on TCM intervention in T2DM. The present article elaborated the understanding of the relationship between T2DM and inflammation in TCM and the seven signaling pathways related to the regulation of inflammation in T2DM by TCM, including nuclear transcription factor-κB (NF-κB) signaling pathway,mitogen-activated protein kinase (MAPK)-dependent signaling pathway,phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) signaling pathway,protein kinase C (PKC) signaling pathway,adenosine 5′-monophosphate (AMP)-activated kinase (AMPK) signaling pathway,Wnt signaling pathway,and transforming growth factor-β (TGF-β) signaling pathway.
9.Comparison of frailty assessment and related tools in elderly hospitalized patients with atrial fibrillation

Objective:We evaluated frailty in elderly hospitalized patients with atrial fibrillation and analyzed the relevance, consistency, and diagnostic power of different frailty tools.Methods:From September 2018 to April 2019, a total of 197 elderly patients with atrial fibrillation aged ≥ 65 years in Beijing Hospital, Chinese PLA General Hospital, and Beijing Tsinghua Changgung Hospital were prospectively enrolled.Five frailty tools, including the clinical frailty scale(CFS), FRAIL scale(FRAIL), Fried frailty phenotype(Fried), Edmonton frail scale(EFS), and comprehensive geriatric assessment-frailty index(CGA-FI), were used for frailty assessment.Results:A total of 197 hospitalized elderly patients with atrial fibrillation were enrolled, with an average age of(77.5±7.1)years old(57.4% male). The prevalence of frailty, according to the five frailty tools, were 25.4%(FRAIL), 27.9%(EFS), 34.5%(Fried), 40.6%(CFS), and 42.6%(CGA-FI), respectively.CFS had a good correlation(correlation coefficient 0.80)and and consistency(Kappa value 0.71, 95% CI 0.61~0.81)with CGA-FI.The combined frailty index was used as the gold standard for frailty diagnosis.The results showed that CFS and CGA-FI had high diagnostic sensitivity(95.9 % and 98.0 %, respectively)and specificity(77.7 % and 75.7 %, respectively). Conclusions:Frailty is common in elderly hospitalized patients with atrial fibrillation, showing multidimensional features, and physical weakness is not prominet.CFS and CGA-FI are recommended for the assessment of frailty in patients with atrial fibrillation, which had good correlation and consistency.
