Chinese Journal of Orthopaedics 1999;0(04):-
Objective To compare the diagnostic value of four signs for superior labrum anterior and posterior (SLAP) lesions of the shoulder. Methods The physical examination was performed randomly on 81 cases with abnormalities of the shoulder. There were four tests, including Kibler anterior sliding test, Liu crank test, O'Brien active compression test and Kim biceps load test Ⅱ. The arthroscopic examination were also performed. The result of the arthroscopic examination was considered as a golden standard, so that we could estimate the diagnosis value of the four tests according to the method of evaluation of diagnosis test on clinical epidemiology, their sensitivity, specificity, positive and negative predictive value, accuracy. Results There were 21 cases diagnosed as SLAP lesions by arthroscopy. The diagnosis value of Kim biceps load test Ⅱ was the highest among the four tests, in which 19 of true positive, 59 of true negative, 1 of false positive, only 2 of false negative case; while the sensitivity was 90.48%, specificity was 98.33%, positive predictive value was 95.00%, negative predictive value was 96.72%, and accuracy was 96.30%. However the sensitivity, specificity, positive predictive value, negative predictive value and accuracy of Kibler anterior sliding test were 76.19%, 96.67%, 88.89%, 92.06%, 91.33%; and those of Liu crank test were 85.71%, 93.33%, 81.82%, 94.92%, 91.35%; those of O'Brien active compression test were 80.95%, 91.66%, 77.27%, 93.22%, 88.89%. Conclusion Kim Biceps load test Ⅱ may be the best for clinical diagnosis of SLAP lesions of the shoulder.

BACKGROUND: Non-adherent mesenchymal stem cells (NA-MSCs) can form colony forming unit of fibroblasts and induce the differentiation into adipocytes, osteoblasts, and chondrocytes. OBJECTIVE: To determine the effect of epithelial growth factor (EGF) on colony formation of fibroblasts and differentiation into neuron-like cells from NA-MSCs.METHODS: Bilateral femur and tibia as well as total MSCs were separated, and repeated-transfer was employed to purify NA-MSCs. The fifth-passage total MSCs and NA-BMCs were induced in a medium containing EGF and basic fibroblast growth factor (b-FGF) for 2 weeks. Colony unit formation of fibroblasts, effect of EGF on colony-forming unit of fibroblasts, and relative protein expression detected by toluidine blue and immunocytochemical staining were observed. RESULTS AND CONCLUSION: Both total MSCs and NA-BMC could generate colony-forming unit of fibroblasts. After treatment of EGF, colony-forming unit of fibroblasts from NA-BMC was increased significantly. Immunocytochemical staining demonstrated that two weeks later both neuro-specific NeuN and NF-200 were observed in total MSCs and NA-BMC; while, toluidine blue staining indicated that neuron-specific Nissl body was observed in some cells. EGF can effectively promote colony-forming unit of fibroblast from NA-BMC, and repeated-transfer NA-BMC can induce differentiation into neuron-like cells.

BACKGROUND: Non-adherent mesenchymal stem cells (NA-MSCs) can form colony forming unit of fibroblasts and induce the differentiation into adipocytes, osteoblasts, and chondrocytes.OBJECTIVE: To determine whether non-adherent mesenchymal stem cells (NA-MSCs) in adult mouse bone marrow could differentiate into neuron-like cells in cerebral ischemic region.METHODS: Bilateral femur and tibia of β-Gal transgenic mice was separated, and repeated-transfer was used to collect the fifth-passage purified NA-MSCs which were adjusted at concentration of 1×10~(12)/L Middle cerebral artery occlusion was established in the two groups. After 7 days, 3 μL fifth-passaged NA-MSCs suspension was injected into cerebral ischemic region in the transplantation group, while an equal amount of saline was injected into model group. Survival, distribution, and differentiation of donor cells in cerebral ischemic region were observed at 8 weeks after transplantation.RESULTS AND CONCLUSION: LacZ staining showed that donor cells could express β-Gal protein after 8 weeks and survived in the ischemic region. Simple and double immunohistochemical staining indicated that β-Gal-positive donor cells were detected in necrotic region and at necrotic edge of ischemic model. Additionally, partial cells could express neuro-specific NeuN protein and glial cell-specific GFAP. NA-MSCs are able to survive and migrate in cerebral ischemic region; moreover, partial NA-MSCs can differentiate into mature neuron-like cells or glial cells which participate in repairing brain injury.

Objective:To investigate the biological characteristics of cultured salivary gland cells in vitro. Methods: Submandibular gland cells of rat were primarily cultured and subcultured in DMEM supplemented with ?=10% fetal bovine serum. The growth of the cells was studied by cell counting and MTT assay. The morphological characteristic was observed with microscope and transmission electron microscope. The specific protein expression was investigated by immunohistochemical technic. Results:Morpholgy of the cells was epidermoid with well developed organella and zymogen granules. The cells could subultured to 4~5 passages and kept in culture for 30~40 days. The cells of primary culture and passage 2 grew faster than those of passage 4.The cells were CK8.13, keratin and ?-SMA positive. Conclusion:Rat submandibular gland cells can be primarily cultured and subcultured in vitro. The primarily cultured and first subcultured cells may be more potential in growth.

Objective To study the effect of vasopressin (VS) on the small bowel circulation and the safety of emboliotherapy for the small intestinal hemorrhage by DSA. Methods Ten dogs were divided into three groups. Vasa recta were ligated 30min after VS infusion ended in group A( n =4), and 2h after VS infusion ended in group B( n =4), they were ligated without VS infusion in control group ( n =2). DSA were performed before and after VS infusion, before and after the ligation. The tested parts of intestine were resected to make the pathologic examination a week late. Results All branches of mesenteric arteries contracted and the contrast developed light in the intestinal wall after VS infusion. Branches contraction recovered but the contrast developed still slight in the intestinal wall about 30min after infusion ended. All manifestation of DSA recovered to normal 2h after infusion ended. In all groups, the blood vessel net can be seen but is fewer and scattered in the area of ligation. The collorate presented soon after the ligation. The pathologic examination proved that there was only mind mucosal ischemia in all groups. Conclusion The repressive effect of VS to the circulation of intestine weakened and then disappeared rapidly after the infusion ended. VS infusion had no significant effect on the safety of emboliotherapy for small intestinal bleeding when the infusion has been finished for more than 2hr. DSA can demonstrated the circulation state of the intestine before and after embolization.
