1.Analysis of nursing scientific research project in China

Objective To analyze the situation of nursing scientific research project in order to provide a basis in promoting nursing research.Methods The literature method was used to analyze the materials officially published from 1999 to 2008 from a number of fund projects about the funding sources,research content and undertakers.And the results were compared with scientific project in other medical field.Results The results showed that the number of nursing research project had an increasing trend year by year.Sources of funding scientific research were being broadened.The pattern which hospital as the main research pattern had been broken.The nursing research projects had been covering a wide range,but teaching reform project was still make up the main body.Conclusions We should deepen the reform of nursing education,strengthen innovation and awareness,carry out horizontal joint,to promote the combination of theory and clinical practice,play a team advantage to avoid doing it alone,actively carry out scientific research and encourage policy in a bid to promote nursing research to sustainable development.
2.Comparison of effects of different enteric-coated aspirin tablets on upper gastrointestinal tract in the elderly

Objective To study the effects of different enteric-coated aspirin tablets on upper gastrointestinal tract in the elderly. Methods 404 inpatients aged more than 65 years in last three years who took enteric-coated aspirin tablets (100mg/d) for more than three months were analyzed.They were divided into two groups: bayaspirin-treated group (232 cases) and control group with ordinary enteric-coated aspirin tablet treatment (172 cases). The clinical data and the results of gastroseopy were compared between two groups, and the effects of two kinds of enteric-coated aspirin tablets on upper gastrointestinal tract in the elderly were analyzed by Chi-square test. Results The upper gastrointestinal bleeding rates were 20.3% in Bayaspirin-treated group (47/232) and 32.0% in control group (55/172), and there was significant difference between two groups(χ2=7.19,P<0.01)Gastroscopy indicated that gastroduodenal inflammation occurred in 16 patients (6.9% and peptic ulcer occurred in 8 patients (3.5%) in bayaspirin-treated group, while gastroduodenal inflammation occurred in 12 patients (7.0%) and peptic ulcer occurred in 36 patients(20.9%)in control group. The difference of the incidence of peptic ulcer between two groups had statistical significance(χ2= 31.10,P<0.01). The incidences of gastrointestinal adverse reactions were 8.6% (20/232) in bayaspirin-treated group and 23.3% (40/172) in control group, and there was a statistical difference between two groups. Conclusions Compared with ordinary enteric-coated aspirin tablet, bayaspirin has less adverse effects on upper gastrointestinal tract in the elderly and is more safe for treatment.
3.Construction of a core competency training indicator system for high-level public health talents

Objective:
To construct a core competency training indicator system for high-level public health talents using the Delphi method, so as to provide insights into optimization of high-level public health talents training.
Methods:
Based on review of publications pertaining to core competency training for high-level public health talents, in combination with public health priorities and development needs in China, a preliminary indicator system was constructed. Twenty public health experts were invited for two rounds of Delphi expert consultation. Indicators were screened based on the frequency of full marks, mean score and coefficient of variation (CV), and the weight of indicators was determined using the Entropy weight method. The effectiveness of the Delphi expert consultation was evaluated using the active degree, authority coefficient and the consistency degree.
Results:
Twenty experts participated in the consultation, including 12 men, and there were 6 experts at ages of 30 to 39 years, 8 at ages of 40 to 49 years, and 6 at ages of 50 years and older. There were 17 experts with an educational level of master and above, 19 with senior professional titles and all experts had working experiences for 10 years and longer. The active degrees were 95.24% and 100.00% for two rounds of consultations, and the overall authority coefficient was 0.87. Following two rounds of consultations, all CVs were reduced to below 0.25, and the coordination coefficients were all statistically significant (P<0.05), with a higher coordination coefficient in the second round of consultation than in the first round. The constructed core competency training indicator system for high-level public health talents included four primary indicators, including health protection, health service improvement, health promotion, and essential quality, with weights of 0.388, 0.310, 0.122 and 0.180, 11 secondary indicators, with high weights seen for protection of human health from threats (0.178), monitoring and assessment of health service utilization (0.157) and promotion of health service quality and access (0.112), and 70 tertiary indicators, with high weights seen for Chinese writing (0.038), capability for health policy suggestions (0.034) and global perspective (0.030).
Conclusion
The core competency training indicator system for high-level public health talents constructed in this study may provide insights into training of high-level public health talents.
4.A study of downregulation of miR-93 on the suppression of human glioma cell growth and invasion

