3.Different activities of antimicrobial combinations against muitidrug resistant Acinetobacter baumannii in vitro

Objective To evaluate the activities of 18 pairs of antimicrobials combinations against non - duplicate clinical isolates of multidrug resistant Acinetobacter baumannii (MDRAB) in vitro.Methods Collect isolates of Acinetobacter baumannii from different patients from October 2009 to May 2010,which were isolated in Clinical Laboratory Center of Beijing Friendship Hospital,Capital Medical University.Use broth microdilution method to detect MIC of mono-antimicrobial,and checkerboard broth microdilution method to detect combinatied MIC,and calculate fractional inhibitory concentration (FIC) index to determine drug combinations effects.When the performance of the same drug combinations conflicted,appropriate strains were selected for screening of drug-resistant mechanisms by polymerase chain reaction( PCR),including efflux pump genes.Results In tests in vitro,rifampicin and polymyxin B,imipenem and gentamicin,cefepime and levofloxacin showed synergy at high proportion,68.1%,45.5%,40.9%,respectively.Minocycline and rifampicin,ampicillin/sulbactam and tobramycin.Ceftazidime and ciprofloxacin showed additive effect at high proportion,81.8%,68.2%,68.2%,respectively.There were several combinations which appeared the opposite effects to tested strains.Strains No.19 corresponding reaction was synergy and No.21,No.26 corresponding reactions were antagonism.The three strains above were selected for screening resistant mechanisms.The difference is that genotypes of adeS were negative in No.19 and positive in No.21 and No.26.Conclusion Rifampicin and polymyxin B combination showed synergy against the MDRAB in vitro,which can be considered as the treatment choice for critical infections caused by MDRAB.Imipenem and gentamicin,cefepime and levofloxacin also showed synergy in vitro,but in some isolates showed antagonism.This phenomenon may be due to the gene adeS activated by certain antibiotics,and the activated adeS drived efflux pump express or overexpress,which made the drugs in bacterial cells pumped out,causing antagonistic effect.The individual differences in strains should be considered when clinic strain apply these two combinations above.
4.The regulatory effect of endothelial cells of pulmonary arteriole on the tone of pulmonary artery

The regulatory effect of endothelial cells of the rabbit pulmonary arteriole(PAECs) on the tone of pulmonary artery was studied. The conditioned medium (CM) ofPAECs, cultivated under normal and hypoxic condition for 24 hours, did not induce con-striction of pulmonary artery ring. The aqueous phase was able to induce the constriction.The constriction of pulmonary artery rings was reduced by the heated aqueous phase, but it was disappeared when the heated aqueous phase was treated with trypsin. The constric-tion caused by heated aqueous phase was stronger in hypoxic endothelial cells than innormal cells. Treatment of PAECs with indomethacin and captoril, and administration ofFPL 55712, BN 52021, chlorpheniramine and prazozin to contraction pool did not affectthe constriction of pulmonary artery rings. It was concluded that PAECs might producelong half-life lipid substances which could induce the dilatation of pulmonary artery.Hypoxia inhibted the release of long half-life aqueous heat-unstable vasoconstrictive sub-stances. but enhanced that of the heat-stable fraction which was probably a peptide.
5.Research of self-management self-efficacy and health-related quality of life in hemodialysis patients

Objective To investigate the self-management self-efficacy,health-related quality of life of hemodialysis patients and explore the relationship between them. Methods Self-management self-efficacy questionnaire and SF-36 were used,and 55 hemodialysis patients participated in this study. Pearson correlate and regression analysis were used to test the relationship between them. Results 55 questionnaires were available. Positively associated between self-management self-efficacy and HRQOL(r=0.268,P<0.05);significantly positively associated between physical activity self-efficacy and HRQOL(r=0.409,P<0.01). Regression analysis showed that physical activity self-efficacy is the factor of HRQOL (t=3.261,P<0.01).Conclusion Physical activity self-efficacy is the factor of HRQOL in hemodialysis patients,increasing self-management self-efficacy is benefit to their HRQOL.
6.A QUANTITATIVE METHOD FOR HESPERIDIN (VITAMIN P)DETERMINATION

