Atomic force microscopy is a powerful new tool to investigate the structure and measure the relationship between structure of surfaces and functions in microorganisms. Comparing with conventional electron microscope, atomic force microscope has higher resolution and can image real-time structures from atomic to molecular scale in three-dimensional mode under physiological condition. Therefore, atomic force microscope is being used in almost every aspect of microbiology and has gotten many exciting findings.
2.Combind liver and hilar vascular resection for hilar cholangiocarcinoma

Objective To investigate the value of partial hepatectomy and vascular resection in the treatment of hilar cholangiocarcinoma. Methods Seventy four patients with hilar cholangiocarcinoma who underwent hepatectomy of Chinese People' s Liberation Army from January 2008 through December 2011 were analyzed retrospectively.Results Of the 74 patients,33 underwent radical resection and 19 palliative resection,22 received internal or external drainage.In the radical resection group,the median survival time was 27 months,and the overall survival rate at 1,2 and 3 years were 79%,64% and 49%.In the palliative resection group,the median survival time was 14 months and the overall survival rate at 1,2 and 3 years were 56%,25%,and 19%.In the drainage group,the median survival time was 9 months and the overall survival rate at 1,2 and 3 years were 23%,15%,0.Conclusions Hepatectomy combined with hilar vascular resection helps increase survival rate of patients in radical excision of hilar cholangiocarcinoma and Surgical resection is the most elective method for treatment of hepatic hilar cholangiocarcinoma,and the radical resection might improve the prognosis of the patients with hilar cholangiocarcinoma.
3.Efficacy and safety of laparoscopic radiofrequency ablation therapy for hepatocellular carcinoma

Objective To evaluate the feasibility,safety and efficacy of laparoscopic radiofrequency ablation (RFA) therapy for hepatocellular carcinoma (HCC).Methods Sixty-eight cases of liver cancer lesions were underwent laparoscopic radiofrequency ablation,and their postoperative recovery state,focal necrosis rate were observed.Results All the 68 cases were successfully performed operation,114 lesions were treated including 20 missed lesions at preoperative imaging diagnosis.There were no serious postoperative complications,the average hospital stay was (2.5 ± 1.2) days,focal necrosis rate 3 months after operation was 85.9%,lesion recurrence rate 6 months after operation was 12.2%,the 1-year survival rate was 76.47%.Conclusions Laparoscopic radiofrequency ablation in treatment of hepatocellular carcinoma has high security,few complications,short hospital stay and remarkable clinical effects.It's well worth clinical outreach.
4.Current situation and countermeasures of smoking cessation service system construction in China

Abstract
In order to achieve the goal of reducing the smoking rate among Chinese adults at ages of 15 years and above to below 20% by 2030, smoking cessation services require to be reinforced. The current Chinese smoking cessation service system includes short-term smoking cessation interventions and smoking cessation hotlines at the population level, and smoking cessation clinics at the individual level; however, these smoking cessation services suffer from problems of low awareness, low accessibility and low utilization rate. Based on the publications pertaining to smoking cessation services and released policy documents in China until June 2022, this review analyzes the current status, problems and causes of smoking cessation services, and proposes suggestions for improving the smoking cessation service system construction in China.
6.Ultrasmall superparamagnetic iron oxide-enhanced MRI in a rabbit model of antigen-induced arthritis:a preliminary study

Objective To investigate the feasibility of ultrasmall superparamagnetic iron oxide- enhanced(USPIO)-enhanced MR imaging for monitoring synovitis of antigen-induced arthritis in rabbit model and explore the optimal MR imaging sequences.Methods Nine female white rabbits with antigen(0.5 ml mBSA,2 mg/ml)induced arthritis of the right knees were used in the study.The left knees of these rabbits and both knees of another 3 rabbits served as the control.Nine to 28 days(mean 21.3 d)after successful model induction,all knees were imaged before and 24 h after intravenously injection of USPIO (0.3 ml/kg),among which 2 rabbits were also imaged at 48 and 72 h after administration of USPIO respectively.The MR protocol included spin-echo(SE) T_1WI,fast spin-echo(FSE)T_2WI,gradient echo (GRE)T_2~* WI and short tau inversion recovery(STIR).Images were analyzed quantitatively and qualitatively based on signal characteristics and patterns of the synovium.Paired t-test was used for the analysis of the signal intensity of inflammatory synovial membrane before and 24 h after injection of USPIO. MR findings were correlated with histopathology.Results Arthritis was successfully induced in all 9 right knees with intraarticular injection of mBSA.Pathological examination revealed hyperplasia of synovium with infiltration of USPIO-loaded-macrophages.MR depicted synovial thickening(thickness 2.07?0.97 mm) and joint effusion.Synovium and joint fluid appeared as slightly hypo- or iso-intense on T_1 WI and hyper- intense on T_2 WI or T_2~* WI.Twenty four hours after USPIO injection,significant T_1 enhancement(ASNR 41.91%?27.94%),negative T_2 and T_2~* enhancement(△SNR -34.92%?11.77% and -57.24%? 16.05%)were demonstrated in the region of synovial inflammation respectively.The signal at 48 h and 72 h changed less than that at hour 24.No signs of arthritis occurred in all left knees and in all knees of the artificial model group.Conclusion Iron oxide phagocytized into macrophages can be a root cause resulted in signal change on USPIO-enhanced MR images.The gradient echo sequence should be the optimal sequence to be used in USPIO-enhanced MR imaging in antigen-induced arthritis.
7.Effect of adenovirus-mediated TK/GCV gene therapy in combination with TNF-alpha against murine bladder cancer cells in vitro.

