1.Significance of KAI1 protein expression in tissues of papillary thyroid cancer and follicular thyroid cancer

Objective To investigate the expression of KAI1 protein in tissues of papillary thyroid cancer (PTC) and follicular thyroid cancer,and the effect of KAI1 protein in invasion and metastasis of PTC.Methods The clinical data of 58 patients with PTC (PTC group),17 patients with follicular thyroid cancer (follicular cancer group) and 30 patients with benign thyroid tumor (control group) were retrospectively analyzed from January 2006 to August 2009.The expression of KAI1 protein in PTC tissues,follicular thyroid cancer tissues and benign thyroid tumor tiasues were detected by immunohistochemistry method.Results The positive expression rate of KAI1 protein in PTC group was 48.3%(28/58),in follicular cancer group was 47.1%(8/17),in control group was 86.7 %(26/30).There was significant difference between PTC group,follicular cancer group and control group (P< 0.01).There was no significant difference between PTC group and follicular cancer group (P > 0.05).There was no significant difference in the positive expression rate of KAI1 protein of PTC tissues among the different sex,age and size of the primary lesion (P> 0.05).The positive expression rate of KAI1 protein in the TNM Ⅰ was 88.9%(8/9),the TNM Ⅱ was 60.9%(14/23)and the TNM Ⅲ-Ⅳ was 23.1% (6/26),there was significant difference in the positive expression rate of KAI1 protein among the different TNM stage (P < 0.01).The positive expression rate of KAI1 protein in the lymph node metastasis patients was 32.3% (10/31),the non-lymph node metastasis patients was 66.7% (18/27),there was significant difference (P < 0.05).Conclusion The expression of KAI1 protein may reflect the PTC metastasis and the clinical evolution of tumors,the KAI1 protein may inhibit the invasion and metastasis of PTC.

3.Early efficacy of arthroscopic posterior cruciate ligament reconstruction with tendon through remnant fiber slit

Objectives To introduce arthroscopic posterior cruciate ligament (PCL) reconstruction with the tendon through the middle gap of remnant fiber and evaluate the recovery of postoperative knee function and the preliminary proprioception. Methods 17 cases of eligible patients of posterior cruciate ligament ruptures were performed arthro-scopic Single-beam type anatomical reconstruction with the tendon throung the middle gap of remnant fiber from August 2012 to March 2014. The tendon is fixed in operation through suspension of end-button and extrusion of bioabsorbable interference screw. The recovery of Knee function is assessed with comparative analyzing the preoper-ative and postoperative Lysholm and The International Knee Documentation Committee knee nation form (IKDC) subjective scores and KT-1000 (Joint stability measuring apparatus, PMENT NO 4.563 555, MEDmetric Corporation, San Diego, California, America) scores, and the postoperative proprioception of knee is assessed through measureing the time threshold to detection of passive motion(TTDPM) and the passive re-positioning (PRP) scores. Results There was no serious complications for all patients during the postoperative 12 month follow-up. Subjective scores showed:Lysholm scores from an average of (54.41 ±8.00) scores increased to (90.12±2.69) scores for 12 months followed up ( <0.05);IKDC knee scores from preoperative average (60.29 ±3.48) scores increased to (87.00 ±1.46) scores af-ter follow-up ( < 0.05). All patients reached the pre-injury level of daily life. The results appear as objective scores: preoperative KT-1000 measured mean scores (8.71 ± 2.17) mm, followed up 12 months measured the mean scores(1.88±0.78) mm, proprioception tests follow up 12 months showed, the knee PRP mean Angles respectively in 15o, 45o, 75oand the mean TTDPM Angles test at 45oCompared with the contralateral showed no significant differ-ence ( > 0.05). Conclusion Arthroscopic posterior cruciate ligament reconstruction with tendon throughing remnant fiber can effectively restore the stability of the knee and improve knee function, preserving remnant stubs and syn-ovial of PCL may be beneficial recovery of proprioception and healing of reconstruction tendon.
4.Role of Fas Mediated Cells Apoptosis in Pathogenesis of Human Colonic Cancer Tissue

Objective To explore the role of Fas gene expression in pathogenesis of human colonic cancer tissue.Method Expression of Fas gene protein in benign and malignant colonic tissue was detected by using flow cytometry in forty patients.Results Expression rate of Fas in normal control mucosas was(12 17?3 68)%.The expression rate in colonic cancer tissue was lower than both normal control group and benign tissue.The expression rate in inflammatory tissue was higher than the normal mucosal tissue,and the expression rate was correlated with the tumor differentiation.Conclusions Fas-mediated apoptosis may play an important role in carcinogenesis of colonic tissue.
5.Efficacy of endoscopic dense ligation on bleeding esophageal varices(EV)

