To compare the growth factors mRNA expressions in human skin fibroblasts cultured in two (2D) or three dimensional systems (3D). Human skin fibroblasts were isolated and cultured in collagen jelly (3D) or the regular culture plates (2D). After 72 hours, the total RNA of fibroblasts were extracted. Specific oligonucleotide primers for the growth factor VEGF and bFGF were synthesized and reverse transcriptase polymerase chain reaction (RT PCR) was utilized to amplify the mRNA of those growth factors. RT PCR productions were separated by electrophoresis in 1% wt/vol agarose gels and photographed. Semi quantitative data were statistically analyzed. Although the morphology of fibroblasts in 3D culture was different from that in 2D culture, the RT PCR analysis revealed the same expression and concentration of VEGF mRNA and bFGF mRNA in human skin fibroblasts cultured in different systems.

Objective To evaluate the feasibility of CT staging for esophageal cancer. Methods A retrospective analysis of 304 patients treated,from Jan. 1996 to Dec. 1998 chiefly with radiotherapy(126 conventional radiotherapy,55-65 Gy/27-35 fx/5.5-7.0 w;178 late-course accelerated hyperfractionated radiotherapy ,55-60 Gy/33-36 fx/ 4.5 -5.5 w)complete with CT data before treatment was done. The long survival was compared with pre-therapy CT findings and CT staging. ResultsThe survival rate of stage T1+T2 was very significantly different from those lesion with stage T3,T4(? 2=12.90,P0.05).The survival rates of patients positive for lymph nodes or distant metastasis were lower. Conclusion CT staging is quite optimal for non-operable esophageal cancer in clinical staging,as it is conducive to predict the prognosis.

Objective To study dectin-1 mRNA expression in lung of different immune status mice infected with Aspergillusfumigatus, and to explore the influence of immunosuppressive agent on the expression of dectin-1 and its relationship with the progression of disease. Methods Mice were divided into four groups which were normal control, immuncompromised, immuncompromised with A.fumigatus inoculation and immuncompetent with A.fumigatus inoculation groups. We explored the kinetic mRNA expression of dectin-1 in lung of different groups by real-time quantitative PCR. Pulmonary fungal burden assessment was performed to reflect the progressing of disease during the experimental time course. Results On day 3 after inoculation, pulmonary fungal burden of the immuncompromised mice was higher than that of the immuncompetent group. On day 1 and 3, dectin-I mRNA expression in lung of the immuncompromised group was much lower than that of the normal control. On day 3, dectin-1 mRNA expression in lung of the immuncompetent mice infected with A.fumigatus was much higher than that of the immuncompromised with A.fumigatus inoculation and the normal control groups ( P < 0.05 ). Conclusion During the infection, expression of dectin-1 in lung of the immuncompetent group was strikingly increased, which may play an important role on the defence to A.fumigatus invasion. Cyclophosphamide inhibited the expression of dectin-1 in lung of mice which may be one of the mechanisms of the invasive pulmonary aspergillosis development induced by eyclophosphamide.

Objetive: To study the result of manipulation therapy(MT) for frozen shoulders. Method: Before and after MT, the parameters of involoved shoulder extensors and flexors , abductors and adductors were isokinetically recorded on 20 patients with frozen shoulder twice. Result: Compared with the parameters measured pre-therapy most of those factors measured increased significantly afterwards (P＜0.01 ). Conclusion:Effect of MT on frozen shoulder are satisfactory and ROM are accurate and reliable.

Perepherial blood lymphocytes from normal donors were isolated by Haemonetice V50 Ampheresis System, and incubated in RPMI 1640 contained 1000U/ml rIL-2 and 10%AB serum at 37癈, 5%CO2. Five days later, these cells were harvested and tested their LAK activity. Then 10 patients with malignant metastases were treated by transfer of allo-LAK cells. After 2 and 6 months, the PEL from patients were seperated for the test of response to IL-2, proliferation to ConA and production of IL-2. The result showed that no significantly reducing of these value after patients being treated by Allo-LAK cells. Combined with clinical observation and pathological biopsy, it was concluded that GVHD couldn't be caused by transfer 6f allogenic LAK cells.

