1.Optimization for condition of adsorbing hepatitis A virus antigen with aluminum hydroxide adjuvant and evaluation of immunogenicity of adsorption products

Objective To optimize the adsorption condition of hepatitis A virus(HAV)antigen by aluminum hydroxide[Al(OH)_3]adjuvant and evaluate the immunogenicity of the adsorbed products. Methods The final molar concentration of buffer(0. 01,0. 02 and 0. 04 mol/L),pH value(5. 5,6. 0,6. 5,7. 0,7. 5 and 8. 0),final molar concentration of sodium chloride(NaCl)(0. 075,0. 13,0. 15,0. 17,0. 45,0. 75 and 1. 0 mol/L),mixing mode(mixing after dilution and mixing directly without dilution)and dropping order[HAV stock solution into Al(OH)_3 solution,and Al(OH)_3 solution into HAV stock solution]were selected for single factor screening. Taking Al(OH)_3 adsorption rate as the evaluation index,the selected factors were optimized by orthogonal test. NIH mice were immunized intraperitoneally with adsorbed products prepared under optimal and suboptimal combination conditions with 0. 5 mL/mouse,10 mice in each group with half male and half female. After 28 d of immunization,the blood samples were collected from eyeball or heart,and the serum was separated. The HAV antibody level was detected by ELISA,and the immunogenicity of adsorbed products was evaluated by the seroconversion rate of mouse serum antibody. Results The results of single factor screening test showed that the final molar concentration of buffer,pH value and final molar concentration of NaCl had significant influence on the adsorption effect(F = 6. 582,14. 007 and 8. 612,respectively,each P < 0. 05),while the mixing mode and dropping order had no effect(t =-0. 696 and 1. 153,respectively,each P > 0. 05). The final molar concentration of buffer was 0. 01 and 0. 02 mol/L,the pH value was 5. 5 ~ 7. 0,and the final molar concentration of NaCl was 0. 13 ~ 0. 17 mol/L. The orthogonal test showed that the order of the influence on adsorption effect was the final molar concentration of buffer > pH value > NaCl final molar concentration. The optimal combination was pH 6. 3 + final molar concentration of buffer 0. 01 mol/L + NaCl molar final concentration 0. 15 mol/L,and the suboptimal combination was pH 5. 8 + final molar concentration of buffer 0. 02 mol/L +NaCl final molar concentration 0. 17 mol/L. The seroconversion rate of serum antibody in mice immunized with the adsorbed products prepared by the optimal and suboptimal combination was both 100%. Conclusion The adsorbed products prepared under the optimized conditions have good immunogenicity,which lays a foundation of the research of inactivated hepatitis A vaccine
2.The Functions of the Middle Ears of Children with Adenoid Hypertrophy

Objective To explore the influence of adenoid hypertrophy on the state of the middle ear in children.Methods Two hundred and seventy two adenoid hypertrophy patients,aged from 2 to 12 years old,were examined with otoscopy,tympanometry and temporal bone computered tomography before adenoidectomy.The average age of the patients was 6.3 years old.These patients were divided into two groups:Group A contained 94 normal hearing patients(188 ears),Group B 178 patients(356 ears) containing of hearing loss.Results Out of the temporal bone computed tomography (CT) revealed that 209 patients(396 ears) had middle ear effusions in(72.79%,396/544ears),and 37 patients(65/188 ears,34.57%) hydrotympanum in Group A and 172 patients(331/356 ears,92.98%) in Group B.All the 396 ears effusion were confirmed by operation.Three hundred seventy three ears effusion were confirmed by the CT and operation in the two groups,377 ears with B type tympanogram(373/377,98.94%),93.85%(61/65 ears) in Group A,100%(312/312 ears) in Group B,respectively.The positive predictive value for middle ear effusion of the B-type tympanogram was higher in Group B than in Group A(P<0.01).Fourteen ears with peak pressure <-200 daPa revealed hydrotyrnpanum and 23 ears with normal acoustic stapedius reflex revealed no fluid by CT and operation in 73 ears with C-type tympanogram.CT also revealed two patients suffered from large vestibular aqueduct syndrome and one with cochlear malformation in Group B.Conclusion Type B tracing tympanogram has a high positive predictive value for middle ear effusion in adenoid hypertrophy children.Type C tympanogram could not exclude effusion when the peak pressure is<-200 daPa.CT is the best tool for identifying hydrotympanum.
3.Analysis of the therapeutic effect of interventional embolization and surgical clipping for the treatment of posterior communicating artery aneurysms

