1.The application of combined detection of autoantibodies in primary Sj(o)gren's syndrome

Objective To compare the specificity and sensitivity of antibodies against SSA and SSB,anti-M3 receptor polypeptide antibodies,anti-α-fodrin(IgG)antibodies in primary SjSgren's syndrome (pSS).Methods One hundred and ten pSS patients(mean age was 49.2±14.8.mean disease duration was 5.6±4.6),80 systemic lupus erythmatosis(SLE)patients(mean age was 25.5 4-4.6,mean disease duration was 2.5±1.2)and 80 rheumatoid arthritis(RA)patients(mean age was 44.6±3.5.mean disease duration was 4.2±1.1)were studied.Enzyme-linked immunosorbent assay was used to measure these antibodies.Results The seropositive rates of anti-SSA,anti-SSB antibodies,anti-M3 receptor polypeptide antibodies and anti-α-fodrin(IgG)antibodies were 45.5%,30.9%,78.2%and 77.3%,respectivelv in pSS.They were much higher than those in RA and SLE patients(P<0.05).Specificities of SSA、SSB antibodies,anti-M3 receptor antibodies and anti-α-fodrin IgG were 83.8%,97.7%.92.0%and 90.O% respectively.With the combination of these antibodies in the diagnosis of pSS.the Sellsitivity can be increased at least to 88.2%and the specificity was not decreased significantly.Conclusion Combination of these antibodies can significantly improve the sensitivity of these antibodies in the diagnosis of pSS.Anti-SSA and SSB antibodies,anti-M3 receptor antibodies and anti-α-fodrin(IgG)antibodies are specific antibodies for the diagnosis of pSS.

Aquaporins (AQP), abundant in diversity, are the member of the major intrinsic proteins (MIPs) family with extraordinary ability to transport water, and thus, play important roles in modulation of water relations in plants. The recent advances in classification, structural characteristics, and physiological functions during plant growth and development as well as the various regulation modes of the activity and the effects of several abiotic stresses (water stress and salt stress) and phytohormones (ABA, GA and ethylene) on the gene expression of AQP were reviewed.
4.Pathogens Isolated from Biles of Cholelithiasis Patients:Analysis of Their Distribution and Drug Resistance

OBJECTIVE To survey the distribution of the pathogenic microorganisms in biles collected from cholelithiasis patients and their susceptibility to antibiotics,to guide the selection of reasonable antibiotics.METHODS Totally 546 bile samples were cultured and tested for antibiotics susceptibility.RESULTS Bacteria were cultured from 332 samples with a positive rate of 60.81%,the first three were Escherichia coli(32.96%),Enterococcus faecalis(12.74%),and Klebsiella pneumoniae(9.97%).The pathogenic microorganisms were more sensitive to the third and fourth generation cephalosporins,antibiotic/?lactamase inhibitor combination,carbopenems and aminoglycosides antibiotics,while less sensitive to the first and second generation cephalosporins,penicillins,macrolide antibiotics and fluoroquinolones.CONCLUSIONS The pathogenic microorganisms in bile are very various,E.coli,E.faecalis and K.pneumoniae are usually cultured,fungous and anaerobic infections cannot be ignored.Doctors should pay more attention to analyze the bacterial resistance profile and use the antibiotics properly.
5.The value of anti-M3 receptor polypeptide antibody in the diagnosis of sj(o)gren's syndrome

Objective To establish the enzyme-linked immunosorbent assay (ELISA)method in detecting the anti-M3RP antibody in Sj(o)gren's syndrome (SS) patients.and to explore the significance of this autoantibody in the diagnosis of SS.Methods The synthesized M3 receptor polypeptide was used as antigen in EUSA to detect the anti-M3RP antibody in sera of patients with SS.other CTDS and healthy controls.and the association between the clinical features of SS and anti-M3RP antibody was analyzed.Results Antibodies against M3RP were detected in 84.6%patients with pSS and 81-3% with sSS.8.8%with other CTD and 1%in heahhy controls.The positive rates of antibodies in pSS and sSS were higher than those in other CTDs and healthy controls.The presence of anti-M3RP antibodies had no significant correlation with clinical manifesta-tions and internal organ involvement.Furthermore.the positive rates of anti-M3RP antibodies in anti-α-fodrin.SSA,SSB,and ANA antibodies negative SS patients were 85%.89-3%,88.9%and 95.2%,respectively.body is a complementary parameter in the diagnosis of antibody-negative SS.
6.Protective effects and machanisms of pharmacological preconditioning induced by sodium ferulate on isolated rat heart

