1.Advances of citrin deficiency

Citrin deficiency,caused by mutations in SLC25A13,is an autosomal recessive genetic disorder with two age-related phenotypes:adult-oneset type Ⅱ citrullinemia and neonatal intrahepatic cholestasis.Recently,it has been found mostly in individuals of East Asian ancestry.In south China,there is a high mutation carrier frequency especially.There is still a lack of criteria for clinical or biochemical diagnosis of this disease,gene analysis is the main basis of the current diagnosis consequently.Surging number of case reports indicate that Citrin deficiency is not a self-limited disease.Early diagnosis and proper treatments may improve the prognosis.This paper focuses on the current researches in order to make further comprehensiom.
2.Clinical efficacy of Xuezhikang maintenance treatment for senile hyperlipidemia

Objective To observe the clinical curative effect of Xuezhikang maintaining treatment for senile hyperlipidemia.Methods According to the digital table,96 elderly patients with hyperlipidemia were selected and randomly divided into the three groups.The control group 1 with 32 cases took orally rosuvastatin before sleeping 10mg per time,1 time /day,12 weeks one treatment course.The control group 2 with 32 cases took orally Xuezhikang capsule before sleeping 0.6g 2 times/day,12 weeks one treatment course.The treatment group with 32 cases took first orally rosuvastatin before sleeping 10 mg/time,1 time /day for 4 weeks,then changed to use Xuezhikang capsule 0.6g 2 times/day for six weeks.The total treatment course was 12 weeks.Results After treatment,TC,TG,LDL-C in three groups all declined significantly and HDL-C increased.The curative effect was similar in treatment group and control group 1 with no statistical difference(P >0.05).Conclusion Xuezhikang has similar efficacy with rosuvastatin 10 mg/time in hyperlipidemia,and recommand Xuezhikang clinical use for its lower price.
3.Effect of Fish-oil fatty emulsion on immunity of postoperative patients with malignancy tumors

Objective: To study the effect of fish-oil fatty emulsion on immunity of post-operation patients with malignancy tumors.Methods: Forty-three post-operation patients with malignancy tumors were randomly divided into control group and study group and both groups were provided with parenteral nutrition treatment [104.6 kJ(25 kcal/kg)].Fish-oil fatty emulsion was given to the study group.The level of serum albumin,immunity function,jaundice index,aminotransferase,blood lipid and morbidity rate of infectious complications of the two groups were compared.Results: After five days of post-operation treatment,in the study group,the immunity function was significantly improved and morbidity rate of infectious complications declined.The levels of serum albumin,the blood lipid,jaundice index,and aminotransferase had no significant difference between the two groups.Conclution:Fish-oil fatty emulsion is beneficial to the immunity function and can emilsion decrease infectious complications of post-operation patients with malignancy tumors.

7.Determination of Dolasetron Mesylate by HPLC

OBJECTIVE:To establish a HPLC method for the determination of dolastron mesylate.METHODS:The sample was separated on Diamonsil C18 column.The mobile phase consisted of acetonitrile-water-1mol? L-1 ammonium formate(450∶ 440∶ 110)(pH=8.0 adjusted with trithylamine)with a flow rate of 1.0mL? min-1.The UV detection wavelength was set at 285nm and the sample size was 20? L.RESULTS:The linear range of dolasetron mesylate was 24~ 56? g? min-1(r=0.999 6),with average recovery at 99.7%(RSD=0.74%).CONCLUSION:The established is simple,sensitive and reproducible,and suitable for the quality control of dolasetron mesylate.
8.Effects of Wuling Powder on Insulin Resistance of Mice Induced by High Lipid Diet

Objective To investigate the effects of Wuling Powder on insulin resistance of C57BL/6J mice induced by high lipid diet, and discuss the mechanism. Methods C57BL/6J mice were randomly divided into six groups:normal group, model group, rosiglitazone group, Wuling Powder in low, middle, and high dose groups, 10 mice per group. Besides the normal group, other five groups were fed with high fat and sugar diet, with a purpose to establish insulin resistance model. Normal group and model group were given pure water. Rosiglitazone group received a gavage with rosiglitazone of 0.75 mg/kg. Wuling Power low, middle, and high groups received gavage with Wuling Power of 1.23, 3.69, 11.07 g/kg, respectively, the does volume was 0.2 mL/10 g, once a day. The weight and abdominal girth were detected every week. At the end of the sixth week, mice were given 12-hour fasting, and their eyeball were taken for blood. The body weight, length, and fat in abdomen and both kidneys were detected. Paraffin section was made with HE staining. FPG and FINS of each group were detected. ISI and IRI were calculated, and TC and TG were detected. Results Compared with the model group, Wuling Powder can significantly reduce the body weight and abdominal girth of mice (P<0.01, P<0.001), improve liver fatty degeneration, lower the FPG, FINS, TCH, TG, IRI, and increase the ISI in mice (P<0.05, P<0.01). Conclusions Wuling Powder has the effect of preventing insulin resistance of C57BL/6J mice induced by high lipid diet.
9.Differentiation of rat bone marrow mesenchymal stem cells into neuron-like cells induced by astragalus mongholicus

