Magnetic mediated particles can be located within tumor volume through special ways are can be heated while exposed to an externally alternating magnetic field,thus kill the tumor cells.Compared with othen hyperthermia a major advantage of magnetic mediated hyperthermia is that of targeting tumor specially and selectively.The evidences of clinical research demonstrate that magnetic mediated hyperthermia will have a promising future and play an important role in tumor therapy.
2.An in vitro blood compatibility assessment on the adsorptive resin in hemoperfusion filter cartridges.

OBJECTIVETo investigate necessity of the further processing for adsorptive resin material.
METHODSAccording to GB/ T 16886.4, coagulation tests, haematology tests, in vitro spontaneous platelet-adhesion test and complement activation tests were conducted respectively to assess the in vitro blood compatibility of adsorptive resin material.
RESULTSIn comparison with polypropylene, he adsorptive resin material appears to be a very slight activator to the endogenous blood coagulation system, it can inhibited the exogenous blood coagulation system very significantly (P < 0.01), it can adhere to fibrinogen, red blood cells, reticulocytes very significantly (P < 0.01), and platelet significantly (P < 0.05). The adsorptive resin material is also revealed to be a very significant activator (P < 0.01) to the complement system.
CONCLUSIONSThe in vitro blood compatibility assessment witnesses that it is very necessary for the adsorptive resin material to go through the certain special processing(such as the surface modification and the microencapsulation technology, etc) before its application to human being.

4.The expression and significance of matrix metalloproteinases and neutrophil gelatinase-associated lipocalin in serum and placental tissue of preeclampsia maternal

Objective To investigate expression and significance of matrix metalloproteinase-9 (MMP-9) and neutrophil gelatinase-associated lipocalin (NGAL) in serum and placental tissue of preeclampsia maternal.Methods 74 cases of preeclampsia pregnant women as research subjects,which 39 cases of severe preeclampsia,35 cases of mild preeclampsia,selected 30 cases of normal late maternal as the control group.Three groups of maternal serum levels of MMP-9 and of NGAL were detected by enzyme-linked immunosorbent assay.RT-PCR was used to test placenta MMP-9 mRNA,and NGAL mRNA level.Results Preeclampsia maternal serum levels of MMP-9 and NGAL level were significantly higher than those of severe preeclampsia pregnant women serum level of MMP-9 and NGAL were significantly higher than mild preeclampsia (t =3.127,2.846,3.508,3.284,all P < 0.05).Preeclampsia placentas of MMP-9 mRNA and NGAL mRNA levels were significantly lower than the control group (t =-2.842,-2.728,-3.532,-3.276,(a)ll P < 0.05),severe preeclampsia placentas of MMP-9 and NGAL were significantly higher than mild preeclampsia(t =2.732,2.741,all P < 0.05),severe preeclampsia placentas of MMP-9 mRNA and NGAL mRNA levels were significantly lower than in patients with mild preeclampsia (t =-2.681,-2.587,all P < 0.05).Preeclampsia maternal serum levels of MMP-9 and NGAL was positively correlated with placental tissue mRNA levels of MMP-9 and NGAL mRNA level was positively correlated (P < 0.05).Conclusion Preeclampsia maternal serum MMP-9 and NGAL were significantly elevated,placental MMP-9 mRNA and NGAL mRNA levels were significantly decreased,suggested that they may be involved in the pathogenesis of preeclampsia.
5.An Observation of Experimental Autoimmune Uveitis in Guinea Pigs by Light and Electron Microscope

The soluble antigen was isolated from pig retinal extract by precipitation with ammonium sulfate followed by molecular sieve. 50 ?g S antigen with equal volume complete Freund's adjuvant was injected into double hind foodpads of guinea pigs. Two weeks later, experimental autoimmune uveitis developed. Its pathologic feature is that a great number of lymphocytes infiltrated into chroid which was thickened gradually from anterior to posterior. Inflammatory variation was relative to disease phase. Ultrastructure pathology was characterized by swelling and necrosis in the inner and outer segments of photorecepter cells. This result suggests that S antigen has significant effect on inducing experimental autoimmune uveitis and the outer layer of retina is the sensitive target organ of S antigen.

