OBJECTIVETo explore the effect of Danshensu on the lipid metabolism of hyperlipidemic rats.
METHODSixty clean male SD rats were selected. Twelve of them were selected in the basic control group and fed with common foods, and the remaining rats were fed with the high-fat feeds. After the successful modeling, they were randomly divided into the high-fat control group and low dose (10 mg x kg(-1) x d(-1)), medium dose (20 mg x kg(-1) x d(-1)) and high dose (40 mg x kg(-1) x d(-1)) Danshensu (dissolved in saline) groups. Both of the two groups were abdominally injected with the same volume of normal saline once a day for consecutively 30 days. The serum TG, TC, HDL-C and liver ACC1, FAS, HMGR, CPT-I mRNA expressions were detected.
RESULT AND CONCLUSIONDanshensu could inhibit the LDL-C level, timely clear redundant cholesterol and effectively regulate the lipid metablism of hyperlipidemic rats by reducing the TC content, decrease the fatty acid by reducing the FAS mRNA expression, and reduce the synthesis levels of endogenous cholesterol by inhibit the HMGR mRNA expression.

Objective To compare the prophylactic effect of laminar closure between titanium miniplate and anchor fixation in open-door cervical laminoplasty. Methods Between January 2010 and December 2010,63 patients with cervical spondylotic myelopathy were treated by open-door laminoplasty.Of them,30 patients underwent laminoplasty by titanium miniplate fixation and 33 by anchor fixation.During follow-up,multi-detector CT was performed preoperatively,at 1 week and 6 months after surgery. At each level, the anteroposterior diameter (APD) of the spinal canal and opening angle (OA) were measured. And the spinal canal expansion rate are calculated.MRI was performed preoperatively and 1 year after surgery to evaluate the severity of cord compression. Results All incisions healed by first intention. The incidence of postoperative axial symptoms in miniplate fixation group and anchor fixation group were 33.3% (10/30) and 39.4% (13/33),respectively.The OA,APD,and the spinal canal expansion rate of patients in both groups improved significant postoperatively, but differing from miniplate fixation group. The OA,the APD and the spinal canal expansion rate in anchor fixation group after 6 months were reduced than one week after surgery, and the difference between the groups was statistically significant.Lamina close in two groups was not found.CT images at 6 months showed complete fusion of the hinge area by mature bone or callus in two groups,by cervical sagittal MRI assessment. The severity of spinal cord compression was improved after 1 year.Preoperative and 1 year after the surgery, the severity of spinal cord compression between the two groups showed no significant difference. The severity of spinal cord compression after 1 year in both groups were no more than three grade. Conclusion Open-door cervical laminoplasty by anchor fixation or titanium miniplate can effectively prevent the occurrence of postoperative lamina closure, which can help patients to do functional exercises early, but improvement of spinal cord compression has no significant difference between both of them. However, titanium miniplate fixation for maintenance of the expansive spinal canal is better.

AIM: To identify the novel nuclear export signal by analyzing the DNA sequences and detecting the cell localization of different adenovirus ElA - associated protein BS69 isoforms. METHODS:BS69 DNA sequences in Emsebl database were blasted and the sequence of amino acids was aligned with the typical nuclear export signal. Different BS69 isoform fragments were cloned into pcDNA3.1 vector and transfected into Cos7 cells. The BS69 localization was observed by immunostaining and the function was verified by Western blotting. RESULTS: A novel nuclear export signal was found in BS69 isoform 2 but not in isoform 1.The isoform 2 was localized in cytoplasm and isoform 1 in nucleus, which was also consistent with the DNA sequence. The isoform 2 was involved in LMP1 - activated JNK phosphorylation whereas the isoform 1 was not. CONCLUSION: Different BS69 isoforms have different cellular localization. BS69 isoform 2 is localized in cytoplasm, interacting with Epstein - Barr virus latent membrane protein 1 and may be involved in nasopharyngeal carcinoma development. However, the isoform 1 is localized in nucleus and plays important roles in transcription.

Objective To study the effect of heat stress on the permeability, cytoskeleton and cell cycle of human skeletal muscle cell (HSKMC).Methods The HSKMC membrane permeability was detected by calcium ion inflow with flow cytometer, the cytoskeleton was stained by CBB 250,and the cell cycle was determined by flow cytometer. Results After 1 h of heat stress on human HSKMC cells under different temperature gradient, the median level of calcium ion was 91.63 in 43 ℃ heat stress group compared with 22.98 in 37 ℃ control group. As temperature increased, thicker and shorter cytoskeleton and stress fiber were shown under the high power lens of microscope. The DNA expression of skeleton cells at G0/G1 stage was 44.13-62.98 in groups under heat stress.Compared with normal control group,DNA expression was much higher in heat stress group, when HSKMC was cultured under 37 ℃ temperature for another 18 h,it kept decreasing DNA expression to a similar level as control group. Conclusions Heat stress can cause calcium iron inflow resulting in intracellular calcium overload, and affect the cytoskeleton leading to loss of normal web ordered arrangement and increased gap in HSKMC cells, which give rise to blocking cell cycle into G0/G1 stage.
