Objective To observe the influence of telephone follow-up and home follow-up on compliance behavior and activities of daily living (ADL) of stroke patients during rehabilitation. Methods 50 cases were as telephone follow-up group and 50 cases were as home follow-up group extracted from 355 stroke patients who discharged the hospital during August 1, 2010 to July 1, 2012. They were followed up 2 weeks, 1 month and 3 months after discharge respectively. Their ADL were assessed with Bathel index (BI) at discharge and 3 months after discharge, and the compliance behavior were assessed 3 months after discharge. Results There was no significant difference in the score of BI between 2 groups at discharge (P>0.05), and the score of BI improved in both groups 3 months after discharge (P<0.05), but it was higher in the home follow-up group than in the telephone follow-up group (P<0.05), as well as the rate of compliance behavior (P<0.01). Conclusion Regular home follow-up can improve the compliance behavior of stroke patients during rehabilitation.

Genomic instability is one of the most pervasive characteristics of cancer cells, and DNA damage response (DDR) pathway plays a crucial role in genomic stability. The DDR pathway is a complex signaling network, which involves cell DNA repair, apoptosis and cell cycle regulation.Deficiencies in these repair pathways can result in several different genetic disorders, including cancer.Targeted therapy based on inhibiting the DDR pathway in cancers offers a novel therapy strategy for patients with tumors lacking specific DDR functions. Many small-mole-cule compounds targeting DDR pathway are typically developed for solid cancer therapy. The poly (ADP-ribose) polymerase (PARP) inhibitor is a kind of DDR inhibitors which exploits the principle of synthetic lethality to selectively kill cancer cells. This review highlights the molecular mechanisms of PARP inhibitor action, the progress of PARP inhibitors in cancer therapy, drug resistance and the challenge of PARP inhibitor in the future.

Objective To investigate the role of norepinephrine in the down-regulated visceral sensitivity of rats deprived of rapid eye movement(REM)sleep. Methods Twenty-four male Sprague- Dawley rats were randomly divided into 3 groups:cage-yoked rats as control(YC),rats with REM sleep deprivation(SD)and rats with yohimbine administered intraperitoneally after REM sleep deprivation(YSD).Flower pot technique was employed to make sleep deprivation model.YSD group was given vohimbine intraperitoneally at the 48th hour after REM sleep deprivation. After both SD and YSD groups had completed these processes, rats of all the three groups were given colorectal distension(CRD)and electromyogram (EMG)was recorded at the same time. The number of discharges of EMG and the threshold of Dain perception of the rats were observed to evaluate the visceral sensitivity. The thalamus, rectum and distal colon were taken after CRD;MAO-mRNA and TH-mRNA in three tissues were detected with RT-PCR.Resuits On 48th hour, the number of discharges of EMG in 10 seconds responding to CRD in group SD was significantly less than that in group YC and the threshold of pain perception in group SD was higher than that in group YC(P<0.05).The number of discharge of EMG in group YSD was significantly more than that in group SD(P<0.05).The expression of MAO-mRNA in group SD was lower than that in group YC(P<0.05)and the expression of TH-mRNA in group SD was higher than that in group YC(P< 0.05).Conclusions The visceral sensitivity in rats is down-regulated by REM sleep deprivation, which can increase synthesis of norepinephrine.Norepinephrine can modulate visceral sensitivity.

To provide accurate information on geographic distribution of crude drug Sailonggu in the plateau, we identified zokor species (Eospalax spp.) in Qinghai-Tibet Plateau using molecular methods. Based on the mitochondrial cytochrome B (cytb) gene sequences, we then extracted haplotypes from these sequences and reconstructed phylogenetic trees for the haplotypes using both maximum likelihood (ML) and Bayesian inference (BI) methods. Based on the trees, the species of each sample were determined. Five hundred and three samples from 35 populations were sequenced and their whole cytb sequences (1140 bp) were obtained. From these sequences 150 haplotypes were detected, in which, 126 were Eospalax baileyi, 20 were E. cansus, and 4 were E. smithi of the 35 populations, 28 were E. baileyi type, 5 were E. cansus type, and the remaining 2 were mixed of E. baileyi + E. cansus (DT2) and E. baileyi + E. smithi (ZN3). The results showed that, the regions around the Qinghai lake and near the upper stream of Yellow River started at Guide could be viewed as the producing area of authentic Sailonggu, and also, the cytb gene is a powerful molecular marker to determine the species of zokors as well as for the authentication of geographic distribution of Sailonggu.