Objective To evaluate the efficacy and complication of endoscopic dense ligation for bleeding esophageal varices(EV). Methods 128 cases of EV experienced varcies ligation, and 16~22 points(average 18.8 points) of the varices were ligated by using dense ligation, while only 5~6 points were ligated in common ligation (common method) for varices. All patients got one to two times of ligation therapy. Results In dense ligation group, the rate of prompt hemostasis was 96.9%, rate of elimination of EV was 92.2% with one therapy, and 96.9% with twice therapy, while the rate of bleeding was 0.8%. 6.Relationship between carotid artery intima-media thickness and renal function in patients with diabetes mellitus

Objective To investigate the relationship between carotid artery intima-media thickness and renal function in patients with diabetes mellitus.Methods 424 patients of type 2 diabetes without dialysis were enrolled in a cross-sectional study.According to their artery intima-media thickness (IMT),the patients were divided into normal group and higher IMT group.All patients according to UAER or 24h urinary protein were divided into normal proteinuria group,micro-proteinuria group and clinical proteinuria group.The biochemical examination,eGFR,and atherosclerotic plaque of different groups were compared.Pearson or spearman correlation was used to analyze the relationship between eGFR,IMT and other parameters.Risk factors for eGFR decline were analyzed by binary logistic regression.Results Compared with normal group,patients in the higher IMT group were older [(63.3±10.2) year vs (52.5 ± 10.6) year,P < 0.05],and underwent longer duration of diabetes [(8.9±6.7) year vs (6.2±5.7) year,P < 0.05].Their level of eGFR was decreased [(75.92±28.00) ml/min vs (91.64±24.05) ml/ min,P < 0.05],while plaque incidence (71.3% vs 18.3%,x2=112.42,P < 0.01) and prevalence of hypertension (56.4% vs 29.6%,x2=27.22,P < 0.01) increased.Correlation analysis showed that IMT was positively correlated with age (r=0.503,P < 0.01),duration of diabetes (r=0.204,P < 0.01),24 h urine protein (rs=0.175,P < 0.05),plaque (rs=0.562,P < 0.01),and hypertension (rs=0.193,P < 0.01),but negatively correlated with eGFR (r=-0.307,P < 0.01).Logistic regression analysis showed that age,serum uric acid,24 h urine protein and carotid artery intima-media thickness were independent risk factors for eGFR decline [OR=1.115,95%CI(1.053,1.165),P < 0.001;OR=1.008,95%CI (1.002,1.014),P=0.006;OR=1.492,95% CI(1.170,1.903),P=0.001;OR=1.619,95% CI(1.121,2.339),P=0.010].Conclusion Carotid artery intima-media thickness is an independent risk factor for kidney function decline in patients of diabetes.
7.Effects of isocorydione on cell proliferation in SiHa human cervi-cal carcinoma cell lines

Objective:This paper aimed to investigate the effects of isocorydinone on cell proliferation in SiHa human cervical carcinoma cell lines. Methods:Different concentrations of isocorydione (100, 200, 400, 800, and 1200 μmol/L) were used to treat SiHa human cervical carcinoma cells in vitro for 24, 48, and 72 h. Methyl thiazolyl tetrazolium (MTT) assays were conducted to determine the inhibitory action of isocorydione. Flow cytometry was performed to detect the cell cycle in SiHa human cervical carcinoma cells af-ter treatment with 400 μmol/L isocorydione. Hoechst 33342 staining was used to observe the micro-morphological changes of SiHa cell nucleus after the treatment. The expression of Bcl-2, Bax, and caspase-3 proteins in cervical carcinoma SiHa cell lines was determined using western blot analysis. Results: MTT assays showed that isocorydione inhibits the proliferation of SiHa cells in a dose- and time-dependent manner (P<0.05). The flow cytometry results showed that SiHa cervical carcinoma cells treated with different concen-trations of isocorydione exhibited increased cell cycle. Compared with the control group, Hoechst 33342 staining showed that SiHa cells became narrow, with nuclear pyknosis and fragmentation, and formed an apoptotic body after treatment with 400 μmol/L isocoryd-ione for 48 h. Furthermore, western blot analysis proved that isocorydione significantly inhibited the proliferation of SiHa cell lines, and the expression of Bax protein was increased. By contrast, the expression of Bcl-2 protein decreased gradually. Consequently, the ra-tio of Bax/Bcl-2 increased, as well as the expression of caspase-3 protein. Conclusion:Isocorydione exhibited an overt inhibitory ac-tion on SiHa cells. Isocorydione promoted the occurrence of cell apoptosis, which may be associated with related proteins of mitochon-drial apoptotic pathway.
8.Residue Analysis on Heavy Metals in Medicinal Leeches

