1. Analysis of the composition of monosaccharides and the contents of polysaccharides in four components of radix hedysari polysaccharide 3

OBJECTIVE: To isolate and purify radix hedysari polysaccharide 3 (HPS-3) and determine the monosaccharide composition and the contents of polysaccharide in its four components. METHODS: Radix Hedysari polysaccharide was extracted by water, precipitated with ethanol, treated with Sevag method, decolored with H2O2, then precipitated with ethanol and dialyzed. By using DEAE-cellulose 52 and Sephadex G-100, HPS-3-A, HPS-3-B, HPS-3-C and HPS-3-D were purified. Their purity were determined by high performance gel permeation chromatography (HPGPC). By TLC and GC, the monosaccharide composition of four components was analyzed, and then the modified phenol sulfuric acid method was used to determine the content of polysaccharide of the four fractions. RESULTS: HPS-3-A was composed mainly of glucose, and the content of polysaccharide was 99.2%. But HPS-3-B, HPS-3-C and HPS-3-D were all composed mainly of arabinose and galactose. The contents of polysaccharide of them were 96.5%, 95.3% and 95.2%, respectively. CONCLUSION: This study provides the foundation for the research of structure-function relationship of Radix Hedysari polysaccharide 3. Copyright 2012 by the Chinese Pharmaceutical Association.
2.The diagnosis and treatment of cytomegalovirus (CMV) pneumonia after renal transplantation

Objective To explore the diagnosis and treatment of cytomegalovirus (CMV) pneumonia in patients who received immunosuppressive therapy after renal transplantation. Methods 15 patients out of the 110 recipients of renal transplantation were diagnosed as to have CMV pneumonia, during a period of the 2nd to 4th month after renal transplantation. All the 15 patients were treated with a systematic strategy, including rational use of antibiotics, withdrawal of immunosuppressant drugs and rebuilding the body′s immunity function, timely application of mechanic ventilator, and continuous high volume hemofiltration (CVVHF). The clinical responses of the 15 patients were reviewed and retrospectively analyzed. Results 15 recipients contracted CMV pneumonia out of 110 cases (13.6%), and it occurred on a mean of 93.3 days after renal transplantation. All of them were febrile with cough, and showed manifestations of interstitial pneumonia on chest films. CMV-IgM were positive in 13 cases, CMV DNA positive in 9 cases, and 4 patients presented signs of acute respiratory distress syndrome. The condition developed quickly in all the patients. 9 patients recovered and 5 patients died, including one who gave up treatment for financial reason. Conclusion CMV infection is a serious complication after renal transplantation. Early diagnosis with detection of CMV DNA and serum antibody of CMV-IgM should be emphasized. The comprehensive protocol principally composed of anti-virus therapy with cymevan or ganciclovir, monitoring of peripheral lymphocyte subgoups, and adjustment of dosage of immunosuppressive agent may improve recovery rate of CMV pneumonia.

Objective To investigate the effects of alcohol extract of Kunmu decoction on proliferation and apoptosis of rheumatoid arthritis fibroblast-like synoviocytes (RA-FLS). Methods Synovial tissues were obtained from patients with active RA received joint replacement or arthroscopy. The surface antigen and the amount of apoptotic cells were determined by flow cytometry. The inhibitive effect was detected by MTT assay. Results The CD90+surface antigen of synoviocytes was (94.78 ± 0.98)%. The inhibitive effect on the proliferation in all treatment groups were in a time-and dose-dependent manner. The apoptosis rate was increased in a dose-dependent manner among all dosage alcohol extract groups. Conclusion Kunmu decoction might inhibit proliferation and induce apoptosis of RA-FLS.

Objective To evaluate the targeted killing of malignant melanoma cells by aclarubicin liposomes conjugated with vascular endothelial growth factor(ADM-VEGF-SSL)in vitro.Metheds To detect the binding abilitv of liposomes to malignant melanoma(MM)cells,the human malignant melanoma cell line A375 was cultured in the presence of ADM-VEGF-3H-SSL or ADM-3H-SSL for 2 days followed by the detection of radioactivity of these cells.Then.A375 cells were cultured with various concentrations(0.01,0.1,1,10,100 mol/L)of ADM-VEGF-SSL,ADM-SSL or free ADM for 48 hours in the 48-hour cytotoxity test,or for 0.5 hour followed by another 48-hour culture in drug-free medium in the 0.5-hour cytotoxity test.After that,MTT assay was used to detect the survival rate of these cells.Results ADM-VEGF-SSL could specifically bind to and kill A375 cells.The binding rate of ADM-VEGF-SSL was 2.15 folds as high as that of ADM-SSL.The survival rate of A375 cells after being treated with ADM-VEGF-SSL for 48 hour was similar to that with flee ADM(P>0.05).but lower than that with ADM-SSL(P<0.05),while the survival rate of melanocytes treated with ADM-VEGF-SSL was higher than that with free ADM or ADM-SSL(both P<0.05).As shown by the 0.5-hour cytotoxity test.shortening the treatment course did not attenuate the effect of ADM-VEGF-SSL on A375 cells.Conclusions ADM-VEGF-SSL can specifically recognize A375 cells.efficiently deliver adriamycin into tumor cells,markedly inhibit the proliferation of A375 cells,and eventually,a targeted kill of these cells is realized.
6.Effect of Gambogic acid on proliferation and invasion in colon cancer LoVo cell line

