China Pharmacy 2016;27(5):657-659
OBJECTIVE:To evaluate therapeutic efficacy and safety of chemotherapy combined with Xiaoaiping injection in the treatment of elderly patients with advanced gastric cancer. METHODS:60 elderly patients with advanced gastric cancer were collected from our department during 2012-2014,and then divided into union group and single group with 30 cases in each group. Single group was given irinotecan(CPT-11)150 mg/m2 added into 0.9%Sodium Chloride injection, and union group was addition-ally given Xiaoaiping injection 80 ml,qd,on the basis of single group. A treatment course lasted for 14 days, and both groups re-ceived 4 courses of treatment. The tumor markers, disease single rate,progression-free survival (PFS) and the incidence of ADR were compared between 2 groups. RESULTS:After treatment, the levels of CEA,CA199 and CA242 in 2 groups were significant-ly lower than before;the union group was significantly lower than the single group, with statistical significance(P<0.05). The dis-ease single rate of union group was 70.00%,which was significantly higher than that of single group (33.33%). PFS of union group was(11.23±2.17)months,which was significantly longer that of single group [(9.36±0.84)months],with statistical signif-icance(P<0.05). The incidence of leucopenia, alanine aminotransferase elevation, diarrhea, stomatitis and hand-foot syndrome in union group was significantly lower than in single group, with statistical significance(P<0.05). CONCLUSIONS:Xiaoaiping in-jection combined with irinotecan chemotherapy for advanced gastric cancer has obvious effect with a low incidence of ADR.

Seventy percent of the patients were identified by general practitioners. Follow-up was initiated by general practitioners(60%)or other healthcare providers(40%).Our findings suggested that traditional disease management model focused mainly on acute conditions, while community health center-based model Was expected to meet the need for chronic disease prevention and treatment and play an important role in minimizing economic burden and improving doctor-and-patient relationship.
5.Construction and identification of small interfering RNA expression plasmid target to angiotensinogen
Chinese Journal of Tissue Engineering Research 2010;14(11):1951-1954
BACKGROUND:In mammalian cells, introduction of double-stranded small interfering RNA(19-25 bp)can cleave and destroy the cognate RNA,which can result in suppression of gene expression.OBJECTIVE:To construct siRNA expression plasmid for interference angiotensinogen(AGT),thereby,to resist AGT expression in adipose cells.METHODs:The mRNA sequence of AGT gene was searched from NCBI(NM000029).Utilize of GenScript siRNA technology,AGT-siRNA oliaonucletides were chemically synthesized and inserted into pRNAT-U6 1/Neo vector after annealing, then transformed into TOP10.The recombinant plasmid was identified by restriction endonuclease and DNA sequencing.RESULTS AND CONCLUS1ON:The recombinant plasmid psiRNAT-U6.1/Neo-AGT was obtained by connecting 19 bp segment containing AGT-mRNA sequence to pRNAT-U6.1/Neo After EcoR Ⅰ and Hind Ⅲ digestion.351 bp segment was obtained from empty vector.and 397 bp fragment band was obtained form recombinant plasmid, which was coincidence to the expectation.DNA sequencing showed Targeting siRNA oligonucleotides were correctly inserted into the eukaryotic expression vector pRNAT-U6.1/Neo without base mutation. The interference vector psiRNAT-U6.1/Neo-AGT was successfully constructed.
6.Construction of alienation scale in adolescent students and assessment of its theory
Chinese Journal of Tissue Engineering Research 2005;9(24):218-221
BACKGROUND: Alienation is derived from dissimilation in philosophy. It is practically defined as the subjective state of individuals who develop negative feelings such as social isolation, uncontrollable and meaningless sense, oppression, and self-alienation because they are alien to others, the.society, nature and themselves, and even feel controlled.OBJECTIVE: To investigate alienation from psychological perspective, aiming to construct and verify the theoretical hypothesis of alienation, thereby working out alienation scale for adolescent students.DESIGN: Cross-sectional investigation.SETTING:National Key Research Center of Basic Psychology and National Key Liberal Arts of Southwest Normal University.PARTICIPANTS: The investigation was carried out from March to June 2000. The method of cohort stratified random sampling was adopted and the questionnaire was administered to adolescent students in Chongqing City and Lanzhou City. Inclusion criteria: volunteer students of grade one in junior high school to grade four in colleges. Exclusion criteria: Those who had incomplete data or had differences of more than 6 points in the total scores for three matched lie-testing questions in the questionnaire. A total of 1502 pieces of questionnaire were collected (including 1 066 questionnaires used for exploratory factor analysis and 436 questionnaires for verifying factor analysis); the age of the students covered the whole range of adolescence, and the boy-to-girl ratio in each grade was matched.METHODS:We combined literature analysis, investigation research and interview. The details are as follows: First, theoretical hypothesis was put forward based on literature analysis; then related variances were collected through interviews and open questionnaires, which were compared with the items in scales used home and abroad at present so as to work out predicting questions for adolescence alienation questionnaire. Finally, 52 items were screened out and used to construct the preliminary questionnaire. The formal alienation scale was made after standardized designation of behavioral variances, and verified for alienation analysis in adolescent students.MAIN OUTCOME MEASURES: Behavioral apparent variances were collected based on interviews and open questionnaires, and used as psychological potential variances after statistical processing.RESULTS:Data of 1502 pieces of questionnaire were analyzed.① The correlation coefficient of each item in the scale was over 0.35. ② The correlation coefficient of each item with the dimension was over 0.30, with statistical significance(P < 0.01). ③ The reliability coefficient of each item was over 0.39 with great significance(P < 0.01). ④ Alienation was displayed in three dimensions: social alienation, interpersonal alienation and surrounding alienation, which were associated with the correspondingdimensions of related scales (P < 0.05-0.01). ⑤Alienation could be divided into 9 levels: sense of meaninglessness, sense of self alienation, isolation, nature alienation, parent alienation, environmental alienation, sense of uncontrollability and social alienation. Parameters that accorded with the theoretical hypothesis and used for verifying analysis were proved rational.Based on the analysis of the 9 first-rank factors, second-rank structural model of 3 unrelated factors were obtained using main component analysis and the maximum variance cross rotation with the optimal coefficients of simulated parameters of: X2/df=4.87;GFI=0.94;A GFI=0.90;RMR=0.0S;NFI=0.93;NNFI=0.92;CFI=0.95;IFI=0.95.CONCLUSION: Alienation scale has been proved to be reliable and valid. It can be used to assess alienation in adolescent students and has potential value of application in healthy psychological development and psychological education.