Aim To study the protective effects of pharmacological preconditioning induced by sodiumferulate(SF)on isolated rat heart and its machanisms.Methods Isolated SD rat hearts were divided randomly into 5 groups : Control group (hearts were perfused by oxygenated perfusate for 100 minutes); Ischemia/Reperfusion(I/R) group (hearts suffered from 40 min global ischemia/30 min reperfusion after oxygenated perfusate for 30 minutes); Ischemia preconditioning group(hearts were preconditioned by 3 periods of 5-minute global ischemia/5-minute reperfusion before it suffered from I/R); SF group (previously hearts were perfused with oxygenated perfusate administered with 1.69 mmol?L-1 SF then subjected to I/R); Glibenclamide group (previously hearts were perfused with oxygenated perfusate administered with 1.69 mmol?L-1SF and 30 ?mol?L-1 Glibenclamide before it suffered from I/R). Results Compared with I/R group, 1.69 mmol?L-1 SF precondition improved significantly heart function, reduced the incidence and severity of ventricular arrhythmias, alleviated calcium overload in myocardial cell; furthered activities of Na+,K-ATPase and Ca2+-ATPase in myocardium. These effects were attenuated by 30 ?mol?L-1 Glibenclamide.Conclusions SF precondition can improve heart function and resist arrhythmia and Ca2+-overload. The cardioprotective effects of SF precondition maybe related with the opening of ATP-sensitive potassium (KATP) channels.
7.The Effect of the Simvastatin Administration on the Expression of Connective Tissue Growth Factor in Fibrotic Lungs of Rats

Objective To explore the effect of the simvastatin administration on the expression of connective tissue growth factor (CTGF) in fihrotic lungs of rats. Methods The cats were treated with single intratracheal instillation of bleomycin (BLM) or instillation of the same volume of normal saline (NS) as a control. The administration of simvastatin(20 mg/kg)began once a day immediately or 7 days later after intratracheal BLM instillation respectively with the same volume of NS was given as a vehicle control. The rats were killed on day 7,14 and 28 respectively. Pathological alteration of lung tissue was observed hy HE staining and Masson staining. Hydroxyproline(HYP)content in lung tissue was used to determine the severity of pulmonary fibrosis. The expression of CTGF in lung tissue was exanfined by immunohistochem- istry staining and photodeusitometry. Results Histopathological changes of pulmonary fibrosis emerged gradually after the instillation of BLM. The expression level of CTGF was increased in lungs of rats after intratracheal instillation of BLM, compared with the control. The administration of simvastatin immediately or 7 days after intratracheal instillation of BLM, attenuated the histopathological changes of bleomycin- induced puhnonary fibrosis and prevented the increased expression of CTGF in lung tissue on day 28. Conclusion The adntinistration of simvastatin, immediately or 7 days after intratracheal BLM instillation, prevented the up-regulation of CTGF in fibrotic lungs of rats, which ntight be one of the mechanisms of the anti-fihrosis of simvastatin in lungs.
8.The study on CASP 8-652 6N I/D polymorphism and susceptibility of 2 type diabetes mellitus

