1.Effects of seven sevoflurane and propofol anesthesia on heart rate and myocardial enzymes in elderly patients with coronary heart disease

Objective To investigate the effects of seven halothane and propofol anesthesia on heart rate and myocardial enzymes in elderly patients with coronary heart disease (CHD), and to analyze the safety of clinical application of the two kinds of anesthetic drugs.MethodsA total of 70 cases of elderly patients with coronary heart disease treatment January 2012 to December 2014 general anesthesia surgery as the research object, randomly divided into A group and B group were 35 patients in.A group were treated with seven sevoflurane anesthesia, group B patients were given propofol anesthesia, observe the change of the two groups of patients before and after anesthesia hemodynamic index and myocardial enzyme etc.index.ResultsA group of patients with HR 5min after induction and 10min were significantly lower than before the surgery, and the difference was statistically significant (P<0.05);HR stabilized after the induction of 15min and 20min, and compared with before surgery had no significant difference;patients in group B HR and 10min 5min after induction there was no obvious change compared with that before operation, no statistical significance;and after the induction of 15min and 20min HR increased, the difference was statistically significant (P<0.05).A group and B group in patients with systolic and diastolic blood pressure were lower than before anesthesia in varying degrees, and group B more significantly, the difference was statistically significant (P<0.05).A group and B group of patients after 6 h, cTnl and CK-MB in 12h and 24h than 15min before surgery has increased significantly, and A group cTnl and CK-MB were lower than B group, the difference was statistically significant (P<0.05).ConclusionThe clinical application of seven sevoflurane anesthesia for elderly patients with coronary heart disease during propofol is feasible, but seven halothane on blood pressure and heart rate of patients affected with propofol, improve the myocardial enzyme level, and better reduce the damage of perioperative myocardial, more suitable for anesthesia in elderly coronary heart disease.
2.Study on the preparation of mouse embryonic stem cell feeder layer

Objective:To establish a stable and effective system of mouse embryonic fibroblasts(MEFs) feeder layer in order to separate and culture mouse embryonic stem cells(ESC) in vitro.Methods:the MEFs were isolated from the mouse embryos at 13.5~14.5dpc(days post-coitus) through primary tissue digestion and tissue piece culture.The optional planting density of cell was determined according to the cellular growth curve detected by MTT and the morphology in the culture dish under inverted microscope.The optional concentration of mitomycin C and the time treating with mitomycin on MEFs were determined by MTT assay.Murine blastcysts(3.5dpc) were cultured on the feeder layer treated by mitomycin C and ESC growth was observed.Result:The method of tissue digestion and tissue piece culture is an efficient way to obtain primary MEFs;1?105~2?105/ml cells is an optional planting density;MEFs proliferation could be effeciently repressed after being treated with mitomycin C 10?g/ml for 3.5 hours or 20?g/ml for 2.5 hours,and the quantity of MEFs could maintain at least for 7 days.ES clone could be obtained from the mouse blastcysts(3.5dpc) after being cultured on the feeder layer.Conclusion:MEFs feeder layer could effectively support mouse embryonic stem cells to grow.
3.Experimental study on IL-3 gene expression induced by TSPG

Objective:To explore the effect of TSPG (total saponins of panax ginseng) on gene expression of IL-3 which is one of the important hematopoietic growth factor at early stage of hematopoiesis.Methods:Techniques of RT-PCR and in situ hybridization were used.Results:A band corresponding to IL-3 was observed after RNA extraction from TSPG stimulated Jurkat cells were subjected to RT-PCR,while such band was not observed after RNA extraction from unstimulated Jurkat cells were subjected to RT-PCR.Comparing with the control group,both quantity and intensity of IL-3mRNA expression in fetus thymocytes and splenocytes induced by TSPG were obviously promoted.Conclusion:TSPG can induce human lymphocytes to express IL-3 mRNA,thus promote human hematopoiesis at early stage.
4.Practice of applying PBL teaching method to experimental teaching of histology and embryology

5.EXPERIMENTAL STUDY FOR THE EFFECT OF TSPG ON THE EXPRESSION OF IL-3 IN HEMATOPOIETIC STROMAL CELLS

Objective Panax ginseng is a well-known Chinese traditional medicine functional in “invigorating qi”,and saponins are one of its main effective fractions.Our study investigates the effect of total saponins of panax ginseng(TSPG)on human hematopoiesis at early stage and its possible regulative mechanism to clarify the hematonic mechanism of Panax ginseng. Methods The techniques of culturing hematopoietic progenitor cells in vitro,bioassay of hematopoietic growth factor(HGF),immunocytochemistry and nucleic acid in situ hybridization were used to study the effect of TSPG on IL-3’s expression in hematopoietic stromal cell and its possible mechanism. Results TSPG directly added into culture system in vitro can markedly increase the colony forming yields of CFU-Mix;the different conditioned culture media prepared with TSPG can promote the proliferation and differentiation of CFU-Mix;the protein and mRNA expression of IL-3 in BMSC and cells of EcV304,THP induced by TSPG has been much intensified.Conclusion TSPG may regulate hematopoiesis at early stage by activating IL-3's gene expression in hematopoietic stromal cell,which is the most important element in hematopietic inductive microenvironment(HIM).
6.Analysis on professor WANG Juyi's crevice theory from Tao Teh King.

