2.Effects of processing on piceid and emodin in Polygonum cuspidatum

Object To investigate the effects on the active components in Polygonum cuspidatum Sieb. et Zucc. and its different processed products. Methods The qualitative analysis of piceid and emodin are taken by TLC, and content of emodin was determined by TLCS and HPLC. Results The contents of emodin significantly increased in samples fried with rice wine, vinegar and salt compared with the crude. Conclusion Different methods and ajuvants made a certain influence on the contents of emotion in P. cuspidatum.

4.Analysis on aleukocytic suspending red blood cell supernatant fragment and particle image changes at different stored periods by microscopic phase contrast technology

Objective To investigate the cellular fragments and particle image changes of inventory aleucocytic suspending RBC produced by the storage time extension ,RBC damage or hemolysis in order to provide the revelatory experimental basis for the transfusion safety .Methods The supernatant was prepared from different stored days (3 ,7 ,14 ,21 d) of stock aleukocytic suspen‐ding RBC .The particles in supernatant were observed and morphologically analyzed by using the microscopic static image analytic technology .Results There were a small amounts of visible particles in the sample supernatant preserved for 3 ,7 d and the parti‐cles′sizes are similar to cells′;the number of particles began to significantly increase from 14 d and the diameter became smaller . The particles filled the entire field until 21 d ,showing fragmentary status .Conclusion The cellular fragments and particles in the supernatants of stock aleukocytic suspending RBC with the storage period exceeding 14 d are significantly increased and have signif‐icant difference compared with those stored for less than 14 d .These exogenous fragments and particles may become antigens and induce the body immune response ,lead to transfusion adverse reactions .It is recommended that the patients should be transfused with stock aleukocytic suspending RBC within a storage period of 14 d .
5.Modified design of biotype self-locking hip joint prosthesis

BACKGROUND:Hip prosthesis needs to meet the good structural compatibility with femoral bone marrow cavity and the biomechanical properties of the original bone tissue. However, the difference of individual anatomical characteristics and the stress concentration in the local area after replacement directly affect postoperative combination of femoral prosthesis stem, force flow transfer and femoral reconstruction, and finaly result in aseptic loosening of hip joint in some patients.
OBJECTIVE:To analyze the biomechanical characteristics of modified biotype self-locking hip prosthesis by the method of finite element analysis, and to provide the reference for the application of self-locking hip prosthesis in hip replacement.
METHODS:Experimental design contained traditional biotype hip joint prosthesis group and self-locking hip joint prosthesis group. In accordance with the finite element models of self-locking hip prosthesis, 28 kinds of screw fixation were established. Three-dimensional models were generated in the Minics system. After the optimization of prosthesis, prosthesis was designed. In the Solidworks software, the femur was re-positioned and set for contacting set in Ansys to analyze the stress distribution and displacement distribution of the femoral-prosthesis-screw by Ansys. We compared the peak of stress and displacement of the femur and prosthesis, and analyzed the biomechanical stability of prosthesis.
RESULTS AND CONCLUSION:(1) By using the Ansys finite element method, we analyzed the stress and displacement distribution of the femoral-prosthesis-screw. The minimum stress peak value of the femur was 15.698 MPa. The minimum stress peak value of prosthesis was 45.491 MPa. The minimum stress peak value of screw was 8.359 MPa. The minimum displacement peak value of femur was 1.125 3 mm. The minimum displacement peak value of prosthesis was 1.039 6 mm. The minimum displacement peak value of screw was 0.566 4 mm. (2) Compared with the traditional biotype hip joint prosthesis group, in 28 kinds of self-locking hip joint prostheses, the stress was minimum in the group E. The displacement was minimum in groups A-F. Regarding comprehensive stress and displacement, groups A-F were the best combination. The maximum stress of femoral-prosthesis-screw was 18.936, 59.494 and 12.382 MPa. The maximum displacement of femoral-prosthesis-screw was 1.125 3, 1.039 6 and 0.626 3 mm. (3) These findings indicated that the stress and displacement parameters of groups A-F was the best parameter combination in 28 kinds of self-locking hip joint prostheses, and it is the recommended index of researching and developing self-locking hip prosthesis.
6.The role of promoter CpG islands methylation of leptin gene in osteoarthritis

