Objective:To elucidate the relationship between HLA-DQB1 allele polymorphism as well as the expression level of Th1/Th2 cytokines with familial clustering of hepatocellular carcinoma ( HCC) to provide some evidence for the seeking susceptibility gene or resistant gene of HCC in Guangxi yao,China. Methods:With the same sexuality, age ±5 year,40 members whose families have had two or more HCC patients( high-occurrence families) were selected as the case group, and 40 members whose families have no any cancer patient were selected as the controls.Peripheral blood samples were collected to extract DNA,PCR-SSP was used to detect HLA-DQB1 alleles and ELISA was used to detect IL-2,IL-4 and IL-10.Results:(1) The gene frequency of the HLA-DQB1*02/09 alleles in the case group was higher than that in the controls(P<0.05);but the gene frequency of the HLA-DQB1*04/05/06/07/08 alleles were never significant difference between two groups ( P>0.05 ).( 2 ) The gene frequency of alleles HLA-DQB1 in HBsAg positive group and HBsAg negative group were never significant difference (P>0.05).(3)The expression levels of IL-4,IL-10 in the case group was higher than that in the control ( P<0.05 ).( 4 ) The expression level of IL-10 in the positive group of the HLA-DQB1*02 allele was higher than that in the negative group of the HLA-DQB1*02 allele ( P<0.05 ).( 5 ) The expression level of IL-4 in the positive group of the HLA-DQB1*09 allele was higher than that in the negative group of the HLA-DQB1*09 allele( P<0.05) .Con-clusion:(1) HLA-DQB1*02/09 seem to be susceptibility genes of hepatocellular carcinoma in high HCC incidence areas of Guangxi yao.(2) There may be not significant correlation bewteen HLA-DQB1 alleles and the susceptibility of HBV infection in high HCC incidence areas of Guangxi yao.( 3 ) The imbalance of IL-4, IL-10 might be associated with familial clustering of hepatocellular carcinoma in Guangxi yao.(4)The imbalance of IL-10 might be due to the carrying of HLA-DQB1*02;the imbalance of IL-4 might be due to the carrying of HLA-DQB1*09.Through the same approaches, these might lead to the phenomenon of familial aggregation of HCC in Guangxiyao.

Objective To enhance the understanding of the quality control of rubber closures and to promote pharmaceutical manufacturers'reasonable selection and effective management of rubber closures. Methods According to the performance needs of drug products for rubber closures, the impacts of the ingredients and manufacturing processes of rubber closures with other possible influence on drug products were analyzed.Complied with the requirements of pharmaceutical lifecycle management, the processes of selection and utilization of rubber closures were divided into three periods, including screening, validation and maintenance, and suggestions on the tasks and goals for these three periods were given. Results Though the actual performance of rubber closures was affected by multiple factors, the processability in the pharmaceutical manufacturing processes and the suitability in the periods of storage, transportation and clinical application could be confirmed by reasonable screening and scientific validation of rubber closures. And by the effective quality control of rubber closures, the suitability could be maintained across the lifecycle of drug products. Conclusion By means of the reasonable selection and effective quality control of rubber closures,it would be benefit for keeping drug safety, efficacy and quality controllability throughout the lifecycle.

Objective To establish methods for detection of water in rubber closures as one of the Chinese Pharmacopoeia series of standards for pharmaceutical packaging material. Methods The formulation process of determination of water in rubber closures was explained, the key contents of the standard were analyzed, and the determination results of water content by oven-drying method and Karl Fischer drying furnace-coulometric titration method were compared. Results The determination results of water content in multiple batches of rubber stopper samples by over-drying method were generally higher than that by Karl Fischer drying oven-Coulometric titration method. The latter method is highly specific and accurate, and can better reflect the true water content of rubber closures. Conclusion Two methods for determining water content in rubber closures have been developed.Enterprises can choose the appropriate method based on the intended use of their products and daily testing needs.

Objective? To explore the effects of ideation and imitation behavior training on rehabilitation in patients with chronic schizophrenia. Methods?Totally 200 schizophrenia patients admitted and treated in 2016 from Fuzhou Neuro-Psychiatric Hospital were selected and equally randomized to treatment group (n=100) and control group (n=100) according to the random number table. Patients in the control group received conventional nursing, while patients in the treatment group received ideation and imitation behavior training for 8 weeks on this basis. Wisconsin Card Sorting Test (WCST), Nurses' Observation Scale for Inpatient Evaluation (NOISE) and Personal and Social Performance Scale (PSP) were used to compare the rehabilitation effects between the two groups. Results? After intervention for 8 weeks, the score for correct response in WCSI in patients in the treatment group was (24.78±7.39), higher than that of patients in the control group, and the score for continuous mistakes in WCSI in patients in the treatment group was (14.78±5.27), lower than that of patients in the control group (t=3.800, 3.640;P<0.01). After intervention for 8 weeks, the scores for NOISE (200.06±26.09) and PSP (79.83±4.21) in patients in the treatment group were higher than those of patients in the control group (t=6.880, 19.040;P<0.01). Conclusions?Ideation and imitation behavior training helps to improve the patients' cognitive performance, including attention, memory, linguistic competence and reasoning ability, as well as self-care ability of daily living and social function, and promotes chronic schizophrenia patients' rehabilitation.

Background and Objectives:
Although human-induced pluripotent stem cells (hiPSC) can be efficiently differentiated into cardiomyocytes (CMs), the heterogeneity of the hiPSC-CMs hampers their applications in research and regenerative medicine. Retinoic acid (RA)-mediated signaling pathway has been proved indispensable in cardiac development and differentiation of hiPSC toward atrial CMs. This study was aimed to test whether RA signaling pathway can be manipulated to direct the differentiation into sinoatrial node (SAN) CMs.
Methods:
and Results: Using the well-characterized GiWi protocol that cardiomyocytes are generated from hiPSC via temporal modulation of Wnt signaling pathway by small molecules, RA signaling pathway was manipulated during the differentiation of hiPSC-CMs on day 5 post-differentiation, a crucial time point equivalent to the transition from cardiac mesoderm to cardiac progenitor cells in cardiac development. The resultant CMs were characterized at mRNA, protein and electrophysiology levels by a combination of qPCR, immunofluorescence, flow cytometry, and whole-cell patch clamp. The results showed that activation of the RA signaling pathway biased the differentiation of atrial CMs, whereas inhibition of the signaling pathway biased the differentiation of sinoatrial node-like cells (SANLCs).
Conclusions
Our study not only provides a novel and simple strategy to enrich SANLCs but also improves our under-standing of the importance of RA signaling in the differentiation of hiPSC-CMs.
