Objective To select a safe,low-cost basal medium in combination with platelet lysate(PL),suitable for the growth of human umbilical cord mesenchymal stem cells(hUC-MSCs)with no influence on the functions of cells during the cultivation process,and to use it for the culture of a large number of high-generation hUC-MSCs.Methods Three different basal media,MSCBM,α-MEM and IMDM,were mixed with PL UltraGROG Advanced respectively to culture hUC-MSCs from P0 to P8. The cell morphology,cell expansion quantity,proliferation function,differentiation function and cell surface markers of different generations were compared to determine the optimal basal medium suitable for hUC-MSCs cultivation.Results The morphology of P1-P5 cells cultured in the three media was uniform and radial,with no significant difference between them and the standard cells(t_(IMDM VS α-MEM)= 0. 159,t_(MSCBM VS α-MEM)= 0. 147,t_(IMDM VS MSCBM)= 0. 161;each P > 0. 05). The result of doubling time(DT)was consistent with the proliferation function. The number of adipoblasts differentiated from P8 cells cultured in MSCBM was higher than that from P8 cells cultured in α-MEM(t =0. 036,P < 0. 05),and the number of adipoblasts differentiated from P8 cells cultured in α-MEM was higher than that in IMDM(t = 0. 031,P < 0. 05).Conclusion In this study,the optimal culture system of hUC-MSCs was MSMBM with 5%UltraGROG Advanced,followed by α-MEM with 5% UltraGROG Advanced,and these two serum-free culture systems were suggested to be selected for large-scale culture of MSCs.

Objective:To observe adjuvant curative effect and adverse reactions of montelukast in the treatment of children with bronchial asthma and its effect on recurrence. Methods:Totally 180 cases of children with bronchial asthma were randomly divided in-to the control group and the observation group with 90 ones in each. The control group was treated with oxygen and budesonide terbutal-ine inhalation, intravenous methylprednisolone sodium succinate and the other conventional treatment methods, and the observation group was additionally treated with montelukast sodium tablets 5 mg, po, qd. The treatment course was three months, and then expira-tory dyspnea, disappearance time of asthma and wheeze and adverse drug reactions in the two groups were evaluated. After 3-6-month follow-up, the recurrence was compared between the two groups as well. Results: The disappearance time of dyspnea, asthma and wheeze in the observation group was shorter than that in the control group (P<0. 05), the total effective rate in the observation group was 96. 67%, which was significantly higher than that in the control group (73. 33%, P<0. 05). The mean follow-up duration was (5. 1 ± 0. 3) months, and the recurrence rate was 8. 51% in the observation group, which was significantly lower than that in the con-trol group (31. 43%, P<0. 05). During the treatment, no significant adverse drug reaction was appeared in the two groups. Conclu-sion:The conventional drug treatment combined with montelukast can rapidly improve the clinical symptoms and reduce recurrence with promising safety, which is worthy of clinical application.

Objective To screen the relative risk and protective factors of children patients with asthma after capillary bronchiolitis.Methods The clinical and follow-up data of 220 children patients with capillary bronchiolitis were collected.Sex,treatment method,immunization vaccination,idiosyncrasy,feeding way,family history were investigated and analyzed.The risk factrs of asthma were screened.Results Univariate analysis showed that the incidence of asthma between different genders had no significant difference (P ＞0.05).The asthma incidence in patients with BCG vaccination on time and large-dose immunoglobulin treatment were lower than those in patients without BCG vaccination on time and large-dose immunoglobulin treatnent (P＜ 0.01 ).The asthma incidence in patients with idiosyncrasy,family history was significantly higher than that in patients without idiosyncrasy,family history( P ＜ 0.05 ).The asthma incidence in patients with obesity was higher than that in other children (P ＜ 0.05).The asthma incidence in patients with more than 4 months breast feeding was lower than that in patients with other feeding ways (P ＜ 0.05 ).Multi-factors regression analysis showed that family history,obesity,idiosyncrasy were the risk factors of asthua,and large-dose immunoglobulin treatment,BCG vaccination on time,breast feeding ＞ 4 months were the protective factors.Conclusions Family history,idiosyncrasy,obesity are the risk factors of asthma in children patients with capillary bronchiolitis,and should be focused on olservation.Immunoglobulin treatment and procedural BCG vaccination are the protective factors to reduce the asthma incidence.They are worthy of application especially in children with risk factors.

The reversible phosphorylation plays an important regulatory role in various pathological and physiological processes. Src Homology 2 domain containing protein tyrosine phosphatase (SHP2),a featured member of the protein tyrosine phosphatase family,is identified as the first PTP proto-oncogene. SHP2 is associated with breast cancer,leukemia,lung cancer,liver cancer,gastric cancer,laryngeal cancer,oral cancer and other types of cancer. Signalling pathways involving SHP2 have also been discovered. It has been recently reported that the high expression of SHP2 in lung cancer regulates cancer cell proliferation,metastasis and drug resistance. SHP2 may be a potential thera?peutic target because it is implicated in many diseases. This article focuses on the research progress in functions of SHP2 in lung cancer.

Objective To evaluate the efficacy of the combination interventional oviduct recanalization with traditional chinese medicine for infertile women with oviduct obstruction.Methods 38 cases with 76 oviduct obstruction were treated with FTC-900 oviduct recanalization apparatus guided with DSA,the apparatus was supplied by COOK company,before recanalization,assisted with side fornix vaginae and enema with traditional chinese medicine.Results Of 76 obstructive oviduct,71 oviduct were recanalization,the successful rates of recanalization were 93.4%;Of 38 infertile women ,25 cases become pregnant,the pregnant rates were 67.3%;Of 13 unsuccessful cases,8 cases turn out re-obstructived.Conclusion Combination interventional oviduct recanalization with traditional chinese medicine for infertile women with oviduct is a simple,safe,and more effective method,it has a high successful rates than either method solely,should be spreaded.

