Objective Study on the traceability of serum enzyme ass ays by testing enzyme reference material in clinical laboratories. Methods 50 laboratories were involved in this survey. One enzyme reference material was send to each participate lab. The reference material was tested by use of routine method and the results were recorded. All lab data were processed with computer. Results Compare with the target values, the bias of alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatine kinase (CK) and lactate dehydrogenase (LD) assays were 6 3%, 5 5%, -5 9% and -5 0% respectively, the bias of alkaline phosphatase (ALP) assays was -36 6. The inter laboratory coefficient variation of the amylase assay was 29 2%. Conclusions The results of enzyme assay in clinical laboratories could be traced to the international reference material.

Objective Using the frozen human-pooled serum samples,with values assigned by the enzyme reference procedure in laboratory network to explore the way of quantity value transfer from enzyme reference measurement system to routine ones.Methods The study is about quantity value transfer.The frozen human-pooled serum sample was assigned by the enzyme reference laboratory network including six domestic reference procedure laboratories,namely Beijing Chaoyang Hospital affiliate of Capital University of Medical Sciences,Beijing Aerospace general Hospital,Beijing Shijitan Hospital affiliate of Capital University of Medical Sciences,Peking Union Medical College Hospital,Beijing Leadman Biochemistry Co.,Ltd.and Sichuan Maker Biotechnology Co.,Ltd.,which used the reference measurement procedures of creatine kinase (CK) and lactate dehydrogenase (LDH) from the International Federation of Clinical Chemistry and Laboratory Medicine (IFCC).First,the assigned serum was used to calibrate or verify trueness of routine measurement systems from the 49 laboratories of tertiary hospitals in Beijing in Feb,2013 by Beijing Centerfor Clinical Laboratories.Second,the assigned serum and two levels of patient samples were tested in duplicate by laboratory 's measurement systems after calibration with manufacturer's calibrator and with assigned serum.During this procedure,all laboratories data were collected.Harmonization of results was observed after calibration with the assigned serum according to the statistics.Results After calibration,the interlaboratory coefficients of variation (CVs) of two patient samples were decreased from 4.67％ and 5.96％ to 1.86％ and 1.47％ for CK and from 5.72％ and 5.04％ to 3.67％ and 3.62％ for LDH.For peer group of A and B instrument,after calibration,both CVs of the assigned serum and two patient samples were less than 2％ for CK,and less than 4％ for LDH ; For C instrument group,which was not calibrated with the assigned serum,the biases were-4.75％ for CK and-13.1％ for LDH,while the biggest CVs were 3.64％ for CK and 7.57％ for LDH.For peer group of reagents,after calibration,the CVs were decreased from 1.60％-4.44％ to 0.23％-1.62％ for CK and from 0.93％-6.07％ to 1.06％-2.72％ for LDH.Conclusions It is one of the feasible ways for quantity value transfer by using the frozen human-pooled serum samples,which were assigned by the enzyme reference laboratory network for calibration or verification.It can effectively solve the problem of trueness and harmonization of enzyme testing results.In addition,for those measurement systems which are not applicable for this calibration.

AIM: To study the effect of natural killer(NK)-cells on graft-versus-host disease(GVHD), graft rejection, engraftment and reconstituting of hematopoiesis in mouse allogeneic bone marrow transplantation. METHODS: Lethally irradiated BALB/c(H-2 d) mice were transplanted with C57/6j(H-2 b) bone marrow containing donor peripheral T cells and/or NK cells. Recipients CD 34 + cell counts and the expression of H-2K b+ cell were detected by flow cytometry, peripheral white blood cell(WBC) was detected by auto-cytometry, and the recipients survival rates, GVHD, engraftment and hematopoiesis recovery were then observed. RESULTS: In the group of transplanted with NK cells infusion, the incidence of GVHD was evidently lessened, the survival rates, WBC and CD 34 + cell counts and the expression of H-2K b+ cell were significantly high than that without NK cells infusion. CONCLUSION: In mouse allogeneic bone marrow transplantation, alloreactivity NK cells prevent GVHD, reduce graft rejection, and promote engraftment and reconstituting of hematopoiesis. [

Objective To investigate the expression of CD40-CD154 co-stimulatory pathway in peripheral circulation and intestinal mucosa in patients with inflammatory bowel disease (IBD),the difference between the expression of CD40-CD154 in patients with IBD and that in healthy controls,and the correlation between CD40-CD154 levels and disease activity. Methods A total of 62 patients with Crohn's disease (CD),64 patients with ulcerative colitis (UC) and 56 healthy controls were enrolled. Enzyme-Linked Immuno Sorbent Assay (ELISA),SYBR-green real time PCR and immunohistochemical assay were respectively applied to evaluate expression of CD40-CD154 in plasma,mononuclear cells of peripheral blood and intestinal mucosa. Results Levels of CD40 (P=0. 000) and CD154 (P=0. 001) in plasma,mononuclear cells of peripheral blood and intestinal mucosa were significantly higher in patients with CD and UC than in healthy controls. However,no correlation between disease activity and CD40-CD154 expression in peripheral circulation or intestinal mucosa was detected (P ＞ 0. 05 ). Conclusion CD40-CD154 pathway activation is found in plasma,peripheral blood mononuclear cells and intestinal mucosa in patients with IBD,but is not correlated with disease activity.

