1.Significance of mutant p53, Ki-67, ER and PR in differential diagnosis of uterine endometrial carcinoma and uterine serous carcinoma

Objective To investigate the expression of mutant p53,Ki-67,ER and PR in uterine endometrial carcinoma and uterine serous carcinoma,and explore their significance in differential diagnosis.Methods The samples including 37 cases of uterine endometrial carcinoma and 37 cases of uterine serous carcinoma were analyzed.The expression of mutant p53,Ki-67,estrogen receptor and progesterone receptor were performed by using the immunocytochemical (IHC) EnVision system.Data were analyzed with SPSS 11.5 statistic software.Results The positive rate of mutant p53 in uterine endometrial carcinoma was statistically lower than that in uterine serous carcinoma [21.62 % (8/37) vs 64.86 % (24/37) (P < 0.01)].The positive rate of Ki-67 in uterine endometrial carcinoma was statistically lower than that in uterine serous carcinoma [37.84 % (14/37) vs 70.27 % (24/37) (P < 0.01)].The positive rate of estrogen receptor in uterine endometrial carcinoma was statistically higher than that in uterine serous carcinoma [78.38 % (29/37) vs 32.43 % (12/37) (P < 0.01)].The positive rate of progesterone receptor in uterine endometrial carcinoma was statistically higher than that in uterine serous carcinoma [75.67 % (28/37) vs 29.73 % (11/37) (P < 0.01)].Conclusions The expression of mutant p53 and Ki-67 are higher in uterine serous carcinoma.The expression of estrogen receptor and progesterone receptor are higher in uterine endometrial carcinoma.Combined detection of mutant p53,Ki-67,ER and PR has important significance in screening and preventing uterine endometrial carcinoma and uterine serous carcinoma.

3.The expression of thanatos-associated protein-8 in ovarian cancer tissue and its clinical significance

Objective To investigate the expression of thanatos-associated protein-8 (THAP-8) in ovarian cancer tissue,and explore its clinical significance.Methods A total of 86 patients with ovarian cancer were enrolled.The gene and protein expression of THAP-8 of the ovarian cancer tissue and adjacent tissue were detected by fluorescent quantitation polymerase chain reaction and immunohistochemistry.Results The gene expressions of THAP-8 of ovarian cancer tissue and adjacent tissue were 0.304 ± 0.104 and 0.615 ± 0.152.The protein expressions of THAP-8 in ovarian cancer tissue and adjacent tissue were 30.16 ± 12.34 and 67.34 ± 22.37.The gene and protein expressions of THAP-8 in ovarian cancer tissue were significantly lower than those in adjacent tissue (P< 0.01).The gene and protein expressions of THAP-8 in the low and middle degree of tumor differentiation of ovarian cancer tissue were significantly lower than those of the high degree of tumor differentiation of ovarian cancer tissue (0.216 ±0.085 vs.0.457 ±0.124 and 24.22 ± 9.21 vs.46.24 ± 17.52,P < 0.01 or < 0.05).The gene and protein expressions of THAP-8 in lymph node metastasis of ovarian cancer tissue were significantly lower than those of no lymph node metastasis of ovarian cancer tissue (0.247 ± 0.087 vs.0.476 ± 0.133 and 23.44 ± 8.57 vs.49.60 ± 18.35,P < 0.01 or <0.05).The gene and protein expressions of THAP-8 in clinical TNM pathologic Ⅲ,Ⅳ stage of ovarian cancer tissue were significantly lower than those of Ⅰ,Ⅱ stage of ovarian cancer tissue (0.232 ± 0.114 vs.0.424 ±0.125 and 25.64 ±8.12 vs.44.78 ± 16.84,P <0.05).Conclusions The downregulation-expression of THAP-8 has a closed relationship with the development of ovarian cancer.The THAP-8 might be used as a potential marker to assess the degree of rumor grade and prognosis in ovarian cancer.
4.Temperament characteristics and its determinants in children with epilepsy

