1.Relationship between ERCC2 single nucleotide polymorphisms and male idiopathic infertility in Ningxia.

OBJECTIVETo explore the influence of the DNA repair gene ERCC2 single nucleotide polymorphisms (SNPs) rs13181, rs1618536, and rs1799793 on male idiopathic infertility in Ningxia, China.
METHODSUsing MassArray, we conducted a case-control study and genotyped three ERCC2 SNPs rs13181, rs1618536, and rs1799793 for 351 males (aged 31.0 +/- 4.2 years) with idiopathic infertility and another 327 normal fertile men (aged 33.0 +/- 5.9 years) as controls.
RESULTSThe ERCC2 AnyG-anyA-anyA genotypes were significantly associated with an increased risk of idiopathic infertility (OR 0.414, 95% CI 0.176 - 0.970), while the three single ERCC2 SNPs rs13181, rs1618536, and rs1799793 showed no significant differences between the cases and controls (P > 0.05).
CONCLUSIONThe ERCC2 SNPs rs13181, rs1618536, and rs1799793 play a role of interaction in male idiopathic infertility in Ningxia, contributing to the risk of the disease.

2.Main Pathogenic Bacteria and Their Influencing Factors about Food-borne Diarrhea in Yunnan Province from 2012 to 2016

Objective To investigate the incidence and its main pathogenic bacteria infection status of food-borne diarrhea and analyze their influencing factors in Yunnan province from 2012 to 2016. Me thods 1743 cases of food-borne diarrhea were collected, which were supervised from 11 hospitals covering from 2012 to 2016 years in Yunnan province.We gathered and tested the biological samples. Meanwhile, we analyzed the main pathogenic bacteria and their influencing factors. Re s ults 65 positive strains samples were checked out in 1743 cases of food-borne diarrhea positive samples, the positive rate was 3.73% (65/1743). Salmonella and Shigella strains were the main pathogenic bacteria, the main suspect food was meat and its products, as well as fruits and their products. In this study, Professions, methods of processing and clinical diagnosis were considered as the main factors of main pathogenic microbe detectable rates of food-borne diarrhea cases in Yunnan province. Conclus ion We should carry out public health education widespreadly in nursery, scattered children, farmers and migrant workers, students and other special crowds, provide intervention measures, attach great importance to food processing, reduce the possible contamination of food during processing, improve the level of clinical diagnosis and treatment and the recognization ability of food-borne diseases, and propose targeted prevention and control measures.Thus, food-borne disease can be prevented and controlled effectively.
4.Plaster combined with splint for the treatment of gartland type III humeral supracondylar fractures in children.

OBJECTIVETo study clinical effects of plaster combined with splint for the treatment of Gartland type 1I humeral fractures.
METHODSFrom March 2002 to May 2006, 24 children with humeral supracondylar fractures of Gartland type ill were reviewed. Among the patients, 14 patients were male and 10 patients were female, ranging in age from 4 to 12 years, averaged 6.6 years. Ten patients had injuries in the left and other 14 patients had injuries in the right limb. Firstly, the patients were treated with manipulative reduction to maintain the length of humerus without emphasis on anatomic reduction. Then, the patients were treated with external fixation using plaster for 5 to 7 days, and secondary manipulative reduction after swelling disappeared. Lastly, the patients were treated with external fixation using splint for 4 to 5 weeks until fracture union.
RESULTSAll the patients were followed up, and the duration ranged from 5 months to 2 years, with an average of 1.2 years. All the patients had no complications such as neurovascular injury, myositis ossificans, forearm compartment syndrome and Volkmann contracture. According to ZHU Xiao-ting evaluation criteria for humeral supracondylar fractures in children, 12 patients got an excellent result, 8 good, 3 poor and 1 bad.
CONCLUSIONTreatment of child humeral supracondylar fractures with plaster and splint has several advantages such as avoiding serious soft tissue injuries around fractures due to many times reduction, fracture dynamic correction to obtain satisfactory reduction, reducing complications, obtaining good reduction, and restoring elbow function in a relatively short period of time.

5.Effects of partial portal vein arterialization on liver regeneration after hepatectomy in minipigs with obstructive jaundice.

