1.Establishment and validation of a universal test method in vitro for SARS-CoV-2 mRNA vaccine potency

Objective To establish and verify a universal and stable potency test method in vitro for SARS-CoV-2 mRNA vaccine,so as to use it for the quality control of SARS-CoV-2 mRNA vaccine.Methods ELISA kits that could bind well to S protein of SARS-CoV-2 variants,as well as transfected cells,cell plating concentrations and doses for transfection were screened,and then a potency test method for SARS-CoV-2 mRNA vaccine in vitro was established and verified.Results An ELISA kit was found with good binding ability to S protein of each variant,and HEK293T cells were determined as the transfection cells,with the plating concentration of 2.5 × 10~5 cells/mL and the transfection dose of 4 μg/well in the 6-well plate.An universal and stable potency test method for SARS-CoV-2 mRNA vaccine in vitro was established.The verification results showed that the method met the quality control needs.Conclusion The established potency test method in vitro for SARS-CoV-2 mRNA vaccine has good relative accuracy,linearity,intermediate precision and range,and can be applied to the quality control of SARS-CoV-2 mRNA vaccines

3.Experimental study of correlation between the focus-to-flat panel detector distance and entrance surface dosed in digital radiography of lumbar spine

Objective To investigated the correlation between the focus-to-flat panel detector distance and entrance surface dosed in digital radiography of lumbar spine.Methods The spine phantom used for bone mineral density(BMD) calibration was radiographed at different value of FFD (100 ~ 240 cm with the interval 10 cm,totally 15 times) using 70 kV and 90 kV with the automatic exposure control (AEC) mode without anti-scatter grid.The parameters measured by QUART dido 2100K type X-ray dose meter.So record the value parameters of the tube current (mAs) and entrance surface dose (ESD).The relation curve were counted between FFD,ESD,mAs and Exposure index (EI) use excle.Pearson correlation analysis and related coefficient difference analysis were performed using the SPSS 19.0 software (P <0.05 for the statistically significant level).The FFD and ESD values of regression equation were obtained with regression analysis.Results (1) When the tube voltage was 70 kV:FFD and ESD were negative correlation (r =-0.922,P < 0.001),FFD and mAs were positive correlation (r =0.991,P <0.001),FFD and EI negative correlation (r =-0.938,P < 0.001).(2) When the tube voltage was 90 kV:FFD and ESDwere negative correlation (r=-0.955,P <0.001),FFD and mAs were positive correlation (r=0.994,P <0.001),FFD and EI showed no significant correlation (r=-0.523,P>0.05).(3) Different position correlation coefficient analysis of the difference was not statistically significant difference.(4) The regression analysis results:The regression equation of the lumbar anteriorposterior position FFD and the ESD at 70 kV:y=0.805 x + 319.731 (F=74.175,P <0.001).The regression equation of the lumbar lateral position FFD and the ESD at 90 kV:y =3.152 x + 457.613 (F =133.524,P < 0.001).Conclusions The FFD and ESD in the lumbar spine X-ray radiography were relevant and highly negative correlation.It can effectively reduce the radiographic ESD of lumbar spine with the increasing of the FFD.
4.Study on endoscopic treatment of choledocholithiasis

Objective To investigate the clinical value of endoscopy in the treatment of choledocholithiasis. Methods All 227 patients underwent endoscopic treatment. 14 of them with stones less than 1. 0 cm in diameter were treated with endoscopic papillary balloon dilation; 194 patients with stones 1. 0-1. 5 cm in diameter were treated with endoscopic sphincterotomy; and 19 patients with stones up to 1.5 cm in diameter were treated with endoscopic mechanical lithotripsy. Results Choledocholithiasis were not confirmed by choledochography in 34 cases ( 15% , 34/227) , who underwent exploration of common bile duct through EPBD or EST, but no stones were found. In 187 of the 193 choledocholithiasis patients their stones were removed, the overall success rate with complete stones clearance was 96.9% ( 187/193). The incidence of complication was totally 5. 29% ( 12/227) , including acute cholangitis in 3 patients, acute pancreatitis in 8 patients, and bleeding in 1 patient. Conclusions Endoscopic treatment should be the first choice of patient with choledocholithiasis due to its advantages of safety, effectiveness and with less complication. EPBD or EST was not recommended in case of the common bile duct stones were not confirmed by ERCP, in order to avoid the risk of papilla injury.