OBJECTIVETo investigate the cell-killing effect of adenovirus-mediated TK-ganciclovir (GCV) gene therapy in combination with tumor necrosis factor-alpha (TNF-alpha) against murine bladder carcinoma cells in vitro.
METHODSMurine bladder carcinoma MB49 cells were transfected with the adenoviral vector containing TK gene and green fluorescent protein (GFP) gene. The transfection efficiency was observed and the TK gene expression in the transfected cells was detected by RT-PCR. The survival rate of MB49 cells in response to TNF-alpha treatment and that of the TK gene-transfected cells after treatment with GCV and GCV+TNF-alpha were determined by MTT assay. The apoptosis of the cells after the treatments was analyzed by flow cytometry.
RESULTSIn cells transfected with TK gene, the cell inhibition rate increased gradually with the increment of GCV and TNF-alpha concentration. GCV in combination with TNF-alpha resulted in significantly increased killing efficiency of the cells as compared with GCV or TNF-alpha treatment alone, and the effect of the combined treatment was enhanced as the TNF-alpha concentration increased. GCV treatment (50 microg/ml) alone produced a cell killing rate of (24.39-/+1.10)%, and when combined with 5 microg/ml TNF-alpha, the rate was increased to (40.05-/+0.97) %, and further to (65.47-/+0.67) % when TNF-alpha concentration increased to 20 microg/ml. Flow cytometry revealed obvious apoptosis of the cells 8 h after treatments with TK/GCV, TNF-alpha, or TK/GCV+TNF-alpha, and the combined treatment resulted in the highest cell apoptotic rate.
CONCLUSIONTK/GCV in combination with TNF-alpha can enhance the effect of suicide gene therapy against murine bladder carcinoma cells and effectively induce apoptosis of the cells.

9.Correlation of MMP-9, GA, HbA1c, and adipokines levels with DR

·AIM: To investigate the correlation of matrix metalloproteinase -9 (MMP-9), glycated albumin (GA), glycosylated hemoglobin ( HbA1c ) and adipokines ( including visfatin, resistin and leptin ) with diabetic retinopathy ( DR) .·METHODS:From March 2015 to March 2017, 74 patients with DR were treated in our hospital, including 40 patients ( 80 eyes ) with non proliferative diabetic retinopathy ( NPDR ) and 34 patients ( 68 eyes ) with proliferative diabetic retinopathy ( PDR ) , and diabetes mellitus 40 patients ( 80 eyes ) with non DR ( NDR ) and 40 healthy volunteers (80 eyes) were selected as controls, the levels of MMP-9, GA, HbA1c, visfatin, resistin and leptin in each group were detected.·RESULTS: PDR group visfatin was 4. 41 ± 0. 82ng/mL, was significantly lower than the NPDR group, NDR group and control group ( P<0. 05 ) , while, resistin, leptin and MMP- 9 were 9. 01 ± 1. 04ng/mL, 17. 96 ± 2. 03μg/L and 740. 06 ± 84. 43μg/L, GA and HbA1c were 26. 14%± 4. 57%and 17. 60% ± 1. 91%, significantly higher than those of NPDR group, NDR group and control group ( P<0. 05 ) . NPDR group visfatin was 6. 44 ± 0. 79ng/mL, was significantly lower than that of NDR group and control group (P<0. 05), while, resistin, leptin and MMP-9 were 7. 80±0. 87ng/ml, 15. 68±1. 98μg/L and 634. 12±80. 22μg/L,GA and HbA1c were 22. 06%± 4. 38% and 12. 46%± 1. 69%, significantly higher than those of NDR group and control group (P<0. 05). MMP-9, GA, HbA1c were positively with DR levels ( rs = 0. 523, 0. 461 and 0. 414, P<0. 05 );visfatin was negatively correlated with DR levels ( rs = -0. 433, P < 0. 05 ), resistin and leptin were positively correlated with DR levels (rs=0. 401 and 0. 460, P<0. 05).·CONCLUSION: MMP-9, GA, HbA1c, and adipokines may play a role in the development and progression of DR, in which MMP-9 is associated with adipokines, both are not significantly related to the levels of GA and HbA1c.
10.Ranibizumab with photodynamic therapy in the treatment of wAMD

· AIM:To investigate the clinical effect of Ranibizumab combined with photodynamic therapy (PDT) for wet type age-related macular degeneration (wAMD) and the effect on the serum neovascularization factors.· METHODS:Totally 68 cases (68 eyes) of wAMD patients treated with PDT in our hospital from January 2014 to June 2016 were analyzed retrospectively.Among them,34 patients were treated by photodynamic therapy (control group) and 34 patients were treated by combination of photodynamic therapy and ranibizumab (treatment group).Comparison of BCVA,mean retinal thickness and central macular thickness (CMT) and serum neovascularization regulatory factors before and after treatment were taken between the two groups.· RESULTS:Before treatment,there was no significant difference on the BCVA,the average retinal thickness and the CMT value between the two groups (P>0.05).At 3,6 and 12mo after treatment,the BCVA,average retinal thickness and CMT in both groups were significantly lower than those before treatment (P<0.05).At 3,6 and 12mo after treatment,the BCVA,the average retinal thickness and the CMT value of the patients in the treatment group were significantly lower than those in the control group (P<0.05).There was no significant difference of vascular endothelial growth factor (VEGF),platelet derived growth factor (PDGF),matrix metallo-proteinase inhibitor (TIMP)-1 and endostatin (ES) between the two groups before treatment (P> 0.05).Three months after treatment,VEGF,PDGF and ES in both groups were significantly lower than those before treatment (P<0.05).Three months after treatment,the levels of VEGF,PDGF and ES in the treatment group were significantly lower than those in the control group (P<0.05).· CONCLUSION:The wAMD patients treated with ranibizumab combined with photodynamic therapy can achieve a more significant clinical effect,and more effectively reduce the level of serum neovascularization regulators.
