Objective To investigate the method of constructing bioartificial renal tubule assist device(RAD) and its transport function to creatinine,glucose,sodium and asparagine,theonine,serine,glutamic acid in vitro.Methods The cultured LLC PK1 cells were seeded into the inner space of the hemofilter(FH66)and the hemofilter was incubated for two weeks.Then the filtration in vitro was simulated to detect the function of the RAD.The transport rates of sodium,creatinine,glucose and four kinds of amino acids were measured.Meanwhile,the effects of oubaine and phlorizin on the transport rates of sodium,glucose and amino acids were determined and compared with no LLC PK1 device.Results The creatinine transport rate of RAD was significantly decreased as compared with control group(P

Three antibodies were used for immunohistochemical study of 15 cases of adenoid cystic carcinoma(6 cases contained normal salivary gland tissue). The results indicated that myoepithelial cell of the normal salivary glands revealed positive reaction of S-100 protein; The basal cells of the straited ducts and excretary ducts revcaled strong positive reaction of keratin. In adenoid cystic carcinoma positive reaction of S-100 prctein and vimentin were found in luminal cells of cribriform pseudocyts and peripheral cells of tumor nests, and the outer layers'cells of tubular stucture and the border cells of trabucula. But the positive reaction of keratin was found in inner cells of tubular and central cells of trabecular patten. It indicated that adenoid cystic carcinoma probably stems from the basic cells of the striated ducts and excretary ducts of normal salivary glands.

Object To investigate the chemical constituents from the roots of Glycyrrhiza uralensis Fisch. Methods The constituents were isolated on normal and reversed silica gel column chromatography and their structures were identified by spectral evidence. Results A new oleanane-type triterpenoid saponin and twelve known compounds, including two triterpenoid saponins, two cumarins and eight flavonoids, were isolated. Conclusion The new compound was elucidated as 3-O-[β-D-(6-methyl) glucuronopyranosyl (1→2)-D-glucuronopyranosyl]-24-hydroxy-glabrolide on the basis of ESI-MS, 1HNMR,13CNMR, HMQC and HMBC spectral evidence.

Objective To study the effects of bioartificial kidney(BAK)treatment on cytokines interleukin-10(IL-10)and tumor necrosis factor-α(TNF-α),biochemistry indexes,and survival time in acute uremic pigs with multiple organ dysfunction syndrome(MODS).Method Pigs with MODS and acute renal failure(ARF)were treated with BAK(group A,n = 6)or sham BAK containing no cells(group B,n = 6)or received no treatment(group C,n = 5).Data on blood pressure,hepatic and renal functions,serum IL-10 and TNF-α levels,arterial blood gas and survival time of all the pigs were recorded.Comparisons of values were done using Student's t-test or repeated-measures analysis of variance(ANOVA).ResultsMean arterial pressure(MAP,mmHg)responsed more rapidly and reached higher values in group A (91.82±5.73)compared with group B and C at 24 hours(P＜0.01).The peak level of serum EL-10(pg/mL)ingroupA(249.57±43.51)was significantly higher than that in group B and C(132.06± 17.53,104.25 ±13.42,P＜0.01).Serum TNF-a level(pg/mL)in group A dropped gradually to(402.91 ± 32.47)at 24 hours,and had significant discrepancy compared with that before the treatment(537.16 ± 38.45)and that in groupB(P＜0.05).The average survival time(hours)in group A(113.01 ± 14.32)was significantly longer than that in the group B and C(P＜0.01),which was prolonged by 35.93%and 63.90%,respectively.ConclusionsTreatment with BAK can prolong the survival time of uremic pigs with MODS,possibly through ameliorating the MAP,increasing the level of IL-10 and reducing the concentration of TNF-α.

Simple and effective methods are needed for the identification of Chinese medicinal material species and their variety. Lonicera japonica Thunb. is one of Chinese herbal medicines widely demanded. A total of 3 705 EST-SSRs of L. japonica and 2 818 EST-SSRs of L. japonica var. chinensis Thunb. were identified from EST database in our lab. In average, there was one EST-SSR per 4.05 kb in L. japonica ESTs and per 7.49 kb in L. japonica var. chinensis ESTs, separately. The identified SSRs in L. japonica were consisted of 51.98% dinucleotide and 34.61% trinucleotide repeats, while SSRs in L. japonica var. chinensis had 57.45% dinucleotide and 30.09% trinucleotide. The results reviewed that the classes AG/TC and GAG/TCT were predominant in the dinucleotide motifs and the trinucleotide motifs, respectively. Total 87 EST-SSRs were identified of significant difference between L. japonica and L. japonica var. chinensis. PCR products were obtained from 52 L. japonica samples in 13 out of 15 SSR markers tested. The polymorphism in L. japonica, L. japonica var. chinensis and other honeysuckles could be distinguished by three markers (jp.ssr4, jp.ssr64 and jp.ssr65) tested.

