Objective To study the roles of dengue (DEN) virus specific T cells in the pathogenesis of DEN virus infection. Methods 2D42 cells, DEN virus specific CD8 + cell clones, were employed to investigate their in vivo function in DEN virus infection using an animal model. HepG2 was implanted into mice with severe combined immunodeficiency disease (HepG2-SCID) for the establishment of HepG2-SCID model. The animals were divided into 3 groups: Group A: HepG2-SCID mice were inoculated with 2D42 cells and then infected with DEN virus type 2 (DEN2) intraperitoneally; Group B: HepG2-SCID mice were inoculated with normal mouse thymuscytes (NMT) and then intraperitoneally infected with DEN2; Group C: HepG2-SCID mice were intraperitoneally infected with DEN2 alone. The mortality, viremia, and frequency of histopathological changes in the major organs of mice in the three groups were observed after infection. Results After inoculation of 2D42 cells, 80% infected mice showed severe clinical signs and died at the average 12.8 d after infection. The others only had transient manifestations, and then recovered from the disease and survived for more than 3 months. In contrast, after inoculation of NMT and /or DEN2 alone, 100% mortality rate was noted in these two groups. High viremia and frequency of histopathological changes in the major organs were observed in the mice in groups A and B. Conclusion Our data support both protective and pathogenic roles for DEN-specific CD8 + T cells in DEN virus infection.

BACKGROUND:Finding of neural stem cells(NSCs)brings new hope for repairing central nervous system(CNS)injury. However, the influence of internal environment after brain injury on the survival and differentiation of NSCs is a complexand variable process.OBJECTIVE:To observe the survival and differentiation of human embryonic NSCs following implantation into rats with fluid percussion brain injury.DESIGN:Open experiment.SETTING:Department of Neurosurgery,Guangdong Provincial Hospital of Traditional Chinese Medicine;Beijing Institute of Neurosurgery.MATERIALS:This experiment was carried out In the Laboratory of Neural Stem Cells,Beijing Institute of Neurosurgery from September 2002 to March 2003.Twenty-four female SD rats, aged 7 weeks, with body mass of(250±10)g,were provided by the Experimental Animal lnstitute, Chinese Academy of Medical Sciences[License Nd. SCXK(Jing)2002-2003]. Cerebrum of 8-week aborted fetus was obtained (Informed consents were obtained from parturients and their relatives). Fetal survival was monitored by B ultrasonic wave during abortion. BrdU monoclonal antibody(Sigma Company),rabbit anti-nidogen polyclonal antibody(Chemicon Company),mouse anti-microtubule-associated protein 2 (MAP-2)monoclonal antibody(Neomarkers Company),rabbit anti-gliaI fibrillary acidic protein(GFAP)polyclonal antibody (Biogenex Company).METHODS:① CerebraI cortex cells of 8-week aborted human fetus was harvested and cultured in vitro for obtaining human embryronic NSCs.②Rat models of hydraulic impact injury were developed.Bone window of motor sensory area of cerebral cortex was set at 2.5mm posterior to bregma which was zero point and 3.0 mm lateral to midline.Hydraulic impact injury parameters were set as impact pressure 0.3 MPa. impact time 25 ms and impact time once. ③BrdU-labeled human embryronic NSCs were implanted into injured area al 24 hours after injury. After 1 and 4 weeks.rats were sacrificed.Adjacent sections were doubly stained bv BrdU/MAP-2 and BrdU/GFAP.MAIN OUTCOME MEASURES:①Survival and immigration of implanted human embryonic NSCs.②Differentiation of implanted human embryonic NSCs.RESULTS:①BrdU-positive cells were oval and brown. At 1 and 4 weeks after implantation,BrdU-positive cells survived and migrated, and they migrated more widely at 4 weeks after implantation. ②At 1 week after implantation, more BrdU-positive cells were found in the subcortical granular layer and subcortex, and BrdU/MAP-2-positive cells were more than BrdU/GFAP-positive cells; At 4 weeks after implantation,BrdU-positive cells were significantly reduced, and found in choroid plexus and blood capillary, and BrdU/GFAP-positive cells were more than BrdU/MAP-2-positive cells.CONCLUSION:Implanted human embryonic NSCs can survive in the region of brain injury.gradually differentiate into astrocytes during rehabilitation and are easily digested by endothelial phagocytes. It indicates that immunological rejaction possibly influences the survival of human embryonic NSCs.

Chinese Journal of Marine Drugs 1994;0(03):-
Sodium succinate (0.8 g?kg~ -1,ig) could significantly shorten afterdischarge duration (ADD), lower Racine’s score on amygdala electrical kindling in rat.Additional sodium succinate capsule (0.25g,po) was effective on patients with epilepsy administrated with common antiepileptic drugs which showed little effectiveness on these patients. The results supposed sodium succinate would be effective in epilepsy therapy.

OBJECTIVE:
To evaluate the effect of two surgical methods on treatment of allergic rhiniti complicated with nasal septum deviation.
METHOD:
Eighty-seven cases of allergic rhiniti complicated with nasal septum deviation were divided into 2 groups according to the degree of mucosal hypertrophy and hyperplasia of bone in inferior turbinate. They were treated by resection of nasal septum deviation combined with temperature-controlled radio-frequency, or combined with partial submucoperiosteous resection of inferior turbinate bone. The Lanzhou standard (2004) and nasal airway resistance were used to evaluate the efficacy.
RESULT:
After one year follow-up time, the nasal resistance was significantly decreased and the effective rates were greater than 88% in each group.
CONCLUSION
Both of the two surgical methods can significantly depress the nasal resistance and improve the allergic symptoms, which shows good effect.

Chinese Journal of Behavioral Medicine and Brain Science 2013;(2):110-112
Objective To investigate the effects of partial hepatectomy on spatial learning and memory abilities and the expression of spinophilin, which was used as a marker to quantify changes in dendritic spine density in the hippocampus of APP/PS1 transgenic mice. Methods 3-month-old and 6-month-old male APP/PSI transgenic mice and 3-month-old wild-type male littermates were divided into five groups based on their genetic background:3-month-old control (group A,n =8),anesthesia (group B,n =32),surgery (group C,n =32),littermates control(group D,n =8),6-month-old control (group E,n =8).According to the different time points at day 1,3,7,14 after anesthesia or surgery, group B and C were divided randomly into 4 subgroups:B1,B3,B7,B14 and C1,C3,C7,C14.The spatial learning and memory abilities were detected by Morris water maze test in each group at the corresponding time points.Meanwhile, spinophilin expression was detected by Western blot and Immunocytochemistry in the hippocampus. Results Compared with the same time point subgroup B,the latency to find a hidden platform was longer and the frequency of passing through the platform was decreased in Group C at day 3,7,14(P < 0.05).No significant differences were found among group A,group D,and group B (P > 0.05).The grey value and Western blot relative integrated optical density of spinophilin in group A(1 609 453 ±92 801/0.471±0.015),approached the Group D(1 329 398.4±77 783/0.434±0.008)(P>0.05),were significantly increased compared with that(801 678 ± 10 505/0.053 ± 0.003) in group E (P < 0.01).Compared with the Group A,the same time point subgroup B and subgroup C14,the grey value and Western blot relative integrated optical density of spinophilin were decreased in Group C at day 1,3,7 (P < 0.05).Conclusion Partial hepatectomy can impair spatial learning and memory abilities of the APP/PS1 transgenic mice, which is closely related to the decrease expression of spinophilin in hippocampus.
