1.Development of Multi-function Blooding Device for Blood Sucking Insects

Objective To develop a blooding device for blood sucking insects. It's designed for researches, such as artificial breeding of indoor medium insects, evaluation of vermicides'effect, and artificial infection of pathogenic microorganisms. Methods This device was developed with AT89C51 MCU. Results The desired instrument could provide blood which could be adjusted from 30℃to 38℃. The error of temperature could be controlled within 0.5℃. Conclusion The instrument is easily-operated and stable. It can replace the artificial blood-breeding.
2.Effect of Herba Houttuyniae on Urinary Albumin and Insulin Resistance of Diabetes Mellitus Rats

Objective To observe the effect of Herba Houttuyniae(HH) on urinary albumin and insulin resistance of diabetes mellitus(DM) rats.Methods Three rat model of diabetes mellitus was induced with streptozotocin(STZ) and high glucose-lipoids animal feeds,then treated with HH for 8 weeks.The change of biochemical parameters such as 24-h urine volume,urinary albumin,fasting insulin(FINS) level,fasting glucose and insulin sensitivity index were observed.Results After treatment,FINS level was lower in HH group than that in the model group,rosiglitazone group and losartan group(P

4.Reconstruction surgery of knee by using free patella grafts to repair block defects

[Objective]To discuss the clinical results of reconstruction of block bone deficit knee(lower femur and upper tibia) by transplantation of patella and present surgery protocols.[Methods]There were 7 patients who have giant cell tumor of bone in the lower femur or upper tibia at the knee in this report.The case history was from 4 to 12 months,average 5.8 months.Four were male and three were female.The age ranged from 18 to 40 years,average 31.8 years.Four had lesions at the lower femur and the other three had lesions at the upper tibia.According to Ennecking grade,five cases were in grade 2 and two cases were in grade 3.When the cartilage of the knee was damaged by bone tumors,the author supplemented it by autogenous patella to fill the defect.[Results]All cases of patella grafts were presented,with follow-up from 3 to 10 years(average 4.9 years),three cases were followed up more than 5 years.During the follow up,there was no case with recurrence、metastasis or necrosis and infection of graft.All the grafts had good union.According to the function evaluation standard of the international protecting extremities institute,3 excellent,4 good.The main complications were less range of movement of the knee(movement range 40~110 degree,average 88 degree).Weakness of the extensor apparatus was not obvious.One case had slight collapse of tibia plateau and another case sufferd from joint pain 6 years after operation.[Conclusion]When the cartilage of the lower femur and the upper tibia is damaged by bone tumors,the bone tumors and its affected condyle cowld be incised,then apply patella grafts to repair and reconstruct the defect of the knee.
5.The effect of captopril on fibrinolytic system during the intimal hyperplasia after arterial balloon injury

Objective To study the effect of captopril on fibrinolytic system during the arterial intimal hyperplasia after ballon injury and the prevention of restenosis after transluminal angioplasty. Methods 36 rabbits were divided randomly into three groups:sham operation group (control group),injury group,medical treatment group (captopril+injury) each n =12.Captopril (2 mg?kg 1 ?d 1 p.o.) was given to 12 rabbits in medical treatment group from 1 day before to 30 days efter balloon injury.The injury group,12 rabbits underwent same ballon injury without medication.The tissue plasminogen activator (tPA),plasminogen activator inhibitor 1 (PAI 1) antigen level were measured by ELISA before,and 1,3,5,7,14,30 days after vascular injury.30 days after angioplasty pathomorphological examination was carried out.To observe the intimal thickness and extent of lumen stenosis.Results (1) The PAI 1 levels in the injury group were significantly higher than those in the medical treatment groups.(2)The intimal thickness and extent of lumen stenosis in medical treatment group was significantly lower than that in injury gropu.Conclusion Impaired fibrinolysis after ballon injured artery affect the repair process after vascular injury,which leads to restenosis.Captopril can prevent restenosis after angioplasty.
6.Inhibition of HER-2 expression by RNA interference in osteosarcoma cells

