1.Evaluation of growth-promoting effect of PL-NBCS instead of FBS on BHK-21, Vero and HEK293T cells

Objective To compare the growth-promoting effects of platelet lysate-containing newborn calf serum(PL-NBCS) and fetal bovine serum(FBS) on cultured BHK-21,Vero,HEK293T cells,and to provide experimental basis for exploring the feasibility of culturing cells with PL-NBCS instead of FBS.Methods BHK21 cells,Vero cells and HEK293T cells were cultured with PL-NBCS and FBS,respectively.The growth-promoting effect of PL-NBCS on cells was detected by kinetic statistical analysis of morphological observation,average cell proliferation density,viability and doubling time of continuously subcultured cells.Results There was no significant difference in morphological characteristics between three kinds of cells cultured in PL-NBCS and those cultured in FBS.Under the two culture conditions,the cells grew in an "S" shape.The average proliferation density,viability and doubling time of BHK-21 cells cultured in PL-NBCS showed no significant difference with the BHK-21 cells cultured in FBS(t=0.734,0.432 and 0.029,P=0.472,0.671 and 0.977,respectively).The average proliferation density and viability of Vero cells cultured in PL-NBCS were higher than those in FBS group,and the average doubling time was significantly higher than that in FBS group(t=2.228,P=0.042).The average proliferation density,viability and doubling time of HEK293T cells cultured in PL-NBCS were significantly higher than those in FBS group(t=2.309,2.848 and 2.682,P=0.033,0.011 and 0.015,respectively).Conclusion PL-NBCS has better growth-promoting effect on cells,and can replace FBS to subculture cells.
2.Observations on the Efficacy of Acupuncture plus Chinese Herbal Medicine in Treating Allergic Rhinitis

Objective To investigate the clinical efficacy of acupuncture plus Chinese herbal medicine in treating allergic rhinitis. Methods One hundred and thirty-five patients with allergic rhinitis were randomly allocated to combination, acupuncture and Chinese herbal medicine groups, 45 cases each. The combination group received treatment with acupuncture plus Chinese herbal medicine; the acupuncture group, treatment with acupuncture alone; the Chinese herbal medicine group, treatment with Chinese herbal medicine alone. IgE, IL-4, IL-5 and TNF-αlevels and clinical symptom and sign total score were observed in the three groups before and after treatment. The clinical therapeutic effects were compared between the three groups. Results There were statistically significant pre-/post-treatment differences in IL-4, IL-5, TNF-αand IgE levels in the three groups (P<0.05). There were statistically significant post-treatment differences in IL-4, IL-5 and IgE levels between the combination group and the Chinese herbal medicine or acupuncture group (P<0.05). There was a statistically significant pre-/post-treatment difference in clinical symptom and sign total score in the three groups (P<0.05). There was a statistically significant post-treatment difference in clinical symptom and sign total score between the combination group and the Chinese herbal medicine or acupuncture group (P<0.05). The total efficacy rate was 91.1%in the combination group, 71.1%in the Chinese herbal medicine group and 66.7%in the acupuncture group. There was a statistically significant difference in the total efficacy rate between the combination group and the Chinese herbal medicine or acupuncture group (P<0.05). Conclusions Acupuncture plus Chinese herbal medicine has a better therapeutic effect on allergic rhinitis. It can effectively relieve the clinical symptoms and signs and reduce IgE, IL-4 and IL-5 levels in the patients.
3.Study on extraction and characteristics of ?-glucosidase inhibitor from Chlorella vulgaris

Objective To study on extraction and characteristics of ?-glucosidase inhibitor from Chlorella vulgaris.Methods The extracts were prepared by drying,degreasing and silica gel columniation washing with different organic solvents,and its pure degree and components were identified.The activities of the inhibitor and its effects on ?-glucosidase under different conditions were studied.Results The results showed that ?-glucosidase inhibitor from Chlorella vilgaris may be a kind of substance of hydroxybenzene.Conclusion The inhibitor showed potent inhibitive effection on ?-glucosidase,and belong to a mechanism of noncompetitive inhibition
4.The role of immunity mediated by TLR2 in macrophages against tuberculosis in elderly adults

Objective Macrophages are important innate immune cells,and are also the main target cells of Mycobacterium tuberculosis.Macrophages express many kinds of Toll like receptors.TLR2 is a structural component of macrophages in the identification of Mycobacterium tuberculosis.Activation of TLR2 signaling pathway can activate cell signal transduction,induce specific gene expression and secretion of cytokines.Immune function decline in old people and the function of macrophages was also changed.In this article,The role of immunity mediated by TLR2 in macrophages against tuberculosis in the elderly is reviewed.
5.Processing for Arctium Lappa with orthogonal test

