1.Effects of Andrographitis Paniculata extracts on the expression of CD40 in endothelial cells.

In order to investigate the expression of CD40 in endothelial cells (ECs) in a variety of injured conditions and the interventional role of Andrographitis Paniculata isolate (API(0134)), the thoracic aorta ECs of guinea pigs were cultured in vitro until the third passage, incubated in the presence of media containing xanthine oxidase (XO) and xanthine (Xan) which produced oxygen free radical (OFR group); oxidized-LDL (ox-LDL group); XO, Xan and API(0134) (OFR+API(0134) group); or ox-LDL and API(0134) (ox-LDL+API(0134) group). The expression of CD40 in ECs was detected by immunofluorescence assay and reverse transcription-PCR (RT-PCR). The results showed as compared with the control group, the expression of CD40 in ECs in OFR group and ox-LDL group was increased (P<0.01), but attenuated significantly in OFR+ API(0134) group and ox-LDL+API(0134) group (P<0.05). It was suggested that API(0134) could protect atherosclerosis by inhibiting the expression of CD40 molecule in injured ECs.

4.Effect of ultraviolet rays on the re-activation of the physicochemical properties and bioactivity of aging titanium surface

BACKGROUND:The physicochemical properties and bioactivity of titanium surface will degrade with time because of the aging phenomenon,affecting the efficiency of implant-bone osseointegration.Therefore,the re-activation of the implant surface physicochencical properties and bioactivity is important.OBJECTIVE:To investigate the re-activation effect of ultraviolet rays on aging titanium surface.METHODS:Sand-blasted and acid-etched titanium discs were divided into three groups randomly:Group A was new titanium surface,Group B was stored in a sealed container for 4 weeks,Group C was treated with ultraviolet rays for 15 minutes after stored in the sealed container for 4 weeks.The surface roughness,elemental composition and surface energy of the titanium surface were examined by optical profilometer,X-ray photoelectron spectroscopy and contact angle measuring device,respectively.The bioactivity of the titanium surface was examined by cell culture experiments.MG63 osteoblast-like cells were cultured on the different titanium surfaces.After 30 minutes,1,2,4 hours of incubation,the cells were strained with Hoechst33342 fluorescence,and initial attachment of cells was evaluated by measuring the amount of cells attached to the titanium surface.The proliferation of cells was quantified in terms of cell density at 1 and 3 culture days using tetrazolium salt (MTS)-based colorimetry.RESULTS AND CONCLUSION:There was no obvious difference in the surface roughness of the three groups,and ultraviolet treatment did not change the surface morphology of titanium surface.X-ray photoelectron spectroscopy showed that the C element content of group A and group C was lower than that of group B (P < 0.05),and the content of Ti,O,N elements was significantly higher than that of group B (P < 0.05).No difference in the surface element composition between group A and group C was found.Both of the surface contact angles of group A and group C were Oo,but in group B,the value was 115°.The number of initially attached cells of group A and group C was significantly higher than that of group B after incubated 30 minutes,1,2 and 4 hours (P < 0.05),and no difference between group A and group C was found.The proliferation of cells of group A and group C was significantly higher than that of group B after incubated 1 and 3 days (P < 0.05),but no difference between group A and group C was found.In conclusion,ultraviolet rays show good effects on the re-activation of the aging titanium surface,which can reduce hydrocarbon contamination and recover the high surface energy to increase cell adhesion and proliferation.
5.New model of biliary perfusion systemin vitro

ObjectiveTo establish a new model of biliary peffusion in vitro,and to explore the feasibility of comparative biliary studies with the model.MethodsA set of ten-text-area model of biliary peffusion was designed in vitro.Saline,glycerol and pig bile were put into this model.The outflow of the first tube was set at 8 drops/min,and these liquids were collected through the ten sample test areas and measured at 0.5 hour,1.0 hour,1.5 hours,2.0 hours,12.0 hours and 24.0 hours.Equality of liquid amount was evaluated from each sample test area.Rods containing delayed release EDTA of ten different levels were placed in the test areas.The liquid was collected and EDTA concentrations were evaluated from the ten tubes at 24.0 hours.EDTA concentrations of the same rod in the three liquids were compared.ResultsA(1) the same time point,the amounts of saline,glycerol,pig bile flowing through the ten sample test areas showed no significant difference (P > 0.01 ) ; but EDTA concentrations of the same liquid at 24.0 hours were significantly different (P < 0.01 ) ; and EDTA concentrations of the same rod in the three liquids were also obviously different ( P < 0.01 ).ConclusionThe same liquid flow rates and the same experimental environment can be achieved.Different test results can be obtained from different test areas in different experiments.The test results of the same kind of sample change with the changes of experimental conditions,which simulates biliary duct.
6.The value of BAEF in early diagnosis of acute bilirubin encephalopathy in newborn

