1.Antiangiogenic therapy for PTCL
Journal of International Oncology 2012;39(3):190-193
Peripheral T-cell lymphomas (PTCLs) are insensitive to traditional chemotherapies which lack of effective treatments and have worse prognosis than B-cell lymphomas.Angiogenesis plays an essential role in the malignant tumor growth.Recently, studies have shown that the expression level of VEGF is related to the clinical characteristic and prognosis of PTCL.Novel drugs based on anti-angiogenic theory show curative effect in the preliminary clinical trials for the treatment of PTCL.
2.Preparation of three types soluble chitosan
International Journal of Biomedical Engineering 2011;34(1):30-33,49
Objective Although chitosan has excellent biocompatibility and biodegradability, it is only soluble in acid solution that is not feasible for application in tissue engineering. The current study is aimed to prepare the carboxymethyl chitosan (CMC) for the purpose of improving the solubility of chitosan to meet the requirement of development of tissue engineering scaffolds. Methods The preparation methods and process conditions, such as the proportion of reactant, reaction temperature, reaction time and so on for three types of carboxymethyl chitosan were studied. Results The results indicated that optimal process conditions of three types of water soluble chitosan were obtained. Conclusion Water solubility of chitosan is greatly improved through carboxymethyl modification, which provides possibilities and foundation for further research.
3.Diagnosis and clinical management of tension-type headache
Chinese Journal of Practical Internal Medicine 2001;0(06):-
Episodic tension-type headache is the most common headache in the general population and is usually self managed.Chronic tension-type headache may be highly disabling and often prompts medical consultation.Diagnosis is clinical, based on widely accepted and validated criteria and algorithm.Peripheral factors are implicated in episodic tension-type headache, whereas central factors probably underlie chronic tension-type headache.Simple analgesics, such as aspirin, are used for acute treatment.Amitriptyline and biofeedback assisted relaxation training have the best evidence of effectiveness for headache prevention.

Chinese Journal of Cancer Biotherapy 1995;0(03):-
Retroviral vectors are the most used gene delivery vehicle in human gene therapy. When the therapeutic gene is packagedfrom producer cell line, one has to determined whether thereplication-competent retrovirus(RCR) is being generated. In this study, two methods, namely the marker rescue assay and RT/PCR, were employed to detect RCR in the cells. While the marker rescue assay can detect RCR with a limit at 6?102CFU/ml, RT/PCR can be used to detect RCR at as low as lCFU/103ml. Combining the specificity of the marker rescue assay and the sensitivity of RT/PCR, both assays together should serve as an adequate test for detecting the generation of RCR in retroviral producer cell lines.

PURPOSE: Metalloproteinases (MMPs) and tissue inhibitors of metalloproteinase (TIMPs) have been linked to the angiogenic process in general. In order to understand the potential roles of MMPs and TIMPs in corneal neovascularization process, we examined the expression and activities of MMP-2, MMP-9, TIMP-1 and TIMP-2 during the course of suture-induced corneal neovascularization in rat model. METHODS: Corneal neovascularization of rat cornea was induced by suturing. The expression of MMP-2, MMP-9, TIMP-1 and TIMP-2 in sutured corneas was examined by immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR). The activities of MMP-2 and MMP-9 were measured before and after suture by gelatin zymography. RESULTS: MMP-2 proenzyme, and TIMP-1, -2 were expressed in normal corneas, predominantly in corneal epithelium. After injury, expression of MMP-2, MMP-9, TIMP-1 and TIMP-2 increased, notably in healing corneal epithelium, infiltrating inflammatory cells, stromal fibroblasts, and ingrowing vascular endothelial cells. The intensity of immunostaining and enzymatic activities of MMP-2 and MMP-9 paralleled the magnitude of inflammatory cell infiltration, which peaked around day 7 after suture. Immunoreactivity of MMP/TIMP decreased significantly two weeks after suturing. At day 35 after suture, staining of MMP-2, TIMP-1, -2 remained visible only in corneal epithelium and vascular endothelial cells. CONCLUSIONS: MMPs as well as TIMPs were upregulated during suture-induced corneal neo-vascularization, suggesting that both may take part in extracellular matrix remodeling in the corneal wound healing, inflammatory, and neovascularization processes.

Journal of Chinese Physician 2010;12(2):175-177
Objective To investigate the expression of matrix metalloproteinases-7 (MMP-7) in benign and malignant thyroid tissues and its clinical significance. Methods The expression of MMP-7 in 50 thyroid cancers, 45 adenoma and 20 adjacent noncancerous portion tissues were studied by microwave-LSAB immunohistochemical method. Results The positive rate of MMP-7 in thyroid cancer was 64. 0%, which was significantly higher than 37. 0% in adenoma and 25.0% in adjacent noncancerous portion(X_2=8. 72,6. 52, P < 0.01 ). No correlation was observed between the expression of MMP-7 and histological grading of thyroid cancer. The expression of MMP-7 in lymph node metastasis and Grade Ⅲ~Ⅳ were both higher than that in negative cases and in Grade Ⅰ~Ⅱ ( P < 0. 05 ). The percentage of recurrence and death in MMP-7 positive cases were notably higher than that in negative cases( P <0. 05). Conclusion The expression of MMP-7 can be regarded as a parameter for evaluating the degree of malignancy, biological behavior and prognosis of thyroid cancer.

Objective To investigate the relationship of the activation of nuclear factor kappa B(NF-kB) with myocardial neutrophil infiltration and injury in human open heart surgery, and to observe the inhibiting effect on the activation of NF-kB and protecting effect on myocardium of the coenzyme Q10,a scavenger of oxygen free radicals. Methods Forty-seven adult patients undergoing open heart surgery were randomly divided into two groups, the control and the treatment.CoenzymeQ10 tablets were given to the treatment group 5 days before operation.Biopsy of right atrium for myocardial pathology, activated NF-kB detection and ultrastructure observation were done prior to cardiopulmonary bypass,45 minutes of ischemia and 45 minutes of reperfusion. The dynamic indexes, vasomotor drug dosage, and outcomes were observed postoperatively. Results Upon 45 minutes of ischemia and 45 minutes of reperfusion,in control group there were neutrophil accumulation and adhesion of vascular endothelium, ultrastructural damages, and positive expression of NF-kB both in nuclei and cytoplasm,in myocardium. In treatment group, there were only mild neutrophil infiltration and ultrastructural damages, and weak positive expression of NF-kB both in nuclei and cytoplasm. However, the dynamic indexes, vasomotor drug dosage, and outcomes of two groups were not significantly different. Conclusion NF-kB plays an important role in pathophysiological process of myocardial ischemia and reperfusion in open heart surgery.CoenzymeQ10 has obvious inhibiting effect on activation of NF-kB and protecting effect on myocardium.