AIM Protective action of TMP on hearts and brains under the condition of the acute hypoxia was studied. METHODS On the device of mice acute hypoxia ,ability of tolerancing hypoxia of mice was investigated,and superoxide dismutase(SOD) and malondialdehyde(MDA) and nitric oxide(NO) contents in hearts and brains were measured.RESULTS TMP could make the resisting hypoxia action in mice strengthen significantly,and the action had a does dependent increase. TMP made SOD contents in hearts and brains in does dependent increase;MDA contents in hearts and brains were not notable in low does ,but TMP could decreased MDA contents in hearts and brains in middle and high does ( P
7.Therapeutic Observation of Xing Nao An Shen Jie Yu Needling for Depression After Cerebral Stroke

Objective To observe the clinical efficacy of Xing Nao An Shen Jie Yu needling method (brain-awakening mind-calming and anti-depression needling method) in treating depression after cerebral stroke. Method Totally 120 patients with depression after cerebral stroke were randomized into two groups: the acupuncture group was intervened by An Shen Xing Nao Jie Yu needling method, while the Western medication group was by oral administration of Fluoxetine Hydrochloride capsules. The Hamilton Depression Scale (HAMD), National Institute of Health Stroke Scale (NIHSS), and Barthel Index (BI) were adopted to evaluate the depression degree, neural function deficit, and activities of daily living after 4-week and 8-week treatments. Result The total effective rate was 96.5% in the acupuncture group, versus 83.9% in the Western medication group, and the difference was statistically significant (P<0.05); the acupuncture group was superior to the Western medication group in improving the HAMD, NIHSS, and BI scores (P<0.01), and its therapeutic efficacy present with a feature of time-effect. Conclusion Xing Nao An Shen Jie Yu needling can significantly improve the symptoms of depression after cerebral stroke, promoting the recovery of neural function and enhancing the quality of life.
8.Biological characteristics of G666-1 nasopharyngeal cancer stem cells in Survivin gene silencing

BACKGROUND:Survivin gene is the most potent anti-apoptosis factor, which can inhibit tumor cel proliferation and migration. It has become a hot topic related to cancer research in recent years.
OBJECTIVE:To explore the effect of Survivin gene silencing on the biological characteristics of G666-1 nasopharyngeal carcinoma cel s.
METHODS:siRNA sequence of Survivin was designed and synthesized and then transfected into G666-1 cel s using LipofectamineTM 2000. After Survivin gene silencing, Survivin mRNA and protein expressions were detected by RT-PCR and western blot assay, respectively. Expression of CD44+CD133+cel s was detected by flow cytometry. In addition, the effects of Survivin gene silencing on the proliferation, migration and invasion of G666-1 cel s were evaluated through cel counting kit-8 test, scratch healing assay and cel invasion assay.
RESULTS AND CONCLUSION:After Survivin gene silencing, the expressions of Survivin mRNA and protein were significantly reduced (P<0.05);the growth of G666-1 cel s was slowed dramatical y, indicating the proliferation ability was inhibited, and the migration and invasion abilities were both decreased significantly (P<0.05);the expression ratio of CD44+CD133+cel s was significantly lowered (P<0.05). These findings indicate that the proliferation, migration and invasion of G666-1 nasopharyngeal cancer stem cel s can be suppressed after Survivin gene silencing, and the proportion of CD44+CD133+cel s is reduced, thus indicating Survivin gene silencing may be a new effective target for treatment of nasopharyngeal carcinoma.
10.Affinity Selection of Human ScFv Against Botulinum Neurotoxin Serotype A

In this work, rare specific binders to botulinum neurotoxin antigen were selected from human phage immunized library through three rounds of panning in vitro. Of them, clone ScFvB17 is of high affinity and specificity to target antigen. B17 is of 750bp encoding 250 amino acid. Analysis results of gene structure showed V-genes of B17 belonging respectively to VH4 or? chainⅡ family are new antibody Fv genes. More important result showed that recombinant ScFvB17 expressed in E.coli could bind specific for neurotoxin competing with antiserum. Successful selection of human phage antibody library and obtaining specific ScFv against botulinum neurotoxin serotype A provide useful materials for further study on detection of neurotoxin and human engineering therapeutic antitoxin.