The crevice theory proposed by professor WANG Juyi is analyzed in this article. In the crevice theory, it is believed that the meridians are located among "skin, pulse, flesh, tendon and bone", and the crevice is the path for the transportation of qi-blood and body fluid in the body, which is the essential condition for vital movement. This is in agreement with the idea of "action through inaction" from Tao Teh King. Based on crevice theory, the abnormalities of meridians can reflex the deficiency and excess of diseases, and regulating meridians is a significant way to treat diseases, and maintaining smooth meridian is important for healthcare.

7.Application of Molecular Biology in Investigation of Resistance in Enterococci

Objective This article aims to provide mechanisms and recent developments of molecular biology pertaining to re-sistance of Enterococci,providing rapid approaches for detecting resistant strains.Methods This article reviewed recent lit-eratures on resistance of Enterococci and a systemic analysis was conducted.Results Common detecting methods include polymerase chain reaction (PCR),pulsed field gel electrophoresis (PFGE),multilocus sequence typing (MLST)and South-ern blot.There also exist less widely-used methods such as pyrosequencing and genechip technique,which may prove effi-cient in some aspects.Conclusion Every method has its advantages and disadvantages.This article discussed how to utilize these methods to achieve their maximum capabilities.
8.Reflection on practice of progressive assessment on comprehensive quality of undergraduates

Progressive assessment is one of the methods adopted by colleges and universities to evaluate the comprehensive quality of undergraduates.It can provide objective references for various awards and selections.Most colleges used static and transverse methods; based on these methods,advantages and disadvantages of individual undergraduates were compared with general groups,but feedback and forecast functions of assessment can not be realized.Therefore it is necessary to transform our conceptions and practice progressive assessment on comprehensive quality of undergraduates.We can learn from the relevant conceptions and methods in enterprise management and use data envelopment analysis to exclude the influence of different basic conditions of undergraduates.These undertakings are of great importance to truly reflect undergraduates' effective progress degree,guide them to test and evaluate themselves correctly and to advance comprehensive quality continuously.
9.Comparative study of in vitro anti-tumor activities of three different extracts of Pedicellus MeIo

Objective To study the antitumor effects of three different extracts from Pedicellus Melo on the proliferation of cancer cells in vitro, and discuss the possible mechanism. Methods Pedicellus Melo was extracted by water, 75%ethanol and 95%ethanol, respectively. The anti-cancer activities in vitro were detected by MTT assay on five tumor cell lines SGC-7901, NCI-H460, SMMC-7721, K562 and L929 cells. Flow cytometry was used to detect the cell cycle and apoptosis of different cell groups. Results After 72-hour treatment with water, 75%ethanol and 95%ethanol extracts, inhibitory effects were found in five tumor cell lines, which showed dose-dependent manner on the proliferation of the cells. The IC50 values were significantly lower in the two ethanol extracts than those of water extract (P<0.05) . And the IC50 values of 95%ethanol extract on SGC-7901, SMMC-7721, K562 and L929 cells were significantly lower than those of 75% ethanol extract (P <0.05). All the extracts were sensitive to SGC-7901 cells. Apoptotic rates of SGC-7901 cells treated with 95%ethanol extract of the concentrations of 0.4, 0.6 and 0.8 mg/L for 48 hours were (8.32 ± 0.55)%, (11.06 ± 1.15)%and (12.25±1.51)%. The cell cycle was arrested in S and G2/M phases. Conclusion Results demonstrate that 95%ethanol extract of Pedicellus Melo has significant inhibitory effect on SGC-7901 cells in vitro, which may be related to the induced apoptosis and the inhibition of cell division.
10.Effect of dietary oils on pulmonary vascular reactivity and chronic hypoxic pulmonary hypertension in rats

The effect of dietary oils on pulmonary vasoreactivity and chronic hypoxic pulmonary hypertension were assessed in rats fed on soybean oil, pig oil, soybean oil plus pig oil or regular diets for two months before the start of normobaric hypoxia(10?0.5%O_2 ventilation, 8 hours per day, 14 days). Mean pulmonary arterial pressures and pulmonary vasoreactivity to acute hypoxia were higher in the chronically hypoxic rats fed on 15% soybean oil than in the rats fed on 15% pig oil, 7.50% soybean oil plus 7.5% pig oil or regular diets. Hypertrophy of the right ventricle in the rats fed on 15% soybean oil were more serious as well. Blood viscosity and hematocrit were not different among the four groups of chronic hypoxia. The findings suggest that dietary soybean oil can potentiate hypoxic pulmonary vasoconstriction, and thereby promote the development of pulmonary hypertension and right ventricular hypertrophy.