Objective To investigate the effects of gambogic acid on proliferation,invasion and protein expression of MMP-2 in LoVo human colon cancer cell line.Methods The CCK-8 assay was used to determine the anti-proliferation ability.Transwell chamber assay was used to study the invasion of LoVo colon cancer cells.Western Blot was applied to examine the protein expression of MMP-2.Results CCK-8 assay showed that after cells treated with 1,2,4 μmol/L GA for 24 h,48 h the inhibition rates were(0.16±0.11)%,(0.24±0.08)%,(0.58±0.01)%,(0.67±0.03)%,(0.79±0.01)% and (0.27±0.05)%,(0.69±0.09)%,(0.85±0.01)%,(0.87±0.01)%,(0.89±0.01)%,and there had statistically significant differences between control group (0 μmo/L) with experimental group (P<0.05).Gambogic acid can effectively inhibit LoVo cells invasion at a low-dose concentration,which the numbers of cells that digested the matrigel in control group (0 μmo/L)was 120.50± 8.69 and 69.83 ±4.75 in experimental group (1 μnol/L),P< 0.05).Western Blot revealed that Gambogic acid can down-regulate the expression of MMP-2,which the levels of down-regulating the expression of MMP-2 after treated with 1,2,4 umol/L GA were 48.67%,74.72%and 82.70% (P<0.05) Conclusion Gambogic acid can effectively inhibit the growth and invasion of the LoVo cells,which may be related with down-regulating the expression of MMP-2.
7.Development of three dimensional facial measurement system based on structured light projection

Objective To construct a system for three dimensional face scanning and measurement. Methods The measurement system was based on the principle of triangulation and the combination of gray-code and phase-shift structured light projection. The system software was developed for Windows XP with the aid of tools such as Visual C++ and Hoops. Results A three dimensional measurement system based on structured light projection was developed. The system hardware was composed of fringe projection unit, image gathering unit, system control unit and mechanical appearance, and the system software was composed of point cloud display and editing module. The lamp house of the system was 12V, the working distance was 900 mm, the scanning time was 5.5 s and the scanning field was 500 mm×400 mm. Conclusion The three dimensional measurement system based on structured light projection is a refined machine with safe light to eyes, and the accuracy and scanning speed are suitable to face scanning.
8.Evaluation of transcatheter arterial chemoembolization combined with radiofrequency capacitive heating on clinical therapeutic effect of metastatic hepatic carcinoma

Objective To evaluate clinical therapeutic efficacy and adverse effecacy of transcatheter arterial chemoembolization(TACE)combined with radiofrequency capacitive heating(RCH) for metastatic hepatic carcinoma(MHC). Methods Thirty-nine cases of MHC were enrolled in this study and divided into two groups: study group(n = 19)and control group(n = 20). Before therapy, the Karnofsky's score of the patients was all beyond 60. Results The carcinoma growth rate of the study group was -(0.38 ? 0.22), while that of the control group was -(0.13 ? 0.25), showing significant statistical difference(P 0.05). Conclusion The therapeutic effect of MHC can be further improved by the treatment of TACE combined with radiofrequency capacitive heating without increase of adverse side effects.
9.AFLP analysis on genetic diversity of Haloxylon ammodendron in China.

To determine the genetic diversity of Haloxylon ammodendron collected from 14 sites in 5 provinces, 103 H. ammodendron samples of 12 wild populations and 2 cultivated which collected from 14 sites in 5 provinces were analyzed by amplified fragment length polymorphism (AFLP) DNA markers. PopGen32 and NTSYSpc2.1 was applied to evaluate genetic diversity of H. ammodendron populations. The average percentage of polymorphic loci (PPL) of total H. ammodendron populations was 94.13%, the average Nei's gene diversity index (H(e)) from 14 populations was 0.308 0, and the Shannon's genetic diversity index (I) was 0.467 6. The results indicated that the genetic diversity of H. ammodendron populations was high. Genetic differentiation index (G(st)) was 0.313 8, and the gene flow (N(m)) was 1.093 5 at the population level. The level of gene flow of H. ammodendron showed it possessed the feature of wind-pollinated outcrossing plants. AMOVA analysis indicated that genetic variation of H. ammodendron was much higher within groups (89.34%) than that among groups (10.66%), moreover genetic variation within groups mainly occurred among populations in different producing areas (84.80%). Cluster analysis (UPGMA) was applied to generate dendrogram based on Nei's genetic distances of 14 populations. Samples from Xinjiang and Qinghai were clustered respectively as a clade for their distant genetic relationship, while Samples from Gansu, Inner Mongolia and Ningxia were clustered together for their close genetic relationship. Genetic diversity of H. ammodendron populations is high in China, and genetic differentiation among regions is small, thus abundance within this specie is high at this stage. Therefore, wild nursery and artificial cultivating in different areas are effective measures for the conservation and sustainable utilization of H. ammodendron resources.