Objective To investigate the temperament of children with epilepsy and the effects of various epilepsy-related variables on it. Methods The temperament of 60 pairs of epilepsy and normal children were assessed by temperament scale. The results of the two groups were compared. Epilepsy-related variables were analyzed. Result The scores of all dimensional variables in the epilepsy group were higher than that in normal group. The differences in adaptability and intensity of response between the two groups was statistically significant ( P <0. 05), and the differences in rhythmicity, approach-withdrawal toward new situations, emotional nature and persistence were more notable ( P <0.01). The temperament of epilepsy children was incline to difficult temperament( X2 = 10. 91, P <0.01). Age at onset of epilepsy, main caregiver and maternal education level had impaction on the temperament of epilepsy children..Conclusions The temperaments of epilepsy children tend to be negative. Children's temperaments are inclined to easy when their mothers are well educated and their main caregivers are parents, while they tend to be difficult temperament when the age of onset is early.
5.Comparison of the effect of two prophylactic regimens for postoperative analgesia after shoulder arthroscopic surgery

Objective To observed the analgesia and adverse reactions of continuous patients-controlled in-terscalene analgesia with patient-controlled intravenous analgesia for the shoulder arthroscopic surgery.Methods 40 patients with ASA Ⅰ ~Ⅱ anesthesia in general anesthesia for the shoulder arthroscopic surgery were selected and randomly divided into the two groups,20 cases in each group.They were continuous patients-controlled interscalene analgesia(PCNA)and patient-controlled intravenous analgesia (PCA).PCNA with 0.2% ropivacaine +saline to 100mL,background parameters 4ml/h,a single dose of 0.5mL,lock 20min.24 hours after dosing.PCA with fentanyl lmg +tropisetron 5mg +saline to 100mL,background parameters 2mL/h,a single dose of 3.5mL,lock 20min.Record the VAS score 6h,12h,24h,48h after surgery,and adverse side effects of postoperative dizziness,nausea,vomiting and itchy skin.And the patient's analgesic satisfaction survey 48h after surgery.Results PCNA after surgery 6h,12h, 24h,48h in VAS score was significantly lower than the PCA(P <0.05).PCNA were dizziness,nausea,vomiting and the incidence is lower than the PCA,the difference was statistically significant(P <0.05).The PCNA of patients sat-isfaction was 100%,the PCA of patients satisfaction is 75%,the difference was statistically significant(P <0.05). Conclusion In the postoperative analgesia of shoulder arthroscopic surgery,the analgesic effect and patient satisfac-tion of continuous patients-controlled interscalene analgesia are better than patient-controlled intravenous analgesia. And the dizziness,nausea,vomiting is lower.
6.Mediating effects of adolescent loneliness and depressive symptoms on interparental conflict and adolescent non-suicidal self-injury

Objective:
To explore the mediating effects of adolescent loneliness and depressive symptoms on interparental conflict and adolescent non-suicidal self-injury, so as to provide insights into the prevention and interventions of adolescent non-suicidal self-injury.
Methods:
Students from 7 middle schools in Tianjin, Henan, Sichuan, Zhejiang and Inner Mongolia were selected using convenient sampling method. Their loneliness, depressive symptoms and non-suicidal self-injury occurrence were investigated using the Three-Item Loneliness Scale, the Center for Epidemiological Studies Depression Scale and Non-Suicidal Self-injury Scale, and interparental conflict was evaluated by adolescents and their mothers respectively, using the adolescence revision of Co-parenting Scale and the Co-parenting Relationship Scale. The mediating effects of loneliness and depressive symptoms on interparental conflict and non-suicidal self-injury was examined using a structural-equation model.
Results:
A total of 544 students were recruited, with a mean age of (14.19±; 1.43) years. There were 241 males (44.30%) and 303 females (55.70%); 452 junior high school students (83.10%), 46 high school students (8.45%) and 46 vocational high school students (8.45%); and 150 students with non-suicidal self-injury (27.57%). The scores of loneliness, depressive symptoms, parental conflict evaluated by students and their mothers were (4.38±; 1.87), (7.32±; 6.06), (14.10±; 8.03) and (3.04±; 3.19), respectively. The results of structural equation model analysis showed that parental conflict had no direct effect on non-suicidal self-injury, but loneliness (β=0.074, 95%CI: 0.038-0.114) and depressive symptoms (β=0.096, 95%CI: 0.061-0.136) played indirect effects on interparental conflict and non-suicidal self-injury, and they also exerted a chain mediating effect (β=0.064, 95%CI: 0.037-0.095) (all P<; 0.05).
Conclusion
Interparental conflict can impact adolescent non-suicidal self-injury through the mediating effects of adolescent loneliness and depressive symptoms.
7.Diagnostic value of galactomannan detection for invasive aspergillosis infections in pediatric patients with Meta-analysis

