1.The relationship between the timing of intervention and rehabilitation efficacy after cerebral infarction : a retrospective analysis

Objective To conduct a retrospective analysis of the relationship between the time of rehabilitation intervention and its effectiveness among hemiplegic patients after cerebral infarction.Methods Fifty-four hemiplegic stroke patients within 1 week of onset were randomly divided into three equal groups of 18:In the A group rehabilitation was begun 1 to 2 weeks after onset; in the B group it was started between 2 and 4 weeks; in the C group rehabilitation was begun after four weeks.The patients all received comprehensive hemiplegic limb training,electric standing bed training,functional electrical stimulation and medical gymnastics for hemiplegia.Motor function was assessed using the Fugl-Meyer balance function assessment and the modified Barthel index before treatment,in the course of the first week of treatment and 3 months later.Results A group and B group made similar progress,but the curative effect in group C was significantly weaker.Conclusion Rehabilitation should be started within 1 month after the onset of cerebral infarction.
2.Influence of Different Follow-up Ways on Compliance Behavior of Stroke Patients during Rehabilitation

Objective To observe the influence of telephone follow-up and home follow-up on compliance behavior and activities of daily living (ADL) of stroke patients during rehabilitation. Methods 50 cases were as telephone follow-up group and 50 cases were as home follow-up group extracted from 355 stroke patients who discharged the hospital during August 1, 2010 to July 1, 2012. They were followed up 2 weeks, 1 month and 3 months after discharge respectively. Their ADL were assessed with Bathel index (BI) at discharge and 3 months after discharge, and the compliance behavior were assessed 3 months after discharge. Results There was no significant difference in the score of BI between 2 groups at discharge (P>0.05), and the score of BI improved in both groups 3 months after discharge (P<0.05), but it was higher in the home follow-up group than in the telephone follow-up group (P<0.05), as well as the rate of compliance behavior (P<0.01). Conclusion Regular home follow-up can improve the compliance behavior of stroke patients during rehabilitation.
3.Targeting DNA damage response pathway: recent progress of PARP inhibitors in cancer therapy

Genomic instability is one of the most pervasive characteristics of cancer cells,and DNA damage response (DDR) pathway plays a crucial role in genomic stability.The DDR pathway is a complex signaling network,which involves cell DNA repair,apoptosis and cell cycle regulation.Deficiencies in these repair pathways can result in several different genetic disorders,including cancer.Targeted therapy based on inhibiting the DDR pathway in cancers offers a novel therapy strategy for patients with tumors lacking specific DDR functions.Many small-mole-cule compounds targeting DDR pathway are typically developed for solid cancer therapy.The poly (ADP-ribose) polymerase (PARP) inhibitor is a kind of DDR inhibitors which exploits the principle of synthetic lethality to selectively kill cancer cells.This review highlights the molecular mechanisms of PARP inhibitor action,the progress of PARP inhibitors in cancer therapy,drug resistance and the challenge of PARP inhibitor in the future.
4.Molecular authentication of Sailonggu and its resource distribution in Qinghai-Tibet Plateau.

To provide accurate information on geographic distribution of crude drug Sailonggu in the plateau, we identified zokor species (Eospalax spp.) in Qinghai-Tibet Plateau using molecular methods. Based on the mitochondrial cytochrome B (cytb) gene sequences, we then extracted haplotypes from these sequences and reconstructed phylogenetic trees for the haplotypes using both maximum likelihood (ML) and Bayesian inference (BI) methods. Based on the trees, the species of each sample were determined. Five hundred and three samples from 35 populations were sequenced and their whole cytb sequences (1140 bp) were obtained. From these sequences 150 haplotypes were detected, in which, 126 were Eospalax baileyi, 20 were E. cansus, and 4 were E. smithi of the 35 populations, 28 were E. baileyi type, 5 were E. cansus type, and the remaining 2 were mixed of E. baileyi + E. cansus (DT2) and E. baileyi + E. smithi (ZN3). The results showed that, the regions around the Qinghai lake and near the upper stream of Yellow River started at Guide could be viewed as the producing area of authentic Sailonggu, and also, the cytb gene is a powerful molecular marker to determine the species of zokors as well as for the authentication of geographic distribution of Sailonggu.

