1.Effect of glutaminase 1 inhibitor on carbon tetrachloride-induced liver fibrosis in mice

Objective To investigate the effect of glutaminase 1(GLS1)specific inhibitor BPTES[bis-2-(5-phenylacetamido-1,3,4-thiadiazol-2-yl)ethyl sulfide]on the liver fibrosis in the mouse model of liver fibrosis induced by carbon tetrachloride(CCl4).Methods Male C57BL/6J mice were intraperitoneally injected with olive oil(control group),10%CCl4(10 μL/g,model group)or 10% CCl4(10 μL/g)+ BPTES(10 mg/kg,treatment group),with 10 mice in each group,two doses a week for four weeks to establish liver fibrosis model. Collagen deposition in mouse liver tissue was observed by Sirius red staining. The expression levels of actin alpha 2(Acta2),collagen typeⅠalpha 1(Col1a1)GLS1 and GLS1 protein were detected by qRT-PCR and immunohistochemical staining.Results Compared with the control group,the liver tissue of mice in the model group was generally enlarged,the surface was not smooth and granular,and the ratio of liver mass to tibia length significantly increased(t = 2. 979,P < 0. 05);The Sirius red positive area of collagen deposition increased signifi-cantly in the liver tissue of mice in the model group(t = 7. 661,P < 0. 01),the relative expression levels of Acta2 and Col1a1 significantly increased(t = 4. 335 and 5. 319,respectively,each P < 0. 01),and the mRNA and protein levels of GLS1 significantly increased(t = 5. 319 and 9. 725,respectively,each P < 0. 01). However,compared with the model group,the BPTES treatment group had a reduction in liver mass,a significant reduction in the Sirius red positive area of collagen deposition in liver tissue(t = 7. 427,P < 0. 01),and a significant reduction in the relative expressions of Atca2 and Col1a1(t = 3. 713 and 2. 628,respectively,each P < 0. 05).Conclusion Inhibition of GLS1activity can significantly improve the degree of liver fibrosis induced by CCl4,providing a new idea for the treatment of liver fibrosis.

4.Analysis of the clinical diagnosis and therapy and misdiagnosis for congenital middle ear cholesteatoma

[ABSTRACT]OBJECTIVETo investigate the methods of diagnosing and treating congenital middle ear cholesteatoma
and causes of misdiagnosis.METHODSThis was a retrospective review of 9 cases of congenital middle ear cholesteatoma in this hospital.RESULTS9 patients all received operative treatment and definite diagnosis was made by histopathologic investigation. The ossicular reconstruction was performed to 6 patients. All of the cases were followed-up for 3 months-2.5 years. A-B gap was 20 dB. After 1 year, no residual or recurrence of cholesteatoma was found in CT scan. All patients had been misdiagnosed. Misdiagnosis rate was 100%.CONCLUSIONCongenital middle ear cholesteatoma is a clinically rare and diagnosis is usually delayed in clinical practice due to the silent nature of the disease in its early stage. Preoperative diagnosis may be based on Levenson diagnostic criteria and CT imaging. Early surgical treatment can obtain a good effect of hearing reconstruction. To avoid misdiagnosis and incorrect treatment, radiological evaluation and careful local examination are really important, which may also avoid serious complications.
5.Study on the Quality Standard for Gardenia jasminoides Commercially Available

OBJECTIVE:To establish the quality standard for Gardenia jasminoides. METHODS:The moisture,total ash and extract were determined. HPLC was used for contents determination of geniposide,rutin,crocinⅠand crocinⅡ:the column was Waters Xbridge-C18 with mobile phase of acetonitrile-0.2% phosphoric acid(gradient elution,gardenia and rutin),methanol-water (45∶55,V/V,crocinⅠ,crocinⅡ)at a flow rate of 1.0 ml/min;detection wavelength was 256 nm for gardenia and rutin,440 nm for crocinⅠ,crocinⅡ;column temperature was 30℃;injection volume was 10μl for gardenia and rutin,5μl for crocinⅠ,cro-cinⅡ. RESULTS:The moisture total ash and ethanol-soluble extract of G. jasminoides were 5.8%-8.4%,3.7%-5.9% and 29.5%-37.9%,respectively. The linear range was 162.08-1 620.84 μg/ml for geniposide(r=0.999 9),2.07-20.72 μg/ml for rutin (r=0.999 9),8.04-80.41 μg/ml for crocinⅠ(r=0.999 9)and 1.05-10.53 μg/ml for crocinⅡ(r=0.999 9);RSDs of precision,sta-bility and reproducibility test were lower than 2%;recoveries were 99.33%-101.43%(RSD=1.09%,n=6),97.97%-101.83%(RSD=1.39%,n=6),97.97%-101.30%(RSD=1.36%,n=6) and 98.65%-103.04%(RSD=1.84%,n=6). CONCLUSIONS:The established standard can be used for the quality control of G. jasminoides commercially available.
6.Research Progress of Acupuncture in Treating Cervical Vertigo