To determine the effect of CX43 expression on gastric mucosa apoptosis and proliferation under stress conditions, a water immersion-restraint stress(WRS) model was performed with SD rats. Apoptotic cells were quantitated in gastric mucosa by terminal deoxynucleatidyl transferase-mediated dUTP nick and labelling (TUNEL) techniques. Cell proliferation was detected by immunohistochemical staining for proliferating cell nuclear antigen (PCNA). The expression of CX43 mRNA was detected by in situ hybridization. The results revealed that in control rats not exposed to WRS, apoptotic cells were seen only occasionally at the surface epithelium, The PCNA cells occurred in the proliferation zone of the gastric glands. The CX43 mRNA predomination occurred in the zone of the gastric glands. In contrast, the apoptotic quantities rose significantly 2h to 5h after the termination of WRS and reach a peak gradually, while PCNA proteins and the CX43 mRNA decreased significantly. At 8～12h after WRS the apoptotic cells gradually declined, while the PCNA cells increased to reach a peak. The CX43 mRNA almost returned to normal level and then increased to reach a peak. At 24h after WRS, the apoptotic cells remained significantly higher than normal level while the expression of PCNA proteins and CX43 mRNA returned to the values observed in the intact rats.These results suggested the expression of CX43 might associate with cellular differentiation, proliferation and the stabilization of glandular zone and affect the apoptosis of the surface epithelium.

Objective To investigate the effect of antiulcer agents Omeprazole, Misoprostol and Talcid on the gastric mucosa lesion by analyzing the gastric mucosa cell apoptosis and proliferation and CX43 transcriptional expression. Methods Mucosa lesion were evaluated by UI, 32 mice were randomly divided in equal numble into Omprazole (O), Misoprostol (M), Talcid (T), Comparison (C). Apoptosis cells in gastric mucosa were quantitated by terminal deoxynucleatidyl transferase mediared dUTP nick and labelling (TUNEL) techniques, while the expression of PCNA proteins were detected by immunohistochemical staining, CX43 mRNA was detected by In situ hyberdization. at 2h after WRS. Results We found that in comparison with control group, the group of pretreatment with Omeprazole, Misoprostol and Talcid showed a significant reduction in damaged mucosa and epithelial cell apoptosis and increase in the expression of PCNA proteins, and the effect of Omeprazole and Misoprostol was better than Talcid. All three antiulcer agents increased CX43 mRNA expression, the effect of Misoprostol and Talcid was better than Omeprazole. Conclusions Omeprazole, Misoprostol and Talcid can attenuate the multiple gastric mucosa lesion induced by WRS. It may be the ultimate way to prevent and cure stress ulcer by exerting direct cytoprotective effect to improve the cell ability and inhibit stress injury.