5.The role of norepinephrine in down-regulation of visceral sensitivity in rats deprived of rapid eye movement sleep

Objective To investigate the role of norepinephrine in the down-regulated visceral sensitivity of rats deprived of rapid eye movement(REM)sleep.Methods Twenty-four male Sprague- Dawley rats were randomly divided into 3 groups:cage-yoked rats as control(YC),rats with REM sleep deprivation(SD)and rats with yohimbine administered intraperitoneally after REM sleep deprivation(YSD).Flower pot technique was employed to make sleep deprivation model.YSD group was given vohimbine intraperitoneally at the 48th hour after REM sleep deprivation.After both SD and YSD groups had completed these processes,rats of all the three groups were given colorectal distension(CRD)and electromyogram (EMG)was recorded at the same time.The number of discharges of EMG and the threshold of Dain perception of the rats were observed to evaluate the visceral sensitivity.The thalamus,rectum and distal colon were taken after CRD;MAO-mRNA and TH-mRNA in three tissues were detected with RT-PCR.Resuits On 48th hour,the number of discharges of EMG in 10 seconds responding to CRD in group SD was significantly less than that in group YC and the threshold of pain perception in group SD was higher than that in group YC(P<0.05).The number of discharge of EMG in group YSD was significantly more than that in group SD(P<0.05).The expression of MAO-mRNA in group SD was lower than that in group YC(P<0.05)and the expression of TH-mRNA in group SD was higher than that in group YC(P< 0.05).Conclusions The visceral sensitivity in rats is down-regulated by REM sleep deprivation,which can increase synthesis of norepinephrine.Norepinephrine can modulate visceral sensitivity.
6.Hard-soft tissue changes with the Twinblock appliance in Class Ⅱ malocclusion patients

BACKGROUND:Twin block appliance has been shown to effectively promote mandibular bone growth of Class Ⅱ malocclusion patients and improve lateral appearance of mandibular retrusion.The other improvement of Twin block appliance on hard-soft tissues remains unclear.OBJECTIVE:To assess the hard-soft tissue changes following Twinblock appliance treatment.METHODS:Atotal of 48 adolescent patients,30 males and 18 females,aged 11.6 years (range 9.58-12.37) with Class Ⅱ malocclusion were selected from Department of Orthodontics,Affiliated Stomatology Hospital of Hebei Medical University.Of them,27 were treated with Twinblock appliance as the treatment group,and 21 adolescent patients without treatments were selected as the control group.Cephalonetric analysis was used to compare the changes before and after treatment.RESULTS AND CONCLUSLON:Comparative analysis showed that Twinblock could effectively adjust or improve the effect of the natural growth,reduced upper lip convexity and mentolabial furrow angle.Moreover,Twinblock inhibited facial vertical distance reduction and facial convexity increase to decrease the potential differences between maxillary and mandibular bones in sagittal plance.Results show that Twinblock could effectively improve the profile of soft-tissues of Class Ⅱ malocclusion cases,
7.Pharmacokinetic-pharmacodynamic study on antipyreticeffects of coptisine on endotoxin-induced pyrexia of rats

Aim To establish the pharmacokinetic-pharmacodynamic(PK-PD) modeling to characterize the antipyretic effects of coptisine, an active component in coptis chinensis on rats.Methods Nine healthy male Sprague-Dawley(SD) rats were randomly divided into three groups, each with three.The rats in the first group were injected intravenously with lipopolysaccharide(LPS,100 μg·kg-1) alone.The second and third group rats were given coptisine high-dose(3.87 mg·kg-1) and coptisine low-dose(1.93 mg·kg-1) by tail vein injection at 30 min after LPS injection, respectively.Body temperature was measured at different time points, and blood samples from tail vein were collected simultaneously.The blood concentration of coptisine was determined by ultra performance liquid chromatography.Monolix software was used to model PK-PD of coptisine mean plasma concentration and temperature effects,by population computation with non-covariates.Besides.the model with advantage was selected by the fitting goodness.Results Coptisine could inhibit body temperature of endotoxin-induced fever in rats significantly.Two-compartment linear elimination model was used to describe the final PK model.Gaussian function, an input function of body temperature changes, which was used to depict PD model, the PK and PD models were connected by the Emax model.At last, the final model was fitted better;the fitting results indicated that the EC50 of antipyretic effect of coptisine was 89.7 μg·L-1, and the Emax was 1.88℃.Conclusions Coptisine has a powerful anti-pyretic effect on endotoxin-induced pyrexia of rats with high potency, Low in vivo distribution and quick clearance.
8.Effect of chemotherapy on serum ABCG2 level of non-small cell lung cancer

