1.Investigation in prevention related knowledge, attitude and behavior of high risk population of cerebral stroke

ObjectiveTo understand the status of knowledge,attitude,behaviors about stroke prevention among the high risk population,so as to provide scientific basis for health education. MethodsA selfdesigned questionnaire about knowledge,attitude,behaviors of stroke prevention was used to investigate 106 cases from clinics and communities.The contents included general data,risk of cerebral stroke,score of related knowledge,attitude,behaviors about stroke prevention. ResultsThe mean scores of knowledge,attitude,and behaviors about stroke prevention were(46.96±10.07),(70.71±12.37) and (64.29±16.23),individually.Correlation analysis showed there was significant positive correlation among the scores of knowledge,attitude,and behaviors and their age,educational levels and residence,but was not significantly correlated with sex and risk score of cerebral stroke. ConclusionsPrimary prevention about stroke should focus on the high risk population with lower educational levels,ages under 60 years old,living in suburbs,lack of stroke prevention knowledge.
2.Breast cancer stem cells:separation and resistance protein analysis

BACKGROUND:Breast cancer stem cels are relatively special cels in the body, which have the self-renewal and multi-differentiation ability to promote tumor formation and development, and maintain tumor growth for a long-term. Therefore, it is of great significance to analyze the expression of resistance proteins of breast cancer stem cels.
OBJECTIVE:To isolate breast cancer stem cels from human breast cancer tissues, to observe their differentiation and morphology characteristics and to analyze their resistance proteins.
METHODS:Thirty tumor samples of breast invasive ductal carcinoma were selected to separate single cel suspension using mechanical separation method, and breast cancer stem cels and differentiated cels were sorted with two-step immunomagnetic bead method. Two-step immunocytochemistry method was used to detect the expression of resistance proteins in breast cancer stem cels.
RESULTS AND CONCLUSION:Percentage of breast cancer stem cels had no significant correlations with age, long diameter of the tumor, lymph node metastasis and histological grading (P > 0.05). P-gp and GST-π positive rates in the breast cancer stem cels were significantly higher than those in the differentiated cels (P < 0.05); while TopoII and LRP positive rates in the breast cancer stem cels were significantly lower than those in the differentiated cels (P < 0.05). To conclude, breast cancer stem cels show stronger drug resistance than the differentiated cels by highly expressing P-gp and GST-π and lowly expressing TopoII and LRP, which may be the key reason for chemotherapy resistance in breast cancer.
3.Evolution and status of unicompartmental knee prostheses

BACKGROUND:Unicompartmental knee arthroplasty has been prevailed in orthopedics,but there is a lack of overview addressing unicompartmental knee prostheses.OBJECTIVE:To review the evolution and current situation of unicompartmental knee prostheses and to discuss its classifications,characters and clinical outcomes,thereby providing guidance for clinical application.METHODS:PubMed and WanFang databases were retrieved for the articles related to unicompartmental knee prostheses and its clinical outcomes.Totally 177 articles were retrieved,and finally 35 eligible articles plus other 4 reports were included after excluding repetitive studies.RESULTS AND CONCLUSION:(1) It has been more than 60 years since the birth of unicompartmental knee prostheses.Despite the difficult in the past,it comes to mature and the survival rate is more than 90% at 10 years.(2) At present,the most popular prostheses include mobile-bearing (represented by Oxford 3) and fixed-bearing (represented by ZUK and Link).There is no significant difference in clinical outcomes between two designs.However,their differences have been noted in their modes and timing of failures.(3) Aseptic loosening and lateral osteoarthritis progression remain the most common failure modes in unicompartmental knee prostheses.(4) Cementless prostheses and individual unicompartmental knee prostheses based on CT or MRI scan maybe promote the long-term efficacy of unicompartmental knee prostheses.
4.Application of the detection of heart fatty acid-binding protein to early diagnostic of myocardial injury

Heart fatty-acid-binding protein(H-FABP) is small molecular weight cytosolic protein that is abundant in the heart and has tissue specificity. Concentration of H-FABP changes regulation in serum after the myocardial damage. In addition it has a positive correlation with the degree of myocardial damage. It is an early diagnostic biochemical marker of acute myocardial ischemia. The advance of H-FABP in the research on physiochemical nature,and its application for myocardial injury and prospective study of sudden cardiac death in forensic science were reviews.
5.Effects of midazolam on the response of parafascicular nucleus to noxious stimuli

Objective To investigate the effect of intravenous midazolam on the response of parafascicular nucleus(Pf) to noxious stimuli in rats. Methods Twenty SD rats of both sexes weighing 260-300g were used in the study. The animals were anesthetized with intraperitoneal urethane(1.2 g/kg) . A hole was drilled in the skull. Under stereomicroscope micro-glass electrode was inserted into Pf nucleus and the characteristic discharge of nociceptive neurons was recorded. The experiment was then performed in four steps. The discharge was recorded in the basal state (A), when hind paw was pinched by a pair of strong forceps: noxious stimuli (B) , 2 min after midazolam 0.2 mg/kg was given intravenously , step B was repeated (C) and flumazenil 0.05mg was given iv and 2 min later step B was repeated (D) . There was a pause of 5 min after each step. Results The characteristic discharge of nociceptive neurons was recorded only in 9 animals and the data from 6 animals were complete and could be analyzed. Noxious stimuli induced significant increase in spike of discharge of nociceptive neurons. Midazolam significantly depressed the response of nociceptive neurons to noxious stimuli. There was no significant difference in the mean spikes of discharge of nociceptive neurons between step A and C. After flumazenil the mean spike of discharge increased again in response to noxious stimuli. Conclusion Midazolam can inhibit the activity of the nociceptive neurons located in Pf nucleus, probably through GABA-BZ receptor.
6.Next-generation sequencing in molecular diagnosis of hereditary hearing loss.