6.Effects of high-frequency oscillatory ventilation on brain injury in preterm infants with respiratory distress syndrome

Objective To investigate the association between high-frequency oscillatory ventilation (HFOV) and the incidence of brain injury in premature infants(BIPI) with respiratory distress syndrome (RDS).Methods A total of 136 premature infants who were diagnosed as RDS and treated with mechanical ventilation between January 1,2014 and June 30,2016 were enrolled.Atter stratified by gestational age of 36-34 weeks,33-32 weeks,31-28 weeks and ≤ 27 weeks,the neonates were randomly divided into two groups (68 cases each):conventional mechanical ventilation (CMV) and HFOV groups.Duration of ventilation and changes in blood gas parameters following 24,48 and 72 hours of ventilation were monitored and compared between the two groups.Incidences of BIPI and complications in the two groups were calculated and their associations with gestational age and birth weight were analyzed.Moreover,incidences of cure rates in the two groups were comparatively analyzed.Independent samples t-test,two-way analysis of variance,Chi-square test or Fisher's exact test was used for statistical analysis.Results (1)Significant difference was observed neither in the perinatal factors (prenatal glucocorticoid usage,and incidences of premature rupture of membrane and gestational diabetes mellitus),nor in the severity of RDS between the two groups (all P>0.05).(2) The average duration of ventilation in the CMV group was higher than that of the HFOV group [(68.44±10.3) vs (64.7±8.5) h,t=2.285,P<0.05].No significant difference in the values of pH,partial pressure of carbon dioxide (PaCO2) or partial pressure of oxygen (PaO2) before and after 24,48 or 72 hours of ventilation treatment was found between the two groups (all P>0.05).(3) Neither the incidence of hemorrhagic brain injury nor that of non-hemorrhagic brain injury showed any significant difference between the CMV and HFOV groups [36.8% (25/68) vs 39.7% (27/68);16.2% (11/68)vs 14.7% (10/68),both P>0.05].The total incidence of BIPI showed no significant difference [44.1%(30/68) vs 45.8%(33/68),22=0.266,P=0.606].The smaller gestational age at birth and the lower birth weight,the higher incidence of BIPI,although no significant difference was shown in the incidence of BIPI when compared among different gestational age groups and different birth weight groups (all P>0.05).(4) The incidence of complications in the CMV group was higher than that in HFOV group [25.0%(17/68) vs 11.8%(8/68),22=3.970,P=0.044],while the cure rate of RDS was similar [94.1%(64/68) vs 95.6%(65/68),x2=0.151,P=0.703].Conclusions HFOV is a safe and reliable therapy for preterm infants with RDS.Compared with CMV,HFOV can shorten the duration of ventilation and reduce the incidence of complications without increasing the risk of BIPI.However,the cure rate of RDS is not increased by HFOV.
7.Risk factors for extrauterine growth retardation in moderate and late preterm infants

Objective To study the incidence and risk factors for extrauterine growth retardation ( EUGR) at discharge in moderate and late preterm infants. Methods A retrospective analysis was performed on 607 premature infants who were admitted to the neonatal intensive care unit between January 1st,2016 and December 31st,2016. These subjects were classified into EUGR (n =159) and non-EUGR groups (n=448) based on the body weight at discharge. The risk factors for the occurrence of EUGR were studied by multivariate logistic regression analysis. Results Based on the body weight, the incidence of EUGR at discharge was 26. 2% ( 159/607 ) . The incidence of EUGR in intrauterine growth restriction (IUGR) infants was significantly higher than in non-IUGR infants (P<0. 001). The very low birth weight (VLBW) infants had a higher incidence of EUGR than non-VLBW infants (P<0. 05). Compared with the non-EUGR group,the fasting time,the age to achieve full enteral feeds,the length of mechanical ventilation, oxygen therapy and the length of hospital stays were significantly greater in the EUGR group ( P<0. 05 ) . The percentages of caesarean section,multiple gestation,pregnancy-induced hypertension,intrahepatic chole-stasis of pregnancy and umbilical cord abnormality in the EUGR group were higher than in the non-EUGR group(P<0. 05). The incidences of septicemia,hypoalbuminemia,anemia,retinopathy of prematurity,brain injury in premature infants and mechanical ventilation in the EUGR group were higher than in the non-EUGR group(P<0. 05). The logistic regression analysis showed that birth weight,IUGR,multiple gestation,abnor-mality of umbilical cord and the length of hospital stays were the independent risk factors for EUGR. Conclu-sion The causes of EUGR are multi-factorial. To strengthen pregnancy care,aggressive and reasonable nutri-tional strategy,prevention and treatment of complications may facilitate to reduce the occurrence of EUGR.
8.Analysis on the status quo of spiritual nursing needs and its relationship with self-perceived burden and perceived social support among elderly patients with primary glaucoma