Objective To observe the change of the serum ABCG2 level of patients with non‐small cell lung cancer(NSCLC)be‐fore and after chemotherapy ,and explore its clinical significance .Methods Venous blood specimens of 15 healthy adults and 50 pa‐tients with NSCLC were collected before and chemotherapy ,the serum ABCG2 level of these specimens were detected by ELISA . the relation between the ABCG2 level and the chemosensitivity was investigated .Results The serum ABCG2 level of patients with NSCLC before chemotherapy was significantly higher than that in healthy adults(P<0 .05);the serum ABCG2 level was not related with TNM stage in NSCLC(P<0 .05);The serum ABCG2 level of patients with NSCLC after chemotherapy was significantly high‐er than that of before(P<0 .05);in chemosensitive patients ,the different of serum ABCG2 level was not significantly before and af‐ter chemotherapy(P>0 .05);among chemoresistant patients ,the serum ABCG2 level of patients with NSCLC after chemotherapy was significantly higher than that of before(P<0 .05);the serum ABCG2 level was higher in patients with pleural effusion than the patients without pleural effusion ,but the different was not significantly(P>0 .05) .Conclusion The serum ABCG2 level of patients with NSCLC is higher than that of healthy adults ;serum ABCG2 level may become a useful indicator in predicting the effect of NSCLC chemotherapy .
9.Short-term influence of LASIK on anterior and posterior corneal asphericity

Background Laser in situ keratomileusis (LASIK) is frequently performed to reduce or eliminate myopic refractive error.Some patients complain of a loss of visual function after surgery even when they have 20/20 visual acuity.One of the reasons is the change of asphericity of the cornea.Objective This study attempts to investigate the short-term changes of the anterior and posterior corneal asphericity after LASIK.Methods One hundred and seven myopic eyes of 54 subjects with the equivalent spherical diopter of -1.30 to -7.50 D who have received LASIK were enrolled in this prospective study.The Q-values of the posterior corneal surface for different corneal diameters (6mm,7mm,8mm and 9mm) were measured with Pentacam preoperatively and 1 month postoperatively.The correlations between Q-value,Q change (△Q),and the mean preoperative spherical equivalent refraction (SE),central corneal thickness (CCT),central ablation depth (AD) and residual bed thickness were investigated.Written informed consents were obtained from all the subjects prior to the clinical trial.Results The Q-values of the anterior and posterior corneal surfaces gradually decreased to negative values with the increase of corneal diameter in myopic eyes.Weak correlations were found between the asphericity of the anterior and posterior corneal surfaces from diameters of 6mm,7mm,8mm and 9mm (r=0.227,0.288,0.303,0.389;P=0.019,0.003,0.002 and 0.000,respectively).No statistically significant correlation was found between Q-value and the diopter of refractive error (P>0.05).Both the anterior and posterior corneal Q-values varied toward the positive direction except that in the 9mm area of the posterior corneal surface at postoperative 1 month(t=-1.495,P=0.138).The increase of the anterior corneal asphericity (△Q) was more obvious than that of the posterior corneal surface and showed a positive correlation with ablation depth and a negative correlation with residual bed thickness.However,no statistically significant correlation was seen between △Q and these two parameters in the posterior corneal surface (P>0.05).Conclusion The shape of the anterior and posterior corneal surface shows more prolateness as the increase of corneal diameter in myopic eyes.Both the anterior and the posterior corneal surfaces have an oblate shift within the ablation zone 1 month after LASIK.
10.Differential expression profile in the progeny of normal human liver cells irradiated by 60Co γ-rays

Objective To investigate the differential expression profile in the progeny of human liver cells surviving from ionizing radiation.Methods Complemental DNA gene chip was used to measure the transcriptional profile in progeny of HL-7702 cells exposed to 0, 2, 4, and 6 Gy of 60Co γ-rays, and the differentially expressed genes HAVCR2 and RAN were further identified by real-time PCR.Results The transcription level of 262 genes, 2746 genes and 3406 genes changed in the progeny of survival cells at 2, 4 and 6 Gy, respectively.A total of 71 common differentially expressed genes were screened, most of which were associated with transduction, cell cycle regulation, cellular immunity, cytoskeleton and movement, cell replication and repair mechanism.Conclusions Ionizing radiation could induce the expression changes of many genes, which might reveal the molecular mechanisms of gene expression in radiation induced genomic instability.
