1.The importance of appling laboratory tests in diagnosing hemophagocytic lymphohistocytosis

It is a challenge in diagnosis of hemophagocytic lymphohistocytosis (HLH).Rapid diagnosis is the key to make decisions in clinical management of HLH.The timely and reliable diagnosis of HLH can be assisted by laboratory tests including nature killer (NK) cell activity, sCD25, hemophagocytosis, ferritin, surface CD107a, and genetic testing.Here is introduced lab assays and evaluate the clinical applications in HLH.The aim is to raise awareness of the values of laboratory evaluations in diagnosing HLH.
2.Effect of recombinant bovine basic fibroblast growth factors combined with new rehabilitative liquid on neonatal diaper dermatitis

Objective To investigate the effects of recombinant bovine basic fibroblast growth factors combined with new rehabilitative liquid on neonatal diaper dermatitis. Methods One hundred and twenty neotates with neonatal diaper dermatitis from August 2013 to July 2014 were randomly divided into the control group and experiment group, 60 cases in each group. The control group was treated with conventional care, while the observation group with recombinant bovine basic fibroblast growth factors combined with new rehabilitative liquid. The total effective rate and the treatment time were compared between the two groups. Result The total effective rate of the observation group was significantly higher than that of the control group (P<0.05) and the treatment time of the observation group was statistically significantly shorter than that of the control group (P<0.05). Conclusion Recombinant bovine basic fibroblast growth factors combined with new rehabilitative liquid is effective in the treatment of neonatal diaper dermatitis and it is worth promoting and using clinically.
3.Quality Evaluation of Acetylspiramycin

Objective:To evaluate the quality condition and problems of acetylspiramycin at present. Methods:According to the standard described in Chinese Pharmacopoeia (2015 edition), acetylspiramycin and its preparations were determined, and the overall quality level of domestic acetylspiramycin was evaluated. The related substances, components and dissolution were analyzed to further explore the quality status. Results:In 258 batches of samples, the dissolution of only one batch did not meet the standard with the passing rate of 99. 6%. The results of exploratory research showed that the content of impurities in raw material was determined by the synthesis process, and the impurity profile of domestic and foreign formula was different. The dissolution behavior of domestic and for-eign formula in four kinds of media was not similar as well. Conclusion: The quality of domestic acetylspiramycin is promising. The research of related substances provides the reference for controlling the target impurities. The consistency assessment of dissolution pro-vides the reference for the formula process improvement and clinical application of acetylspiramycin tablets.
4.A survey on reference ranges for serum pituitary-thyroid hormones of 2-6 years old healthy Chinese children in Hangzhou

Serum concentrations of TSH, total and free thyroxine, and total and free triiodothyronine in 592 healthy children aged 2-6 years were determined by means of ADVIA Centaur. The normal reference ranges were calculated. The reference ranges of TSH, T3, T4, FT3, and FT4 were 1.09-4.60 mIU/L, 1.17-1.95 μg/L,57.43-99.60 μg/L, 5.35-7.50 pmol/L, 13.96-19.70 pmol/L, respectively. There was no significant difference in the interval of thyroid hormones between males and females as well as among different age groups.
6.Influencing factors of seizures in glioma patients and its relationship with recurrent epilepsy

Objective To analyze the influencing factors of epilepsy in patients with glioma,and to explore the relationship between the epileptic factors and the recurrence of epilepsy after surgery.Methods From January 2012 to December 2015,clinical data of 95 glioma patients in the Fifth Affiliated Hospital of Zhengzhou University were retrospectively analyzed,which were divided into two groups,including 54 cases with epilepsy and 41 cases without epilepsy,all patients were underwent tumor resection,to search and resect epilepsy lesions in patients with epilepsy,Follow-up using outpatient examination and telephone interview was performed to record seizure recurrence after operation,sort and analyze the clinical data of patients,to investigate the influencing factors of epileptic seizures,and the relationship between the factors associated with epileptic seizures and postoperative recurrence of epilepsy.The epileptogenic factors were analyzed using x2 test and multifactor Logistic regression model,epileptogenic factors and recurrent epilepsy after operation were analyzed usingx2 test.Results Logistic regression model showed that peritumoral edema > 2 cm(OR =6.905,95% CI:2.787-17.106),low tumor pathological grade (0R =5.032,95% CI:1.696-14.931) were independent risk factors for epileptic seizures;Follow-up situations:16 cases (13 cases of epilepsy group,3 cases of no epilepsy group) showed epileptic seizure in different time after the operation,preoperative epilepsy history,peritumoral edema were related to the recurrence of epileptic seizures in epilepsy group and the patients without epilepsy (x2 =2.940,3.049,all P < 0.05),the pathological grade was related to the recurrence of epileptic seizures in epilepsy group (x2=4.774,P < 0.05),however,it was not associated with the patients without epilepsy (x2 =0.060,P > 0.05),and there was a correlation between the preoperative history of epilepsy and the postoperative recurrent seizures (x2 =4.672,P < 0.05).Conclusions Peritumoral edema > 2 cm and low tumor pathological grade are the factors that lead to epileptic seizures and postoperation epilepsy recurrence in glioma patients.It's expected to take some positive treatment to reduce epileptic seizures,we should pay special attention to glioma patients with epilepsy.
7.Selection and therapeutic effect evaluation of chemotherapy regimens for pancreatic cancer

