5.Tetracycline inhibits Staphylococcus aureus growth in bacterial prostatic tissues of rats
Basic & Clinical Medicine 2006;0(09):-
Objective To probe the methods of experimental research for the drug activities of the rat prostates. Methods With androgen and Staphylococcus aureus injected into the prostates, the bacterial prostatitis (BP) models and benign prostatic hyperplasia with bacterial prostatitis (BHP-BP) models were developed. The animals were fed with tetracycline and then the drug activities in the serum and the prostatic tissues, the quantities of the bacteria and the histopathlogical changes in the prostateic tissues were separately detected in different time. ResultsAfter ten days, the histopathlogical changes of the benign hyperplasia and mesenchyma were found in the prostatic tissue from BHP animals and acute suppurative inflammation were dicegnosed in prostatic tissue from the BP group after infected for two days. The drug activities of 22.44 mg/g in BP group and 20.26 mg/g in BPH-BP group were found until 3.5~4 hours and the values were higher than MBC of the tetracycline and the antibacterial activity of the sera at the same time. After 8 to 11 days of treating, the bacteria would not be isolated from the prostates of the animals with BP or BHP-BP. Stopping treatment a week 11 days after treating acute or chronic inflammation histopathlogical change did not be found in the prostates of animals with BP. 30 CFU of bacteria could be found in per gram tissue of prostate and both of the acute and chronic suppurative inflammation could be found in the BP group without treatment at the 28 days afterinfected. Conclusion The tetracycline activities in the prostates of rats with BP or BPH-BPis same as or even higher than that of serum and the MBC of the drug. The quantity of sensitive bacteria in prostates will be diminished and disappeared during treatment.
6.Cloning of rat B cell lymphoma-2 gene and construction of its lentiviral vector
Medical Journal of Chinese People's Liberation Army 1982;0(01):-
Objective To clone the full-length cDNA of rat B cell lymphoma-2(bcl-2) gene, then construct and identify the cytomegavirus-mediated lentiviral expression vector of bcl-2 gene, and assess the gene expression in 293T cell, which is a human embryonic kidney cell line. Methods The full-length bcl-2-cDNA fragment was amplified by reverse transcription polymerase chain reaction(RT-PCR) from the kidney tissue of a Wistar rat.The double-stranded oligonucleotides(dsOligoe) were then cloned into the pMD18-T plasmid. After confirmation of a correct construction by sequencing, the positive clone was subcloned into pGC-FU vector with enhanced green fluorescent protein(EGFP),and then transformed into DH5a competent cells. The restricted endonuclease and T4 DNA ligase were used to construct the lentiviral expression vector plasmid pGC-FU-bcl-2 which, combined with the lentiviral packing materials(pHelper 1.0,pHelper 2.0),was then transfected into 293T cell line to form the recombinant lentivirus pGC-FU-bcl-2,and it was used to transfect the 293T cells. The expression of pGC-FU-bcl-2 was further verified by detecting EGFP and bcl-2.Results 1) It was verified by DNA sequencing that the sequence of rat bcl-2 gene was consistent with reported sequence in GenBank.2) The bcl-2 gene was successfully combined in pGC-FU-bcl-2 recombinant plasmid which could be transfected into human embryonic kidney cells.3) The recombinant virus pGC-FU-bcl-2 could be obtained from the 293T cells by co-transfection of pGC-FU-bcl-2 and packing plasmids.4) Targeting gene could be cloned into 293T cells by the recombinant lentivirus with steady expression. The fluorescent protein could be observed under microscope and the expression of bcl-2 protein was detected by Western blotting. Conclusions The lentiviral expression vector containing EGFP and bcl-2 gene has been successfully constructed, with which the transfected 293 T cells can lead to a steady expression of bcl-2 protein. The present study provides a basis for the further study of the function of bcl-2 gene and a potential therapy for the diseases relating to apoptosis.

8.The Important Role of Three Auxiliary Factors of Mammalian Aminoacyl-tRNA Synthetase Complex in The Cellular Signaling Network
Progress in Biochemistry and Biophysics 2006;0(04):-
Aminoacyl-tRNA synthetase catalyzing the first reaction of protein biosynthesis. In mammalian cells, eight aminoacyl-tRNA synthetases (aaRSs) and three auxiliary protein factors form a macromolecular aminoacyl-tRNA synthetases complex (aaRS complex). The three nonsynthetase protein factors, namely, p43, p38, and p18 were found to be involved in many other important life activities besides their roles in the complex. The auxiliary factor p43 was the precursor of endothelial monocyte activating polypeptideⅡ (EMAPⅡ), which involved in angiogenesis and apoptosis. The auxiliary factor p38 was crucial for the development of lung, and its abnormal accumulation in neuron would be related to the Parkinson’s disease. The auxiliary factor p38 and p18 could promote the repair of DNA damage via different pathways in a highly organized way. All these breakthroughs enhance our understanding about the interaction between the aaRS complex and the macromolecular signaling network and promote the studies on this field.
9.The effect of dental arch on dental panoramic radiography
China Medical Equipment 2014;(5):71-72,73
Through the analysis on principles of dental panoramic radiography, we analyzed the influence factors of the dental arch, and provided the main principles for optimizing design to improve the image quality. Through the analysis on the human dental arch’s form, we optimized the radiographic procedure. Many factors affect the quality panoramic image, but the focal trough’s design is one of the most important factors, especially for canine regions. Optimizing design of focal trough can lessen degrees of horizontal and vertical magnification, also can reduce geometric distortion and overlapped images of teeth.
