1.The expression of PIN 1 and Cyclin E in gastric carcinoma tissue after radical gastrectomy and its significance

Objective To explore the expression of PIN1 and Cyclin E in gastric carcinoma tissue after radical gastrectomy and its significance.Methods 46 patients received radical gastrectomy were selected as study subjects.Cancer tissues were harvested as the gastric cancer group,and the adjacent normal tissues of tumor tissue were harvested as the control group.The expression of PIN1 and Cyclin E was observed by immunohistochemistry,and the relationship with tumor infiltration,differentiation,lymphatic and distant metastasis were analyzed.A five year followup study was also applied.Results The expression of PIN1 (3 949.66 ± 157.40) and Cyclin E (5 443.07 ±240.15) was significantly higher than those of the control group[(247.67 ± 75.63),(354.92 ± 93.88)] (all P <0.05),and the expression of PIN1 was related with tumor differentiation,lymphatic and distant metastasis,and the expression of Cyclin E was closely related with tumor infiltration and differentiation.The 5-year survival rate of patients with the simultaneously increased expression of PIN1 and Cyclin E was 50.0%,which was significantly lower than the expression of the two separate elevated or not elevated.Conclusion The expression of PIN1 and Cyclin E in cancer tissues after radical gastrectomy is high,and could better reflect the pathological parameters and prognosis.
2.Analysis of the prognosis of curative resection of spontaneous rupture with hemorrhage of primary hepatic carcinoma

Objective To investigate the prognostic factors of curative operation for spontaneous rupture with hemorrhage of primary hepatic carcinoma (SRHC).Methods The clinical data and followed up results of 41 patients with SRHC treated by curative operation,were retrospectively analyzed.The prognostic factors affecting survival were studied by univariate and multivariate analysis.Results The survival rate of 1-year,3-year,5-year was respectively 46.34%,21.95%,9.76%.Univariate analysis showed that Child-Pugh class,degree of accompanying cirrhosis,tumor size,operation time,blood loss,tumor staging,serum albumin were significant prognosis factors.The use of Coxs multivariate proportional hazard model indicated that significant prognosis factors by curative operation for SRHC were degree of accompanying cirrhosis,Child-Pugh class,blood loss,tumor staging for cumulative survival rate.Conclusion The factors affecting prognosis were degree of accompanying cirrhosis,Child-Pugh class,blood loss,tumor staging.
3.Changes of serum IL-6 levels and pulmonary IL-6 mRNA expression after high-volume hemofiltration in pigs with multiple organ dysfunction syndrome

Objective:To investigate the effect of high-volume hemofiltration(HVHF) on serum levels of IL-6 and pulmonary expression of IL-6 mRNA in pigs with multiple organ dysfunction syndrome(MODS).Methods: Twenty-five pigs were randomly divided into model group,control group and HVHF group.Animals in control group were injected with normal saline,and those in the other 2 groups were treated with "a second hit" to establish MODS model,then animals in HVHF group was treated with HVHF for 3 days.Serum IL-6 levels were determined by ELISA and Real-time quantitative PCR was used to determine the expression level of IL-6mRNA.Lymphocyte apoptosis was detected by Annexin-Ⅴ/PI flow cytometry.Results: After HVHF,the serum indices associated with organ-function and the morbidity and mortality were much lower than those in MODS model group(P

A efficient and convenient method for covalent binding of antibodies to liposomes was de-scribed.Antibodies were attached to the aldehye groups on the gangliosides by the periodate ox-idation to form schiff—base and the complex was stabilized by reduction.Separate the liposome —IgG complex from unbound IgG using discontinous dextran T40 gradient centrifugation.AKP and HRP were coupled to liposomes,and both enzymatic activity was remained more than 80%.and compared this with glutaraledhyed method.

7.Inhibition of corneal neovascularization by vascular endothelia growth inhibitor gene

AIM: To evaluate the effect of EffecteneTM lipofectine mediated plasmids encoding human pcDNA4-vascular endothelia growth inhibitor (pcDNA4-VEGI) gene on corneal neovascularization (CNV).METHODS: Forty New Zealand albino rabbits were sutured by 5- 0 silk on the superior cornea to induce CNV and divided into 4 random teams, ten per each team: team A: transfected by pcDNA4-VEGI gene mediated by EffecteneTM lipofectine transfection; team B: by plasmid pcDNA4; team C: by EffecteneTM, and team D: by normal saline. Length and area of CNV were observed under slit lamp every day after transfection. Immunohistochemistry was performed to detect the expression of VEGI protein in corneas at day 3, 7, 14 and 21. RESULTS: 1) Average occurrence of CNV was 6.3 days in team A, 3.1 days in team B, 3.2 days in team C, and 3.2 days in team D. Difference was significant between A and other teams (P<0.01); 2) Length and average area of CNV in each period in team A was significantly different from those in team B, C and D (P<0.01); 3) VEGI expressions were observed in epithelium, stroma, endothelium and the cliff of CNV in team A at 3 days after transfection by immunohistochemical staining. None VEGI positive cells were found in the control teams (team B, C and D) all the time.CONCLUSION: EffecteneTM lipofectine transfection technique can effectively transfect pcDNA4-VEGI gene into rabbit cornea and the length and CNV areas can be inhibited by VEGI gene.
9.Effects of p90 ribosomal S6 kinase pathway in breast cancer and its mechanism

p9O ribosomal S6 kinases(BSKs)are overexpressed in 50% human breast cancer. RSKs enhance the proliferation of breast cancer cells by regulating several key breast cancer-related proteins. They also improve survival of the breast cancer cells through regulating translocation and translation of mRNA. In addition. RSK promotes tumor angiogenesis by interaction with estrogen receptor. However, RSK4 may play an inhibitory role in breast cancer. In general, except RSK4,RSKs may become promising targets of breast cancer therapy.
10.Platycodin D protects oral epithelial cells against infection of Candida albi-cans

AIM: To investigate the effect of platycodin D on Candida albicans infection in oral epithelial cells.METHODS:The viability of the oral squamous carcinoma KB cells was detected by MTT assay after treated with different concentrations of platycodin D .The KB cells were infected with Candida albicans, and then were incubated with platycodin D at different concentrations .Adherent numbers of the Candida albicans were counted by Gram staining , and the bacterial activity and conversion were measured by Trypan blue staining .Furthermore , the protein levels of IL-18 and hu-man β-defensin 2 ( HBD-2) were analyzed by ELISA , and the expression of HBD-2 at mRNA and protein levels was deter-mined by RT-qPCR and Western blot , respectively .RESULTS:The viability of the KB cells was not affected by platycod-in D at the concentrations used .The adherent numbers , bacterial activity and conversion were decreased by treatment with platycodin D in a dose-dependent manner .In addition, the protein level of IL-18 in the culture supernatant and the mRNA expression of HBD-2 in the KB cells were also reduced after platycodin D treatment .CONCLUSION:Platycodin D has a bacteriostasis effect and prevents oral epithelial cells from Candida albicans infection.