9.Relationship between fasting plasma glucose in early pregnancy and gestational glucose metabolic disorders
Chinese Journal of Perinatal Medicine 2011;14(3):166-169
Objective To investigate the relationship between early pregnancy fasting plasma glucose (FPG) and gestational glucose metabolism disorders. Methods Six hundred and fifty-six pregnant women who were singleton, non-diabetes before pregnancy and had FPG examined during 5-13 weeks of pregnancy were admitted into this study from January 1, 2009 to May 31, 2009. All these subjects had routine prenatal examination and finally delivered in the Department of Obstetrics of Peking University First Hospital. The FPG, 50 g glucose challenge test (GCT) after 24 weeks of pregnancy, 75 g oral glucose tolerance test (OGTT), gestational diabetes mellitus (GDM),gestational impaired glucose tolerance (GIGT) were analyzed with receiver operating characteristic (ROC) curve. Results (1) Relationship between FPG and GCT were analyzed with ROC curve. The maximum area under curve was 0. 539 (95% CI: 0. 493-0. 586) and there was no correlation between the FPG and GCT results(P=0. 057). (2) Relationship between early pregnancy FPG and abnormal FPG examined after 24 gestational weeks were also analyzed . The maximum area under curve was 0. 796(95% CI: 0. 672-0. 920). If 5. 05 mmol/L was taken as the cutoff value, the sensitivity and specificity was 54. 5% and 83. 2%, respectively. There was significant relationship between the two values (r=0. 432, P=0. 000). (3) There were no relationship between early pregnancy FPG and the blood glucose value of 1, 2 and 3 h in 75 g OGTT (r=0. 093, 0. 036 and 0. 107, P=0.122, 0. 549 and 0. 074 respectively). OGTT 0 h value was positively related to OGTT 1, 2 and3 h glucose level (r=0.493, 0.421 and 0.368, P=0.000, respectively). (4) All early pregnant FPG values in this study were under 6.1 mmol/L. Twenty-two GDM and 27 GIGT patients were diagnosed in this study. Early pregnancy FPG did not relate to the GDM and GIGT diagnosis. Conclusions Early pregnancy FPG could not replace 50 g GCT as an early screening for glucose metabolic abnormality in pregnancy, but FPG during early pregnancy is necessary.
10.Anti-lipotoxic action of sesamin on renovascular hypertensive rats fed with a high-fat, high-sucrose diet.
Acta Pharmaceutica Sinica 2012;47(1):58-65
This study is to observe anti-lipotoxic effect of sesamin on renovascular hypertensive rats fed with a high-fat, high-sucrose diet. Thirty-four complex model rats were induced by two-kidney, one-clip method and on high-fat and refined-carbohydrate diet for thirteen weeks. From the fifth week, intragastric administration of sesamin (120, 60 and 30 mg x kg(-1) x d(-1)) lasted for eight weeks. Blood pressure (BP), blood fat (BF), blood glucose (BG), free fatty acids (FFA), insulin (Ins), tumor necrosis factor (TNF)-alpha and interleukin (IL)-6 were determined. Pathological changes of pancreas, perirenal fat and liver were semiquantitatively analyzed. In sesamin (120 and 60 mg x kg(-1) x d(-1)) group, it was found that there were decrease of levels of BP, BF, BG, TNF-alpha, IL-6 and FFA, improvement of insulin resistance and glucose tolerance, alleviation of body weight, humid weight of fat, liver and pancreas and their organ index, and reduction of islet cell hyperplasia and amount of lipid droplet vacuoles in lipocyte and hepatocyte. It is implied that sesamin had anti-lipotoxic effect and its mechanism may be closely associated with the amelioration of insulin resistance via reducing lipidoses in hepatocyte and inflammatory adipokines such as TNF-alpha and IL-6.