Objective:To understand the current situation of spiritual nursing needs in elderly patients with primary glaucoma and its influencing factors, and to analyze its relationship with self-perceived burden and perceived social support, so as to provide a theoretical basis for reducing their spiritual distress and pain and meeting their spiritual nursing needs.Methods:A total of 218 elderly patients with primary glaucoma who were hospitalized in the department of ophthalmology of the First Hospital of Jilin University from March to September, 2022 were selected by convenience sampling. They were investigated by using general information questionnaire, the Nurse Spiritual Therapeutics Scale, the Self-Perceived Burden Scale, and the Perceived Social Support Scale. And multiple linear regression was used to analyze the influencing factors of spiritual nursing needs among elderly patients with primary glaucoma.Results:The total score of spiritual nursing needs of elderly patients with primary glaucoma was (30.73 ± 4.85). The highest dimension of item average score was "building a good atmosphere" (3.02 ± 0.59), and the lowest dimension was "helping religious practice" (1.95 ± 0.63). The total scores of spiritual nursing needs and self-perceived burden were negatively correlated ( r=-0.423, P<0.01), and positively correlated with the total scores of perceived social support ( r=0.515, P<0.01). Multivariate linear regression analysis showed that age, religious belief, educational level, treatment duration, residence, self-perceived burden, and perceived social support were the main influencing factors of spiritual care needs of elderly patients with primary glaucoma, ( t values were -5.23-7.04, all P<0.01), which could explain 44.5% of the total variation. Conclusions:The spiritual nursing needs of elderly patients with primary glaucoma were at a medium level. Therefore, it is recommended that nurses should carry out targeted spiritual nursing for them according to the differences and characteristics of different patients, to reduce their self-perceived burden, and to improve perceived social support level, in order to meet their spiritual nursing needs.
9.Efficacy evaluation of low-level laser therapy on temporomandibular disorder.

OBJECTIVETo evaluate effectiveness of low-level laser therapy (LLLT) on temporomandibular joint (TMJ) pain.
METHODSThe patients with TMJ pain were randomly assigned laser group (n=21) or control group(n=21), once a day for 6 consecutive days of treatment. TMJ pain and function were measured at baseline, just after treatment course, 1 month and 2 months after the treatment.
RESULTSThe changes of visual analogue scale (VAS) were appearing over time in both groups but presented statistically significant differences between groups (P<0.001). VAS of laser group decreased faster than that of control group. The same tendency occurred for painless maximum vertical opening (MVO), left lateral excursion (LLE) and right lateral excursion (RLE), which increased faster in laser group. There were no statistically significant differences between groups and evaluation times for protrusion excursion (PE), but an interaction between group and evaluation times existed and should be explored further.
CONCLUSIONLLLT is an appropriate treatment for TMJ pain.

10.Association of maternal adverse childhood experiences and parent child relationship in preschool children

Objective:
To explore the relationship of maternal adverse childhood experiences(ACEs) with mother child relationship, so as to provide reference for parent child relationship and child health promotion.
Methods:
In June 2021, children aged 3-6 years old of 36 kindergartens in three areas in Anhui Province were selected by stratified cluster sampling method, follow up data were collected in December 2021, and a total of 6 111 children were included in the study. Maternal ACEs and mother child relationship were respectively assessed using the Adverse Childhood Experiences International Questionnaire(ACEs-IQ)and the Child Parent Relationship Scale (CPRS). A multiple linear regression model was established to analyze the association of maternal ACEs and mother child relationship in preschool children.
Results:
History of maternal childhood sexual abuse, physical neglect and peer bullying were negatively associated with mother child intimacy ( r =-0.03, -0.03, -0.03, P <0.05). Maternal emotional abuse, physical abuse, sexual abuse, emotional neglect, physical neglect, peer bullying, community violence, and total family dysfunction were positively associated with mother child dependence and mother child conflict ( r =0.09, 0.08, 0.05, 0.14, 0.06, 0.11, 0.08, 0.04; 0.18, 0.17, 0.07, 0.20, 0.11, 0.16, 0.12, 0.10, P <0.01). There was no statistically significant between all types of maternal ACEs and mother child intimacy in boys( P >0.05). Mothers with a history of physical abuse, sexual abuse and peer bullying had a statistically significant relationship between mother child intimacy in girls( β =-0.17, -0.62, -0.19, P <0.05). All types of maternal ACEs were positive predictors of mother child conflict between boys and girls( β =0.37-1.96, P <0.05). There was statistical significance between maternal childhood sexual abuse and mother child dependence of boys( β =0.53, P <0.05), but no statistical significance between maternal childhood sexual abuse and mother child dependence of girls( P >0.05). All other types of maternal ACEs were positive predictors of mother child dependence( β =0.09-0.41, P <0.05).
Conclusion
Maternal ACEs are associated with poor mother child relationship among preschool children, and maternal ACEs should be actively followed, which is of great significance for improving the parent child relationship and promoting child healthy development.