Pancreatic cancer is a highly malignant tumor and its treatment is still a challenge.Recent studies have shown that medication plays an important role in preoperative and postoperative adjuvant therapy for locally advanced pancreatic cancer and is also a major thera-peutic method for advanced pancreatic cancer.It can improve the survival time and quality of life in patients with pancreatic cancer.Tradi-tional chemotherapy regimens based on gemcitabine and fluorouracil have limited effects in the treatment of advanced pancreatic cancer,and studies on molecular targeted therapies have achieved some progress in recent years.With reference to related guidelines or consensus on the diagnosis and treatment of pancreatic cancer and important clinical trials of the treatment of pancreatic cancer,this article elaborates on the selection and therapeutic effect evaluation of chemotherapy regimens for pancreatic cancer.We believe that with the research and develop-ment of new drugs and the application of new techniques,the treatment of pancreatic cancer will achieve new breakthroughs in future.
8.Effect of glutaraldehyde on biological characterstics of vincristine-loaded intact human erythrocytes

Objective To explore the optimal method and condition for controlling release of vincristine (VCR) from VCR-loaded erythrocytes and enhancing the stability of carrier erythrocytes. Methods Erythrocytes were loaded into human erythrocytes using the modified hypotonic pre-swelling and isotonic resealing technique, and then with VCR, and they were treated with glutaraldehyde in concentration of 0.16% and 0.25% respectively. After storing at 4℃ for indicated time, the amount of VCR was determined with HPLC to compare their drug-release rate, and the degree of hemolysis in the supernatants was observed to compare the fragility of VCR-loaded erythrocytes treated with 0.16% glutaraldehyde with that with 0.25%, VCR-loaded erythrocytes treated with PBS, and VCR-loaded erythrocytes exposed to different hypotonic sodium chloride in different concentrations. Results The VCR-accumulated release rate increased in glutaraldehyde-treated VCR-loaded erythrocytes as well as untreated group during preservation. The VCR-accumulated release rate of 0.25% glutaraldehyde-treated group decreased by 71.67?4.20%, which was significantly slower than untreated VCR-loaded erythrocytes and 0.16% glutaraldehyde-treated group. After storing at 4℃ for 21 days, no significant changes at erythrocyte morphology was found in VCR-loaded erythrocytes treated with 0.25% glutaraldehyde, whereas untreated VCR-loaded erythrocytes and those treated with 0.16% glutaraldehyde can be stored only for 5d and 7.5d respectively. As compared to un-treated VCR-loaded erythrocytes, no significant change of osmotic fragility was seen in 0.25% glutaraldehyde-treated group, but significant increase in osmotic fragility was seen in 0.16% glutaraldehyde-treated group. Conclusion 0.25% glutaraldehyde could optimally block the membrane of VCR-loaded human erythrocytes and enhance their stability.

Objective To study the function of immunocytes in synovial fluid and the mechanisms of immune function disorder in patients with rheumatoid arthritis(RA).Methods The synovial fluid of 20 patients with RA and 10 healthy amputees because of traumatism in the affiliated hospital of Zhejiang university were collected in this syudy.Monoclonal antibodies against CD28,CD154(CD40L),CD80,CD86,CD40,CD69,CD25,HLA-DR,CD71 were used for flow cytometry and the expressions of costimulatory molecule CD28,CD154,CD80,CD86,CD40 on immunocytes and lymphocyte activation markers CD69,CD25,HLA-DR,CD71 in synovial fliud were studied in these patients.We also used enzyme linked immunosorbent assay(ELISA) to detect the cytokines level of Th1 cell including interleukine 2(IL-2),interferon-gamma(IFN-?) and those of Th2 cell including interleukine 6(IL-6),interleukine 10(IL-10) in the synovial fliud.Results Compared with control group,the expressions of CD80[(2.09?0.59)%vs(0.67?0.57)%,P
10. Mechanism of hanshi bi granules in the treatment of ankylosing spondylitis based on network pharmacology

BACKGROUND: Hanshi Bi granule has been used to treat ankylosing spondylitis in the clinic, but the pharmacological mechanism B is still unclear. OBJECTIVE: To screen the compound and drug targets of Hanshi Bi granules based on network pharmacology, and construct a compound-target network to systematically investigate the pharmacological mechanism for treating ankylosing spondylitis. METHODS: The compounds and drug targets of Hanshi Bi granules were collected from TCMSP database. Ankylosing spondylitis targets were from DRUGBANK, GeneCards, Home-OMIM-NCBI, and PALM-IST databases. The obtained drug targets and disease targets were corrected by Uniprot database and common genes were obtained using Draw Venn Diagrams analysis tool. PPI analysis was performed on the common gene in combination with the STRING database and Cytoscape 3.6.1. GO enrichment analysis and KEGG pathway enrichment analysis were performed on the common gene by using Cytoscape 3.6.1 plugin ClueGO. RESULTS AND CONCLUSION: (1) Totally 69 active ingredients and 142 drug targets were obtained from TCMSP, and 595 ankylosing spondylitis targets were obtained using DRUGBANK, GeneCards, Home-OMIM-NCBI, and PALM-IST databases. (2) Using the Draw Venn Diagrams tool to analyze all targets, 39 common genes were obtained. Through PPI analysis, tumor necrosis factor, interieukin-6 and prostaglandin-in-peroxidase 2 were highly connected in the PPI network. (3) Through GO enrichment analysis, it mainly involved biological functions such as regulation of reactive oxygen metabolism, fatty acid metabolism, acute inflammatory reaction, neurotransmitter biosynthetic process, and metabolism of arachidonic acid. (4) Through KEGG pathway enrichment analysis, it mainly involved in AGE-RAGE signaling pathway, fluid shear stress and atherosclerosis, irtterieukin-17 signaling pathway, nuclear factor-KB signaling pathway, tumor necrosis factor signaling pathway, and Toll-like receptor signaling pathway. (5) This study preliminarily predicts the pharmacological mechanism underlying the treatment of ankylosing spondylitis with Hanshi Bi granules, and provides new ideas for secondary development of old drugs and experimental research.
