1.Development of the General Module for the System of Quality of Life Instruments for Patients with Chronic Disease: Items selection and structure of the general module.

Objective: To develop a general module of self-administered quality of life instrument for Chinese patients with chronic disease. Methods: Using the structured group (nominal group and focus group) methods and the qualitative and quantitative theory and methodology in developing rating scales Items were preliminary screened, evaluated and modified, and the QoL data measuring from 201 cases of patients of 8 different chronic diseases were analyzed by the methods of coefficient of variation?factor analysis, clustering analysis and correlation analysis. Result: General module of quality of life instrument for patients with chronic disease (QLICD-GM) was developed and evaluated. It consists of three domains: physical function, psychological function and social function, 32 items. Conclusions: QLICD-GM possessed a reasonable good content validity for it reflect WHO 's definition and connotation of QoL, and also common issues of patients with chronic disease.
2.Identification of bacterial strain ge15 and its controlling effect on ginseng diseases.

Based on previous results of 16S rDNA sequence homologuous and results of physic-biochemical indexes and morphological characteristics in the present work, bacterial strain ge15 isolated from roots of ginseng plants was identified as Stenotrophomonas rhizophila. Confronting incubation results showed that, strain ge15 inhibited the growth of Alternaria panax, Phytophthora cactorum, and Cylindrocapon destructans significantly, and the width of inhibition zone was 13.3, 24.0, 12.0 mm, respectively. Further results showed that the emergence rate and seedling survive rate of ge15 treatment was significantly higher than those of the control, and which was similar to pesticide carbendazol treatment. The ge15 strain has good application potential in ginseng diseases control without contamination.

4.Correlation between hyperalgesia and nuclear factor-kappa B expression in spinal dorsal cord of rats with complete Freurd's adjuvant arthritis

BACKGROUND: Nuclear factor-kappa B (NF-κB), as a promoter of inflammatory reaction, stimulates injured parts or transcription of local inflammatory gene, promotes generation of inflammatory factors, and induces pain onset; however, the mechanism on chronic inflammatory pained spinal cord has been less reported.OBJECTIVE: To explore the NF-κB expression in spinal dorsal horn and behavioral hyperalgesia by preparing rat models of complete Freurd's adjuvant arthritis.METHODS: A total of 24 SD rats were randomly divides into sham-surgery group and complete Freund's adjuvant group, with 12 rats in each group. Adjuvant arthritis model was produced by injection of 50 pL complete Fraund's adjuvant (CFA) to the right ankle joint after anesthesia. The same volume saline was injected to the rat right ankle joint in sham-surgery group. The mechanical pain threshold, paw withdrawal thermal latency (PWTL), the diameter of ankle, and NF-kB expression in spinal dorsal horn were investigated 2 days before and 4, 7, 14, 21, and 28 days after CFA injection.thermal symptoms were not obvious. The inflamed symptoms significantly appeared on right ankle joint and developed to food to before injection and sham-surgery group, the mechanical pain threshold was significantly decreased at 4 days after CFA injection, and reached the lowest value at 21 days (P < 0.01). The PWTL was significantly decreased at 4 days after CFA injection significantly increased in Ⅰ-Ⅵ in spinal dorsal horn in the complete Fraund's adjuvant group, which was higher than sham-surgery group (P < 0.01). The results indicated that we could gain stable monoarthritis model by injecting CFA with oil-contained water intorat ankle joint space, and the model shown prolong and significant hyperalgesia to radial thermal and mechanical pressure;meanwhile, the NF-kB expression increased significantly in lamber Ⅰ-Ⅵ in spinal dorsal horn after the ankle joint arthritis.
5.The changes of ultrastructural microcirculation of small intestine in canines with portal hypertension

Objective To examine the changes of ultrastructural microcirculation of small intestines of portal hypertension (PHT) canines. Methods PHT canine models were established by coarcating a half main portal vein with silk line chronic emboliztion. The ultrastructural changes of small intestine epithelium, mucous membrane and submucosa microcirculation were examined. Results The characteristics of ultrastructural changes of small intestine epithelium, mucous membrane and submucosa microcirculation were as follows: the number of blood vessels was increased and the diameter of them was expanded significantly; the lumen of arteriole was decreased, and the wall was thickened; arteriole collagen fibers were hyperplastic and confused; the lumen of venule was increased, the wall was thinned; basement membrane was damaged; microcirculatary endothelial cell was damaged generally; leukocytes was infiltrated; epithelial cells and basement membrane of intestinal mucosa were damaged; smooth muscle cell nucleus of ileum were deformed. Conclusion Small intestine epithelium, mucous membrane and submucosa microcirculatary ultrastructral showed obvious changes in PHT canines.
6.Debridement and suturing combined with double vacuum sealing drainage technology for treatment of pressure ulcer

