1.Influence of COX-2 inhibitor on expression of Ang-1 and Ang-2 protein in human tongue squamous cell line Tca-8113

Objective To investigate the influence of Nimesulide(NIM),a selective Cyclooxygenase-2 (COX-2) inhibitor, on the expression of Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) protein in human tongue squamous cell carcinoma Tca-8113 cell lines. Methods Tca-8113 cell were cultured in vitro, and exposed to a range of concentrations of nimesulide (NIM) (0, 25, 50, 100, 200 μmol/L) for 24 h, 48 h, 72 h, and the PGE2 level in the cells cultured supernate was quantitated by the radioimmunoassay, and the expression of Ang-1 and Ang-2 protein in human tongue squamous cell line Tca-8113 were measured by the SABC immunocytochemistry staining simultaneously. Results The PGE2 level in the cultured supematant and expression of Ang-2 in Tca-8113 cells decreased by NIM in a concentration and time dependent manner, but not Ang-1. Conclusion NIM can decrease PGE2 level and Ang-2 expression, which may be involved in the inhibition of cancer angiogenesis of COX-2 dependence mechanisms.

4.An animal experimental study of the loading time on anchorage implant stability.

OBJECTIVEThis study aimed to evaluate the influence of loading in different healing periods on implant stability and to establish an animal model of anchorage implant screws.
METHODSAnchorage implant screws were implanted in sheep alveolar bone to establish the implant screw animal model. The animals were randomly divided into four groups: group A, sheep without loading; group B, sheep loaded immediately after implant; group C, sheep loaded after two weeks; and group D, sheep loaded after four weeks. The maxillary and mandibular tissue specimens with implants were dissected. The maxillary tissue specimens were used to make undecalcified bone grinding slices, and the healing mode of the implant-bone interface was observed by light microscope. The maximum shear strength of loose mandibular specimens was measured by a material testing machine. The differences in group data were statistically analyzed. RESULTS 1) A general difference in shear strength exists among the four groups. The shear strengths in groups A and B were lower than those in groups C and D. 2) No significant difference in healing mode was found between the pressure side and the tension side of the longitudinal-grinded bone slices with implants. Both fiber combination and osseointegration of transverse-grinded slices could be found in the interface in all four groups. The major integration mode of the implant-bone interface of groups A and D was osseointegration, whereas that of the implant-bone interface of groups B and C was fibrous tissue transformation. The surrounding bone tissue regenerated toward the implants of group C.
CONCLUSIONResults show that loading time influences the stability of implant anchorage.

5.Application of refining management in sterilization and supply center

Objective To strengthen the management of contaminated area of sterilization and supply center, increase the cleaning quality of instruments and control nosocomial infection. Methods We executed the following measures: paying attention to the decontamination work, changing concepts, refining regulations, establishing programmed decontamination process, specification of work, thinning pest responsibility, strengthening infection mangement of contaminated area, effect examination and training and checking. Results The bacteria removing rate reached 95.3%. Conclusions Refining management could result in perfect cleaning effect which was applicable in the management of contaminated area of sterilization and supply center.

7.Effect of Xingding Injection on expression of heat shock protein 72 in vascular endothelial cells

OBJECTIVE: To study the effect of Xingding Injection on the expression of heat shock protein 72 (HSP 72) in vascular endothelial cells after ultraviolet radiation. METHODS: Porcine aortic endothelial cells were cultured for 72 hours in culture mediums with different concentrations (0.1, 0.5, 1.0, 5.0, 10 mg/ml) of Xingding Injection. Ultraviolet radiation was administered to the cultured cells for 30 minutes. Western-blot assay was used to measure the expression of HSP 72 in the vascular endothelial cells. RESULTS: There was no expression of HSP 72 in the cultured vascular endothelial cells without ultraviolet radiation, but there was some expression of HSP 72 after ultraviolet radiation. Xingding Injection of different concentrations could significantly improve the expression of HSP 72. The expression of HSP 72 in the vascular endothelial cells cultured in culture medium with 1.0 mg/ml Xingding Injection was the highest, and there was no more increase of expression when the concentration was higher, instead the expression decreased. CONCLUSION: Xingding Injection can protect the vascular endothelial cells from injury during stress. It may be one of its mechanisms in preventing and treating cardio-cerebrovascular disorders.

Objective To evaluate the effect of total parenteral nutrition (TPN) on hyperemesis gravidarum (HG) and pregnancy ending.Methods 27 cases of HG were treated with TPN and 30 cases with the routine infusion to adjust water electrolyte and acid base balance for the purpose of observation of vomiting stopping,time of turning ketonic negative and pregnancy ending.Results After 3 day treatment with TPN,stopping vomiting rate was 55.6%,time of turning ketonic negative was (3.1?0.84) days and neonatal weight was (3.4?0.38) kg,which was 13.3%,(5.2? 1.06) days and (2.9?0.45) kg respectively in control group.There was significant difference between the two groups (P
9.Laparoscopic Ligation and Resection for Interstitial Tubal Pregnancy

Objective To study the efficacy of laparoscopic ligation and resection for the treatment of interstitial tubal pregnancy.Methods Totally 32 patients with interstitial tubal pregnancy were enrolled into this study.The cases were randomly divided into groups A and B(16 in each).Laparoscopic ligation and resection was performed in group A,while the patients in group B received traditional salpingectomy to remove the embryo after local electrical coagulation.The operation time,intraoperative blood loss,rates of conversion to open surgery and postoperative complications,the 24-hour change of ?-hCG,and the recovery time of ?-hCG level were compared between the two groups.Results Compared to the group B,group A had significantly shorter operation time and less blood loss (40?16)min vs(60?19)min,t=-3.178,P=0.004;and(40?12)ml vs(95?26)ml,t=-7.644,P=0.000].The 24-hour change and recovery time of the level of ?-hCG were similar between the two groups (9889?7945)mIU/ml vs(9696?7883)mIU/ml,t=0.068,P=0.946;(10.1?3.2)d vs(9.8?2.4)d,t=0.294,P=0.771].Conclusions Laparoscopic ligation and resection is effective for the treatment of interstitial tubal pregnancy.
10.Clinical study of t-PA and u-PA expression in patients with gastric or intestinal cancer

Purpose:To study the changes of t-PA ?u-PA expression and fibrinolysis molecular mark ers in patients with gastric or intestinal cancer in order to elucidate their cl inical significance. Methods:The plasma levels of t-PA ?u-PA ?u-PAR and PAP were measured by ELISA . Gen e transcription of t-PA?u-PA mRNA were detected by Real-time RT-PCR. Results:The plasma levels of u-PA?u-PAR and PAP were elevated in gastric or intestina l cancer patients ,while u-PA was markedly elevated in patients with local infi ltration?lymph node involvement or distal metastasis. u-PA mRNA was higher and t-PA was lower in gastric or intestinal cancer compared to normal tissue. Conclusions:Hyperfibrinolysis was an important factor related with m etastasis potential of gastric or intestinal cancer. t-PA may be a character of well differentiated tissue.