Objective To analyze the complications and clinical effects of interventional embolization and surgical clipping for the treatment of posterior communicating artery aneurysms, and to compare the results between the two methods.Methods A total of 90 patients with confirmed posterior communicating artery aneurysm, who were admitted to the Second Affiliated Hospital of Zhengzhou University during the period from August 2013 to March 2015, were enrolled in this study. Of the 90 patients, 49 received interventional embolization therapy (interventional embolization group) and 41 underwent surgical clipping treatment (surgical clipping group). Before the treatment the patient's condition was evaluated according to Hunt-Hess classification, after the treatment the therapeutic effect was assessed with Glasgow prognosis scale (GOS). The postoperative GOS scores and complications in patients with different Hunt-Hess classification were compared between the two groups. Results In patients of Hunt-Hess 0-Ⅲ grade, no statistically significant differences in postoperative GOS scores existed between the interventional embolization group and the surgical clipping group (t=0.842,P>0.05), while the postoperative GOS scores in patients of Hunt-HessⅣgrade of the interventional embolization group was remarkably higher than that in patients of Hunt-HessⅣgrade of the surgical clipping group, the difference was statistically significant (t=1.713,P<0.05). The incidence of complications in the interventional embolization group was significantly lower than that in the surgical clipping group (x2=1.036,P<0.05). Conclusion For the treatment of Hunt-Hess 0-Ⅲgrade posterior communicating artery aneurysms, the interventional embolization and the surgical clipping show no difference in their therapeutic effects; while for the treatment of Hunt-Hess Ⅳ grade posterior communicating artery aneurysms, the interventional embolization in superior to the surgical clipping, as the interventional embolization carries lower operation risk and complication incidence, and it also has reliable effect.

Objecave The effects of PGF2α on the reactive oxygen species generation and cardiomyocyte hypertrophy were examined in experiments on the cultured neonatal rat cardiomyocytes.It is to study the role of ROS in the signaling pathway of cardiomyocyte hypertrophy induced by PGF2α.Methods The level of intracellular ROS wag measured by the ROS-specific probe 2',7'-dichlorofluorescin diacetate (DCF-DA).Cardiomyocyte hypertrophy was determined by total protein content of the cells and the cell diameter.Results In the cardiomyocytes treated with PGF2α(1nmol/l,10nmol/l,100nmol/l),the fluorescence intensity of intracellular DCF-DA increased by 38.99%,61.76% and 93.55% respectively compared with control group(F=195.69,P<0.01).It is indicated that PGF2α can induce intracellular ROS generation on the cultured neonatal rat cardiomyocytes in a dose-dependent manner.Compared with control group,the total protein increased by 39.51%, 69.93%and 139.06%respectively(F=74.014,P<0.01),and the cell diameter increased by 29.02%,60.79%and 127.40%respectively(F=721.02,P<0.01).It is indicated that PGF2α induce cardiomyocyte hypertrophy on the cultured neonatal rat cardiomyocytes in a dose-dependent manner.Conclusion PGF2α can induce intracellular ROS generation and cardiomyocyte hypertrophy on the cultured neonatal rat cardiomyocytes in a dose-dependent manner.
6.Effect of Extract of Ginkgo Biloba(EGb)on Cell of Neonatal Rat Cardiocyte Hypertrophy Induced by Prostaglandin F2 Alpha

Mandibular molar loss, which is in a high ratio, is found accompanying with alveolar bone resorption frequently while being restored in clinic. Therefore, due to the existence of mandibular neural canal, the success ratio of implant denture in this area can be directly affected by the height reduction of alveolar bone. In clinic, there are some achievements obtained to improve the survival rate of implant denture, such as increasing the implant support area, enhancing alveolar bone height through surgery, optimizing the design of the superior part of implant. This article summarizes the research progress on the mandibular molar implant denture with alveolar bone absorbed and aims to search a selective design of mandibular molar implant denture under the high absorption of alveolar bone.

10.Protection of Grateloupia filicina polysaccharide against hepatotoxicity induced by Dioscorea bulbifera L.

The present study was designed to observe the protection of Grateloupia filicina polysaccharide (GFP) against hepatotoxicity induced by Dioscorea bulbifera L in mice and its underlying mechanism. GFP was intragastrically (ig) given to mice at various doses. After 6 days, the mice were treated with ethyl acetate extract of Dioscorea bulbifera L (EF, ig). Serum levels of alanine/aspartate aminotransferase (ALT/AST), alkaline phosphatase (ALP), total bilirubin (TB) were measured, and liver histological evaluation was conducted. Furthermore, reductions of liver glutathione (GSH) amount and glutamate cysteine ligase (GCL) activity were tested. The expressions of GCL-c, GCL-m, and HO-1 (heme oxygenase-1) in liver were observed by Western-blot. The results showed that GFP (600 mg x kg(-1)) decreased EF-induced the increase of serum ALT, AST and TB, and GFP (400, 600 mg x kg(-1)) inhibited EF-induced the increase of serum ALP. Liver histological evaluation showed that the liver injury induced by EF was relieved after treated with GFP. GFP further increased liver GSH amount and reversed EF-induced the decrease of GCL activity. The Western-blot result showed that GFP augmented EF-induced the increase of HO-1, and reversed EF-induced the decrease of GCL-c. In conclusion, GFP can act against the oxidative stress liver injury induced by Dioscorea bulbifera L in mice.