Objective To explore the clinical efficacy of ebridement and suturing combined with double vacuum sealing drainage (VSD) technology for the treatment of pressure ulcer. Methods Totally 32 patients of pressure ulcer (from July 2011 to October 2015) re-ceived debridement and suturing after infection control. Then F14 silicone ventricular drainage tube was placed in the wound for drainage while VSD was placed outside the wound. Both tubes inside and outside wound were connected to the central vacuum (20~40 kPa). Three days after the drainage,F14 silicone ventricular drainage tube was removed,and VSD material outside wound was replaced to continuous vacu-um treatment for another 3 to 4 days. Results All of 32 cases were healed up without hematoma,dead space and flap margin necrosis,inclu-ding 4 cases of poor healing,which were healed after active dressing. Conclusion Small area of the pressure ulcer can be closed up by de-bridement and suturing combined with double VSD technology,which is a simple operation with little injury and high clinical application value.
7.Factors Affecting the Clinical Probation Teaching and the Countermeasures

8.CT perfusion imaging study of perihematomal cerebral blood flow in experimental intracerebral hemorrhage

ObjectiveTo study the mechanism of hemodynamic alternation surrounding the hematoma in the acute stage of intracerebral hemorrhage (ICH) in rats.MethodsSeventy male Sprague Dawley rats were randomly divided into ICH group and sham operated group, which were microinjected with 40 μl fresh autologous blood or saline into the right caudatum respectively. The each group was divided into 7 subgroups at 1h,3h,6h,12h,24h,48h and 72h after the ICH. CT perfusion imaging in measurements of regional cerebral blood flow adjacent to hematomas was performed. The ratios of side to side were measured at the regions around the hematomas by personal computer aided mapping. So the parameters of regional cerebral blood flow(rCBF), regional cerebral blood volume(rCBV) and mean transit time(MTT) were calculated respectively.ResultsThe rCBF and rCBV adjacent to the hematomas were lower than those of the outer region pronouncedly. The alternation of rCBF around the hematoma were fluctuated, which reduced to the valley at 1h after ICH, and then gradually returned to the peaks at 6h and 24h after ICH. In the meantime, the rCBV around the hematoma reduced to the valley at 1h after ICH, and then gradually increased to the peak at 24h after ICH.ConclusionThe abnormal hemodynamic changes can be found in the perihematomal region after ICH. The alternation of rCBF around the hematomas are fluctuated, but the changes of rCBV remain continuous increase. The mass effect of hematoma, intracranial hypertension caused by the mass effect of hematoma, and the autoregulation of cerebral blood flow motivated by the initial depression of cerebral blood flow play a very important role in the changes of cerebral blood flow.
9.Migration and Survival of Rat Neural Stem Cells Marked by Green Fluorescent Protein after Transplanted to Injured Spinal Cord

Objective To investigate the migration and survival of neural stem cells(NSCs)in vivo.Methods NSCs cultured in vitro were transfected by lentiviral vectors expressing green fluorescent protein(GFP)to construct GFP-NSCs,then trans-seeded into lactide-co-glycolide(PLGA)scaffold and implanted into the injured site of T9 spinal cord in rat.One month after transplantation,the migration of NSCs in spinal cord was examined by fluorescence microscope,and the survival rate of NSCs was counted out.Results NSCs labeled GFP had strong expression of green fluorescence.One month after transplanting,part of NSCs expressing GFP could be seen in PLGA scaffolds and rostral,caudal spinal core.The survival rate counted out was 1.4911±0.0313%.Conclusion NSCs marked by GFP and transplanted to rat injured spinal cord could migrate into the spinal cord tissues and the minority of them could survive.
10.Dissolution testing combined with computer simulation technology to evaluate the bioequivalence of domestic amoxicillin capsule.

Re-evaluation of bioequivalence of generic drugs is one of the key research focus currently. As a means to ensure consistency of the therapeutic effectiveness of drug products, clinical bioequivalence has been widely accepted as a gold standard test. In vitro dissolution testing based on the theory of the BCS is the best alternative to in vivo bioequivalence study. In this article, the conventional dissolution method and flow-through cell method were used to investigate the dissolution profiles of domestic amoxicillin capsules in different dissolution media, and the absorption behavior of the drugs with different release rates (t85% = 15-180 min) in the gastrointestinal tract was predicted by Gastro Plus. The flow-through cell method was thought better to reflect the release characteristics in vivo, and amoxicillin capsules with regard to the release rates up to 45 min (t85% = 45 min) were having a satisfied bioequivalence with the oral solution according to the C(max) and AUC. Although two different dissolution profiles of domestic amoxicillin capsules were found by flow-through cell methods, prediction results revealed that domestic capsules were probably bioequivalent to each other.