BACKGROUNDHilar cholangiocarcinoma is a malignant tumor that is difficult to cure. The aim of this study was to observe the effects of flow-controlled partial portal vein arterializations (PPVA) on liver regeneration after hepatectomy in minipigs with chronic obstructive jaundice.
METHODSEight minipigs were made into chronic obstructive jaundice models. United semi-hepatectomy, which imitates extended radical surgery for treatment of hilar cholangiocarcinoma, was then performed. The eight minipigs were randomly divided into groups A and B (n = 4 minipigs each). PPVA was performed in Group A but not in Group B. The effects of flow-controlled PPVA on live regeneration after hepatectomy were observed for 30 days after hepatectomy.
RESULTSThe portal vein PO(2) at the immediate time point and on postoperative day 30 was higher in Group A ((47.33 ± 2.43) and (48.50 ± 4.44) mmHg) than in Group B ((35.38 ± 4.06) and (35.55 ± 2.55) mmHg respectively, all P < 0.01). The mitotic index of liver cells on postoperative days 14 and 21 was higher in Group A (12.55% ± 2.85% and 15.25% ± 1.99% respectively) than in Group B (6.85% ± 2.10% and 11.88% ± 1.15% respectively, all P < 0.05). The regeneration rate of residual liver on postoperative days 14 and 21 was higher in Group A (24.56% ± 6.15% and 70.63% ± 9.83% respectively) than in Group B (11.96% ± 5.43% and 44.92% ± 7.42% respectively, P < 0.05 and P < 0.01 respectively).
CONCLUSIONFlow-controlled PPVA can promote liver regeneration after hepatectomy and prevent liver failure in minipigs with chronic obstructive jaundice.

6.Analysis on Foodborne Disease Outbreaks in Yunnan Province From 2013 to 2017

Objective To provide a besis of the control measures for foodborne disease outbreaks by analysing the laws and the popular features of foodlorne disease outbreaks in yunnan from 2013 to 2017.Methods The data of epidemic situation and field investigation of foodborne disease outbreaks from 2013 to 2017 in Yunnan province was analyzed. Results From 2013 to 2017,a total of 2 519 cases of foodborne disease outbreaks were repeatedin Yunnan Province,Among 18681 cases, 257 patients died with mortality 1.38% . Foodborne disease outbreaks occur in the third quarters.Outbreaking and outbreaking from a few number of households, followed by the work unit Employer Unit; outbreaking deaths in the family is also the most. Primary causal factor is plant. Pathogen the pathogenic bacteria main pathogen are Salmonella (38.89%), Bacillus cereus intoxication bacillus (38.89%),staphylococcus aureus (11.11%), Diarrheagenic escherichia coli (5.56%), Proteus Proteus species Proteus vulgaris (5.56%),et al. Conclusion From 2013 to 2017,foodborne disease outbreaking in yunnan is mainly due to plants and micro-organisms and mosty oceurs at home and canteens. It is necessary to strengthen the food hygiene management at home and canteens,to avoid foodborne disease outbreaking incidents.
7.Unrelated cord blood stem cell transplantation for high-risk/refractory childhood acute myeloid leukemia: a clinical analysis of 160 cases

Objective:To retrospectively analyze the clinical outcomes of single unrelated cord blood transplantation (UCBT) in children with high risk and refractory acute myeloid leukemia (AML) .Methods:Between June 2008 and December 2018, a total of 160 consecutive pediatric patients with AML received single UCBT (excluding acute promyelocytic leukemia) . Myeloablative conditioning (MAC) regimen were applied. All patients received a combination of cyclosporine A (CsA) and mycophenolate mofetil (MMF) for the prophylaxis of graft -versus- host disease (GVHD) .Results:The cumulative incidence of neutrophil cells engraftment at day +42 and platelet recovery at day +120 was 95.0% (95% CI 90.0%-97.5%) at a median of 16 days after transplantation (range, 11-38 days) and 85.5% (95% CI 83.3%-93.4%) with a median time to recovery of 35 days (range, 13-158) , respectively. Incidence of grades Ⅱ-Ⅳ and Ⅲ-Ⅳ acute GVHD and chronic GVHD were 37.3% (95%CI 29.3%-45.2%) , 27.3% (95% CI 20.0%-35.0%) and 22.4% (95% CI 15.5%-28.7%) , respectively. The transplant-related mortality (TRM) at 360 day was 13.1% (95% CI 8.4%-18.9%) . The 5-year cumulative incidence of relapse was 13.8% (95% CI 8.5%-20.3%) . The 5-year disease-free survival (DFS) and overall survival (OS) were 71.7% (95% CI 62.7%-77.8%) and 72.2% (95% CI 64.1%-78.7%) , respectively. The 5-year GVHD and relapse free survival (GRFS) was 56.1% (95% CI 46.1%-64.9%) . The 5-year cumulative recurrence rates of CR1, CR2, and NR groups were 5.3%, 19.9%, and 30.9% ( P=0.001) , and the 5-year OS rates were 79.9% (95% CI 70.3%-86.7%) , 71.1% (95% CI 50.4%-84.4%) and 52.9% (95% CI 33.0%-69.3%) ( χ2=7.552, P=0.020) , respectively. Conclusions:For pediatric patients with high risk and refractory AML, UCBT is a safe and effective treatment option, and it is favorable to improve the survival rate in CR1 stage.
8.Berberine Inhibits Atherosclerosis by Regulating Lipophagy Via Targeting Wnt5a/NPC1 Signaling Pathway