Next-generation sequencing (NGS) technologies have improved as well as the costs have gradually decreased in the detections of genetic diseases. This article describes the principle, platform, and data analysis of NGS and the application of NGS technologies to the molecular diagnosis of hereditary hearing loss (HL). The use of NGS technologies makes the discovery of HL genes more feasible than ever. And the data obtained by NGS used in genetic counseling for clinical practice may assist in defining genetic profiles of HL individuals and expedite the pace of personalized medical care.

7.Strategies for elaboration of comprehensive quality standard system on traditional Chinese medicine.

Based on the research goal of "traditional Chinese medicine standards lead in the international standard-setting", scientific strategies for the elaboration of traditional Chinese medicine (TCM) comprehensive quality standard system were introduced. TCM is a complex multi-component system which was used under the guidance of traditional Chinese medical theory. The present paper has put forward the basic principle of " deep research and simplified standard" to construct the quality standards of TCM. " Deep research" refers to systematic, thorough investigations on active constituents and biological mechanisms. On the basis of deep research, "simplified standard" means to establish a scientific and feasible standard, which should be practical and less complicated, to control the quality of Chinese herbs. Three key issues related to the elaboration of the quality standards are suggested. For reference substances, we propose the research methodology using reference extract to develop the qualitative and quantitative determination methods. Compared with that of using reference compounds, the preparation of reference extract is more accessible, economical and practical. Moreover, compared with reference crude drugs, the reference extract showed better batch-to-batch consistency. For identification, in addition to conventional methods, high performance liquid chromatography (HPLC), chemical fingerprints and characteristic chromatogram are proposed, in which more major marker compounds are monitored. And then LC-MS technique is employed to comprehensively analyze and characterize the peaks in the fingerprint. For multi-component quantification, the method of single standard to determine multi-components (SSDMC) is suggested, providing solutions for the lack of reference standards in quality evaluation. The SSDMC method uses a single reference standard to simultaneously determine the content of multiple compounds. In general, the integrate quality control standard of TCM is established based on combining innovative technology with practical applications.

8.Fatigue analysis system with HR and HRV as indexes

In this paper,heart rate (HR) and heart rate variability (HRV) are extracted from ECG,and the five indexes in both time and frequency domains are analyzed.The result shows that the standard deviation (SDNN) of R-R intervals,the value of total power (TP),the power value of low frequency (LF) of HRV and the value of LF/HF increase obviously,while the power value of high frequency (HF) decreases markedly after fatigue.The physical fatigue level is classified according to the rate of increase and decrease of the indexes above.It is suggested that these five ECG indexes be used to reflect and evaluate the degree of physical fatigue quantitatively.
9.Application of 3C bolus wizard on controlling postprandial glucose in diabetic patients

ObjectiveTo explore the influence of 3C bolus wizard on postprandial glucose levels in diabetic patients. MethodFifty-eight patients with type 2 diabetes in our hospital were monitored with continuous glucose monitoring system(CGMS), continuous subcutaneous insulin infusion(CSII)and CareLink(3C for short)for 6 days.The function of “3C” bolus wizard was applied during treatment and all the patients were given dietary nursing and health education.The glucose level was observed.ResultThe level of P2hBG of the 58 patients was between 6.4~10.7mmol/L in 3~6 days.ConclusionsBolus wizard plays an important role in “3C” treatment.It can make the postprandial glucose of patients satisfactorily controlled in a short time only to avoid insulin hypoglycemia because of large doses of insulin.At the same time,health education on patients can achieve remarkable results as well.
10.Effect of Shenqihuang Steady Plaque Caps Combining Simvastatin on Stabilization of Atherosclerosis Plaque in Rabbits

Objective To investigate the role of Shenqihuang steady plaque caps (SQH) and simvastatin on stabilization of atherosclerosis plaque in rabbits whit atherosclerosis (AS). Methods Forty-five rabbits were randomly divided into five groups: normal food group (A), instability plaque group (B), SQH group (C), simvastatin group (D), SQH add simvastatin group. A group was fed with common feeds, and B, C, D, E groups were given high cholesterol feeds. An atherosclerotic rabbit model was established by feeding high cholesterol diet supplemented for 12 weeks in B, C, D, E groups. Then C, D, E groups received corresponding drugs. At the end of 24~(th) week, the drug were injected into the aortic segments rich in plaque. Two weeks later, the concentration in serum of lipids, vascular cellular adhesion molecule-1 (VCAM-1), intercellular cellular adhesion molecule-1 (ICAM-1) and high sensitivity C reaction protein (hs-CRP), and area of plaque, the thickness of endomembrane, ttmica media, fibrous cap were detected. Results Compared with B group, the concentration of VCAM-1, ICAM-1, hs-CRP and area of plaque, the thickness of endomembrane, tunica media in C, D and E groups were lower significantly, the thickness of fibrous cap was thicker significantly (P
