1.The effect of peroxisome proliferator-activated receptor gamma ligand on signaling pathway in pancreatic acinar calls

Objective To investigate the putative relationship between peroxisome proliferators activated receptor gamma (PPARγ) and nuclear factor (NF)-κB in cerulein-treated pancreatic aeinar AR42J cells. Methods The AR42J cells were allocated to control group, pioglitazone group (treated with 40 μmol/L of pioglitazone), pioglitazone + cerulein group (treated with 40 μmol/L of pioglitazone+ 10~(-8) mol/L of cerulein) and pioglitazone + cerulein + PPARγ antagonist (GW9662) group (treated with 40 μmol/L of pioglitazone + 5 μmol/L of GW9662 + 10~(-8) mol/L of cerulein). Activity of NF-κB and PPARγ expression were detected 30 minuts after stimulated by cerulein with or without the presence of pioglitazone. The protein expressions of NF-κB and PPARγ, antibody to IκBα phosphorylation, the differential expression between IκB kinase (IKK)β and IκBa, the IKKβ activity as well as changes of pIκBa were examined by Western blotting. The nuclear accumulation of NF-κB (p65 and p50 subunits) was determined by immunofluorescence and Western blotting. The interaction between NF-κB p65 and IκBα was observed by immunopreeitation. Results Treatment of AR42J cells with pioglitazone attenuated cerulein induced cytosolic activity of IKK protein (1.6 : 3.7)or IκBa phosphorylation (0.9 : 1.5), strengthened the integration of IκBα and NF-κB (0.8:0.3), inhibited transcription activity of p50 and p65 NF-κB dimer and nuclear accumulation (P<0.01). Adversely, the inhibitory effect of pioglitazone on NF-κB activity induced by cerulein was almost reversed by GW9662 (P<0.05). Conclusion These findings provide evidence for the involvement of PPARγ in the activity of NF-κB in cerulein treated AR42J cells.
2.The effects of pioglitazone on the process of oxidative stress in acute pancreatitis induced by cerulein

Objective To investigate the effects of pioglitazone,a peroxisome proliferation activated receptor(PPAR)γ agonist on oxidative stress process and the therapeutic effects on severity of acute pancreatitis(AP).Methods Thirty male Sprague-Dawley rats were randomly divided into five groups with 6 in each:control group,eerulein plus pioglitazone group,cerulein group,cerulein plus vehicle group,cerulein plus pioglitazone and GW9662 group.Rats were sacrificed at 30 min after the induction of pancreatitis.Pathologic changes of the pancreas were observed under light microscope.The ratios of pancreatic wet/body weight of rat were determined in each group.Serum amylase,pancreatic nitric oxide synthase(NOS),inducible nitric oxide synthase(iNOS),malondialdehyde(MDA),and myeloperoxidase (MPO)were determined by chromometry.Results The serum amylase,pancreatic wet/body weight and the score of pathologic damage increased after the induction of pancreatitis,AP samples were characterized by increased pancreatic MDA,MPO,NOS and iNOS(P<0.01).Pioglitazone(20 mg/kg and 40 mg/kg)exhibited a protective effect against oxygen free radicals reflected by lower serum amylase,less severe pancreatic lesions,normal pancreatic MDA,MPO and NOS levels(P<0.05).GW9662 reversed the effects against oxidative stress of pioglitazone(40 mg/kg)(P<0.05).Conclusions Pretreatment with pioglitazone may exert its therapeutic effect on AP by lowering pancreatic oxidative free radicals and reducing pancreatic tissue infiltration of neutrophils.
3.Research of correlative factors between the ultrasonography of trauma-caused deep venous free-floating ;thrombus of lower limbs and the prognosis

Objective To analyze the color Doppler ultrasonography of trauma-caused deep venous free-floating thrombus(FFT)of lower limbs,discuss the relations between sonography and the prognosis. Methods Clinical data and color Doppler ultrasound examination results of 130 cases with trauma-caused deep venous FFT of lower limbs were retrospectively analysed.Univariate analysis was conducted to identify the statistically significant factors of patients' prognosis,non-conditional logistic regression analysis was performed to analyse the independent prognostic factors of FFT.Results The non-conditional logistic regression analysis showed that the position of FFT,thrombosis edge definition,floating degree and the volume of detouring blood around the thrombus were independent factors of patients'prognosis(P <0.05). The FFT prognosis of iliofemoral vein was more likely to be fixed than the FFT prognosis of popliteal vein. The FFT prognosis with blurring edges was more likely to be fixed.The more obvious the floating degree of FFT,the higher chance that the FFT prognosis is recanalized.More detouring blood flow around FFT indicated more possibilities that the prognosis could be recanalized.Conclusions The color Doppler ultrasound is able to evaluate the relations between the deep venous free-floating thrombus of lower limbs and prognosis of the patients,which is beneficial for early detection and early treatment,as well as reducing mortality and disability resulted from deep venous thrombus during the orthopaedic perioperative period.
4.Hydrokinesi-therapy for the treatment of knee osteoarthritis and its application in community-based rehabilitation therapy

Objective The purpose of this article is to expound the treatment models of hydrokinesi-therapy and its effectiveness for the treatment of knee osteoarthritis, then to explore whether hydrokinesi-therapy is suitable for our country's community rehabilitation.The hydrokinesitherapy has been carried out abroad, which improve the function of the patients with knee osteoarthritis as far as possible using efficient resource of community.It has obtained good results, especially for the elderly who can't take exercise on the ground because of pain.Therefore, the hydrokinesi-therapy for the treatment of knee osteoarthritis can be applied to community-based rehabilitation therapy.The geological resources of our country is rich, the thermal spring water can be used to develop community rehabilitation for the treatment of knee osteoarthritis.
5.Development and application of ward adjustable pillow