This study was aimed to detect the heavy metal residues of medicinal leeches, in order to understand the current market circulation situation of medicinal leeches, and to analyze possible factors which may cause heavy metal pollution, and to provide references for the standardized safe cultivation of medicinal leeches. Inductively coupled plasma mass spectrometry (ICP-MS) technique was applied to detect heavy metal residues in medicinal leeches. The results showed that medicinal leech samples tested for heavy metal content severely exceeded the standard, which caused a great threat to the safety of medication. It was conclude that more attentions should be paid on factors for causing heavy metal accumulation within medicinal leeches during the breeding process. The related department should also list safety limits explicitly and separately for animal drugs such as medicinal leech during the development of quality standards of Chinese materia medica.
9.Hydrogen-rich water inhibits mitochondrial oxidative stress and inflammation in the skeletal muscle after eccentric exercise

BACKGROUND:It is unclear whether hydrogen-rich water can be used to protect skeletal muscle injury induced by eccentric exercise, as wel as the relative mechanism. OBJECTIVE:To observe the effect of hydrogen-rich water on the mitochondrial oxidative stress and inflammation in rat skeletal muscle after eccentric exercise, and to investigate the relative signaling pathway of hydrogen-rich water. METHODS:Forty Sprague Dawley rats were randomly divided into four groups: control group, eccentric exercise group, eccentric exercise+saline group, and eccentric exercise+hydrogen-rich water group. Rats in three eccentric exercise groups were exercised on a motor-driven rodent treadmil at a speed of 16-18 m/min and a slope of-16° for 90 minutes per day. Rats in the eccentric exercise+hydrogen-rich water group were subjected to intraperitoneal injection of hydrogen-rich water (10 mL/kg) immediately after exercise; and rats in the eccentric exercise+saline group were administrated with normal saline after exercise. Al the interventions lasted for 5 days. RESULTS AND CONCLUSION:Hydrogen-rich water intervention after eccentric exercise could markedly enhance the mitochondrial Sirtuin-3 expression, improve the mitochondrial membrane potential and activity of manganese superoxide dismutase, down-regulate the mitochondrial reactive oxygen species generation and mitochondrial DNA oxidative damage, thus inhibiting inflammatory cytokines expression, such as NLRP3 and interleukin-1β. The results indicated that hydrogen-rich saline could directly scavenge reactive oxygen species. In addition, hydrogen-rich water could improve mitochondrial energy metabolism and antioxidant capacity through up-regulation of Sirtuin-3, which in turn inhibits eccentric exercise-induced mitochondrial oxidative stress and secondary inflammation in the skeletal muscle.
10.Study on the Content of Copper in Hirudinidae Medicinal Material Influenced by Culture Environment

OBJECTIVE:To explore the trend of cooper enrichment in hirudinidae influenced by culture environment. MET-HODS:The soil containing low-content,medium-content and high-content of cooper groups(30.00,60.00,90.00 μg/g)and water culture control group were set up. Hirudinidae leech were culture for 60 d,and sampled every 15 days. ICP-MS techniques was used to determined and compared the contents of cooper in W. pigra and soil. RESULTS:In first 15 days,the contents of cooper in leech from 4 groups increased greatly,compared with before;in the following 15 days,the content of cooper kept stable in high-content group while decreased in other 3 groups;in the 30-45 day,the contents of cooper increased rapidly in 4 groups,and those of low-content group and high-content group reached the peak in this experiment;in the last 15 days,the contents of cooper in control group and high-content group increased continuously,while those of low-content and medium-content groups decreased to some extent. Compared with before,the contents of cooper in leech from control group,low-content,medium-content and high-content groups increased by 292%,186%,293%,464% respectively;those of soil from latter 3 groups increased by 81.12%,35.98% and 21.28% respectively. CONCLUSIONS:The content of cooper in leech increase with time,and is positively correlated with the content of cooper in soil. It is suggested to control the content of cooper in hirudinidae through controlling cul-ture environment when hirudinidae are cultured as medicinal material,in order to meet the quality standard of heavy metal in medic-inal material.