ObjectiveTo investigate the regulatory effect and molecular mechanism of berberine (BBR) on lipophagy in the prevention and treatment of atherosclerotic (AS) lesions in mice. MethodFifty apolipoprotein E-knockout (ApoE-/-) mice were randomly divided into an AS model group, an atorvastatin group (5 mg·kg-1), and low-, medium-, and high-dose BBR groups (2.5, 5, 10 mg·kg-1). Ten C57BL/6J mice were assigned to the control group. After 12 weeks, hematoxylin-eosin (HE) and oil red O staining were performed to assess the histopathological changes of AS plaques in the aorta. Biochemical analysis was used to measure serum lipid levels, and enzyme-linked immunosorbent assay (ELISA) was employed to measure the levels of inflammatory cytokines interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α), oxidative stress marker reactive oxygen species (ROS), and serum lipophagy marker Beclin1 and microtubule-associated protein 1 light chain 3 Ⅱ (LC3Ⅱ). The xanthine oxidase method was used to measure serum superoxide dismutase (SOD) activity. Immunohistochemistry (IHC) was used to detect the distribution of wingless-type MMTV integration site family member 5a (Wnt5a) and Nieman Pick type C1 (NPC1) in the aorta, and Western blot was used to determine the protein expression of Wnt5a and NPC1 in the aorta. ResultCompared with the control group, the AS model group showed significant AS plaque formation, significantly elevated levels of serum total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), IL-6, TNF-α, and ROS, aortic Wnt5a distribution and protein expression (P<0.01), and significantly reduced levels of serum high-density lipoprotein cholesterol (HDL-C), SOD, Beclin1, LC3Ⅱ, and aortic NPC1 distribution and protein expression (P<0.01). Compared with the AS model group, the atorvastatin group, and high- and medium-dose BBR groups showed a significant reduction in AS plaque area (P<0.05, P<0.01), significantly decreased levels of serum TC, TG, LDL-C, IL-6, TNF-α, ROS, and aortic Wnt5a distribution and protein expression (P<0.05, P<0.01), and significantly increased levels of serum HDL-C, SOD, Beclin1, LC3Ⅱ, and aortic NPC1 distribution and protein expression (P<0.05, P<0.01). There was no statistically significant difference in the above indicators between the atorvastatin group and the medium-dose BBR group. ConclusionBBR can competitively bind to Wnt5a to activate NPC1 expression, upregulate lipophagy levels, reduce blood lipids, and inhibit the release of inflammatory mediators and oxidative stress damage, thereby exerting a preventive and therapeutic effect on AS.

Objective: To investigate the effects of the active ingredients combined therapy on inflammatory factors interleukin 1 beta (IL-1β) and neuropeptide Y (NPY) based on pharmacodynamics in rats. Methods: The animal model was built by transient middle cerebral artery occlusion (MCAO). The method for evaluating the concentrations of the FA-Pr-Al components in rat plasma was established by using HPLC and the expression levels of IL-1β and NPY were determined by ELISA. A new mathematics method of the trend of percentage rate of change (PRC) was used to assess the correlation between pharmacokinetics (PK) and pharmacodynamics (PD). Results: FA-Pr-Al in combination reduced neurological deficits, decreased infarct volume and inhibited the expression levels of IL-1β and NPY (all P<0.05) compared with the model group. FA, Pr and Al all displayed two compartment open models in rats. Clockwise hysteresis loops were obtained by time-concentration-effect curves.