ObjectiveTo confirm the effect of miR-93 inhibitor in glioma cell growth and invasion.MethodsMalignant glioma cells were transfected with miR-93 inhibitor by lipofectamin to downregulate their overexpression of miR-93.Real time-PCR was taken to measure miR-93 expression after transfection.The cell cycle kinetics and cell growth rate were detected by flowcytometry and MTT assay,the cell proliferative ability was evaluated by soft agar assay,and the invasive ability was detected by transwell assay.ResultsThe highlevel expression of miR-93 was downregulated effectively in glioma cells after transfecting the miR-93 inhibitor.Meanwhile,the cell cycle progress was delayed,S phase cells were reduced,the speed of growth was slowed,cloning formation ability was receded,the number of cells through the matrigel was reduced,and invasive ability was significantly repressed.ConclusionDownregulation of miR-93 expression could inhibit the proliferative ability and invasive ability of glioma cells.
5.Comparison of panoramic radiography and cone-beam computed tomography images for detecting external root resorption of mandibular second molar associated with third molar impaction

Objective:
To compare the diagnostic performance between panoramic radiography(PR)and cone beam computed tomography(CBCT)in the assessment of external root resorption(ERR)of mandibular second molars associated with impacted third molars.
Methods:
A total number of 832 patients with 1 074 mesially and horizontally impacted mandibular third molars treated at our institution from January 2019 to December 2020 were retrospectively analyzed. Presence of ERR on the adjacent second molar was investigated with PR and CBCT. Factors affecting the diagnostic accuracy of PR were determined.
Results:
The overall incidence of ERR in second molars was 33.15%(356/1 074)as detected by CBCT images. The accuracy of PR was 66.39%. Multivariate Logistic regression analyses further revealed that middle and Class Ⅲ impaction, crown contact or overlap with the root of adjacent tooth were risk factors for inaccurate diagnosis of PR(P<0.05).
Conclusion
The accuracy of detection on ERR of mandibular second molar associated with impacted third molar using panoramic radiography is lower. CBCT is recommended for this clinical scenario.
6.Influence of high-fat diet in intestinal flora and fecal weight in SD rats and its significance

Objective To investigate the influence of high-fat diet in the intestinal flora,fecal weight and its water content in rats, and to clarify the effect and significance of high-fat diet in the occurrence of obesity forming. Methods Twenty Sprague-Dawley (SD)rats were randomly divided into normal diet (ND)group and high-fat diet (HF)group (n=10).The rats in ND group were fed with normal diet,and the rats in HF group were fed with diet rich in oil and fat.The fresh feces were collected separately on days 1,15,30,and 49 for analysis of weight, water content,and intestinal flora.Results On the 49th day,the wet weight and water content of feces of the rats in ND group were (6.61 ± 0.17)g and (37.07 ± 3.04)%,respectively,while those in HF group were (4.46±0.30)g and (18.04±2.23)% (P<0.05).Compared with ND group ,the fecal pellets in HF group were increased obviously from the 7th day (P<0.05).There were obvious changes in intestinal microbial populations of HF group. The counts of enterococci, bifidobacteria, Lactobacillus, Bacteroides bacteria were significantly decreased on the day 49, but the count of Escherichia coli was increased significantly (P < 0.05 ). Conclusion High-fat diet can result in decrease for weight,water content,and pellets of feces;it can change the structure of intestinal flora. As result, there is a possibility that all of changes above can promote obesity in the future.
7.Influence of PET/CT positioning on the effect of late-course accelerated hyperfractionated and threc-dimensional conformal radiotherapy for stage Ⅲ lung squamous cell carcinoma