Objective To evaluate the diagnostic value of galactomannan(GM) detection for invasive aspergillosis infections in pediatric patients, a Meta-analysis was performed.Methods According to selection criteria, studies indexed by PubMed,EMBASE,Medline,CNKI and Wanfang Data on GM testing in pediatric aspergillosis were included from January 2001 to November 2012. The methodological quality was assessed by QUADAS, sources of heterogeneity investigated, pooled effect quantities evaluated, and summary receiver operative curves(ROC) and subgroup analysis performed.Stata12.0 was used to analyze diagnostic parameters, such as, sensitivity, specificity, positive likelihood ratio, negative likelihood ratio and area under the curve(AUC). Results Thirteen articles with 16 sets of data were included.The weighted sensitivity and specificity with 95% confidence interval were 0.71(0.60-0.79)and 0.94(0.89-0.96), respectively.Under different cut-off values, sensitivities of GM assay in diagnosis of pediatric aspergillosis were 0.71(0.54-0.84) for 0.5, 0.77(0.64-0.87) for 0.8, 0.72(0.58-0.84) for 1.0 and 0.59(0.39-0.76) for 1.5, and its responding specificities were 0.94(0.89-0.97), 0.89(0.82-0.93), 0.89(0.81-0.94) and 0.91(0.88-0.94).Conclusion Under the cut-off value of 0.8, the GM assay is a useful method in the detection of pediatric invasive aspergillosis.
8.Analysis of the relationship between IL-10 SNPs and pulmonary tuberculosis susceptibility

Objective To investigate the relationship between IL-10 SNPs and pulmonary tubercu-losis susceptibility.Methods Collected 104 blood samples of pulmonary tuberculosis patients as case group and 104 blood samples of healthy.To analyze genotype SNPs of IL-10-819 -1082 by genomic DNA extraction,PCR amplification and TOF mass spectrometry.Finally the differences of gene and the risk of pulmonary tuberculosis occurrence were analyzed.Results IL-10-819 C/C, C/T and T/T site genotype frequencies were not different between two groups( P >0.05).IL-10-1082 G/G、G/A、A/A site genotype frequencies were not different between two groups( P >0.05).In logistic multivariate regression analysis found that:be compared with IL-10-819 C/C, the risk carrying IL-10-819 C/T, T/T group respectively of pulmonary tuberculosis were OR =0.001(95%CI =0.511~1.921), P =0.977, OR =2.116(95%CI=0.263~1.219), P =0.146, without statistical significance.Be compared with IL-10-1082 A/A, the risk carrying IL-10-1082 G/A、G/G group respectively of pulmonary tuberculosis were OR =0.113(95%CI =0.540~2.393), P =0.737, OR =1.872(95%CI =0.275~1.258), P =0.171, without statisti-cal significance.Conclusions IL-10-819 and -1082 genotype and allele distribution are not different, the mutation of genotype is not correlation with pulmonary tuberculosis.
9.Human papillomavirus 16 E6 inducing autophagy of the cervical cancer C33A cells by endoplasmic reticulum stress pathway