Objective Understand the variation of serum creatinine measurement in clinical laboratories of some hospitals in Beijing.Methods 8 samples of mixed frozen human serum added different creatinine concentration standard materials(the creatinine concentration were80-1 000 ?mol/L)and 8 samples of mixed frozen serum of patients(contained different creatinine concentration)were distributed to 13 clinical laboratories(31 series of detection systems)with the way of spot investigation.Every clinical laboratories measured the samples followed the standard operating procedure.Results As to the mixed frozen human serum added different creatinine standard materials,the CV of different detection systems results were 5.74%-9.68%;as to the mixed frozen patients' serum,the CV was 5.90%-11.69%. Compared with Beckman closed detection systems,the results of Dade systems(which used the kinetic alkaline pieric acid method)showed the bias were-5.99%-0.35%,and as to the other systems which measured by alkaline picric acid method,when creatinine concentrations were 200 ?mol/L,the results showed negative bias,and the greatest bias was-8.45%.The bias plots revealed negative for all of the detection systems with enzymatic method over the whole concentration range,and the greatest bias was -8.88%.Conclusions The creatinine determination results of Beckman and Dade closed detection systems were consistent.The results of detection systems which used enzymatic method were generally lower than Beckman detection systems.What's more,the creatinine measurement variations of clinical laboratories were very large,especially for the results of unclosed detection systems,so it was urgent need to solve the standardization of creatinine measurement.

AIM:To evaluate the effect of mycoplasma on the rRNA composition of mature ribosomes in human cell lines.METHODS:We isolated ribosome nascent-chain complex ( RNC) from multiple mycoplasma-positive-negative human cell lines.RNC-RNA was acquired from each cell line through RNA extraction, followed by agarose gel separation, fluorescent visualization and gray-scale value measurements on the rRNA bands.Western blot analysis was performed to in-vestigate the MAPK pathway alterations.RESULTS:In various human cell lines derived from different tissues, we found that as compared with mycoplasma-negative cells, mycoplasma-positive cells showed aberrant RNC-rRNA band patterns, featured by the decreases in 28S and 18S eukaryotic rRNAs and the increases in 16S and other unknown prokaryotic rRNAs.When cultured without ciprofloxacin, mycoplasma-negative HBE cells acquired mycoplasma contamination as ob-served with such characteristic abnormal rRNA bands.The ciprofloxacin treatment was able to recover the RNC-rRNA bands of the mycoplasma-contaminated cells to the normal pattern.The results of Western blot analysis on total and phos-phorylated proteins indicated that p38 pathway was significantly deactivated in mycoplasma-infected A549 cells, while ERK1/2 pathway was not significantly altered.CONCLUSION:Mycoplasma contamination severely alters the RNC-rRNA composition via p38 MAPK pathway, thus seriously impacting on the host cell translational behaviors.

AIM: To study c-Myc expression and its relationship with caspase-3 in a dexamethasone (DEX)- induced mouse thymocyte apoptosis model, and discuss the role of c-Myc in cell apoptosis. METHODS: Mouse thymocyte apoptosis was induced by 1 ?mol/L DEX, the apoptotic and necrosis cells were measured by Annexin V-FITC/PI double staining flowcytometry at 30 min, 3 h, 6 h and 9 h . Electron microscopy observation was carried out at 6 h, and c-Myc and caspase-3 contents were tested by Western blot at 0, 30, 60, 180 min. RESULTS: By 1?mol/L DEX treatment, the apoptosis rates of thymocytes at 30 min, 3 h, 6 h, 9 h were (5.70?0.46)%, (35.79?1.13)%, (50.61?2.15)% and (35.52?1.66)%,respectively; in control group, they were (5.97?0.25)%, (10.20?0.71)%, (12.10?0.66)% and (15.45?0.51)% (P

Objective To investigate the measurement of serum calcium by flame atomic absorption spectrometry (FAAS) and to validate the method for use as a candidate reference method. Methods Serum was diluted by 50 fold with 50 mmol/L hydrochloric acid containing 10 mmol/L LaCl3 and analyzed for calcium on an AA 6800 atomic absorption spectrophotometer. Dilution solutions and FAAS conditions were optimized and the performance of the method was evaluated. Results The method showed within-run, between-run and total CVs of 0. 31%～0.38%, 0.16%～0.30% and 0.35%～0.49%, respectively, and analytical recoveries ranging 98.9%～101.1%. No significant interferences were detected. Conclusions A FAAS method for the measurement of serum calcium has been established. The method is simple and accurate and may be used as a candidate reference method for serum calcium.