The research progress was surveyed from the perspectives of filiform needling, moxibustion, acupoint injection, acupuncture and massage,acupuncture and traction. In the treatment of cervical vertigo, acupuncture acts to dilate blood vessels,diminish vessel resistance and increase blood flow, hence to improve micro-circulation and oxygen supply to the brain.
7.Molecular epidemiology of Chlamydia trachomatis in outpatients visiting STD clinics in Fuzhou,China

We investigated the distribution of Chlamydia trachomatis genotypes in the outpatients visiting STD clinics in Fuzhou City ,China .Between 2013 and 2014 ,a total of 2 019 urethral or endocervical specimens from susceptible genital C .trachomatis infections were collected and detected by using real‐time PCR assay kit .The C .trachomatis‐positive specimens were subsequently amplified with nested PCR for Omp1 region and analyzed by sequencing .Genotyping was performed by BLAST similarity search and phylogenetic tree analysis .Eighty‐three of the 86 C . trachomatis‐positive specimens were suc‐cessfully sequenced .Only one specimen was identified as multiple infection .Eight different genotypes were identified .The most prevalent was F (36 .59% ) ,followed by E (24 .39% ) ,D (12 .20% ) ,G (9 .76% ) ,J (9 .76% ) ,H (3 .66% ) ,K (2 .44% ) and B (1 .22% ) .Synonymous mutation rate and non‐synonymous mutation rate among Omp1 genes of different geno‐types were calculated .All genotypes but G showed dN/dS values of<1 .Genotypes F and E were commonest .Synonymous sub‐stitution was the major pattern of variation in the process of Omp 1 gene evolution .
8.The role of botulinum toxin for acute-onset concomitant esotropia:a pilot study

Objective To observe the effectiveness of botulinum toxin A (BTXA) in the treatment of small sample patients with acute acquired concomitant esotropia. Design Retrospective case series. Participants 6 patients (3 female and 3 male) with acute acquired concomitant esotropia aged 6 to 34 years. Methods All the patients were received 2.5 units of BTXA injected into the bilateral medial rectus muscles once under electromyographic control. They were performed alter prism-cover test,synoptophore and stereoscopic charts test. Main Outcome Measures The alignment of eyes and binocular vision. Results The follow-up of patients was 4 to 39 months. The pre-injection angle of deviation was 43.3?1.0△; the angle of deviation at the last follow-up was 3.3?8.2△,and 5 patients achieved alignment. In pre-injection,one patient had distance stereopsis,2 patients had near stereopsis with 600 seconds and 40 seconds; after injection,5 patients demonstrated distance and near stereeopsis (3 patients 40 seconds,1 patient 60 seconds,and 1 patient 200 seconds). Conclusion This small sample study shows that BTXA injection is effective alternative in treating acute acquired concomitant esotropia.
9.The analysis of the risk factors of primary gout

Objective To investigate the risk factors of primary gout.Methods 420 patients with primary gout were analyzed.Results Among the 420 patients,male to female ratio was 21∶1.Average onset age of female was significantly higher than that of the male,whereas,the mean disease duration was significantly shorter in female than that in male.The mean disease duration of patient with positive familial history was significantly shorter than that with negative ones.38% patients had an alcohol abuse history,and 54 percent patients had had high purin food intake.Mean weight of body was (74.7?9.9)kg,and average body mass index(BMI) was (27.3?2.6).Events of inducing acute attack included engorgement,tire,infection,operation and trauma.Conclusion Risk factors of primary gout include age,sex,inherity,food habit,hyperlipemia,renal function failure.Engorgement,tire,infection,operation,trauma and use of diuretics could induce gout acute attack.It should be taken importance to that the onset age shows a young tendency.
10.Regulation of peroxisome proliferator activated receptors-? on inflammatory reaction in left ventricular hypertrophic rats

Objective To investigate the effects of rosiglitazone, an synthetic ligand of peroxisome proliferator activated receptor-?(PPAR-?) on the expression of proinflammatory factors in rats of myocardial hypertrophy. Methods The rat model of hypertrophy were established by coarctation of abdominal aorta in male SD rats, and the rats with myocardial hypertrophy were treated with rosiglitazone or physiological saline (n=10 for each therapy). Some rats underwent sham operation and were given rosiglitazone or physiological saline (n=10 for each therapy). Rosiglitazone at the dose of 4 mg?kg -1?d -1 or saline of 1 ml?kg -1?d -1 was given intraperitoneally for 1 week after postoperative 4 weeks. Another 9 rats served as control. Hemodynamics, ventricular hypertrophic index, TNF-?, PAF, MPD, PPAR-? and NF-?B were detected in all rats at postoperative 5 week. Results Significant hypertrophy was found in the left ventricle in rats undergoing the coarctation of abdominal aorta.