Objective To investigate the association of cysteine aspartic acid specific protease 8 (CASP 8) gene-652 6N Insertion/Deletion polymorphisms and susceptibility of type 2 diabetes mellitus (T2DM). Methods CASP 8 gene -652 6N I/D polymorphisms were detected by polymerase chain reaction-restriction fragment length polymorphism and DNA sequencing in 414 controls and 410 patients with T2DM. Results I/I, I/D and D/D genotype frequency were 56.5%, 38.9%, 4.6%in controls and 58.0%, 32.9%, 9.0%in T2DM group respectively (P<0.05). The risk in D/D genotype people was 1.916 times than I/I genotype (adjusted OR=1.916, 95%CI=1.199~3.054, P<0.05). The fasting blood sugar of D/D genotype people was significantly higher than that of I/D and I/I genotype people (P<0.05). Conclusions CASP 8 gene-652 6N I/D polymorphisms are associated with T2DM outbreak.
9.Effects of genistein sodium sulfonate on liver function and liver tissueα7nAchR and IL-1βexpression in mice chronic hepatic injury

Objective To observe the protective effects of genistein sodium sulfonate(GSS)on mice chronic hepatic injury in-duced by carbon tetrachloride(CCl4 )and its influence on the protein expression of α7 nicotinic acetylcholine receptor(α7nAChR) and interleukin-1 beta (IL-1β)in liver tissue.Methods 60 SPF grade male Kunming mice were randomly divided into 5 groups,in-cluding the control group,model group,low and high doses GSS groups,and positive control group,12 cases in each group.Except for the control group,the other 4 groups were intra peritoneally injected by 10 % CCl4 with a volume of 0.1 mL/10 g for 6 weeks. The mice chronic liver injury was prepared.At the same time,the high and low doses DSS groups were given the different doses of GSS(0.30,0.10 mg/kg),the positive control group was given bifendate(DDB,2.5mg/kg),the control group and the model group were given the equal volume of normal saline for 6 consecutive weeks.The AST and ALT activity was detect and the ratio of ALT/AST was calculated;the Western blot method was used to detect the expression levels ofα7nAChR and IL-1βprotein in liver.Re-sults The serum levels of ALT and AST in the model group were increased obviously,and the expression level ofα7nAChR in the liver tissue was decreased,while the expression level of IL-1βwas increased;after the GSS treatment,the serum AST and ALT lev-els were significantly lower than those in the model group(P <0.05),while the expression level ofα7nAChR was increased (P <0.01)and the expression level of IL-1βwas decreased(P <0.05).Conclusion GSS might increase the expression ofα7nAChR in injured liver tissue,activates the cholinergic anti-inflammatory pathway,thus decreases the expression of inflammatory cytokines and antagonizes the mice chronic liver injury by inhibiting the inflammatory reaction.
10.Effect of previous analgesia of scalp acupuncture on post-operative epidural morphine analgesia in the patient of intestinal cancer.

OBJECTIVETo probe the effect of previous analgesia of scalp acupuncture.
METHODSSixty cases of radical operation of intestinal cancer were randomly divided into 2 groups: a scalp acupuncture plus epidural analgesia (scalp acupuncture group) and an epidural analgesia group, 30 cases in each group. The scalp acupuncture group received scalp acupuncture from 20 min before operation to the end of the operation, and epidural analgesia at the end of the operation. The epidural analgesia group only received epidural analgesia at the end of the operation. Recovery of VAS scores, BCS scores and gastrointestinal function after operation were investigated to probe into effects of scalp acupuncture on epidural morphine analgesia after operation in the patient of intestinal cancer.
RESULTSThe VAS scores at 6 h, 12 h, 24 h and 48 h in the scalp acupuncture group were lower than the epidural analgesia group, with significant differences at 6 h and 12 h between the two groups (P < 0.05). BCS scores at the 4 time points in the scalp acupuncture group were lower than the epidural analgesia group with significant differences between the two groups (all P < 0.05). The scalp acupuncture group in recovery of bowel sound, exsufflation from the anus in the scalp acupuncture group was faster than the epidural analgesia group (P < 0.01).
CONCLUSIONScalp acupuncture has a certain previous analgesic effect, reducing discomfort of the patient after operation and promoting the recovery of gastrointestinal functions.