Based on the cyanidine reaction, a quantitative electrocolorimetric method for the estimation of hesperidin is described. The main steps of the procedure are as follows:Extraction: The sample is washed successively with 95% alcohol and ether until the washings are free from yellow color when alkali is added. The washed sample is then extracted under re fluxing for 90 minutes with 50% alcohol (the best ratio between the sample and alcohol ia 1:100 by weight), filtered while hot, and the filtrates is kept at 70℃ until used.Color producing and comparison: Five ml of the above extract are added into a test tube containing 250 mg of magnesium powder. The tube thus prepared is placed into warm water of 15℃ and 2 ml of cone. HCl are added slowly (0.2 ml for every 2 minutes) with gentle shaking. After the addition of HC1 is completed, the content of the tube is shaken and incubated at 37℃ for 1 hour. The extract is then cooled, filtered and its optical density is determined. The error introduced by the colored sample solution is corrected by setting the optical density reading of the blank at zero before the reading of the unknown. The blank was prepared similarly as the unknown except that the reaction of magnesium powder with added HCl was completed before the addition of sample solution.The hesperidin content of the sample is determined by interpolation of the optical density values on a standard curve prepared by a series of gradient concentration of hesperidin.An amount of 0.025 mg hesperidin in 5 ml solution can be determined quantitatively by this method and the recovery was 97.1-100.3%.
7.RESEARCH INTO THE ACTION AND MECHANISM OF LARGE DOSE OF ViT.C ON COLD TOLERANCE AND ADAPTATION IN GUINEA PIGS

117 guinea pigs were divided into groups, receiving 1mg, 5mg, 50mg and 100mg Vit C per os daily respectively in 2-4 weeks. There were conducted four experiments: (1) the effect of different vitamin C levels on the cold tolerance and adaptation in guinea pigs; (2) effect of large dose of vitemin C on sudden and long time exposure to cold: (3) the action of vitamin C, in large dose and short time in combination with hormones, on cold tolerance of guinea pigs; (4) effect of large dose of vitamin C on the myocardial enzyme activity, during the process of cold adaptation. Rectal temperature, body weight, weight and vitamin C contents of adrenal glands, the excretion of urinary 17-Ketostero'ids, activity of myocardial succinic dehydrogenase and cytochrom C oxidase were observed. The results revealed that large dose of Vitamin C per os might lessen the lowering of rectal temperature and body weight after exposure to cold; promote the activities of myocardial enzymes. Better results were obtained with combination of vitamin C with cortisones. It was noted that large dose of vitamin C apparently increased and accelerated the tolerance and adaptation of organisms to cold.
8.Research of Serial Communication in Multi-Axis Controller Based on .NET

Objective A solution of serial communication in Multi-Axis Controller which uses programmable multi-axis controller (PMAC) as down processor and PC as up processor is introduced. By the PComm32.dll provided with Delta Tau Data Systems Inc., the serial communication between PC and PMAC is realized based on .NET. Methods In order to realize the programming techniques of serial communication, a new method that managed codes invoked the unmanaged codes in Common Language Runtime environment by using Platform Invocation Services was adopted. The serial communication program was designed by using the program language C#. Besides the single axis locomotion,the debugging procession of the multi-axis joint locomotion was also recounted based on the five-DOF robot. Results The results indicated that the serial communication between PC and PMAC was completely realized. Conclusion The developed serial communication software module is of good correctness, stability and reliability, and also shows that the software is convenient and easy to use, which can accord with system requirements.
9.Progress of the relationship between homocysteine and kidney disease

Homocysteine(Hcy) is an intermediate product of methionine metabolism.It is considered that Hcy is an independent etiological factor of cardiovascular disease,and blood Hcy level is closely related to many diseases.Patients with kidney disease may have different levels of homocysteine.The study has found that Hcy may not only be related to kidney damage,but also make the risk of cardiovascular disease in patients with kidney disease increased.The relationship between Hcy and kidney disease is briefly introduced in this article in order to provide a new basis for prevention,diagnosis and treatment of kidney disease and complications.