Objective To investigate the irfluence of PET/CT positioning on the effect of late-course accelerated hyperfractionated and three-dimensional conformal radiotherapy for stage Ⅲ lung squamous cell carcinoma.Methods 79 stage Ⅲ lung squarnous cell carcinoma patients were enrolled and divided into PET/CT group(n =37 cases,PET/CT positioning) and general CT group (n =42 cases,general CT positioning) according to the way of positioning.The conventional fractionated 3 DCRT( about 40 Gy) was used in previous 2/3 time and hyperfractionated( about 1.5 Gy) was used in later 1/3 time.Both groups used chemotherapy as adjuvant therapy.Full course of the treatment was a month or so.And the clinical efficacy and a variety of adverse reactions of the two groups was observed.Results The total rate of remission in PET/CT group were 73.0%,and 71.4% in general CT group,and there was no significant difference in the two groups (x2 =1.347,P > 0.05 ).The major radiation injury were radioactive pulmonary injury and tracheal damage,incidence rates were 56.8%and 57.1% in acute stage,and there was no significant difference in the two groups( x2 =2.178,P > 0.05 ) ; but in late stage 69.0% in general CT group was significantly higher than 62.2% in PET/CT group(x2 =4.142,P <0.05).Ater followed-up 1 year,the recurrence rate of hillar and mediastinal lymph node in PET/CT group was 43.7%,lower than that of the general CT group( 59.4% ) ( x2 =4.732,P < 0.05 ).Conclusion PET/CT positioning in late-course accelerated hyperfractionated and three-dimensional conformal radiotherapy for stage Ⅲ lung squamous cell carcinoma could reduce late stage radioactive pulmonary injury and tracheal damage,lower the recurrence rate of hillar and mediastinal lymph node.
8.Regulation of miR-19a and miR-19b on tumor suppressor gene-SEPT7

objective To identify SEPT7as one of the target genes of miR-19a and miR-19b.MethodsmiR-19a inhibitor and miR-19b inhibitor mediated by lipofectamine2000,were transfected to SNB19 glioma cells for knocking down miR-19a/19b overexpression.Real time PCR was conducted to detect the expression of miR-19a/miR-19b in transfected cells.The expression of SEPT7was determined by Western blot analysis.RT-PCR was used to detect the mRNA expression of SEPT7; and Luciferase reporter assay was used to identify the direct regulation of miR-19a/19b on SEPT7.ResultsIn SNB19 glioma cells transfected with miR-19a/19b inhibitor,the expression of miR-19a/miR-19b was significantly reduced,whereas the protein expression of SEPT7 was upreguhtted; no significant change of SEPT7 mRNA level was found and luciferase activity became stronger as compared to control cells.ConclusionSEPT7 is the target negatively regulated by miR-19a and miR-19b.
9.Yes-associated protein modulation of human glioma cell growth invitro

Objective:This study aimed to explore the effect of Yes-associated protein (YAP) on the growth of the human glioma cell line LN229. Methods:YAPsiRNA was transfected into LN229 cells to knock down the YAP expression. The downregulation of the YAP expression was identified through Western Blot analysis. Colorimetric assay using methyl-thiazolyl-tetrazolium was applied to evaluate cell proliferation ability. Cell invasive activity was examined using Transwell assay. Flow cytometry and AnnexinV were used to detect cell cycle and apoptosis, respectively. The relevant molecules regulating proliferation, invasion, cell cycle progression, and apoptosis were examined through Western Blot analysis. Results:The YAP expression was downregulated after YAPsiRNA was trans-fected into LN229 glioma cells. Reduced YAP expression could arrest the cell cycle at G0/G1 phase, inhibit cell proliferation and inva-sion, and promote apoptosis. The expression of the proliferating cell nuclear antigen (Ki-67), matrix metallopeptidase-9 (MMP-9), cy-clin D1, and Bcl-2 were downregulated. Conclusion:The downregulation of YAP in LN229 cells suppresses cell proliferation and inva-sion, as well as promotes cell apoptosis. This study provides a novel evidence for further study on Hippo-YAP signal pathway in molec-ular pathology of glioma.
10.Genetic mapping and mutation analysis in a family with paroxysmal kinesigenic dystonia

Objective To study the clinical characteristics and genetic cause of a Chinese family affected with paroxysmal kinesigenic dystonia(PKD).Methods The detailed clinical data and the blood samples of the affected patients with PKD and their relatives were collected.After genomic DNA was extracted from blood leukocytes,target linkage analysis Was performed using multiplex PCR by microsatellite marker's located in the reported critical region on chromosome 16.All exons and flanking regions of SCNN1G and ITGAL genes were amplified by PCR-sequence.Results In this three-generation 12 member family,5 individuals have been diagnosed as PKD.Target linkage analysis suggested the disease gene linked to chromosome 16.between D16S3396 and D16S3057 with two-point LOD score of 1.47 at recombination fraction(θ)=0.0.All affected individuals shared a common haplotype which co-segregated with the phenotype.Except for 8 reported SNPs,no pathologic sequence variants were found in candidate genes SCNN1G and ITGAL.Conclusions The studied family is genetically linked to the reported critical locus of PKD on chromosome 16.SCNN1G and ITGAL were ruled out as the causative genes for the studied pedigree.Further genetic analysis in this family may reveal new genetic cause responsible for PKD.