Objective Assembly whole mitochondrial genome sequence of Rongshui miniature pig ( RMP ) breed and analysis the structure of mitochondrion based on the next-generation sequecing method.Comparison of phylogenetic relationship and genetic diversity among different pig breeds.Methods We collected peripheral venous blood sample from RMP and constructed two paired-end sequencing libraries.A whole-genome shotgun sequencing strategy and Illumina Genome Analyser sequencing technology were used in our study.Results The mitochondrial genome of RMP consists of 13 protein coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes and the length of pig is 16888 bp.The GC content of this pig mitochondrial genome is about 44 %.Based on phlogenetic analysis, population genetic analysis, our findings confirmed that the ancestral cluster in East Asia mainly occurred among Diannan 7#pig, Hainan wild boar, Lanyu and RMP.Conclusion RMP, a typical miniature pig breed in China, is an earlier ancestor than Lanyu pig breed.

Objective To investigate the correlation of anthropometric parameters with alanine aminotransferase (ALT) in Qinhuangdao college students.Methods A total of 789 subjects from two colleges in Qinhuangdao City (369 men and 420 women) were recruited using a cluster-stratified sampling method.Anthropometric measurements included height,body weight,waist circumference,hip circumference,blood pressure.and ALT.Body mass index,waist to height ratio,and waist to hip ratio were calculated.Results Elevated ALT was found in 16.3% of male participants and 4.0% of females(P<0.05).For males,those with elevated ALT showed significantly higher levels of anthropometric parameters (P<0.05).In females,the levels of anthropometric parameters were similar between subjects with or without elevated ALT(P>0.05).Receiver operating characteristie curve analysis also showed that obesity indices predicted elevated ALT only in men [area under the curve:0.8-0.9(P<0.05) in men,0.5-0.6(P>0.05) in wonlen].Blood pressure did not predict elevated ALT in both men and women (area under the curve:0.5-0.7).Conclusion Dender difference of obesity indices could be found in Qinhuangdao college students.Obesity indices may be independent predictors of elevated ALT in men.However,blood pressure seems not to be an independent predictor of elevated ALT.

Objective To study the bystander effects and associated mechanisms through irradiated conditioned medium(1CM). Methods Natural kilhr(NK) cells were obtained from peripheral blood samples. ICM irradiated with different doses of 60Coγ-rays was used for culturing K562 cell strain. The degree of injury of K562cells by activated NK cells was observed, as well as the apoptosis frequency of K562 cell was investigated. Results Severe injury was induced in K562 cells cultured in ICM than the control (sham-irradiated) as shown by increased sensitivity to NK cells (P < 0.05). The apoptosis frequency of K562 cell was increased significantly compared with the control cells (P < 0.05). Conclusions The bystander effect induced by irradiation is existent. ICM can trigger the bystander effect on K562 cell strains.

Objective To investigate the relationship between impaired fasting glucose(IFG) and the distribution of body fat in adolescents.Methods Stratified cluster sampling was used to select 3874 adolescents aged 13-18 years for this cress-sectional study.Measurements included height,weight,waist circumference,hip circumference and fasting plasma glucose (FPG).Family history of diabetes was determined by using a serf-administered questionnaire.Participants were divided into normal fasting glucose group(FPG＜5.6 mmol/L,n=3738) and impaired fasting glucose group (5.6 mmol/L≤FPG ＜ 7.0 mmoL/L,n=136) according to their FPG levels. Results (1) After adjusting for age and sex using covariance analysis,the impaired fasting glucose group showed increased levels of body mass index,weight circumference,waist/hip ratio,waist/height ratio,as compared to the normal fasting glucose group (P＜ 0.05).(2) After the age and gender were adjusted,body mass index,weight circumference,waist/hip ratio and waist/height ratio were positively correlated with FPG level (P＜0.05).Among the partial relation coefficients, that between waist/height ratio and FPG(r'=0.0925) was the highest (3) In multiple regression analyses,age (β=-0.102,P＜0.05),family history of diabetes (β=0.186,P＜0.05) and waist/height ratio (β=0.842,P＜0.05) were consistently associated with FPG.Conclusion Central obesity was an important predictor of IFG in adolescents.Waist/height ratio may be an useful index of central obesity and an important predictor of IFG in adolescents.

Objective To observe the effects of modified Jianpi Yishen Decoction on urinary osteopontin (OPN) in calcium oxalate nephrolithiasis patients after percutaneous nephrolithotomy (PCNL) or ureteroscope lithotomy (URL);To clarify the mechanism of modified Jianpi Yishen Decoction on the prevention of calcium oxalate kidney stones. Methods Totally 116 calcium oxalate nephrolithiasis patients were randomly divided into trial group (62 cases) and control group (54 cases). The trial group took modified Jianpi Yishen Decoction every other day, while the control group took potassium sodium hydrogen citrate granules three times a day. The concentrations of OPN, urinary calcium and urinary oxalic acid of the patients in the two groups were observed before treatment and 2 weeks and 4 weeks of treatment. Results The concentration of urinary OPN of 2 weeks and 4 weeks of the treatment in the trial group was significantly increased compared with before treatment (P<0.05), and the difference was statistically significant compared with the control group (P<0.05). The concentration of urinary OPN in the control group had no significant change after treatment (P>0.05). The differences in the concentrations of urinary calcium and urinary oxalic acid of the two groups between before and after treatment were not significant (P>0.05). Conclusion Modified Jianpi Yishen Decoction can effectively restrain the formation of the calcium oxalate stones by increasing the level of urinary OPN, which demonstrates effective prevention in the calcium oxalate nephrolithiasis patients after PCNL or URL.