Objective To investigate the effects of human papilloma virus 16 E6 (HPV16 E6) on endoplasmic reticulum (ER) stress-autophagic response in the cervical cancer C33A cells.Methods Polymerase chain reaction was used for detecting the integration of HPV DNA.The eukaryotic expression vector of HPV16 E6 was constructed and transfected via lipofectamine into C33A cells.Experimental cells were classified into 3 groups:pcDNA3.1--HPV16 E6 group,pcDNA 3.1-group and C33A group.Western blot was used to measure expression of protein of HPV16 E6,Beclin 1,LC3 Ⅱ,IRE1,PERK and ATF6 in transfected cells.Results here was no HPV DNA integration in C33A cells that were confirmed as the intervention cells.Eukaryotic expression vector pcDNA3.1--HPV16 E6 was constructed successfully.The eukaryotic expression vector pcDNA3.1--HPV16 E6 significantly improved the expression of protein of HPV 16 E6 in C33A cells.The protein expression of Beclin 1,LC3 Ⅱ,IRE1,PERK and ATF6 were significantly improved after transfection with vector pcDNA3.1 +-HPV16 E6 (P < 0.05).Furthermore,LC3 Ⅱ protein level was reduced by treatment with ER stress inhibitor.Conclusion HPV16 E6 can improve autophagy through the ER stress pathway,and this response may play an important role in the process of HPV16 E6 inducing cervical cancer,providing one of the new strategies for gene therapy of cervical carcinoma.
10.Epileptiform activities of pyramidal neurons in rat CA1 area induced by low-dosage veratridine

BACKGROUND: The event of paroxysmal deplorizing shift (PDS) is the cellular hallmark of brain neurons of epileptiform activities. Its development used to be considered to be related to abnormal synaptic interactions. Recertly, the intrinsic nature of PDS has received more attention.OBJECTIVE: To observe the characteristics of epileptiform activities of rat hippocampal CA1 pyramidal neurons induced by low-dosage veratridine and investigate its possible ion mechanism.DESIGN: An exploratory and observational trial.SETTING: Institute of Neuroscience, Fourth Military Medical University of Chinese PLA.MATERIALS: This study was conducted at the Institute of Neuroscience,Fourth Military Medical University of Chinese PLA, from October 2002 to October 2004. Forty healthy SD rats of 14 days old were selected. Drugs were provided from Tianjin Drug Company and Sigma Company.METHODS: Rats were anesthetized by intraperitoneal injection, and their brain was removed and cut into slices. Epileptiform activities were induced by 0.5 μ mol/L veratridine. Then 80 nmol/L tetrodotoxin was added into the perfused solution on 6 cerebral slices, and 5 μmol/L phenytoin was used on another 5 cerebral slices. The electrophysiological characteristics of the cells under the effect of different kinds of drugs were observed.MAIN OUTCOME MEASURE: Discharge pattern of cells and tetrodotoxin-sensitive sodium currents under voltage-clamp configuration through Ⅰ-Ⅴ reaction.RESULTS: After perfusion of 0.5 μmol/L veratridine, the rat pyramidal neurons in CA1 area displayed relatively fixed-mode of runs of PDS bursting,followed by the hyperpolarization of cell membrane. Such epileptiform activities were blocked either by 80 nmol/L tetrodotoxin or 5 μnol/L phenytoin. The tetrodotoxin-sensitive sodium currents in epileptic neurons and normal controls under voltage-clamp configuration on holding potential of -55 rmV, -60 rmV, -65 mV. This shows that persistent sodium currents could be improved by low-dosage veratridine in a voltage-dependent manner.CONCLUSION: Low-dosage veratridine may induce runs of PDS like epileptiform activities on rat CA1 pyramidal neurons. Such changes can be blocked by low-dosage tetrodotoxin or phenytoin. Its ion mechanism may be related to persistent sodium currents.