BACKGROUND: In vitro cultured bone marrow mesenchymal stem cells (BMSCs) can differentiate into neural cells. Some inductors are poisonous and cannot be used in human. OBJECTIVE: To induce the differentiation of rat BMSCs into neuron-like cells with astragalus mongholiens. DESIGN, TIME AND SETTING: The randomized control experiment was performed at the Yunnan Key Laboratory of Faculty of Basic Medicine of Dali University and Stem Cell Research Center of College of Preclinical Medicine and Forensic.Medicine of Sichuan University from January 2002 to January 2005. MATERIALS: Healthy SD rats aged 6-weeks old and weighing 120-130 g were used in this study. METHODS: BMSCs were collected from rat bone marrow by density gradient centrifugation, and then cultured. BMSCs were incubated in serum-free L-DMEM medium containing 0.125 volume fraction of astragalus mongholicus to observe morphologic changes in differentiated cells. Expression of the nestin, neuron specific enolase (NSE) and glial fibrillary acidic protein (GFAP) was detected by immunohistochemistry. MAIN OUTCOME MEASURES: Immunohistochemistry of differentiated cells; The expression of Wnt-1 gene and Ngn-1 gene were detected by semi-quantitative reverse transcdption-polymerase chain reaction during cell differentiation. RESULTS: After passage, BMSCs were fibroblast-like, and had high reproductive activity. After cultured with astragalus mongholicus, BMSC morphous changed. Nestin, NSE and GFAP were positive. The expression of Wnt-1 gene and Ngn-1 gene increased after astragalus mongholicus treatment. CONCLUSION: BMSCs can differentiate into neuron-like cells induced by astragalus mongholicus. Wnt-1 gene and Ngn-1 gene play a positive regulatory effect during the differentiation.
10.Reversal effect of isotetrandrine, an isoquinoline alkaloid extracted from Caulis Mahoniae, on P-glycoprotein-mediated doxorubicin-resistance in human breast cancer (MCF-7/DOX) cells

This study investigated the reversal effect of isotetrandrine, an isoquinoline alkaloid extracted from Caulis mahoniae, on P-glyeoprotein-mediated multidrug resistance in human breast cancer doxorubicin-resistant (MCF-7/DOX) cells. RT-PCR assay and immunity histochemistry assay were used to determine the expression level of mdrl gene and P-gp in MCF-7/DOX cells to elucidate resistant character of MCF-7/DOX cells. The activity of isotetrandine to enhance doxorubicin cytotoxicity was tested using MTT (3-(4,5-dimethyhhiazol)-2,5-diphenyltetrazolium bromide) assay and was evaluated by the reversal fold (RF) values. Intracellular accumulation of doxorubicin was assessed by the determination of doxorubicin-assoeiated fluorescence intensity. Effect of isotetrandrine on the expression level of P-gp in MCF-7/DOX cells was then determined by immunity histochemistry assay. The ability of isotetrandrine to inhibit P-gp function was evaluated by detecting the accumulation and efflux of rhodamine123 ( Rh123 )with flow cytometry (FCM). Verapamil was employed as a comparative agent in whole experiment. The results indicated that MCF-7/DOX cells had phenotype of MDR and that the positive expression of P-gp MCF-7/DOX cells and reversal fold (RF) was significantly higher than that of verapamil (P < 0.05 ), butit hardly affected cytotoxicity of DOX in MCF-7 cells and the expression level of P-gp in MCF-7/DOXcells. The ability of isotetrandrine to inhibit P-gp function was reversible, because incubation of MCF-7/DOX cells with isotetrandrine caused a marked increase in uptake and a notable decrease in efflux of Rh123 and a marked increase of intracellular DOX concentrations. In conclusion, isotetrandrine exhibited potent effect on the reversal of P-gp-mediated MDR in vitro, suggesting that it might become a candidate of effective MDR reversing agent in cancer chemotherapy.