Objective By studying clinical features, treatment and prognosis of eosinophilic gastroenteritis of infants resulted from milk protein allergy an, to improve the diagnosis and treatment level of eosinophilic gastroenteritis. Methods 24 cases of infants which diagnosed eosinophilic gastroenteritis were chosen from June of 2010 to January of 2014 in children’s Hospital of XX province and By retrospective analysis clinical manifestations, endoscopic features, histopathology, treatment and prognosis of the 24 cases. Results The 24 cases who were vomiting, paroxysmal crying, abdominal distension (100.00%), which accompanied by haematemesis 23 cases (95.83%), 1case (4.17%) hematochezia, 17cases (70.83%) eczema, 21 cases (87.50%) mild to moderate anemia, 1 cases (4.17%) severe anemia, 19 cases (79.17%) the increasing of peripheral blood eosinophil cells, 8 cases (33.33%)the increasing of IgE of the serum and 4 cases (16.67%) the test of antibody of the Helicobacter pylori in Serum was positive; 3 cases (12.50%) were milk protein allergy by the detecting of food allergen-speciifc IgE antibodies, the endoscopic characteristics were hyperemia, edema, erosion, ulcer of gastric and duodenal mucosa. Among them, 24 cases (100.00%) were gastritis, 5 cases (20.83%) duodenitis and 1 cases (4.17%) duodenal ulcer. The histopathology of the 24 cases revealed that there were gastric or duodenal eosinophils infiltration (> 20/HPF) and were all associated with mast cell infiltration; By antisecretory, protection of the gastrointestinal mucosa and the obviating of milk protein had a satisfactory treatment effect, 24 cases of children with oral general formula milk test conifrmed that the milk protein allergy, The 3 cases of the patients were reviewed by 8~12 weeks after gastroscope, and the mucosa of the duodenum was smooth, Eosinophils were/HPF<8, mast cells were/HPF<5. Conclusion There are no speciifc clinical and endoscopic manifestations in eosinophilic gastroenteritis of infants resulted from milk protein allergy, gastrointestinal mucosa eosinophil inifltration and simultaneously are accompanied by abnormal mast cell inifltration;Mucosal type without the use of corticosteroids, through milk protein avoidance treatment can achieve satisfactory results, But definite diagnosis must rely on biopsy and eosinophils, combined with avoidance stimulation test of milk protein can further confirmed, But the excitation test should be at least 10 days of observation of children, and carefully recorded symptoms, so as not to delay the missed diagnosis of CMPA.

Objective To study the influence of anticipatory and post-event information processing on the memories and perception of the symptom in patients with social anxiety disorder (SAD). Methods A group of 32 SAD patients and a control group of 35 healthy indivisuals were included. Instruments including Self-Rating Depression Scale, Subjective Units Discomfort Scale( SUDS ), Rumination Questionnaire ,Open-ended Recall and Body Sensations Questionnaire were adopted in both groups. Results In the group SAD, no significant difference was identified between the experimental group( (49.68 ± 17.68), ( 19.00 ± 1.25), (0. 54 ±0. 17) ) and the experimental control group( ( 50.43 ± 20.72 ), ( 18.68 ± 1.25 ), ( 0.52 ± 0.17 ) ) when the ratings the memories of body sensations, rumination, and negative self-information score were compared (P＞ 0.05 ). There was a significant positive correlation between the level of rumination ,SUDS, the memories of negative self-information and the body sensations scores( r= 0.72; r= 0.94; r= 0.70, P＜ 0.01 ). The scores of rumination explained 64% of the variation in SUDS scores(β=0.82, P＜0. 01 ). Conclusion This study suggest that social anxiety is affected directly by rumination which can result in more memories of negative self-information and the body sensation. Symptoms are maintained by post-event information processing.

Objective To assess ventricular function of early stage neonates of different gestational ages by tissue doppler imaging(TDI).Methods Pulsed wave TDI velocities were obtained in 36 cases of premature infants with gestational ages of 32 to 36 weeks(premature group) and 33 cases of full-term infants(full-term group) aged 3 to 7 days at the lateral mitral annulus(MA),basal septum,and lateral tricuspid annulus(TA) during ventricular systole(Sa),early diastole(Ea),late diastole(Aa).Tansmitral and transtricuspid inflow were also obtained through pulsed doppler echocardiography.Results Ea and Sa in all of 3 locations were lower in the premature group compared with that of the full-term group,and Ea/Aa in TA was lower in premature group,but Aa and E/Ea showed no difference between 2 groups.Ea and Sa showed a positive correlation with gestational age and birth weight.Conclusions Ventricular systolic and diastolic function in premature infants are poorer than that in full-term infants in the early stage of neonatal period,and ventricular function is related to intra-uterus growth and maturity of the newborn.Diastolic function of the left ventricle in both groups develops rapidly during the early stage of neonatal period.
