2.Resuscitation fluids for the treatment of hemorrhagic shock in dogs:Effects on hemodynamics,oxygen transport and extravascular lung water

Objective:To investigate the effects of resuscitation with different fluids on hemodynamics,oxygen transport and extravascular lung water after acute hemorrhage in dogs. Method:Fifteen dogs were anesthetized and bled to a mean arterial pressure of 4.67 kPa to develop the hemorrhagic shock model.Then the animals were infused with one of three randomly selected fluid:lactated Ringer's solution, blood and polygeline.The terminal point of fluid resuscitation was at the level of PAWP of 2.0 kPa. Hemodynamics, oxygen transport and extravascular lung water values were measured. Result:colloids fluids infusion has more better efficacy and less extravascular lung water than crystalloids fluids infusion in hemorrhagic shock resuscitation,and polygeline infusion could restore hemodynamics better, meanwhile blood infusion could restore oxygen transport better.
3.Clinical value of different surgeries in the treatment of angle closure glaucoma with cataract

AIM: To compare the effects of different surgeries in the treatment of angle closure glaucoma with cataract and the improvement of quality of life.METHODS: The clinical data of 60 cases (60 eyes) with angle closure glaucoma and cataract who were admitted to the hospital between January 2014 and October 2016 were collected.According to the surgical method, they were divided into the control group (simple trabeculectomy, n=28) and the observation group (trabeculectomy combined with phacoemulsification and intraocular lens implantation, n=32).The changes of visual acuity and intraocular pressure before and after surgery were compared between the two groups.The incidence of postoperative complications was statistically analyzed, and the quality of life was compared between the two groups.RESULTS: Before surgery, there was no significant difference in visual acuity, intraocular pressure and anterior chamber depth between the two groups (P>0.05).At 4wk after surgery, the visual acuity of two groups increased and intraocular pressure decreased.The anterior chamber depth of observation group increased while that of the control group decreased (P<0.05).At 4wk after surgery, the visual acuity and anterior chamber depth of observation group were higher than that of the control group but intraocular pressure was lower than that of control group (P<0.05).The success rate of surgery and the opening rate of chamber angle after surgery in the observation group were higher than those in the control group (P>0.05).The incidence of postoperative complications in the observation group was lower than that in the control group (P<0.05).Before surgery, there was no statistically significant difference in quality of life score between the two groups (P>0.05).At 4wk and 3mo after surgery, the scores of the two groups increased (P<0.05), and the quality of life scores of observation group were higher than those of the control group at different time (P< 0.05).CONCLUSION: The success rate of trabeculectomy combined with phacoemulsification and intraocular lens implantation is high in the treatment of angle closure glaucoma with cataract.The regimen can improve the visual function, reduce the incidence of postoperative complications, and improve the quality of life of patients.
5.GROWTH INHIBITION OF COLORECTAL CANCER BY TGF-? ANTISENSE OLIGODEOXYNUCLEOTIDE AND EGFR ANTISENSE OLIGODEOXYNUCLEOTIDE

To investigate the effects of TGF?antisense oligodexynucleotides(ASODN) and EGFR ASODN on human rectal carcinoma cell line HR8348. Cationic liposome, lipofectamine, were selected as delivery vector HR8348 cells were treated with ASODN in vitro. HR8348 cells with or without pretreatment were inoculated into nude mice. The proliferation and DNA synthesis of HR8348 cells treated with TGF-? ASODN and EGFR ASODN were greatly inhibited, and tumorigenic rate was also greatly reduced. TGF-? ASODN and EGFR ASODN could inhibit the proliferation of human colon cancer cell line in vitro as well as in vivo, the results implicate. The potential value in colorectal cancer gene therapy.
6.Effect of Nd:YAG laser treatment for early stage capsular block syndrome

?AIM:To evaluate the clinical outcomes of Nd:YAG laser capsulotomy in the treatment of early stage capsular block syndrome ( CBS) .?METHODS:Eighteen patients (21 eyes) with early stage capsular block syndrome were treated using Nd:YAG laser by anterior capsulotomy only or combined with posterior capsulotomy from January 2010 to July 2015 in Anyang Eye Hospital. Uncorrected distance visual acuity, intraocular pressure, spherical equivalent, depth of anterior chamber were observed preoperatively and 2wk postoperatively.?RESULTS:Seventeen eyes simply underwent peripheral anterior capsulotomy with Nd:YAG laser. Four eyes were combined with posterior capsulotomy. Compared with preoperative, uncorrected distance visual acuity improved, intraocular pressure returned to normal, degree of myopia reduced, depth of anterior chamber had deepened.? CONCLUSION: Nd: YAG laser capsulotomy is an effective treatment for early stage capsular block syndrome.
7.Determination of 10 mycotoxin contaminants in Panax notoginseng by ultra performance liquid chromatography-tandem mass spectrometry.