[Objective]To investigate whether RNA interference(RNAi)could suppress the expression of HER-2 in osteosarcoma OS-732 cells.[Method]OS-732 cells were transsfected using chemically synthesized double stranded RNA(dsRNA)formulated with Lipofectamine 2000.The HER-2 gene expression inhibition was assessed by RT-PCR,Western Blot and immunocytochemistry.The chemosensitivity of transfected cells to cisplatin was determined by MTT measurement.[Result]Sequence specific siRNAs targeting HER-2 down-regulated the expression of HER-2 significantly.Currently,a range of effects on cell physiology,such as growth inhibition or apoptosis,was observed,Introduction of the siRNA into HER-2 positive tumor lines in vitro greatly reduced the expression of the HER-2 mRNA,effects lasted for 10 days,On the 9th day of interference,HER-2 protein expression was down-regulated in interference group.After RNAi,HER-2-siRNA increased thesensitivity of OS-732 to cisplatin by 10-fold.[Conclusion]Sequence specific siRNAs targeting HER-2 can suppress the expression of HER-2,and significantly inhibiting cellular proliferation,increasing the sensitivity of OS-732 to eisplatin.The successful application of HER-2 siRNA for inhibition of proliferation in HER-2 overexpressed cells extends the list of available therapeutic modalities in the treatment of human osteosarcoma.

[Objective] To investigate the relationship between Survivin,Caspase-3 and cell apoptosis in osteosarcoma.[Method]The expression of Survivin and Caspase-3 in osteosarcoma were detected by immunohistoehemistry.The apoptosis of osteosarcoma cell was assessed by terminal deoxynucleotide transferase mediated dUTP nick end labeling(TUNEL).[Result]The results showed that Survivin expression was detected 77% in osteosarcoma tissues and was significantly higher than in the normal tissues.Caspase-3 was detected in 26 of 40(65%)osteosarcoma tissues and in 34 of 38(89%)in normal bone tissues,with significant difference(P

9.Application of Endoscopic Pancreatic Duct Stent Insertion: A Report of 20 cases

Objective To investigate the clinical effects of endoscopic pancreatic duct stent insertion.Methods The clinical data of 20 cases of endoscopic pancreatic stent insertion,including 12 cases of chronic pancreatitis,3 cases of recurrent pancreatitis,2 cases of papilla adenoma after resection,3 cases of pancreatic carcinoma,was retrospectively reviewed from June 2004 to September 2006.All the patients underwent endoscopic pancreatic sphincterotomy and pancreatic stent insertion after endoscopic retrograde cholangiopancreatography(ERCP).Results All the 20 patients received 23 procedures of pancreatic stent insertion.Stent-preserving time was 25-105 days(mean,64.5 days).2 cases experienced stent migration and no postoperative pancreatitis occurred.20 patients were followed up for 6 to 12 months,including 17 patients for 12 months.During a follow-up period of 3 months in the 12 cases of chronic pancreatitis,1 case received repair because of duodenal perforation induced by ERCP,and the symptoms of abdominal pain or discomfort disappeared or were distinctively relieved in 9 and 2 cases,respectively,with weight gain 2-5 kg(mean,3.2 kg).A follow-up period of more than 3 months showed that the remission rate of abdominal pain was 92%(11/12).The jaundice was alleviated(with the average of decrease of 61.8 percent of total bilirubin in serum) and weight gained in the 3 cases of pancreatic carcinoma after the biliary and pancreatic duct plastic stents were inserted at the same time.Patients with recurrent pancreatitis had no relapse in the follow-up period.Conclusions The endoscopic pancreatic duct stent insertion is a safe and minimal invasive procedure,and can prevent postoperative pancreatitis,but the main complication of stent migration should be considered.
10.Teratogenecity of Ethylene Oxide Sterilized Medical Devices on Cells Used for Biotherapy

OBJECTIVE To observe the teratogenecity of ethylene oxide sterilized materials to cells which are used for biotherapy.METHODS Three kinds of cells were cultivated in ethylene oxide sterilized culture flasks which were conserved for different time and high temperature/high pressure sterilized vitric culture flasks.NK/CIK cells induced from cord blood,human mesenchymal stem cell and CD34+ cells were cultivated in ethylene oxide sterilized culture flasks which were conserved for one day(group A),for three months(group B),for more than six months(group C) and cultivated in high temperature/high pressure sterilized vitric culture flasks(group D).After ten days,not only the growth and morphology of cells but also the breakages fragmentations and gaps in chromosomes and replication of chromosome in nucleus were observed.According to these data,the malformation rate was calculated.RESULTS Compared with groups C and D,the inductivity of groups A and B obviously degraded(P0.05).CONCLUSIONS Compared with the control group,the materials which were sterilized by ethylene oxide and conserved for less than one day and three months can bring damage to cells.However,the damage is not discovered in the materials which were conserved for more than six months.Ethylene oxide can induce cell aberrant and mutation.Therefore,it is suggests that materials be conserved for more than six months to ensure safe and effective enforcement of biotherapy.