Objective Acute bilirubin encephalopathy in neonates is the most serious complication of neonatal hyperbilirubinemia, is one of the main causes of neonatal death and disability. Clinical early diagnosis, early treatment can improve the prognosis in children. Methods Brainstem auditory evoked potential (BAEF) was detected on two patients (40 patients with ABE, 40 cases of normal controls, all full-term) in the state of sleep in children and analysis the difference between the two groups ,all testing was completed by experienced Department of ENT full-time technician in charge,SPSS15.0 statistical analysis software was took for data analysis (using rank sum test method). Results There was significant difference between the two groups of neonatal latency of wave I, latency of waveⅤ, interpeak time , acute bilirubinⅠ-Ⅴencephalopathy group was significantly longer than that of the control group. Conclusions The BAEF detection is the sensitive index of brainstem damage , can objectively and sensitively reflect the function of the central nervous system , can reflect the functional status of cochlear and brainstem structures , often brainstem was slightly damaged but no clinical symptoms and signs , BAEP has changed significantly , so the conventional BAEP examination performed on patients with hyperbilirubinemia help to find bilirubin brain damage as early as possible,and prevent the occurrence of bilirubin encephalopathy.
7.Retrograde transposition of composite tissue flap pedicled with peroneal artery to treat bone and soft tissue defects following calcaneal osteomyelitis

Objective To report retrograde transposition of composite tissue flap pedicled with peroneal artery in the treatment of bone and soft tissue defects following calcaneal osteomyelitis.Methods During the period from January 2008 through January 2015,47 patients with bone and soft tissue defects following calcaneal osteomyelitis were treated by retrograde transposition of composite tissue flap pedicled with peroneal artery.They were 43 men and 4 women,aged from 24 to 67 years(average,43.6 years).After radical focus debridement,the areas of skin defect ranged from 2 cm × 2 cm to 5 cm× 5 cm and the bone defects ranged from 1.5 cm × 1.0 cm × 2.0 cm to 3.0 cm × 3.0 cm × 4.5 cm.Results All the 47 patients were followed up for 6 to 72 months (average,25.2 months).Relapse of osteomyelitis occurred in 2 cases but secondary bone graft was needed in none.All the patients recovered their walking function.The texture and color of the composite tissue flaps were similar to those of normal skin,without obvious swelling.The flaps appeared good and imposed no obstacle to wearing shoes.The ankles and hind feet of the patients scored from 58 to 91 points (average,76.6 points) according to the American Orthopaedic Foot and Ankle Society (AOFAS) evaluation system,giving 7 excellent,32 good and 8 fair cases.Hallux flexion contracture without skin ulcer was observed in 10 cases 6 months after surgery,necessitating no surgical intervention.Conclusion Retrograde transposition of composite tissue flap pedicled with peroneal artery can effectively treat bone and soft tissue defects following calcaneal osteomyelitis,leading to a low recurrence rate and good foot appearance.
8.Antiepileptic effect of sodium succinate

Sodium succinate (0.8 g?kg~ -1,ig) could significantly shorten afterdischarge duration (ADD), lower Racine’s score on amygdala electrical kindling in rat.Additional sodium succinate capsule (0.25g,po) was effective on patients with epilepsy administrated with common antiepileptic drugs which showed little effectiveness on these patients. The results supposed sodium succinate would be effective in epilepsy therapy.

Objective:To observe the effect of Xinfeng Capsule (XFC) on synovial membrane PI3K/AKT/mTOR pathway in adjuvant arthritis (AA) rats.Methods:Rats were randomly divided into normal control group,model control group,methotrexate group,tripterygium glycoside group and XFC group.The expressions of IL-6,IL-10,HIF-1α and VEGF-A were detected by immunohistochemical method,and the expressions of PI3K,AKT1 and p-AKT1 were detected by immunoblotting.Results:Expression of AKT1,mTOR and ES proteins in synovial blood vessels.The expression of IL-6,VEGF,HIF-1α and synovial membrane PI3K,AKT1,p-AKT1,mTOR,ES in the model group were significantly higher than those in the control group The expression of PI3K,p-AKT1,mTOR,ES in synovial membrane decreased,and IL-10 in serum increased in XFC group,and the expression of HIF-1α,IL-6 and VEGF-A were decreased.Conclusion:XFC can improve angiogenesis in AA synovium by regulating PI3K/AKT/mTOR signaling pathway,HIF-1α and ES expression.