Objective To explore how Vibrio vulnificus (Vv) invades dendritic cells (DC) and induces acute necrosis of DC via toll-like receptor (TLR) 2 and 4 pathways.Methods Vv 1.1758 strain and DC 2.4 mixed culture model was established,observed the infection rates of DC with optical microscope,the location of Vv and structural changes of DC by transmission electron microscope.The expression levels of TLR2 and TLR4 mRNA were determined by real-time quantitative reverse transcription polymerase chain reaction (RT-qPCR) and tumor necrosis factor-α (TNFα) protein titers were measured by enzyme-linked immunosorbent assay (ELISA).DNA ladder qualitative test was used to detect cell apoptosis,while flow cytometry was used to quantify cell apoptosis and necrosis rates.Statistical analysis was done by chi-square test and one-way ANOVE.Results The infection rates of DC after 0.5,1,2,4 and 6 h of mixed culture were (7.8±0.8) ％,(13.9± 1.1) ％,(34.6±4.9) ％,(77.8± 10.2)％ and (95.8 ± 13.1)％,respectively.Vv was generally located in the internal cell membrane of DC 2.4.After 2 h co-culture,nuclear chromatins of DC became active and intranuclear apoptosis bodies appeared.After 4 h,cytoplasmic vacuoles appeared,chromatin gathered,and cell membranes were seriously damaged.After 6 h,mitochondria was highly swelled and distorted,and cell apoptosis and necrosis occurred.TLR2 and TLR4 mRNA levels reached peak values after co-culture for 0.5 h; TNF-α level began to increase at 1 h (P＜0.05) and reached peak values at 2 h.DNA Ladder electrophoresis presented scouring necrosis after 2 h culture and apoptotic bands appeared between 720 bp and 900 bp after 4 to 5 h culture.Early apoptosis rates of DC after 2,4 and 6 h culture were (3.1±3.8)％,(7.8±4.7)％ and (12.7±8.2)％,and necrosis rates of DC were (16.7±12.5)％,(41.6±25.9)％ and (75.5±33.6)％,higher than that of control group (all P＜0.05).Conclusions Vv infects DC and induce DNA degradation through up-regulated expression of TLR2 and TLR4 and increasing of TNF-α inflammatory mediators.During cell degradation,apoptosis and necrosis coexist,while necrosis is predominant.

Objective To find biological markers to predict the mathematical cognitive ability in order to set patients free from the pain and time-consuming behavioral tests. Methods 86 patients with stroke or brain traumatic injuries were recruited and acquired T1 and rest-ing-state functional MRI imaging data. And a mathematical task (7 calculation items, 2 counting items) and a word-reading task (140 items) was also finished. The partial correlative analysis was made between the score of mathematical task and the amplitude of low frequency fluc-tuation of each voxel of the whole brain with the word-reading performance as controlling task, and AlphaSim correction method was used with corrected P<0.05 (single voxel level:P<0.05;cluster size:>110 voxels). Results There were 5 cerebral regions whose amplitude of low frequency fluctuation significantly correlated with mathematical performance:left inferior parietal lobule (161 voxels, rpeak=0.34), left precu-neus/superior parietal lobule (141 voxels, rpeak=0.31), left middle temporal gyrus (359 voxels, rpeak=0.34), left middle frontal gyrus (491 vox-els, rpeak=0.36), and right middle frontal gyrus (156 voxels, rpeak=0.32). Conclusion The amplitude of low frequency fluctuation of left inferior parietal lobule, precuneus/superior parietal lobule, middle temporal gyrus, middle frontal gyrus, and right middle frontal gyrus could be used as predictors of mathematical cognitive ability for brain-damaged patients.

Objective To evaluate the effectiveness and prospect of innovative interactive follow-up mode for rehabilitation ofpostopera-tive breast cancer patietns in new medical environment. Methods The novel patient-centered medical care was promoted,and Pink Ribbon Clubs was established in different regions. Meanwhile,the patients’ self-support groups were formed, and a specialist for out-patients follow-up of outpatients by telephone or on the internet was arranged to provide psychological assessment,with timely and effective response. All data concerning to the follow-up were recorded and the patients were required to complete the relevant questionnaires. Results Compared with the same period in 2013,the follow-up rate was significantly increased to 80. 25%,and the patients showed higher satisfaction. The incidence of lymphedema in the patients was significantly reduced from 13% in 2013 to 7%. The quality of life was significantly improved and self-rat-ing score on depression was decreased from (51. 90 ± 8. 55) to (35. 81 ± 6. 21). Conclusion The kind of“Trinity” interactive mode is an effective follow-up mode with the clinical benefit in patients’ rehabilitation of physical and mental health,which is worthy of further applica-tion.

Foreign specialist physician system has been implemented for more than 100 years. China piloted its own specialist physician system from 2004. The article compared foreign specialist physician system with China’s pilot system. Combined with TCM clinical characteristics and laws, the article also discussed to set up Chinese medical specialist system which is in accordance with both international conventions and TCM laws of development as well as docking the pilot system.

Objective To investigate the cognitive-communication ability of people aged 20-30 years in China. Methods 139 normal participants took part in the Cookie Theft test. 8 different indexes were used to analyze the language samples. These indexes were total words number, the percentage of incorrect statement, dysfluencies, providing structure support, repetition, content units, elaborations of content units and irrelevancies. Results The rater consistence reliability was 1, 0.89, 0.85, 0.93, 0.92, 0.98, 0.98, 0.62, respectively. Conclusion The norms have acceptable reliability.
