1.Affinity Selection of Human ScFv Against Botulinum Neurotoxin Serotype A

In this work, rare specific binders to botulinum neurotoxin antigen were selected from human phage immunized library through three rounds of panning in vitro. Of them, clone ScFvB17 is of high affinity and specificity to target antigen. B17 is of 750bp encoding 250 amino acid. Analysis results of gene structure showed V-genes of B17 belonging respectively to VH4 or? chainⅡ family are new antibody Fv genes. More important result showed that recombinant ScFvB17 expressed in E.coli could bind specific for neurotoxin competing with antiserum. Successful selection of human phage antibody library and obtaining specific ScFv against botulinum neurotoxin serotype A provide useful materials for further study on detection of neurotoxin and human engineering therapeutic antitoxin.

3.Analysis of cornual pregnancy in 21 patients

Objective:To discuss the diagnosis,treatment of uterus cornea pregnancy.Methods:Retrospective analysis was made in 21 cases of uterus cornua pregnancy is recent 10 years.Results:8 cases were diagnosed uterus cornua,9 cases were misdiagnosed ectopic pregnancy,1 case was misdiagnosed rupture of uterus of artifical obortion,1 case was misdiagnosed pregnancy with uterus septus,1 case was misdiagnosed choriocarcinoma.Surgical treatment was done in 19 patients,and the other 2 cases received conservative therapy.Conclusion:Cornual pregnancy is often misdiagnosed as ectopic pregnancy or other diseases.More careful gynecological and transobdominal ultrasound examination will improve the accuracy of diagnosis.
4.Clinical value of procalcitonin on early diagnosis of sepsis

Objective To study the clinical value of procalcitonin on early diagnosis of sepsis .Methods 60 patients who entered ICU were enrolled in the present study were chosen .The change of procalcitonin , CRP, WBC,ESR,GRANR and the highest temperature was observed dynamically with 1 day and 4 days in hospitalization . The patients were ultimately divided into the two groups according to the diagnostic standard of sepsis ,the sepsis group (SIRS group) and infective systemic inflammatory response syndrome group (NONSIRS group).Results After ana-lyzing the processed data,researchers found that PCT of the sepsis group (6.73 ±1.31)ng/mL was higher evidently than that of the SIRS group(0.25 ±0.05)ng/mL(t=3.10,P<0.05),the area under ROC curve of PCT (0.869) is evidently greater than CRP(0.798),WBC(0.737) and ESR(0.394)(P<0.05),which had statistical significance. Conclusion Procalcitonin is a good index of early diagnosis on sepsis ,therefor,its sensitivity and specificity is higher than the other inflammatory indicators .Finally,procalcitonin can be widely used in clinical trial .
5.Clinical research on preoperative interventional chemotherapy for uterine cervical cancer

Objective:To discuss the clinical effect of preoperative interventional chemotherapy and pathological changes in uterine cervical cancer.Methods:preoperative interventional chemotherapy by femoral intubatton were done in 21 patients with uterine cervical cancer.The project of chemotherapy was bleomycin 45mg and cisplatin or oxaliplatin 80mg/m2.Rusults:21 cases of uterine cervical cancer including 7 cases ofⅠstage and 14 cases of Ⅱstage received one or two times of preoperative interventional chemotherapy.All of the patients successfully completed cervical cancer radical therapy.All of the specimens were sent for pathological examination.A lot of lymphocyte cells infiltration and necrosis were found in the cancer tissues.Conclusion:preoperative interventional chemotherapy for uterine cervical cancer proves to reduce the local cancer size as well as decrease the bleeding of cancer.The patients can receive cervical cancer radical therapy after the interventional chemotherapy.

7.An Observation of Experimental Autoimmune Uveitis in Guinea Pigs by Light and Electron Microscope

The soluble antigen was isolated from pig retinal extract by precipitation with ammonium sulfate followed by molecular sieve. 50 ?g S antigen with equal volume complete Freund's adjuvant was injected into double hind foodpads of guinea pigs. Two weeks later, experimental autoimmune uveitis developed. Its pathologic feature is that a great number of lymphocytes infiltrated into chroid which was thickened gradually from anterior to posterior. Inflammatory variation was relative to disease phase. Ultrastructure pathology was characterized by swelling and necrosis in the inner and outer segments of photorecepter cells. This result suggests that S antigen has significant effect on inducing experimental autoimmune uveitis and the outer layer of retina is the sensitive target organ of S antigen.
8.Protective action of TMPustrazine(TMP) on hearts and brains under the condition of acute hypoxia in mice

AIM Protective action of TMP on hearts and brains under the condition of the acute hypoxia was studied. METHODS On the device of mice acute hypoxia ,ability of tolerancing hypoxia of mice was investigated,and superoxide dismutase(SOD) and malondialdehyde(MDA) and nitric oxide(NO) contents in hearts and brains were measured.RESULTS TMP could make the resisting hypoxia action in mice strengthen significantly,and the action had a does dependent increase.