Objective To investigate the effects of 5-Aza-CdR( methylation transferase inhibitor) on the expression levels of leptin gene in chondrocytes and methylation states of leptin promoter region between osteoarthritis (OA) group and control. Methods The chondrocytes in osteoarthritis group were treated with 5-Aza-CdR with different doses and time-points, and the expression level of leptin was detected by real-time polymerase chain reaction for picking up the optimum dose and time-point. Next, the chondrocytes in 5 osteoarthritis patients and 5 control patients (amputation due to severe trauma) were treated with 5-Aza-CdR. Lastly, leptin mRNA expression levels in the four groups osteoarthritis and control chondrocytes treated with/without 5-Aza-CdR were measured by real-time PCR and the methylation state of promoter region ( - 280- + 79) was detected by epityper quantitative DNA methylation analysis. Results ( 1 ) After treating the chondrocytes in OA groups with 10 μmol/L 5-Aza-CdR for 72 h, the mRNA expression levels of leptin were increased significantly. ( 2 ) The mRNA expression levels of leptin were significantly different among the four groups ( P < 0. 05 ), and the chondrocytes in osteoarthritis groups treated with 5-Aza-CdR showed a marked induction of leptin mRNA expression. (3) Analysis of quantitative methylation data using an unsupervised hierarchical clustering algorithm, showed that methylation patterns of leptin promoter was different between control and osteoarthritis chondrocyte treated with/without 5-Aza-CdR. Conclusion Demethylation of leptin promoter might up-regulate leptin gene expression level and it might contribute to osteoarthritis.
7.Experimental Study on Therapeutic Effects of Xileisan Pessary on Rats with Cervicitis

Objective To study the therapeutic effects of Xileisan Pessary on rats with cervicitis based on the idea of treating different diseases with the same therapy. Methods Vagina perfusion of hydroxybenzene mucilage was used to establish rat cervicitis models. Rats were randomly divided into normal group, model group, positive medicine group, low-, medium-, and high-dose Xileisan Pessary groups. Rats with cervicitis were treated with Xileisan Pessary from the next day after modeling for 10 days. The amount of neutrophilic granulocytes and leukocytes were detected by blood routine analysis meter; cervial histopathologic examination was conducted by HE staining. Results Compared with normal group, the numbers of granulocyte and leucocyte increased significantly in the model group (P<0.01). Compared with the model group, the numbers of granulocyte and leucocyte in the low-dose Xileisan Pessary group showed a decreasing trend (P<0.05, P<0.01); cervical inflammation mucosal epithelium and tissue necrosis were signally alleviated; granulocyte in the medium-dose Xileisan Pessary group showed a decreasing trend (P>0.05);the content of leucocyte hardly changed;granulocyte in the high-dose Xileisan Pessary group increased observably (P<0.01); the number of leucocyte showed an increasing trend (P>0.05), and the numbers of granulocyte and leucocyte in positive medicine group decreased observably (P<0.01), and cervical inflammation mucosal epithelium and tissue necrosis were also signally alleviated. Conclusion Xileisan Perssary can relieve the inflammation of cervix and repair injured mucosa. It has the function of treating different diseases with the same therapy.
8.Preliminary mechanism study of HCoV-OC43 escape from human dendritic cell immune elimination