AIM: To optimize the parameters of processing for Arctium Lappa.METHODS: The contents of arctiin and arctigenin in Arctium Lappa were determined with HPLC,and the total yields of them were chosen as in-dex.The orthogonal test was adopted to acquire the optimum technology for processing Arctium Lappa.RESULTS: The study showed that the best processing parameters for Arctium Lappa consisted of 300 ℃ of the drying tempera-ture and 4-5 minutes of processing time.CONCLUSION: The processing method is reasonable,stable and reliable,and can be used in commercial production.
6.HPLC fingerprint and HPLC-MS analysis for Red Peony Root

AIM:To establish an HPLC fingerprint spectrum of Red Peony Root ( Radix paeonia rubra). METHODS:Ten batches of samples were extracted by 50% ethanol and extract liquor was evaporated to dryness,resolved with methanol. The HPLC analysis was performed on a Diamonsil C18 column with the eluting system of gradient acetonitrile and water. MS was applied to the determination of possible chemical structure. RESULTS:Nine main characteristic peaks were selected from the fingerprint spectrum of 10 batches of samples. Five constituents of which were identified as gallic acid,peoniflorin albiflorin,benzoylpaeoniflorin and paeonol based on the MS spectra. CONCLUSION:The method is valuable for quality conthol of Red Peony Root.
7.Study on Inclusion Technology for Volatile Oil of Pogostemon Cablin

Objective To optimize extraction technology for volatile oil of Pogostemon cablin in the second traditional veterinary drugs anti-stress granule.Methods The single factor was used to investigate the concentration and amount of the ethanol before inclusion of volatile oil.The L9(34) orthogonal design was used to optimize the factors in the process of inclusion.Results The optimal of technology method was that volatile oil was dissolved with 95% EtOH by 1:20(mL/mL),the ratio of oil and ?-CD was 1:8,?-CD was prepared the concentration of 10%,ultrasound pool was at 40 ℃,2.0 h.Conclusion The optimized process is stable and adjustable quality.
8.The analysis of 38 cases of ECG changes after PCI

ObjectiveTo explore the dynamic changes of electrocardiography (ECG) in patients with coronary heart disease(CHD) after the percutaneous coronary intervention(PCI). MethodsTo compare and analyze the regularity of the changes of 12-lead ECG on 38 cases in times,including one week,three weeks,one month,two and three months after the PCI. ResultsCompared with the ECG before the operation, the occurrences of the low-lying or the bidirectional T-wave were significantly increased,and lasting to 2 ~ 3 weeks after tie operation(5.0% and 9.6%va2.6% ,P < 0.05). The ST segment depression( > 1.0mm) and inverted T-wave in the 3 weeks and 2 months after the operation were significantly increased(5.0% ,5.7% vs 2.6% ,P < 0.05 ; 18.0% and 25.2% vs 16.2% ,P <0.01 ). After that,the cases of ST segment depression were reduced to the level before PCI,and there was a significant reduction in the cases of the inverted T-wave ( P < 0.01 ). ConclusionsIn the ECG of the patients with CHD after PCI, there were always early one-off abnormal Q-wave, changes of T-wave (inverted or low-lying, bidirectional)and ST segment depressions,etc. After that ,the changes above would turn back to the level before the operation.
9.Comparative Study on Antibacterial Action of Coumarin Monomer and Ash Bark from Different Origins

6,7-dicho- methoxyl-8-Hydroxycoumarin. Fraxinus rhynchophylla Hance extract had obvious bacteriostasis to 9 kinds of bacterium;monomer mixture and coumarin fingerprint region sample had obvious bacteriostasis to Staphylococcus aureus, Staphylococcus epidermidis, beta streptococcus, aeruginosus Bacillus, Bacillus coli, Klebsiella pneumoniae, catarrh. Unknown component fingerprint region sample had obvious bacteriostasis to Staphylococcus aureus and certain bacteriostasis to Staphylococcus epidermidis, alpha streptococcus, beta streptococcus, Bacillus coli, salmonella. Conclusion The changes of ash bark bacteriostasis were basically identical with known coumarin component content. Aglycone bacteriostasis of ash bark was superior to glycoside, fraxetin and aesculetin bacteriostasis were most powerful in 5 kinds of coumarin monomer.
10.Determination of Berberine Hydrochloride in Shaoyao Decoction by HPLC

Objective To determine the content of berberine hydrochloride in Shaoyao decoction. Methods HPLC was performed on an Agilent XDB C18 column by using acetonitrile-0.05 mol/L potassium dihydrogen phosphate-triethylamine (30∶70∶0.14) as mobile phase at a flow rate of 1.0 mL/min, the column temperature was 25 ℃ and the UV detection wavelength was 345 nm. Results Berberine hydrochloride showed a good linearity redationship in the range of 0.07~1.40 ?g. The average recovery was 98.63% (n=6, RSD=1.21%). The average content was 1.55%. Conclusion The method is simple and accurate, with good repeatability, and can be used for the quality control of Shaoyao decoction.