An adjustable pillow with inflatable cushion and soft polyurethane sponge is introduced.It is not easily deformed and the height is adjustable.In addition,it is easy to carry and can be cleaned and disinfected thoroughly.Though the design is quite simple,this kind of pillow can improve the patients' sleep quality and prevent pollution of pillow as a result of iatrogenic infection.
6.Inhibition effects of sulforaphane-induced autophagy flux to cell proliferation on ex vivo human capsular bags

Background Sulforaphane (SFN) is an effective chemopreventive agent and can regulate the biological molecular mechanisms to inhibit the overgrowth of cells.Autophagy is a biological process of maintaining cellular internal environment.Understanding the affection of SFN to biological behavior of human lens epithelial cells (LECs) and the association of SFN with autophagy is helpful for the prevention and target treatment of posterior capsule opacification (PCO).Objective This study was to investigate the eradication effeccts of SFN on residual lens cell population in vitro posterior capsule opacification (PCO) model and evaluate the mechanism of SFN-induced cell death.Methods In vitro human capsular bag models were generated from fresh donor eyes by phacoemulsification and were cultured in EMEM containing 2% fetal bovine serum (FBS).Different concentrations of SFN (0,1,10 and 100 μ mol) were added in the medium for 30 days respectively according to grouping,and the growth of LECs was observed by optical microscope and immunofluorescence technique.FHL124,a human LEC line,was cultured with EMEM containing 5% FBS and divided into 0,1,10,30 and 100 μmol SFN groups.Lactate dehydrogenase (LDH) release rate in the medium was detected to evaluate cell damage/death.The migration of the cells on capsular bags was assessed by scratch test.The ultrastructure and number of autophagosomes were examined under the transmission electron microscope.The expression of LC3 in the cells were detected using Western blot in the presence or absence of autophagy inhibitors.Results The cell coverage rates on the capsular bags were significantly lower in the 10 and 100 μ mol/L SFN groups than those in the 0 and 1 μmol/L SFN groups,with a statistically significant difference among the groups (F =48.57,P < 0.01).Immunofluorescence showed that the density of F-actin-and Vimentin-positive cells was evidently decreased in the 10 and 100 μmol/L SFN groups compared with 0 and 1 μ mol/L SFN groups.The releasing levels of LDH (absorbancy) were 0.19± 0.03,0.39±0.06,0.56±0.07,0.68±0.08 and 0.89±0.09 in the 0,1,10,30 and 100 μ mol/L SFN groups,respectively,and the releasing level of LDH was gradually increased in the 10 and 100 μ mol/L SFN groups in comparison with the 1 μmol/LSFN group (all at P<0.01).With the increase of SFN concentration,the reduction rate of scratched area decreased with the increase of SFN concentration,and the decrease of scratch area was significantly lower than that of adjacent low mass concentration group and the differences were statistically significant (P<0.05).The relative expressions of LC3-Ⅱ protein were 0.423±0.003,14.543±0.024,0.668±0.024 and 0.576±0.056 in the blank control group,SFN group,SFN + 3-MA group and 3-MA group,respectively,and the relative expressions of LC3-Ⅱ protein were significantly lower in the SFN+3-MA group and 3-MA group than those in the SFN group (all at P<0.01).The number of autophagosomes was 4.07±0.32,4.13±0.34,9.21 ±0.53 and 21.02± 1.34 in the blank control group,and 1,10,100 μmol/L SFN groups,and the number of autophagosomes in the 10 and 100 μ mol/L SFN groups was significantly higher than that in the blank control group and 1 μmol/L SFN group (all at P<0.01).Conclusions SFN mediates LECs death by promoting autophagy in ex vivo capsular bags,and SFN may be a novel agent of potential chemopreventive and target treatment for PCO.

10.Survey of lower urinary tract symptoms in 2811 men in Wuhan City

Objective To assess the prevalence and presentation of low urinary tract symptoms (LUTS) in Chinese men. Methods A random sample was obtained from men dwelling at communities,factories,institutions and colleges in a district in Wuhan City.By completion of the questionnaire and exclusion of individuals with operation and trauma history,2811 men (age range,20~82 years) were assigned into 6 groups according to 10-year age stratum.Frequency of symptoms and severity of discomfort were assessed using the International Prostate Symptom Score (IPSS). Results The 2811 men accounted for 71.7% of the surveyed population. The occurrence of LUTS was as follows.In 20~29 year group,only 5 (0.6%) of 798 men had mild LUTS (IPSS,1~7).In 30~39 year group,55 (12.1%) of 453 men had mild LUTS.In 40~49 year group,143 (26.6%) of 537 men had LUTS;of them 118 were of mild LUTS,20 of moderate (IPSS,8~19),5 of severe (IPSS,20~35).In 50~59 year group,244 (52.5%) of 465 men had LUTS;of them 109 were of mild,117 of moderate,18 of severe.In 60~69 year group,240 (65.6%) of 366 men had LUTS;of them 78 were of mild,138 of moderate,24 of severe.In≥70 year group,162 (84.4%) of 192 men had LUTS;of them 47 were of mild,68 of moderate,47 of severe.Differences between age groups in incidence rates of LUTS were significant (?2=998.385,P