Objective:To study the possible immune escape mechanisms of HCoV-OC43 from human dendritic cells(DC).Methods:HCoV-OC43 was isolated from clinical specimen using BSC-1 cells and identified by Real-time PCR,and the cytopathic effect was observed by phase contrast microscope.DCs were induced in vivo using hu-GM-CSF and IL-4 cytokines,and after 7 days of differentiation,DCs were infected by HCoV-OC43.The morphology of HCoV-OC43 infected DC was observed by transmission electron microscope,and the cytokines related to DC functions were detected by Real-time PCR after infection.DC proportion and function related co-stimulatory molecules were analyzed by flow cytometry.Results:In vitro HCoV-OC43 infected human DC model was successfully built.HCoV-OC43 can infect DC and generate immune response of DC in vitro,but no virus nucleonic acid could be detected in culture supernatant.The DC expression of IFN-α,IFN-β,CCL3 and CCL5 were significant decreased when infected with HCoV-OC43,but the expression of costimulatory molecules including HLA-DR,CD1c and CD86 were not affected by HCoV-OC43 infection.Conclusion:Human DC could be infected by HCoV-OC43 and generate immune response,but could not produce progeny virus.HCoV-OC43 may escape from immune response by suppressing the expression of IFN-α and other inflammatory cytokines and chemokines in DC.
9.The quantitative analysis of S100 in the brain tissue and serum following diffuse brain injury in rats

Objective To investigate the dynamics of the level of S100 in cerebrum, brainstem, and serum following the diffuse brain injury in rats and provide the experimental evidences for estimating injury time. Methods ELISA was used to determine whether S100 protein is changed after diffuse brain injury in rats. Forty rats were sacrificed at 0.5 hour, 2 hours, 4 hours, 12 hours, 24 hours, 3 d and 7 d after diffuse brain injury and normal rats as control. Results The level of S100 in cerebrum, brainstem, and serum increased, followed by a decrease, and then further increased. The level of S100 could be detected to increase at 30 minutes and reached the peak at 4 hours after DBI. The level decreased gradually to the normal at 1d and till 3 d formed the second peak. The level returned to the normal at 7d following injury again. In the postmortem injury groups, there were no significant changes compared to the control group. Conclusion The present study showed that the time-dependent expression of S100 is obvious following diffuse brain injury in rats and suggested that S100 will be a suitable marker for diffuse brain injury age determination.
10.Effect of immature dendritic cells (imDC) induced by sinomenine on the peripheral blood Th1/Th2 cytokines of allogeneic rat renal transplantation

Objective To investigate the effect of immature dendritic cell (imDC) derived from donor-derived bone marrow induced by alkaloid sinomenine (SN) on the Th1/Th2 cytokines in venous blood of receptors,and to probe into the mechanism which imDC induced by SN can lead transplantation immune tolerance.Methods Inbred strain Wistar and Sprague Dawley (SD) rats were selected as kidney transplant donor and recipient,respectively.Vessel sutures of the microsurgery technique were used to build the bilaterally renal transplantation model of rats.By the injection of imDC to the recipient rats preoperatively,enzyme-linked immunosorbent assay (ELISA) was used to determine the level of the interleukin (IL)-2,IL-4,IL-10 and interferon-γ (INF-γ).Results (1) Successful rate of transplantation was 89.5%.Arterial anastomosis time was (12.5 ±5.7)min,and venous anastomosis time was (17.3 ± 3.4)min.(2) The content of the IL-2,INF-γ,IL-4 and IL-10 in SN-imDC 106 group was (17.25 ± 3.41) pg/ml,(239.80 ± 9.06) pg/ml,(337.60 ± 25.07) pg/ml,and (1 432.00 ± 106.39) pg/ml,respectively.Among the same concentration,the level of the IL-4 and IL-10 that stood for the Th2 cytokines was significantly higher in SN-imDC group than imDC group and control group (P < 0.05),and was significantly higher in SN-imDC 106 group than SN-imDC 105 group (P < 0.05).Among the same concentration,the lever of IL-2 and INF-γ that stood for the Th1 cytokines was significantly lower in SN-imDC group than imDC group and control group (P < 0.05),and was significantly lower in SN-imDC 106 group than SN-imDC 105 group (P < 0.05).Conclusions (1) The use of microsurgery for anastomosis could make the model of singel kidney transplantation in rats.(2) Specific imDC induced by SN could induce the migration to Th2 immune response,which proved imDC induced by SN could mediate immune tolerance to the recipient.