Objective To characterize the differential protein expression in the progeny of human liver ceils surviving from ionizing radiation by the proteomic analysis. Methods Two-dimensional electrophoresis gel coupled with mass spectrometry was used to explore the specific protein expression in the progeny of 7702 human liver cells surviving from ionizing radiation.Alterations in expression level of protein spots between the control and the progeny groups were statistically analyzed by ImageMaster 2D Platinum software and mass spectrometry was used to identify the protein spots with significantly altered expression-level.Results The progeny of irradiated ceils were derived from human liver cell line exposed to 0,2,4,6 Gy of 60Co γ-irradiatian.A total of 42 differentially expressed proteins between the control and the progeny of the irradiated cells groups were screened,of which 17 were identified by matrix assistant laser desorption ion-top off light-mass spectrometry(MALDI-TOF MS)analysis, including 4 up-regulated and 13 down-regnlated proteins. Conclusions The differentially expressed proteins profile could be significantly altered in the progeny of irradiated cells. The proteomics approach has the potential to detect the protein changes relevant to radiatian-induced genomic instability(RIGI).Further study of differentially expressed proteins would likely reveal the molecular mechanisms of gene expression in RIGI.

Objective To establish a new traceability pathway of point-of-care testing ( POCT ) of HBA1c by using commutable quality controls, in order to improve the accuracy of POCT HBA1c and the harmonization of testing results with those of central laboratories. Methods The study was about measurement traceability. Human frozen whole blood samples with IFCC assigned values were used to calibrate the G8 HBA1c Variant in June, 2013.According to the CLSI EP9-A2-IR guideline, 50 patient samples and 2-level commercial QC samples were then analyzed by G8 system and DCA Ventage system. The best fitting curves for fresh patient samples and the commercial QC materials were established separately. The patient results tested on the DCA Ventage were modified and verified.Paired t-test and Passing Bablok linear regression were used. Results The linear equation of DCA/G8 before calibration was Y=0.899 5X+0.389 1(R2 =0.991 0).Calibration by fresh patient samples reduced the mean bias of DCA/G8 from -0.40%±0.34%to 0.00%±0.29%.Calibration by QCs reduced the mean bias to 0.15%±0.29%.The linear correlation established by quality controls was stable, which made the bias was lower between DCA and G8 in the consequent six runs. Conclusions The accuracy and the traceability of POC testing could be realized by using commutable QC materials traceable to IFCC assigned values.Through this method, POC testing can become more comparable to the results of clinical laboratory HBA1c instruments.

BACKGROUND:Matrix metal oproteinase-13 is most active in the degradation of col agen type II in the extracel ular matrix of cartilage. Interleukin-1 (IL-1) is thought to be the inducer of matrix metal oproteinases, and participates in the degradation and degeneration of articular cartilage. OBJECTIVE:To study the influence of IL-1βon microRNA-27b (miR-27b) and matrix metal oproteinase-13 expression of chondrocytes in rats. METHODS:Chondrocytes isolated from seven male Wistar rats were cultured and divided into IL-1βstimulation group and control group. No stimulus was given in the control group;10μg/L of serum free medium was used to culture rat chondrocytes in the IL-1βstimulation group. Cel growth was observed at 0, 24, and 48 hours under an inverted microscope. miR-27b and matrix metal oproteinase-13 expression in the cultured chondrocytes were detected. RESULTS AND CONCLUSION:The relative expression of matrix metal oproteinase-13 in rat chondrocytes was gradual y increased when induced by IL-1βat 0, 24, and 48 hours (P<0.05). Expression of miR-27b and miR-31 in rat chondrocytes at 24 and 48 hours induced by IL-1βgradual y decreased (P<0.05);conversely, expression of MiR-26a, miR-26b, miR-23, and miR-204 gradual y increased (P<0.05). After 48 hours of IL-1βinduction, expression of miR-27b was the lowest in rat chondrocytes (P<0.05). These findings suggest that IL-1βinhibits miR-27b expression, strengthens the expression of matrix metal oproteinase-13, and damages chondrocytes, contributing to both the onset and progression of osteoarthritis.