To ensure the quality and safety of Panax notoginseng, a method for the simultaneous determination of 10 mycotoxins in Panax notoginseng was developed using ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). The sample was extracted with acetonitrile and purified by HLB multifunction cleanup column. The separation was performed on a Phenomenex Kinetex XB-C18 column by gradient elution using methanol and 5 mmol·L(-1) ammonium acetate as mobile phase. The targeted compounds were detected in MRM mode by mass spectrometry with electrospray ionization (ESI) source operated in both positive and negative ionization modes. The linear relationships of the 10 mycotoxins were good in their respective linear ranges. The correlation coefficients (r) ranged from 0.9981 to 1.0000. The LOQs of the 10 mycotoxins were between 0.15 and 8.6 μg·kg(-1). The average recoveries ranged from 73.8% to 107.0% with relative standard deviations (RSDs) of 0.10%-10.9%. The results demonstrated that the proposed method was sensitive and accurate, and suitable for the mycotoxins quantification in Panax notoginseng.

8.Carbamylated erythropoietin promotes vascular microcirculation following cerebral infarction

BACKGROUND: Carbamylated erythropoietin (CEPO) cannot only remarkably promote the prognosis of cerebral infraction, but also improve the microcirculation. OBJECTIVE: To explore the underlying mechanism of CEPO promoting the microcirculation following cerebral infraction. METHODS: 150 Wistar rats were selected, and 120 rats were used for establishing the models of cerebral infarction, followed by allotted into four groups. The model rats were treated with 500, 1000 and 2000 u/kg CEPO as experimental groups, and those received no treatment as model group. The other 30 rats were as controls. Vascular endothelial cells were isolated and cultured in vitro, and the cell proliferation was detected by cell counting kit-8 assay. The expression levels of proliferation-related genes (Ki67 and p16) and vascular endothelial growth factor (VEGF) were detected using western blot assay. After selective silencing of VEGF through RNA interference, all above indicators were detected again. RESULTS AND CONCLUSION: Cell counting kit-8 assay results showed that the proliferation ability of vascular endothelial cells was increased with CEPO concentration increasing. Western blot assay results showed a significant upregulation of Ki67, p16 and VEGF. After shRNA-VEFG interference, these indicators had no positive correlation with the increased concentration of CEPO. Our findings indicate that CEPO can improve the proliferation of vascular endothelial cells in an animal model of cerebral infarction via upregulating the VEGF expression.
9.Therapeutic effect of oral exercise method combined with shaker exercise on dysphagia in patients with cere-bral hemorrhage

Objective To investigate the effect of oral exercise method combined with shaker exercise on dysphagia in patients with cerebral hemorrhage.Methods 112 patients with dysphagia caused by cerebral hemor-rhage were randomly divided into control group (28 cases,conventional treatment),oral exercise method group (28 cases,conventional treatment combined with oral exercise method),shaker exercise group (28 cases,conventional treatment combined with shaker exercise)and combination group (28 cases,conventional treatment combined with oral exercise method and shaker exercise).Four groups were assessed with video fluoroscopic swallowing study (VF-SS)before treatment and after treatment.Results (1)There were no differences on VFSS among four groups before treatment (P >0.05).After treatment,there were significant differences on VFSS among four groups(F =9.40,P <0.05).Combination group[(8.68 ±4.74)points]with the highest score,followed by shaker exercise group[(6.52 ± 3.12)points]and oral exercise method group[(6.43 ±2.84)points],control group[(3.96 ±2.02)points]with the lowest score.There were no differences on VFSS between before treatment and after treatment in control groups (F =0.945,P >0.05).In the remaining groups,the scores of VFSS after treatment were significant higher than that of before treatment (P <0.05).(2)The total effective rate in combination group was higher than that of the remaining groups(χ2 =37.76,P <0.01).Combination group (96.43%)combined with the highest total effective rate,followed by shaker exercise group (75.00%)and oral exercise method group (71.43%),control group (21.42%)combined with the lowest total effective rate.Conclusion The application of oral exercise method combined with shaker exercise on patients with dysphagia caused by cerebral hemorrhage could improve the swallowing function.
10.Influence of aerobic exercise training on serum copeptin level in aged patients with coronary heart dis-ease complicated chronic heart failure

Objective:To explore influence of aerobic exercise training on serum copeptin level in aged patients with coronary heart disease (CHD) complicated chronic heart failure (CHF) .Methods :A total of 200 aged CHD + CHF patients were randomly and equally divided into routine treatment group (received cardiotonic ,diuretic etc .routine therapy) and aerobic exercise group (received aerobic exercise training based on routine therapy) .After six months , serum copeptin level was measured ,cardiac function and quality of life were assessed and compared between two groups .Results:Compared with before treatment after six months ,serum copeptin level significantly reduced in both groups ( P<0.05 all);compared with routine treatment group ,there was significant reduction in serum copep‐tin level [ (10.85 ± 2.66) pmol/ml vs .(9.24 ± 3.82) pmol/ml , P=0.036] in aerobic exercise group .Compared with routine treatment group ,there was significant improvement in cardiac function ,significant rise in 6min walk‐ing distance [ (185.3 ± 51.8) m vs .(386.5 ± 62.4) m] ,and significant reduction in score of Minnesota living with heart failure questionnaire [ (32.5 ± 5.5) scores vs .(21.8 ± 4.2) scores] in aerobic exercise group , P<0.05 both ;no patients suffered from adverse events such as aggravated heart failure and malignant arrhythmia etc .during aero‐bic exercise therapy .Conclusion:On the basis of drug treatment ,aerobic exercise training can reduce serum copeptin level ,improve cardiac function and quality of life in patients with chronic heart failure .
